Morphogenetic identification, description and pathogenicity of novel pathogens on Iraqi Wheat Plant (Triticum aestivum) causing head blight and crown rot diseases. Biodiversitas 22: 2999-3005. Wheat (Triticum aestivum L.) plant suffers from several fungal diseases, such as Fusarium head blight, root rot, and crown rot diseases that reduce the quality and quantity of grains. Two Fusarium species (Fusarium brachygibbosum and F. falciforme) have been isolated from wheat roots and rhizosphere regions showing disease symptoms in wheat fields in Al-Hwuir and Al-Qurna districts at Basra province. Each fungal species had been characterized morphologically and molecularly based on DNA sequence data for the translation elongation factor 1- ? (TEF-1 ?). Results of pathogenicity test showed that F. falciforme and F. brachygibbosum at the third week of inoculation induced head bleached symptoms with the severity of 12.46% (with significant differences P > 0.05) and 7.14%, respectively on Abu Ghraib 3 cultivar. Yellowing and necrosis symptoms were recorded in stem and crown region. The mean of disease severity (as necrosis on basal stem and crown) was 5% and 3.34% for F. falciforme and F. brachygibbosum, respectively. The two Fusarium species were identified as new pathogens on wheat plants in Iraq, causing head bleaching, stem and crown necrosis.

Abstract. Cahyo AN, Murti RH, Putra ETS, Nuringtyas TR, Fabre D, Montoro P. 2021. Assessment of factual measurement times for chlorophyll-a fluorescence in rubber (Hevea brasiliensis) clones. Biodiversitas 22: 3470-3477. Chlorophyll-a fluorescence is widely used to determine the stress tolerance levels of some plant species. Measurement of chlorophyll-a fluorescence is accurate if the duration of dark adaptation is well defined and optimal Fv/Fm (maximum quantum yield of primary photochemistry/photosynthesis) is achieved. Leaf clips are usually used to darken the leaf prior to measurement. This procedure takes time and limits the use of chlorophyll-a fluorescence parameter in high-throughput screening of genetic populations. This study aimed to determine the most suitable time for the chlorophyll-a fluorescence measurement. This study was carried out on several rubbers (Hevea brasiliensis Müll. Arg.) clones and consisted of two steps. The first step was conducting the measurements at five different times at night: at 7.30, 8.00, 8.30, 9.00, and 9.30 p.m. The second step was conducting the measurements at daytime, which consisted of two factors. The first factor was the measurement time, which was divided into two categories: 7.30 a.m. and 1.30 p.m. The second factor was the duration of dark adaptation using leaf clips, which consisted of nine levels: 0, 15, 30, 45, 60, 75, 90, 105, and 120 min. Additional treatment (measurement at 9.00 p.m. without using leaf clips to darken the leaf) was used as a control. This study revealed that a dark adaptation time of two hours after the sunset was long enough for the rubber leaves chlorophyll-a fluorescence transient to be measured without using leaf clips for the dark adaptation. If the measurement is conducted by 7.30 a.m., the clone RRIM 600, GT1, and SP 217 required 15 min of dark adaptation, whereas clone PB 260 required 60 min of dark adaptation. Furthermore, measurement of chlorophyll-a fluorescence in the afternoon is not recommended due to the potentially high microclimate fluctuation.

Abstract. Ramadhanti N, Melia S, Hellyward J, Purwati E. 2021. Characteristics of lactic acid bacteria isolated from palm sugar from West Sumatra, Indonesia and their potential as a probiotic. Biodiversitas 22: 2610-2616. Palm sugar is one of the sugar products from palm sap processing. It is made by reducing the water content in the sap to certain water content and is used as a natural sweetener. This study used palm sugar from 3 producers, i.e. Marapalam Gula Semut (MGS), Tetesan Gula Semut (TGS) and Sungayang Gula Semut (SGS) in Sungayang, West Sumatra, Indonesia. This study aimed to isolate and identify lactic acid bacteria (LAB) from palm sugar, potentially as a probiotic, using the 16S rRNA method. The probiotic potential of bacteria was selected based on morphology, biochemical properties, resistance to acidic pH 3 and 0.3% bile salts, antimicrobial activity against pathogenic bacteria. The results showed that lactic acid bacteria isolated from palm sugar are Gram-positive rod-shaped (Bassil), negative catalase, and homofermentative. The total colonies of lactic acid bacteria ranged from 50 x 107 cfu/g - 117 x 107 cfu/g. The MGS lactic acid bacteria had 74.71% viability resistance in acidic pH 3, 65.52% viability against bile salts (0.3% ox gall). The diameter of inhibition against Escherichia coli O157 was 18.20 mm, Propionibacterium acnes (18.00 mm), Acinetobacter baumannii (14.19 mm), and Listeria monocytogenes (16.35 mm). The phylogenetic tree based on 16S rRNA showed that the MGS isolate has a similarity of 99.92% to Lactobacillus fermentum strain 1743. It can be concluded that Lactobacillus fermentum strain 1743 isolated from palm sugar has potential as a probiotic.

