Abstract. Tarigan MRM, Corebima AD, Zubaidah S, Rohman F. 2021. Arthropods discovered in lower and upper pitchers of Nepenthes at Rampa-Sitahuis Hill, North Sumatra, Indonesia. Biodiversitas 22: 5358-5366. This research aimed to identify the families of arthropods trapped in the upper and lower pitchers of Nepenthes in Rampa Sitahuis Hill Area, North Sumatra, Indonesia. This descriptive qualitative research used an experimental method of data analysis. The arthropods were obtained from the upper and lower pitchers of N. rafflesiana, N. gracilis, N. eustachya, N. ampullaria, and N. sumatrana. Consequently, ten families of arthropods, namely Culicidae, Formicidae, Araneidae, Calliphoridae, Rhyparochromidae, Salticidae, Curculionidae, Blattellidae, Coccinellidae, and Tridactylidae were identified. Culicidae and Formicidae were the most abundant arthropods trapped in Nepenthes’ lower and upper pitchers because Nepenthes functioned as a microhabitat and breeding ground for Culicidae (adult mosquitoes) and Formicidae (ant larvae). Thus, these species established a mutualistic symbiosis in which ants used the tendrils of Nepenthes to lay their eggs and collected nectar generated by the honey glands, while the egg-laying process aids in the pitcher’s breakdown process.

Abstract. Madhi QH, Abass MH, Matrood AAA. 2021. The efficiency of some bioagent fungi in reduction of wheat seed decay and seedling damping-off disease with heavy metals interaction. Biodiversitas 22: 3984-3993. Biological control is an ecofriendly efficient measurement for disease control and heavy metals reduction in soils. The use of bioagent fungi such as Trichoderma koningii and Chaetomum globosum reduced the negative effect of pathogenic fungi that cause seed decay and the seedlings damping off wheat alone or by interaction with the concentrations of lead or cadmium, which increases the germination percentage of wheat seeds and reducing seedling damping off. It also reduced the severity index of wheat with pathogenic fungi and reduced the negative effect of interaction between heavy metals and pathogenic fungi on the severity index of the wheat.  Results showed that T. koningii and C. globosum reduced the effect of the interaction of R. solani with 200 mg/kg lead to 57.7 and 55.4%, respectively and R. solani and cadmium 3 mg/kg with 60 and 61.6%, respectively. T. koningii and C. globosum also reduced the effect of the interaction  F.solani with lead 200 mg/kg to 45.4 and 48.5%, respectively and  F. solani and cadmium 3 mg/kg to 46.8 and 52.5% respectively. The bioagent fungi also increased the fresh and dry weight of shoot and root system, T. koningii significantly increased the fresh and dry weight of shoot in the presence of R. solani. The results also indicated that there was a high significant difference in the use of C. globosum in increasing the fresh and dry weight of shoot and root system. T. koningii and C. globosum significantly reduced the effect of interaction between the pathogenic fungi and low concentrations of lead and cadmium leading to an increase in the fresh and dry weight of shoot and root system. They also increased the plant height in the presence of pathogenic fungi as well as reducing the negative effect of the interaction between heavy metals and pathogenic fungi in the height of wheat plants. No significant interaction was observed between the low concentrations of lead and cadmium and pathogenic fungi in the presence of bioagent fungi. The results exhibited that bioagent fungi can reduce the negative effect of interaction of pathogenic fungi with lead and cadmium on the total phenols content of wheat leaves, and no significant difference was recorded in the treatment of low concentrations with the pathogenic fungi. Results showed that bioagent fungi can reduce the negative effect of the interaction of pathogenic fungi with lead and cadmium on the total phenols content of wheat plant leaves. No significant differences were recorded in the treatment of low concentrations with the pathogenic fungi in the presence of bioagent fungi. The two bioagent fungi increased the concentration of chlorophyll a and b, total chlorophyll and carotenoids reduced anthocyanin in leaves, and increased chlorophyll stability index compared to the control treatment.

