Effect of fermentation on sensory quality of Liberica coffee beans inoculated with bacteria from saliva Arctictis binturong Raffles, 1821. Biodiversitas 22: 3922-3928. Fermentation is one of the post-harvest steps that influence the quality of coffee. This study aims to determine the sensory quality of Liberica coffee beans cv. Liberoid Meranti (LiM) fermented using a consortium of bacteria from saliva Arctictis binturong Raffles, 1821 with varying fermentation periods. Fermentation was performed in a wet process for 0, 4, 8, 12-hours in triplicate. The parameters observed were caffeine, protein, and fat content. The sensory quality of brewed coffee was conducted refers to the standard Speciality Coffee Association (SCA). The data obtained were processed with Minitab version 16 for Windows and analyzed using ANOVA with a level of 5% and followed by a post hoc test. The results showed that coffee fermented for 0, 4, 8, 12 hours has protein content of 15.31-15.67%; 15.09-16.62%; 12.71-13.07%; 14.64-14.69%; fat content of 9.48%; 10.20%; 9.96%; 10.21%; and caffeine content of 1.05%; 0.99%; 0.96%; 1.02%, respectively. The length of the fermentation period affects significantly (p<0.05) the content of protein, fat, and caffeine. The final score of cupping test in coffee fermented for 0, 4, 8, and 12 h were 77.63; 78.13; 82; and 80.25, respectively. A fermentation period of 8 and 12 hours improve the flavor score that categorized fermented Liberica coffee as specialty coffee (8.00 - <9.00 = superior).

Abstract. Putri WU, Qayim I, Qadir A. 2021. Invasive species in the soil seed bank of two limestone hills in Bogor, West Java, Indonesia. Biodiversitas 22: 4019-4027. Study on the impact of invasive species on plant communities has been conducted mainly on the aboveground vegetation. The impact on the soil seed bank has received less attention mostly due to practical difficulties in conducting seed bank assessment. Evaluation of seed bank composition is useful in detecting invasive plant species that may have been present as buried seeds. Information on the vegetation composition in Nyungcung and Kapur hills both under and aboveground is available, but specific information on invasive species has not been discussed yet. This paper describes and analyzes the structure (composition and density) of invasive species in the soil seed bank. Eighty soil samples were taken from the study sites. The soil seed banks were analyzed using seedling emergence and seed extraction methods. 2602 and 1280 seedlings emerged from soil seed bank of Nyungcung and Kapur hills, respectively. From that number, approximately 48.96% and 68.51% of the seedlings are invasive species in the soil seed bank of Nyungcung and Kapur hills. The invasive species were dominants in the soil seed bank of the two sites. Cecropia peltata and Clidemia hirta were the most abundant invasive species in the Kapur and Nyungcung hills seed bank, respectively. Species richness and seedling density were higher in Nyungcung hills than Kapur hills, which were invaded for a relatively longer period.

Biodiversitas 22: 2771-2777. Zinnia elegans Jacq. is highly valued as an ornamental plant with a variety of flower colors, sizes, and shapes. Polyploidization has been reported in Z. elegans with pom-pom flowers, nevertheless, the variation in chromosome numbers of various flower shapes has yet to be investigated. This study aimed to analyze the chromosome numbers of Z. elegans Jacq. cultivar “California Giant,” “Lilliput,” and “Cactus Flowered Mix” with single, double, and pom-pom flowers to determine their variations and identify the morphology of the flowers. Chromosomes were prepared using the squashing method, and images were analyzed using the Chromosome Image Analyzing System (CHIAS) IV. The minimum of 5 slides was prepared for each flower type from each cultivar. The results show that the three cultivars are diploid plants (2n = 24) with varying flower morphology. The single and double flowers of Z. elegans “Lilliput” and the single flower of “Cactus Flowered Mix” showed no variation in chromosome numbers. In contrast, chromosome number variation was found in the pom-pom flower of Z. elegans “California Giant” (2n = 22, 24, 48) and the double flower of “Cactus Flowered Mix” (2n = 9, 13, 15, 24). Two cultivars, Z. elegans “California Giant” and Z. elegans “Cactus Flowered Mix,” were successfully analyzed using CHIAS IV. Statistical analysis using a t-test (? = 0.05) showed that the total chromosome length of Z. elegans “California Giant” (2n = 24) was significantly greater than that of Z. elegans “Cactus Flowered Mix” (2n = 24). Chromosome satellites were found in both cultivars.

