Abstract. Ruchin AB, Egorov LV, Polumordvinov OA. 2021. Coleoptera of the Penza region, Russia based on fermental crown trap). Biodiversitas 22: 1946-1960. There are the results of processing the material of the 2019-2020 studies on Coleoptera from the Penza region, Russia. The surveys were carried out using fermental crown traps in various habitats on the territory of 18 districts of the region. In total, 18 traps were installed in 2019 and 96 traps – in 2020. During the research, 5,577 specimens were collected and recorded. Ninety-seven species from 19 families were found, of which 43 species are new to the Penza region. The most diverse families are Cerambycidae (24 species) and Elateridae (11 species). Species from the families Nitidulidae (3281 specimens), Scarabaeidae (1497 specimens), and Cerambycidae (453 specimens) predominated in the traps. A list of species is given, indicating references and information on biology. New data is given for 4 species included in the Red Data Book of the region (Protaetia fieberi, Protaetia speciosissima, Gnorimus variabilis, Purpuricenus globulicollis).

Application of inter-SINE amplified polymorphism (ISAP) markers for genotyping of Cucumis melo accessions and its transferability in Coleus spp. Biodiversitas 22: 2918-2929. An unambiguous characterization of melon (Cucumis melo L.) accessions based on their morphological traits is often laborious and affected by environment when compared with molecular marker genotyping. Short interspersed nuclear elements (SINEs) are highly abundant non-autonomous and non-coding retrotransposons that are widely scattered over all chromosomes of eukaryotes. They can serve as a good molecular marker for routine genotyping in plant breeding and marker-assisted selection. This study aimed to apply inter-SINE amplified polymorphism (ISAP) markers for genotyping of Cucumis melo accessions and its transferability in Coleus spp. Twenty-one C. melo accessions, one C. metuliferus E. Mey. ex Naudin, and 15 accessions of Coleus spp. were used for ISAP marker development. A copy of cucumber-specific long interspersed nuclear element (LINE) and multiple copies of melon-specific SINE were identified and isolated. ISAP primers were designed from the highly conserved region of the SINEs and LINE. The melon and cucumber-specific ISAP markers showed a higher degree of polymorphism (87.5%-100%) than potato ISAP markers (60%-100%) in all the tested melon accessions. The unweighted pair group method with arithmetic average (UPGMA) dendrogram generated from polymorphic ISAP bands clearly distinguish the Cucumis melo accessions from their distantly related wild species C. metuliferus. The transferable nature of Cucumis ISAP marker system was demonstrated in Coleus species, where the marker differentiates the tested accessions.

Abstract. Izzati M, Haryanti S, Hastuti RB. 2021. Effectivity of bulrush (Scirpus californicus) as a soil conditioner  increasing sandy and clay soil fertility. Biodiversitas 22: 3423-3429. Bullrush (Scirpus californicus) is a species of macrophytes that often populates lakesides. This aquatic plant was rarely studied and its use has not been explored yet. This study was carried out to determine the effects of soil conditioner made from bulrush on sandy and clay soil fertility. Collected bulrush from Rawa Pening lake was milled into a powder and used as a soil conditioner in a proportion of 1:1. After a week, soil fertility was evaluated including organic matter content, water retention, the ratio of C/N, and bacteria population. The study was designed using a Completely Randomized Design with two treatments and control. Resulted data were analyzed using a t-test to evaluate the difference between the two treatments. Results showed that bulrush powder application significantly increased sandy and clay soil fertility. The organic matter content significantly increased in both sandy (p<0,01) and clay soils (p<005). The water retention of sandy soil was increased by 74% (p<0,01), while in clay soil was reduced by 27% (p<0,01). The C/N ratio was significantly reduced in both sandy and clay soil (p<0,05), while the bacteria population significantly increased (p<0,01). It is suggested to use the bulrush as a soil conditioner particularly for sandy and clay soils.

