Indigenous Streptomyces spp. isolated from Cyperus rotundus rhizosphere indicate high mercuric reductase activity as a potential bioremediation agent. Biodiversitas 22: 1519-1526. The purification and characterization of mercuric reductase of four indigenous Streptomyces spp. from Cyperus rotundus L. rhizosphere in mercury-contaminated area have been investigated. Cell-free extract was obtained by disrupting cells using sea sand at 4 °C followed by centrifugation. Mercuric reductase was purified by ammonium sulfate precipitation, dialysis, and chromatography column (DEAE Sepharose anion column chromatography). The determination of optimum pH and temperature of mercuric reductase activity was measured based on the number of NADPH2 oxidized to NADP per mg protein per minute using a spectrophotometer. The molecular weight of mercuric reductase was determined using SDS-PAGE. Result showed that the highest specific activity of mercuric reductase was recorded from Streptomyces spp. BR28. The optimum pH and temperature of cell-free extract enzyme mercuric reductase were 7.5 and 80 °C, respectively. The enzyme was purified to 431.87-fold with specific activity 21918.95 U/mg protein. SDS PAGE showed that the molecular weight of mercuric reductase in Streptomyces spp. BR 28  ranged from 50 kDa to 75 kDa. It can be concluded that Streptomyces isolates contain mercuric reductase and have potential as mercury bioremediation agent to overcome mercury contamination in the environment.

Abstract. Gustianingtyas M, Herlinda S, Suwandi. 2021. The endophytic fungi from South Sumatra (Indonesia) and their pathogenicity against the new invasive fall armyworm, Spodoptera frugiperda. Biodiversitas 22: 1051-1062. Maize in Indonesia is currently experiencing attacks and outbreaks of the new invasive fall armyworm, Spodoptera frugiperda. The S. frugiperda larvae emerge from the leaf midrib when eating, after hiding in the maize stalk so that it is difficult to control by contact. This study aimed to find out the endophytic fungi from the roots of maize, banana and chili in South Sumatra and to determine their pathogenicity against S. frugiperda larvae. The endophytic fungi were isolated from the plant roots. Fungal isolates proven to be endophytic were dropped (1 × 106 conidia mL?1) on the second instar larvae. The result showed that the endophytic fungi found were 8 isolates consisting of the genus, Aspergillus sp., Beauveria sp., Chaetomium sp., and Curvularia sp. First report of Aspergillus sp., Chaetomium sp., and Curvularia sp. have insecticidal activity against S. frugiperda larvae. However, the two most pathogenic isolates were JgCrJr and JgSPK isolates of Beauveria sp. with larval mortality of 29.33% and 26.67%, respectively, and could reduce the emergence of S. frugiperda adults up to 44%. So, the two isolates of Beauveria sp. have a high potential to be developed to control S. frugiperda larvae in maize both in the lowlands and the highlands.

Abstract. Mursyidin DH, Nazari YA, Badruzsaufari, Masmitra MRD. 2021. DNA barcoding of the tidal swamp rice (Oryza sativa) landraces from South Kalimantan, Indonesia. Biodiversitas 22: 1593-1599. The tidal swamp rice (Oryza sativa L.) landraces of the South Kalimantan, Indonesia, has been known for hundred years ago with a better adaptation to the local conditions, such as acidity, salinity, and metals contamination. However, the genetic insight of these landraces has not fully understood. Here, the rbcL region of tidal swamp rice from this region was successfully sequenced, aligned, analyzed, and deposited into the GenBank with accession numbers of MT818188–MT818201. The multiple alignments of these sequences showing a barcoding motif with eight mutation or polymorphic sites with both substitutions (transition-transversion) and indels (insertion-deletion). Based on this marker, the tidal swamp rice has a low genetic diversity, only 0.086. However, the UPGMA and maximum likelihood (ML) analyses revealed that this germplasm was grouped into five and twoclusters or clades, respectively. In this case, Bayar Putih is closely related to Siam Panangah and farthest from Lemo. This information might help to develop conservation and breeding programs of the tidal swamp rice landraces in the future.

Mangrove ecosystem in North Sumatran (Indonesia) forests serves as a suitable habitat for mud crabs (Scylla serrata and S. olivacea). Biodiversitas 22: 1489-1496. Mud crabs (Scylla serrata and Scylla olivacea) are often found in muddy substrates associated with mangrove vegetation. The habitat of these crabs ranges from tropical estuaries to sheltered subtropical areas, riverbanks, lower river traits, and intertidal areas. These crabs have an important economic value, and it increases the income of the communities living around the mangrove area. This study aims to obtain more information on the environmental factors of habitat for mud crabs (S. serrata and S. olivacea): insights on the effects of abiotic and biotic factors and potential economic value, in the village of Lubuk Kertang, Langkat, North Sumatra, Indonesia. Data were collected by measuring abiotic factors (temperature, pH, salinity, dissolved oxygen, mud depth, distance from the river) and biotic factors (mangrove stand density through canopy cover). The dataset was analyzed using a binary logistic regression analysis with a stepwise method. Results showed that the abiotic factors that contributed to the presence of the mud crab were dissolved oxygen and a mud depth of 68.7%. Meanwhile, the remaining 31.3% that contributed to the presence of mud crabs were abiotic factors (temperature, pH, salinity, distance from rivers) and biotic factors (stand density through canopy cover). The present study suggests that dissolved oxygen and mud depth have an important function in predicting the presence of mud crabs.

