Abstract. Hamaidia K, Soltani N. 2021. Short communication: New report of Aedes albopictus in Souk Ahras, Northeast Algeria. Biodiversitas 22: 2901-2906. The present paper reports the occurrence of Aedes albopictus (Skuse), an aggressive Asian tiger mosquito in the city of Souk-Ahras (Northeast of Algeria). A “26 April” estate’ inhabitants (Souk-Ahras province) have reported unusual daytime bites by a striped mosquito. On early September 2020, an intensive field inspection was carried out for potential mosquito breeding sites in the locations around the city. A total of 105 specimens (8 larvae, 24 pupae and 73 adults) of Ae. albopictus were collected in a pile of old tyres in a residential garden. After previous records of this species in Algeria, this is the first evidence of its presence in Souk-Ahras province, and the findings enhance combined public participation with professional validation in surveillance of vector borne-diseases programs with emphasis on the need for sensitising citizens about controlling this important vector.

Indonesia is an archipelagic country and is known as one of the world’s megadiverse countries and has three zoogeographic zones. Therefore, proper conservation efforts to face extinction threats are very important, and the fundamental steps to better understanding it’s biodiversity provide valuable baseline information. Amphibians and reptiles (herpetofauna) are main biodiversity components that experience extinction threats but, at the same time, provide an important role in ecosystems and community life. This paper is the first to provide a checklist of the herpetofauna of Peleng Island, Banggai Kepulauan, Central Sulawesi, Indonesia. Data was gathered by field surveys and supplemented by references. Visual Encounter Surveys (VES) were undertaken in 8 sites. A total of 34 species are listed herein, 29 (85%) of them revealed from this fieldwork, and 21 (62%) new for the island. At least two candidate new species of the genus Cyrtodactylus were confirmed by molecular data and one frog of the genus Kaloula also suspected new species.

Distribution and prevalence of scurf (Monilochaetes infuscans) on sweet potato (Ipomea batatas) in West Java, Indonesia. Biodiversitas 22: 2876-2883. Scurf (Monilochaetes infuscans) is a common sweet potato (Ipomoea batatas (L.) Lam.) tuber skin discoloration disease, that can cause significant yield loss, especially when tubers are destined for the export market. Despite the economic importance of the disease in West Java, Indonesia, there is a dearth of information on the distribution and severity of scurf disease. To address this gap, a disease survey was carried out across 28 fields in six districts (Bandung, Sumedang, Garut, Kuningan, Cianjur, and Bogor) of West Java from 2016 to 2018. Scurf disease prevalence was recorded by counting the number of infected tubers from 15 plant samples per field. Disease incidence was recorded when at least one infected tuber per plant was observed. Severity of scurf diseases was estimated by measuring the percentage of infected surface area, using AutoCAD map 2019. Results show that scurf was distributed across all locations surveyed. Disease incidence ranged from 1.68% (Cikadu) to 27.56% (Cilembu) while disease severity ranged from 7.92% (Cikadu) to 57.00% (Cilembu). Based on potato dextrose agar (PDA) culture plates, 68.57 % of tubers were infected with various fungi. Among them, M. infuscans (32.14 %) was the most prevalent and most widely distributed pathogen associated with the scurf disease across sweet potato fields in West Java, Indonesia. To effectively manage the disease and improve farmers’ income, it is imperative to establish effective and sound integrated disease mitigation strategies based on the results of this survey.

Population status and sustainability of Yellow-crested cockatoo (Cacatua sulphurea sulphurea) on Pasoso Island, Central Sulawesi, Indonesia. Biodiversitas 22: 3277-3283. Yellow-crested cockatoo (Cacatua sulphurea sulphurea) is a sub-species of Cacatua sulphurea which is spread in Sulawesi and several surrounding islands, including on Pasoso Island. This cockatoo population is under tremendous pressure until categorized as critical. So far, there is no comprehensive information about population status and sustainable population size. The objective of this research area was to assess the current status or size of the population, study the size of the sustainable population and study the determinants factor of the Yellow-crested cockatoo population sustainability on Pasoso Island. The result showed that the population of the Yellow-crested cockatoo in Pasoso Island was only 3 individuals, consists of 2 adults as the parent, and 1 young individual. From 5 analyzed scenarios of the minimum sustainable population size, the 4.1 scenario is the best scenario to maintain the sustainability of the Yellow-crested cockatoo population on Pasoso Island, which is a minimum population size of four pairs (8 adult individuals). The determinants of population sustainbility analysis with the minimum sustainable population size (8 individuals or 4 pairs of broodstock) showed that inbreeding and predation as determinant factors. These factors need to be controlled through mating management and predator control by both humans and carnivorous animals.