Antibacterial activity of bacteria isolated from earthworm (Pheretimasp.) gut against Salmonella typhiand Staphylococcus aureus: in vitro experiments supported by computational docking. Biodiversitas 23: 1125-1131. Bacteria have a diverse ecology niche as the effect of a long evolutionary process. They can live along with other organisms as endosymbiont. Research into endosymbiont bacteria began because of their ability to increase host resistance, especially to pathogens. This study aimed to determine the antimicrobial ability of endosymbiont bacterial isolates of earthworms Pheretimasp., Bacillus brevis, and Bacillus choshinensis that inhabit Pheretimasp. were isolated.The isolates were grown on tryptic soy broth media for 24 hours. The isolates were then purified using tryptic soy agar and biochemically tested to ensure both isolates are endosymbiotic bacteria. Antimicrobial activity was tested using agar diffusion methods in pathogenic bacteria, Staphylococcus aureus, and Salmonella typhi. Amoxicillin and chloramphenicol were used as positive controls. The inhibitory test results showed that incubation for 15 days effectively assessed pathogenic bacteria growth inhibition, marked by the largest inhibition zone (21.32 mm for Salmonella typhi and 16.88 mm for Staphylococcus aureus). They were very effective at inhibiting Salmonella typhiand Staphylococcus aureus growths.Endosymbiotic compounds’ had a potential as antimicrobial. The in silico test supports the inhibitory test, which concluded that endosymbiont isolates can be an antimicrobial characterized by low-binding affinity values (-8,6 on tyrocidine) on molecular docking analysis.

Aeschynanthus radicans Jack. commonly-known as lipstick flowers, is an epiphytic plant in Gesneriaceae family. It has an attractive morphology and is easy to grow. Therefore A. radicans is used as mother species for hybridization by plant breeders. The flowers of A. radicans are dichogamous and strongly protandrous. The pollen matures before the stigma becomes receptive and bends away and down when the stigma becomes receptive. As a thought-after material for hybridization, little is known about the flower development, pollen viability, and storability. The purpose of this study was to observe the development phase of A.radicans flowers, determine the viability of pollen during flower development, and investigate the pollen storability at -20?C. Flower development was observed in phases from small buds to anthesis. Pollen viability was tested in 10%, 15%, and 20% sucrose solution for 4 hours. Pollen storage was tested at -20?C for 77 days and tested for viability every three weeks. The best sucrose concentration for the germination of fresh pollen was 20%. The highest pollen viability was at H0 was 58±3% and gradually decreased with flowerage. The lowest viability was 18±14% at flower stage H5, coinciding with the first day of stigma receptive. Pollen storage at -20?C decreased pollen viability to 30,2±2% after 42 days and to 13,54% after 77 days.

Abstract. Mukkun L, Lalel HJD, Kleden YL. 2021. The physical and chemical characteristics of several accessions of sorghum cultivated on drylands in East Nusa Tenggara, Indonesia. Biodiversitas 22: 2520-2531. Many sorghums' accessions are widely cultivated in East Nusa Tenggara (ENT) Province, but no information is available on their physical and chemical properties. This study aimed to analyze the physical and chemical properties of several sorghum accessions cultivated in the dryland of ENT province. Seven sorghum accessions were obtained from farmers, namely Numbu, Kawali, Okin, red local, brown local, black local, and white local. Physical properties of the grain were carried out visually, including color, shape, and seed husks. Proximate content, total polyphenols, anthocyanins, and tannins were analyzed to determine each accession's chemical properties. Quantitative data were analyzed by ANOVA followed by Duncan Multiple Region Test (DMRT) at P?0.5. Meanwhile, qualitative data were tabulated and presented in tables and figures. The results showed that all sorghums have almost the same nutritional content as other staple food to develop into a food substitute for rice to support food security. Furthermore, local black sorghum contains high polyphenol compounds with high antioxidant activity, indicating that it has excellent potential to be developed into functional foods. The high anthocyanin content in black sorghum can also be developed into a safe and environmentally friendly food coloring