Abstract. Dakhlan A, Hamdani MDI, Putri DR, Sulastri, Qisthon A. 2021. Short Communication: Prediction of body weight based on body measurements in female Saburai goats. Biodiversitas 22: 1391-1396. Saburai goat is a new composite breed in Lampung Province with little information on its performance. This research aimed to predict body weight based on body measurements, namely body length (BL), chest girth (CG), and shoulder height (SH) in female Saburai goats. This study used 42 female Saburai goats aged 3-4 years. The method used in this study was a survey, namely all-female Saburai goats aged 3-4 years were used for this research. The data obtained were analyzed using correlation and simple and multiple linear regression analysis using R program. Coefficient determination (R2), adjusted R2, residual standard error (RSE), Akaike information criterion (AIC), Bayesian information criterion (BIC), and stepwise regression analysis were used to find the best and most parsimonious regression model to predict BW based on body measurements. The results showed that BL, CG, and SH were positively and significantly correlated with BW of female Saburai goats with correlation coefficients of 0.858, 0.956 and 0.862, respectively. Chest girth was the best predictor for BW if using single predictor with regression equation of ? = -31.17 + 0.93X2. However, combination of BL and CG was the best and most parsimonious regression model in predicting BW of female Saburai goat with regression equation of ? = -36.09 + 0.31X1 + 0.72X2 with R2 = 0.941, adjusted R2 = 0.938, RSE = 2.842, AIC = 216.73 and BIC = 223.78. In conclusion, all body measurements in this study (particularly combination of BL and CG) could be used as predictor for BW with high accuracy of prediction. The result of this study suggested that CG and BL could be as indirect criteria to improve BW of female Saburai goats.

Abstract. Ruslim Y, Sandalayuk D, Kristiningrum R, Alam AS. 2021. Estimation of Above Ground Biomass and carbon stocks of Tectona grandis and Gmelina arborea stand in Gorontalo Province, Indonesia. Biodiversitas 22: 1497-1508. Plantation forest plays an important role to fulfill timber needs, while more recently plantation forest is increasingly acknowledged to sequester and store carbon which can mitigate climate change and also as carbon sequestration for the environment. This study aimed to calculate the stand potential, stand biomass and carbon stocks of teak (Tectona grandis) and gmelina (Gmelina arborea) stands in the context of land after being abandoned in Gorontalo Province, Indonesia. Four plots with size of one hectare each were sampled in which each species (i.e. teak and gmelina) consisted of two plots. In each plot, the diameter at the breast-high (1.3 m) and the height of each individual were recorded. Data analysis included growth parameters of the stands (i.e., Mean Annual Increment/MAI and Current Annual Increment/CAI) and above-ground biomass and carbon sequestered by the stands. Simple linear regression using polynomial trendline was used to determine the relationship between variables and the degree of the relationship. The results showed that the maximum growth of teak stands at Plots I and II reached a maximum point at the age of 32 and 25 years with the total volume of 307.50 and 254.81 m3ha-1, respectively. While the maximum growth of gmelina stands at Plots I and II reached a maximum point at the age of 15 years with the total volume of 190.54 and 251.80 m3ha-1, respectively. The biomass content in teak stands at Plots I and II and gmelina stands at Plots I and II were respectively 267.83; 221.94; 104.03 and 137.48 tons ha-1. Meanwhile, the carbon content in teak stands at Plots I and II and gmelina stands at Plots I and II were respectively 125.88; 104.31; 48.90; and 64.62 tons ha-1. The results of the regression analysis suggest that there was strong relationship between carbon sequestered and the age of the stands as well as total basal area. The results of this study suggest that Tectona grandis is more potential to be developed as plantation forest than Gmelina arborea when aiming at carbon sequestration and biomass production.

Rubus spp. are woody or herbaceous plants that can be used for fruit, ornamental and medicinal purposes. The increasing use of Rubus as a commercial species is highly dependent on the formation of high-quality genetic material. However, the lack of basic biological knowledge is one of the limiting factors in this development. This research aims to describe the morphological characters of Indonesian Rubus flowers at Cibodas Botanical Garden, Indonesian Institute of Sciences, Cianjur, West Java, Indonesia. From January to June 2020, we observed nine Rubus species with five replications, namely R. alceifolius, R. chrysophyllus, R. ellipticus, R. fraxinifolius, R. linneatus, R. moluccanus, R. pyrifolius, R. rosifolius, and Rubus sp (Blackberry) in Cibodas Botanical Garden, Indonesia. The results showed variations in the characteristics of the pistil, stamens, torus shape, and duration of flowering stages between species. The mean value of the stamen-pistil ratio for R. pyrifolius was highest among other species (8.27), and R. fraxinifolius was the lowest (0.16). Furthermore, a correlation analysis between stamens and pistils for nine Rubus species was relatively positive (r = 0.598), similarly distinctly positive between pistils and fruits with r = 0.763. Flower development duration takes ranging 10-12 days from initiation to anthesis, depending on the species.