Abstract. Akbar MR, Purwoko BS, Dewi IS, Suwarno WB, Sugiyanta, Anshori MF. 2021. Agronomic and yield selection of doubled haploid lines of rainfed lowland rice in advanced yield trials. Biodiversitas 22: 3006-3012. Selection process is important step to obtain high yielding variety. This study aimed to obtain information on agronomic performance of doubled haploid lines of rainfed rice in advanced yield trials and select the best lines for multi-location yield trials. Two experiments were conducted in Bogor and Subang using a randomized complete block design for each location and consisted of 35 doubled haploid lines and two check varieties namely Ciherang and Inpari 18 as a treatment. The result revealed that interaction between genotype and environment (G x E) significantly affected all agronomic characters, except on the panicle length. Characters with high heritability value were all agronomic characters except number of productive tillers and productivity. Genotypic correlation analysis showed that number of productive tillers and weight of 1000 grains had significant and positive correlation with productivity. The selection used weighted selection index based on multivariate analysis. Fourteen lines were selected for multi-location yield trials. These lines had characteristics as follows: number of productive tillers (14.0-17.7 tillers), weight of 1000 grains (24.7-32.5 g), and productivity (5.0-6.2 ton ha-1). The selected lines can be evaluated further to obtain superior lines in multi-location trials.

Abstract. Danilova AN, Sumbembayev AA. 2021. The status of the Dactylorhiza incarnata populations in the Kalba Altai, Kazakhstan. Biodiversitas 22: 3180-3195. The article provides a botanical description of the current state of populations of threatened species Dactylorhiza incarnata (L.) Soo in the Kalba. The ecological analysis of the flora, the ecological optimum of the species, the vitality of plant communities, and the degree of anthropogenic load are presented in the research. On the basis of field surveys, four main populations were identified in 13 typical plant communities. Flora of plant communities of D. incarnata on the Kalba Ridge includes 117 species belonging to 30 families and 80 genera. The ecological optimum of D. incarnata can be spotted on the southeastern slopes with diffused lighting and moderate wind impact. The plant communities with optimal conditions are Filipendulo-Sanguisorbo-Thalictrosum and Carexoto-Equiseto-Filipendulo-Festucosum characterized by highly humid meadows and forest edges with low density and competition. Self-maintenance and reproduction of the species are fulfilled mainly by seeds, less often by vegetative reproduction of the finger root. Due to analysis of the age composition of populations and the dynamics and stability of plant communities the recovery index is within the optimum. Typical indicator species are Thalictrum simplex L., Salix viminalis L., Filipendula ulmaria (L.) Maxim., Sanguisorba officinalis L., Geranium pratense L., Festuca pratensis Huds.

Effectiveness of essential oil from the Artemisia herba-alba aerial parts against multidrug-resistant bacteria isolated from food and hospitalized patients. Biodiversitas 22: 2995-3005. The World Health Organization has sounded the warning on the diffusion of multidrug resistance (MDR) bacteria, requiring solutions and alternatives to solve the therapeutic failure that may occur. This study aims to evaluate the antioxidant activity and bactericidal effectiveness against MDR bacteria of Artemisia herba-alba essential oil (A-EO) collected from semi-arid region of Morocco. Chemical composition of the A-EO was determined by Gas Chromatography-Flame Ionisation Detector and Gas Chromatography-Mass Spectrometry, while the antioxidant activity was performed by DPPH scavenging activity and ?-carotene bleaching assay. Antibacterial activity of A-EO, performed by disc diffusion assay and broth dilution method, was tested against: four MDR strains (Escherichia coli, Staphylococcus aureus, Salmonella Typhimurium and Enterococcus faecalis) isolated from food matrices, two (Klebsiella pneumonia and Pseudomonas aeruginosa) from hospitalized patients, and Escherichia coli ATCC 25922 as reference strain. Davanone was the main compound among the 17 identified. An antioxidant activity with IC50 of 1.13±0.02 mg/mL, EC50 of 2.12±0.05 mg/mL and RC50 of 0.87±0.02 mg/mL was observed. A weak activity against P. aeruginosa was observed, while it was intermediate or high against the other bacteria. This study confirms that A-EO could be a suitable alternative to antibiotics in the infection treatment related to MDR bacteria.