Abstract. Kartika EBA, Siregar LAM, Basyuni M. 2021. Distribution of polyprenol and dolichol in oil palms from Pisifera parents and mature plants from tissue culture propagation. Biodiversitas 22: 3423-3436. Oil palm tissue culture is carried out through indirect embryogenesis, which causes somaclonal diversity to occur at the in vitro propagation stage, especially in the callus growth phase. In the cells of all living organisms can be found a group of polyisoprenoid compounds. This study aims to determine variations in oil palm plants resulting from tissue culture based on the presence of polyisoprenoid compounds. Oil palm leaf samples (Elaeis guineensis Jacq.) were collected from the plantation of PT. Socfin Indonesia, North Sumatra, Indonesia. Sample extraction, saponification, isolation of polyisoprenoid compounds, and two-dimensional thin-layer chromatography analysis were carried out to obtain data related to total lipids, polyprenol and dolichol profile, and Carbon (C) chain length polyprenol and dolichol from oil palm leaves of Pisifera parents and their propagated derivatives by indirect embryogenesis. The results showed that the amount of polyisoprenoid mother S24 was 2.41 mg/g higher than that of the parentS8 at 2.17 mg/g dry weight. Total polyisoprenoid fromS8 plants in vitro ranged from 0.71 - 8.53 mg/g dry weight, with the lowest total polyisoprenoid found atS894/22 and the highest was found atS893/2. While for total polyisoprenoid from plant tissue culture of Pisifera S24 ranged from 0.73 - 8.05 mg/g dry weight, with the lowest total polyisoprenoid found at S24H14 and the highest was found at S24H16. The parent plants of Pisifera S8 and S24, as well as plants resulting from tissue culture, were categorized as having lipid pattern type II, which showed a balanced distribution of polyprenol with dolichol. The longest carbon chain was found in vitro plantsS8 93/4 ranged from C50-C110, while the shortest was found in plants produced in vitro S24H7 starting from C45-C55. There were variations in the carbon chain length of polyprenol and dolichol in the leaf samples derived from in vitro propagation of the Pisifera S8 and S24 parents.

The development of unlabeled probes-high resolution melting (UP-HRM) marker on SAD, IAA27 and ACC genes of oil palm. Biodiversitas 22: 3356-3362. The unlabeled probes-high resolution melting (UP-HRM) marker is a useful tool for detecting of single nucleotide polymorphisms (SNPs). The objectives of this study were to develop UP-HRM markers to differentiate specific SNPs patterns on oil palm. The marker was developed and tested with Elaeis guineensis (Eg), Elaeis oleifera (Eo), Eo x Eg (hybrid), and was validated with 53 individuals of BC1F1 populations ((Eo x Eg) x Eg). Four UP-HRM markers were developed based on 2 SNPs in the stearoyl-acyl-carrier-protein 9-desaturase (EgSAD), 1 SNP in the auxin-responsive protein IAA27-like (EgIAA27), and 1 SNP in the 1-amino cyclopropane-1-carboxylate oxidase (EgACC) genes. The SNP discovery result showed that Eg was represented a reference homozygote genotype, while Eo was represented as an alternative homozygote genotype and the Eo x Eg hybrid was represented as a heterozygote genotype in all genes. The typical UP-HRM melt curve graph was successfully generated. This result was consistent with each genotype model for all four markers. The UP-HRM markers can distinguish each genotype according to the single-pass sequencing results. Furthermore, dendrogram analysis on validation divided 53 BC1F1 samples into three cluster groups.

Antiplasmodial activity, biosynthetic gene clusters diversity, and secondary metabolite constituent of selected Indonesian Streptomyces. Biodiversitas 22: 3478-3487. Actinobacteria of the genus Streptomyces are known as the primary candidate antibiotics, but still limited for antiplasmodial drugs. This study aimed to investigate the antiplasmodial activity, the biosynthetic gene clusters (BGCs) diversity, and the secondary metabolites constituent of selected Indonesian Streptomyces. The bacteria were isolated from various habitats: karst soil (GMR22), mangrove sediments (BSE7F and SHP 22-7), and marine sediment (GMY01). Molecular identification by 16S rDNA sequencing were performed for confirmation and morphological characterization by scanning electron microscope (SEM) were performed for identification. In vitro antiplasmodial assay was performed on human Plasmodium falciparum FCR-3. The BGCs which encode secondary metabolites were analysed using antiSMASH version 5 based on available whole genome sequence (WGS) data. The secondary metabolites were obtained from liquid fermentation followed by extraction using methanol and ethyl acetate. The secondary metabolites constituent was determined by liquid chromatography tandem mass spectrometry (LC-MS/MS). The molecular identification showed that GMR22 had similarity to Streptomyces lactacystinicus (98.02%), while BSE7F was similar to Streptomyces althioticus (97.06%), SHP 22-7 was similar to Streptomyces rochei (94.84%), and GMY01 to Streptomyces odonnellii (98.57%). All of isolates had morphological characteristics as the genus Streptomyces bacteria. The highest Plasmodium inhibition (81.84 ± 3.5%) was demonstrated by ethyl acetate extract of marine-derived Streptomyces sp. GMY01 (12.5 µg/mL). Non-ribosomal polyketide synthetase (NRPS), polyketide synthase (PKS) and hybrid of NRPS-PKS were the major BGCs in all Streptomyces. Majority of the Streptomyces produced compounds containing CHON elements with molecular weight approximately 100-400 Da. The active extract of GMY01 bacterium had five major detected compounds, namely kuraramine (C12H18N2O2), laminine (C9H20N2O2) 2-ethylacetanilide (C10H13NO), propoxur (C11H15NO3), and 3-methyl-1,2-diphenylbutan-1-one (C17H18O). This Indonesian marine bacterium is potential for bioassay guided isolation of antiplasmodial compounds in the future studies.