Potency of secondary metabolites of Trichoderma asperellum and Pseudomonas fluorescens in the growth of cocoa plants affected by vascular streak dieback. Biodiversitas 22: 2542-2547. Vascular streak dieback (VSD) is the main disease in cocoa caused by Oncobasidium theobromae. Using secondary metabolites of Trichoderma asperellum and Pseudomonas fluorescens is one of the methods to control disease in cocoa plants. The aim of this research is to evaluate the potency of secondary metabolites of T. asperellum and P. fluorescens to control VSD and study its effect on the growth of cocoa plants. The study used a non-factorial, completely randomized design (CRD) with eight treatments and four replications. The secondary metabolites were applied every week for ten times. The results showed that the secondary metabolites of T. asperellum and P. fluorescens were effective in controlling VSD. The combination of organic fertilizer and secondary metabolite of P. fluorescens and T. asperellum as found to be the best treatment to increase cocoa growth and the number of shoots increased by 43.58 shoots

Tomato (Solanum lycopersicum L.) is a common crop that is widely cultivated and consumed around the world. Demand for tomatoes has increased along with the growth of the human population. Therefore, tomato production has to be continuously developed to meet the ever-rising global demand. Mutation breeding is one of the strategies applied to improve plant yield and quality. This study evaluated a promising mutant breeding line called G6 and compared it to existing varieties. G6 (mutant line), Ratna, Tora, and Intan (commercial varieties) were used as materials and were evaluated for their qualitative and quantitative traits. Overall, G6 showed a slightly flattened fruit shape similar to Ratna. It also had the highest value of locules, long shelf life, and the number of bunches per plant compared to the existing varieties (Ratna, Intan, and Tora). The number of locules is assumed to be linked to fruit size and dimension. Long shelf life is one of the preferred qualities of both consumers and farmers, as it could prevent overmature and rotten tomato fruit during transportation. G6 differed from commercial varieties (Intan, Ratna, and Tora) in terms of the number of locules, shelf life, and the number of bunches per plant.

Effects of copper on the leaf morpho-anatomy of  Rhizophora mucronata: Implications for mangrove ecosystem restoration. Biodiversitas 22: 2058-2065. Mangrove is one of the important yet threatened ecosystems in the Philippines. One of the reported causes of its destruction is heavy-metal pollution. Forest restoration managers recommend species that exhibit functional and adaptive traits in their leaf morpho-anatomy to restore degraded mangrove ecosystem’s ecological integrity. In this study, the morphological and leaf anatomical characteristics of Rhizophora mucronata Lam. growing in mangrove sites in Mamburao, Occidental Mindoro, Philippines with different concentrations of copper element in the soil were examined to determine the species’ adaptation in copper exposed soil and its potential for mangrove ecosystem restoration. Leaf area was measured and leaf anatomical characteristics were determined through histological paraffin technique. The average leaf area of R. mucronata in the copper-rich site was relatively smaller than in site with considerably less copper. Further, anatomical characteristics in the copper-rich site such as thicker cuticular, upper epidermis, and hypodermal layers, thin mesophyll layers and vascular bundle, smaller and fewer xylary elements, and larger salt secretion cells indicate functional and adaptive traits. These structures may help R. mucronata tolerate the high concentration of heavy metals in the plant body and soil, suggesting its potential for mangrove ecosystem restoration especially in copper-polluted sites in the Philippines.

Abstract. Saensouk P, Saensouk S. 2021. Short Communication: Taxonomy and the first report for pollen and leaf surfaces anatomy of the genus Enkleia (Thymelaeaceae) in Thailand. Biodiversitas 22: 1340-1344. Taxonomic of the genus Enkleia in Thailand are revised. Two species, Enkleia malaccensis Griff. and E. thorelii (Lecomte) Nevling, are reported. Enkleia siamensis (Kurz) Nevling and E. siamensis (Kurz) Nevling subsp. andamanica (Hutch. Ex C. E. Parkinson) Nevling are reduced to Enkleia malaccensis Griff.. Therefore, both species are recorded as synonym of E. malaccensis. E. thorelii is reported endemic to Thailand. Vernacular name, distribution data, ecological data, flowering period, fruiting period, conservation status, traditional use of this genus are provided. The pollen and leaf surfaces of the genus are examined and the first studied.