Abstract

Abstract. Arsad S, Putra KT, Latifah N, Kadim MK, Musa M. 2021. Epiphytic microalgae community as aquatic bioindicator in Brantas River, East Java, Indonesia. Biodiversitas 22: 2961-2971. One way to monitor water quality is by using biological indicators, namely epiphytic microalgae (periphyton). This study aims to analyze the epiphytic periphyton community structure and analyze the river health status using a saprobic index. The research location was in the Brantas River, Blitar District, East Java, Indonesia. The method used was a survey with sampling at three sites based on the purposive sampling technique. Periphyton samples were taken using the quadrant transect (5x5 cm²) method and then identified using the Lackey Drop Micro transect Counting Method. The results show that the periphyton community structure is in balance, and there are six divisions of 59 different genera. The six divisions found are Bacillariophyta, Cyanophyta, Chlorophyta, Charophyta, Ochrophyta, and Rhodophyta. The most identified genus is from the Bacillariophyta Division with 28 genera, while the least identified genus is the Rhodophyta division with 1 genus. The abundance ??ranges from 242,800-1,229,174 cells. cm-2, the relative abundance index ??of periphyton ranges from 3-60% (site 1), 9-57% (site 2), 1-62% (site 3), the diversity index ranges from 1.212-2.617 (moderate), the uniformity index ranges from 0.307-2.151 (high), the dominance index ranges from 0.105-0.549 (moderate), and the saprobic index ranges from-0.18 to 0.55. Moreover, supporting water quality parameters are still optimal, except for ammonia and total organic matter parameters that exceed the quality standard. Based on the saprobic value obtained, the water quality of the Brantas River in Tawangrejo Village can be classified into the category of ?/?-mesosaprobic to ?-mesosaprobic saprobic levels with mild to moderate levels of pollution.

Abstract. Widyasari F, Sayuti M, Salampessy RBS. 2021. Short communication: Production, distribution and conservation analysis of Cherax crayfish endemic to Papua and West Papua Provinces, Indonesia. Biodiversitas 22: 3271-3276. Freshwater crayfish is a species from the Parastacidae family that is indigenous to Papua and West Papua Indonesia. This study analyzes the amount of production, economic value, distribution, and conservation of freshwater crayfish endemic to Papua and West Papua. The production data were obtained from Stasiun Karantina Ikan or Fish Quarantine Inspection Agency in Papua and West Papua, Indonesia. Data regarding the price of Cherax crayfish were determined based on interviews with freshwater crayfish sellers. Furthermore, the distribution of freshwater crayfish was explained based on a literature review, while the data of its conservation were retrieved from the IUCN Red List of Threatened Species. Cherax crayfish caught from wild populations in West Papua is known higher than in Papua. Increased sales revenue could support the economy of the local community. There have been 25 species of freshwater crayfish identified, nine of which were from West Papua and 16 from Papua. Three species were under Endangered (EN), Least Concern (LC), Vulnerable (VU) status, respectively. Four species were under Data Deficient (DD) status, while the rest were unidentified. Overfishing of Cherax crayfish causes decline Cherax crayfish stock in the wild, future researchers are expected to conduct more specific studies that include relevant stakeholders regarding the conservation of Cherax crayfish that are endemic to Papua and West Papua.

Abstract. Ratnani DA, Junitha IK, Kriswiyanti E, Dhana IN. 2021. The ethnobotany of Ngusaba ceremonial plant utilization by Tenganan Pegringsingan community in Karangasem, Bali, Indonesia. Biodiversitas 22: 2078-2087. Tenganan Pegringsingan is an ancient village in Bali, Indonesia, which often performs several ceremonies with high intensity. One of them is the Ngusaba ceremony, where many plants are utilized both in species and quantity. Hence, this study aimed to identify the species, family, local names, sources, and parts of plants, used for Ngusaba ceremonies by the Tenganan Pegringsingan community including the Index of Cultural Significance (ICS). Data analysis was qualitative and quantitative. Furthermore, the qualitative method was used to obtain data on the plants’ local names, while snowball sampling was applied to select key informants through in-depth interviews and moderate participation. The results showed that the 130 species distributed in 56 families mostly belonging to the purchased source (34.61%). The Poaceae is the largest family, while the most widely used part of the plant is the leaf. Based on the ICS analysis results, a range of 2-114 values was obtained. The highest value is Base (Piper betle L.) and kangkung (Ipomoea batatas L.) as lowest.