Isolation and identification of thermophilic amylolytic bacteria from Likupang Marine Hydrothermal, North Sulawesi, Indonesia. Biodiversitas 22: 3326-3332. The aims of the research were to isolate and identify the amylase-producing thermophilic bacteria from Likupang Marine Hydrothermal, North Sulawesi, Indonesia. The bacteria were characterized based on the colony and cell morphology and subsequently screened for their amylase activities. The bacterial isolates were identified based on 16S rRNA gene sequences. There were 12 thermophilic bacteria isolates from Likupang Marine Hydrothermal that were able to produce amylase. Two selected isolates (L3 and L9) had an amylolytic index value in the range of 3.04-3.52 at 55oC. The colonies of L3 and L9 are circular, and they are Gram positive, rod-shaped, and motile bacteria. Based on the 16S rRNA gene sequences and phylogenetic tree analysis showed that L3 was closely related to Bacillus caldotenax with 93% similarity while L9 had 99% similarity to B. caldotenax. Hence, we identified L3 isolate as Bacillus sp. strain L3 and L9 isolate as B. caldotenax strain L9. Both of these bacteria were grouped as Geobacillus thermoleovorans.

Abstract. Wisanti, Aloysius DC, Zubaidah S, Lestari SR. 2021. Variation in morphological characters of Marsilea crenata living in floating aquatic, emergent aquatic, and terrestrial habitats. Biodiversitas 22: 2853-2859. Marsilea is a hydrophyte fern that has plasticity often influenced by enviroment. Several of the species with different habitats or geographies show morphological variations. Therefore, this study aims to analyze the variations in morphological characters of M. crenata growing in aquatic and terrestrial habitats. A total of 38 plant samples were collected from their natural habitats including floating aquatic, emergent aquatic, and terrestrial types. Morphological variations data included 4 qualitative characters and 9 quantitative characters. The quantitative characters were analyzed by one-way ANOVA to test for differences, while the combination of both characters was analyzed using Principal Component Analysis (PCA) to determine the distribution between groups. The data results showed a separate grouping pattern between aquatic and terrestrial populations. These two groups showed significant differences in 5 quantitative characters and 2 qualitative characters. However, emergent aquatic samples’ distribution pattern is closer to the terrestrial group. The characters’ similarity of these two populations was in ??the rhizomes air spaces area, the position between leaflets and absence of red streak on the abaxial lamina. Conclusively, the variations in M. crenata’s morphological characters indicate that this plant has experienced morphological adaptations to water availability in its environment.

Abstract

Abstract. Uday UKS, Bhakat RK.. 2021. Short Communication: Assessment of Pteridophytes’ composition and conservation status in sacred groves of Jhargram District, South West Bengal, India. Biodiversitas 22: 3171-3178. Sacred groves are significant as community-preserved areas and have contributed to the conservation of biodiversity, thereby playing a key role in environmental management. The ecological and related cultural values of the species and the activities of local communities would make it possible to understand the importance of the protection of the sacred groves and also to prepare integrated approaches to biodiversity at the ecosystem level. Thus, this study aimed to investigate the status of pteridophyte diversity in the sacred groves of Jhargram district, West Bengal, India. The study showed that 77 pteridophyte species belonging to 44 genera and 15 families were collected and described, of which nine species were classified as Lycopodiopsida and sixty-eight species as Polypodiopsida. The floristic analysis revealed the dominance of the order Polypodiales (70.13%) followed by Aspleniaceae (23.38%), and Polypodiaceae (23.38%). The results also showed the predominance of the genera Selaginella with five species. Of the total species, 76.62% were terrestrial species, 14.29% were epiphytes, and 9.09% were aquatic species. The highest frequency is shown by Class C (25.97%) and major pteridophyte plants (81.81%) were not been evaluated till now.