Mangrove is an important ecosystem located in the coastal tidal zone in tropical and sub-tropical regions. This ecosystem is characterized by the presence of plant species that can survive in brackish and inundated zone of seawater. There are many mangrove forests in Belitung Island, Indonesia, but the data of mangrove flora diversity and zonation in this island have not been described. This study aims to reveal the diversity and analyze the zonation of mangrove flora in Belitung Island. This research was conducted using a cruising method to explore the diversity of mangrove flora species and a line transect to analyze the zonation of mangrove flora. The exploration locations for diversity of mangrove flora were Kuale Tambak Beach, Asam Beach, Belitung Mangrove Park Area, Manggar River, Sentigi Beach, Sabong Beach, and Kembiri Beach. Mangrove flora zonation was observed in Asam Beach (north), Kembiri Beach (south), and Kuale Tambak Beach (east). The result showed that mangrove flora in Belitung Island consisted of 12 families, 16 genera, and 24 species. Species found were dominated by 8 species from the Rhizophoraceae family with the largest number of species were from the genera of Bruguiera and Rhizophora. The species of Bruguiera included Bruguiera gymnorhiza, Bruguiera sexangula, Bruguiera cylindrica and Bruguiera parviflora. Meanwhile, the Rhizophora species that have been found were Rhizophora apiculata, Rhizophora mucronata, and Rhizophora stylosa. The Kembiri beach had the highest number of species of the mangrove flora with 18 species. The zonation of mangrove flora at each research location showed a different zonation pattern. The mangrove vegetation on Kulae Tambak Beach was behind the Casuarina equisetifolia vegetation, while the mangrove vegetation on the Asam and Kembiri Beaches directly faced the sea. The difference in zonation patterns is likely caused by local topography on Belitung Island, substrate conditions and salinity factors on the mangroves of Belitung Island

Abstract

Abstract. Rehman A, Saeed S, Ahmed A. 2021. Genetic diversity and population structure of Seriphidium Sub-genus of Artemisia from different terrains of Balochistan, Pakistan. Biodiversitas 22: 2658-2664. Seriphidium is one of the largest sub-genus of Artemisia growing in different geographical populations and terrains of Balochistan Pakistan. This genus is economically important and used as folk medicine. The genus is highly variable, besides morphological characterization, molecular authentication is also needed for novel drug discovery. Three significant species of the genus viz., S. quettense, S. oliverianum, and S. maritimum were selected for morphological and molecular characterization and phytochemical investigation. Genetic variability assessment was carried out by polymorphic bands analysis by using molecular markers in 18 accessions collected from two sites, namely, Ziarat and Hazarganji-Chiltan National Park, Balochistan, Pakistan. A combined molecular marker system by using 17 Randomly Amplified Polymorphic DNA (RAPD) and 9 Inter-Simple Sequence Repeats (ISSR) amplified 231 loci, of which 111 were polymorphic (48% polymorphism). Phenolic contents and Flavonoids were estimated for phytochemical assessment. Results revealed that the highest phenolic compounds were in S. quettense when compared with other investigated taxa. In conclusion, molecular marker profiling together with phytochemical variation of all three species exhibited genetic diversity and chemical variation. The findings of the current investigation can further be used as a baseline study for the implementation of conservation strategies. In addition, phytochemical assessment can be utilized for modern drug discovery

Inhibition of ?-glucosidase activity, metals content, and phytochemical profiling of Andrographis paniculata from different geographical origins based on FTIR and UHPLC-Q-Orbitrap HRMS metabolomics. Biodiversitas 22: 1535-1542. Ensuring consistency of quality, safety, and efficacy of herbal medicines from raw materials to finished products is important because of the variability in medicinal plants' active components. One of the plants that have been used as an antidiabetic herbal medicine is A. paniculata. This study aims to determine ?-glucosidase inhibitory activity, the content of several metals, FTIR spectrum profile, and putative identification of A. paniculata metabolites using UHPLC-Q-Orbitrap HRMS from different geographical origin. We found that ethanol extract of A. paniculata gave higher inhibition of ?-glucosidase activity compared to water extract. Eight metals were determined using FAAS and FAES, and calcium showed the highest content in all A. paniculata samples. FTIR spectra of A. paniculata showed a similar profile and only differed in the absorbance. We detected the presence of OH, C=O, C=C aromatic, and C-O functional groups in A. paniculata extract. About 32 metabolites were putatively identified in A. paniculata, mainly from phenolic and diterpene lactones class compounds based on UHPLC-Q-Orbitrap HRMS. Using a combination of FTIR spectra and peak area from 32 identified peaks, we can clustered A. paniculata based on its geographical origin. Based on the result obtained, the geographical origin of A. paniculata affected the metals and metabolites composition and level, resulting in different levels of ?-glucosidase inhibitory activity.