Abstract. Iskandar BS, Iskandar J, Mulyanto D, Alfian RL, Suroso. 2021. Traditional market, social relations, and diversity of edible plants traded in Beringharjo Market, Yogyakarta, Indonesia. Biodiversitas 22: 2012-2024. Although the modern markets have rapidly developed in many cities of Indonesia in the last decades, the urban traditional markets still exist in Indonesia, including Beringharjo Market, Yogyakarta. This study illustrated the story and social relation activities, biodiversity of edible plants traded, and market chain system of edible plant supplies from rural areas to Beringharjo Market. The study was conducted between February and March 2019. The method used in this study was qualitative data with an ethnobotanical approach. Some field techniques, including plant surveys and deep interviews with competent informants, were employed to collect the primary data. The result of this study showed that historically the economic activities of Beringharjo Market had been existing very a long time at least documented in 1758, while the market building was constructed by the Sultanate of Yogyakarta Palace, Sri Sultan Hamengku Buwono VIII in 1925. Like other traditional markets in Indonesia, the Beringharjo Market has played an important role in developing local economic activities, as the main place for the transaction of various edible plants and meeting for many ethnics, making the important as center of biocultural diversity at the local level. An ethnobotanical survey revealed that 93 edible plant species belonging to 37 families with a total of 159 landraces were commonly traded. The Beringharjo Market has traditionally traded a high diversity of edible plants that are supplied from local farmers productions of Yogyakarta and Central Java. Therefore, this traditional market has an important role in enhancing local economic development and contributing to promoting and conserving local plant diversity in this region.

Antibacterial activity, antioxidant potential, total phenolic and flavonoids of three plant species of Rubiaceae from Banggai Island, Indonesia. Biodiversitas 22: 2773-2778. The present study was carried out to study the antioxidant and antibacterial potential of different plant parts (stems, leaves, and fruit) of the family Rubiaceae (Timonius celebicus, Psychotria celebica, Gardenia mutabilis) collected from the Banggai Islands. Screening of antibacterial activity against Staphylococcus aureus and Escherichia coli and antioxidant potential was carried out by Thin Layer Chromatography- Bioautography. Determination of IC50 and Minimum Inhibitory Concentration (MIC) was carried out by microdilution in a 96-well microplate. Total phenolic and total flavonoid content was determined by the colorimetric method. Total phenolic (TPC) and total flavonoid content (TFC) was varied from 29.78-380.99 mg GAE/g extract and 105.61-841.18 mg QE/g extract, respectively. The antioxidant capacity of three plant species was varied from moderate to very strong. Antioxidant Activity Index (AAI) was found more affected by TFC than TPC. The antibacterial activity of the extract against E. coli was weak, while some extracts have moderate antibacterial activity against S. aureus

Climate effects on the growth of Pinus latteri and Pinus kesiya at the Intakin Silvicultural Research Station, Chiang Mai Province, Thailand. Biodiversitas 22: 2512-2519. Relationship between the growth of two pine species, Pinus latteri and Pinus kesiya, and past climatic factors are studied. Standard dendrochronological techniques were used on 40 core samples obtained from 20 trees from each species to cross-date tree-ring width and determine the growth patterns of each species. Over a period spanning from 1980-2019 (P. latteri) and 1984-2019 (P. kesiya), the calculated mean ring-width values were 0.596 and 0.430 cm per year, respectively. An analysis of the relationship between tree-ring width of P. latteri and climatic data indicated a positive correlation with the relative humidity in April. However, a negative correlation was observed with the temperature in April and August. The factors limiting the growth of P. latteri the most were the temperature in August (r2=0.228; P<0.01). The index constructed for P. kesiya indicated a positive correlation with extreme minimum relative humidity in December and extreme maximum temperature in March. The factors limiting the growth of P. kesiya with a significant correlation, were the relative humidity in April (r2=0.173) and temperature in May (r2=0.184). These results can be used to plan the management of forest plantations and to select suitable areas for the cultivation of pine trees in Thailand

Abstract. Suryaman GK, Soejoedono RD, Poetri ON, Setiyono A, Handharyani E. 2021. Detection of infectious bronchitis-like virus isolated from psittacines at breeding facilities. Biodiversitas 22: 4261-4265. Captive breeding of parrots has been carried out in Indonesia to fulfill market demand for pets and support conservation efforts. However, there is limited information about Infectious Bronchitis Virus (IBV) in captive parrots. Therefore, this study examines IBV virus in psittacines and observes the pathological changes in inoculated chicken embryos. Cloacal swabs were obtained from 31 psittacine birds consisting of 15 sulfur-crested cockatoos (Cacatua sulphurea), 10 palm cockatoos (Probosciger aterrimus), two blue and yellow macaws (Ara ararauna), two green-winged macaws (Ara chloropterus), and two scarlet macaws (Ara macao). Swabs were also injected for 9 days to obtain specific pathogen-free (SPF) embryonated chicken eggs, and allantoic fluids were collected after 7 days. The residual cloacal swabs and allantoic fluid samples were tested for IBV by reverse transcription polymerase chain reaction (RT-PCR) targeting spike 1 glycoprotein (S1) and nucleoprotein (N) gene by primer set XCE2+/XCE2- and IBVN+/IBVN-, while the harvested inoculated embryos were examined for histopathological lesions by hematoxylin-eosin staining. Three and one allantoic fluid of C. sulphurea and P. aterrimus respectively were found positive for IBV by primer set IBVN+IBVN-. However, there was no significant difference in the inoculated embryo's gross anatomy, and the slight histopathological changes observed were not caused by the virus. Therefore, it is unclear whether the IB-like virus causes clinical disease manifestation in both Psittacine species or the chicken embryo. This is the first study carried out on IB-like viruses for both species.