Evaluation of indigenous fungal entomopathogens and aqueous leaf extract of Annona muricata against Polyphagotarsonemus latus infesting Jatropha curcas in Indonesia. Biodiversitas 22: 2648-2655. The broad mite Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae) has been known to cause a great deal of injury on Jatropha curcas (L.) plantation in Indonesia. Isolation of indigenous fungal entomopathogens from rhizosphere soils, followed by pathogenicity assay was conducted to find an effective isolate for controlling P. latus. Additionally, the potential combination of the selected isolate and soursop (Annona muricata L.) aqueous leaf extract (SLE) was investigated. A total of 24 isolates were obtained by using the insect bait method. Four fungal isolates, namely Paecilomyces sp. 1., Lecanicillium sp., Beauveria sp., and Fusarium sp. 1, showed high conidial viability and were chosen as representatives to assess their pathogenicity against P. latus. Among the tested isolates, Beauveria sp. which had the highest conidial viability among the tested isolates, needed a shorter period to completely kill the tested mites. However, the compatibility test revealed the deleterious effect of SLE on Beauveria sp. SLE at all concentrations showed a very toxic effect on the fungus, therefore the two must be applied separately. Our results provide useful information on the effectiveness of indigenous entomopathogenic fungi Beauveria sp. and aqueous leaf extract of A. muricata as an alternative tool to control the broad mite P. latus on the J. curcas plantation.

Abstract. Wahidah BF, Afiati N, Jumari. 2021. Community knowledge of Amorphophallus muelleri Blume: Cultivation and utilization in Central Java, Indonesia. Biodiversitas 22: 2731-2737. Porang (Amorphophallus muelleri Blume) is a member of the Araceae edible family currently popular on Java island, Indonesia. In the last five years, this plant which has high economic value due to its various benefits as a food plant and medicinal plant has been cultivated in Central Java. This study aims to determine the knowledge of the people in Central Java about porang plant, its cultivation, and its use in people's lives. The method used is a qualitative method by applying data collection techniques through interviews, observation and documentation. Besides general interviews, this study gains more information from some informants whose purposively chosen amongst the respondents as people knowledgeable about porang in general, and in particular A. muelleri. Results showed that people in Central Java do have knowledge about porang plant even though it was not used as food ingredient. Public knowledge about porang cultivation is obtained from various sources including social media and people who strive for porang with success. Therefore, since the community is currently aware of the potential and economic values of porang, they have started to cultivate it. Porang has enormous potential to be developed in the future and provide welfare for the community.

Abstract. Sembiribg M, Sabrina T. 2021. Diversity of non-symbiotic nitrogen-fixing bacteria and their potential in andisols affected by the eruption of Mount Sinabung, North Sumatra, Indonesia. Biodiversitas 22: 3539-3544. Nitrogen is the main macro-nutrient that is very important for plant growth. Nitrogen is absorbed by plants in the form of NO3-or NH4+ ions from the soil. The andisols affected by the eruption of Mount Sinabung alter the chemical, physical and biological characteristics of the soil. As a result, the population of beneficial microbes in the soil decreased, so soil fertility also decreased. The aim of this research is to determine the diversity of nitrogen-fixing microbes in andisol soil affected by the Mt. Sinabung eruption. The soil samples used in this research were collected from andisols affected by the eruption of Mount Sinabung. The isolation of non-symbiotic nitrogen-fixing bacteria was carried out using a nitrogen-free Jensen medium. The results showed that five non symbiotic N-fixing bacteria can increase the N content in andisols affected by Mount Sinabung eruption. Enterobacter cloacae can increase soil N by 111.76% as compared to without microbial application.

The Fourier transform infrared spectroscopy from Diplazium esculentum and Rivina humilis analysis reveals necessary components in oil palm plantations of Ganoderma boninense control. Biodiversitas 22: 3645-3651. The Fourier transform infrared spectroscopy (FTIR) has been widely utilized for biological samples and biomolecular characterization. We aim to identify Ganoderma boninense through FTIR and obtain a functional group that can facilitate early basal stem rot detection. Here, positive control (KP) was not inoculated with G. boninense and negative control (KN) was inoculated with G. boninense. However, the treatment samples, Diplazium esculentum leaf extract, Rivina humilis leaf extract, and fungicide treatment, were not inoculated with G. boninense. The positive control oil-palm leaf samples exhibited spectral bands similar to those in the D. esculentum extract, R. humilis extract, and fungicide treatment. Strong bonds were observed at wavelengths 3379 cm-1, 2927 cm-1, 1639 cm-1, and 1056 cm-1. Others were moderate to weak, except the negative control samples with strong bonds at 2044 cm-1. This indicates amine N-H functional groups, alkane functional group C-H, functional group alkene C=C, C-O, functional group ester, and functional group isothiocyanate N=C=S (C4H5NS or CH2 = CHCH2N=C=S). The FTIR plot result denotes G. boninense through N=C=S Isothiocyanate functional group presence at 2140-1990 cm-1. This unique structure is only found in infected oil-palm leaf tissues of G. boninense. Our study suggests that FTIR spectroscopy is more beneficial than conventional methods in early detection of G. boninense infection in oil palm.

Species identification and prevalence of gastrointestinal helminths in Indonesian native chickens, and its impact on egg production. Biodiversitas 22: 4363-4369. Gastrointestinal parasite (GIP) infection is a severe problem of local chicken production, such as poultry and egg. Hence, the proper strategy to control the parasite invasion should be implemented regarding chicken productivity performance. Moreover, the existing environment is also essential in supporting chicken production. The study's objective was to determine the prevalence rate of the gastrointestinal helminths in four strains of Indonesian native chicken viz. Ranupane, Lokal Putih, Wareng, and Lurik, and the impact of the chicken egg production. A total of 280 chickens which were evaluated consisting of 70 birds from each strain, were tested for the prevalence of helminths eggs in a sample of feces and the effect on hen day production (HDP). The results showed that the percentage rates of infected chicken with helminths reached 56.43%, and the number of eggs per gram (EPG) in all four strains was considered moderate (115 EPG of feces). At the same time, the average of HDP per the four strains at 7 to 12 months was about 34.36-45.80%, which was in a normal range. The majority of helminths species found in examined native chicken were Ascaridia galli, Heterakis gallinarum, Raillietina spp., and Capillaria spp. The prevalence of GIP helminths did not negatively affect egg production in all four strains of chicken by moderately tolerant infections, and the HDP of chickens was normal.

Abstract. Pratasik SB, Lalamentik LTHX, Manoppo L, Budiman J. 2022. Deep sea squid in Sulawesi Sea, North Sulawesi Province, Indonesia. Biodiversitas 23: 1774-1779. This study aimed to identify one of the deep sea squids caught in artisanal fisheries by traditional fishermen in Sulawesi Sea, North Sulawesi Province, Indonesia. Samples were collected using the vertical hand line of traditional fishermen. The specimen was identified using Cytochrome c oxidase subunit 1 (CO1). For morphometric characteristics, all body parts were measured, and each part was compared with mantle size. Results showed that the specimen was identified as purpleback flying squid Sthenoteuthis oualaniensis and had 99.35% similarity to S. oualaniensis (CO1) from China (NCBI - MN101944) with sufficient genetic diversity. Based on the body size, species recorded in the present study belong to the dwarf form, the smallest form of S. oualaniensis groups. The species has the following morphometric characteristics: Head length is 0.28 of mantle length, tentacle length is 1.158 of mantle length, and fin area is 0.5 of mantle length. The fourth arm is the shortest and the second arm is the longest. S. oualaniensis recorded in the present study belonged to dwarf form living in equatorial waters. This finding may contribute to the list of marine resources of fisheries in Indonesian waters.

Abstract. Tarno H, Marsudi EW, Widjayanti T, Setiawan Y. 2021. Short Communication: Nematodes associated with Robusta coffee plantations in Malang District, East Java, Indonesia. Biodiversitas 22: 3306-3312. Coffee (Coffea spp.) is an important commodity in Indonesia. Nematodes have different roles, such as plant parasites and non-parasites, and are commonly found in the soil of coffee plantations. This research determined the ecological role, diversity, and abundance of nematodes in three coffee plantations in Malang District, East Java. We used a Whitehead tray to extract and isolate nematodes from the soil and root samples. Nematodes were identified based on their morphological characteristics. Analysis of variance and diversity indices were used to identify the differences among three coffee plantations. We identified eight nematode genera in the Ngantang sub-district site. In the Jatikerto Agrotechnopark and University of Brawijaya Forest sites, six genera were identified. Criconemoides, Pratylenchus, Xiphinema, Helicotylenchus are plant-parasitic nematodes, and Mononchus, Dorylaimus, Rhabditis, and Aphanolaimus are non-plant parasitic nematodes. Non-plant parasitic nematodes were more abundant than plant-parasitic nematodes in all sites. Non-plant parasitic nematode, Dorylaimus was the dominant genus in this study (272 individuals), ca. 35% of the total nematodes collected. In this study, the differences between coffee plantations and soil pH influence nematode abundance. The nematode abundance increases when the soil pH is lower.