Antibacterial and antioxidant potential of ethyl acetate extract from Streptomyces AIA12 and AIA17 isolated from gut of Chanos chanos. Biodiversitas 22: 3196-3206. Streptomyces has been recognized as a promising and productive source of antibacterial and antioxidant compounds. The phenotypic and genomic characterizations indicated AIA12 and AIA17 are closely related to the S. globisporus ARGB01 and S. misionensis S1-SC15, respectively. Incubation on yeast-malt extract broth for nine days (AIA12) and 11 days (AIA17) proved to be the best growth medium and optimum antibacterial production time for these two strains. Crude-extract of extracellular secondary metabolites, obtained by ethyl acetate extraction, demonstrated broad-spectrum inhibitory activity against Pseudomonas aeruginosa InaCC B52, Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 10876, Escherichia coli ATCC 25922, Salmonella Typhimurium ATCC 14028, and Listeria monocytogenes with minimum inhibitory and minimum bactericidal concentrations from 2.5 to 0.31 and 5.0 to 0.31 mg mL-1, respectively. Evaluation of antioxidant showed AIA17 crude-extract had moderate DPPH scavenging and antioxidant activities of 65.122% ± 0.56 and 28.178 ± 0.24 mg ascorbic acid equivalent antioxidant capacity (AEAC g-1), respectively. The identification of compounds through profiling with RP-HPLC showed optimum absorbance at 210 and 214 nm, which showed the presence of peptide groups in the constituent compounds' molecular structure. These findings indicate that Chanos chanos-derived Streptomyces produces valuable bioactive compounds with various promising biological activities.

Peptide profiling of goat milk fermented by Lactobacillus delbrueckii ssp. delbrueckii BD7: Identification of potential biological activity. Biodiversitas 22: 3136-3145. This study investigated the angiotensin-converting enzyme (ACE) inhibitory activity in fermented goat milk by Lactobacillus delbrueckii ssp. delbrueckii BD7, characterizing the peptide and its potential as a bioactive peptide. The starter culture (2%) was inoculated into pasteurized goat skim milk (11%), then incubated at 37 °C until it reached pH 4.6. Centrifugation at 6000 g x 10 minutes at 4 °C was applied. The supernatant obtained was then ultrafiltrated using a membrane cut-off with a molecular weight of 3 kDa, and the fraction obtained was analyzed to determine the inhibitory activity of ACE. Peptides were characterized using Nano LC / MS / MS, and identification as bioactive peptides was carried out based on a literature review. ACE inhibitory activity of fermented goat milk of Lb. delbrueckii ssp. delbrueckii BD7 was 55.98 ± 3.53%. A total of 157 peptides were released with molecular weights ranging from 770.78 - 2081.12 Da and having 7-19 amino acid residues. The main peptide was hydrolyzed from casein (72.6%), cleavage in the parent protein, specific for aliphatic and aromatic amino acids. Identification of bioactive peptides based on the similarity of amino acid residues at C-terminal obtained 28 ACE inhibitor peptides, 19 antioxidant peptides, and ten antimicrobial peptides. Some of these peptides have homologous sequences with previously reported peptides. Lb. delbrueckii ssp. delbrueckii BD7 has the potential as a starter culture to produce fermented milk, which is rich in biological activity.

Reproductive activity and morphometric assessment of three commercial species of sea cucumber (Echinodermata) from Karimunjawa National Park, Indonesia. Biodiversitas 22: 3333-3341. Sea cucumbers are benthic animals that have economic value and play an important role in the environment. Exploitation on large scale to fulfill high market demand, caused overexploitation. The number of sea cucumbers in Karimunjawa decreased as a result of limited management. There is no study conducted on the commercial sea cucumbers (Holothuria edulis, Pearsonothuria graeffei, and Bohadschia vitensis) in Nyamuk Island, Karimunjawa. The objectives of the research were to determine the morphometric assessment and the reproductive cycle of the three species. The morphometric assessment of weight showed that the weight of H. edulis ranges 247.3-2539g, P. graeffei 415.3-428.1g, and B. vitiensis 672.4-678.1g, respectively. The first maturity size of gutted body weight (GWB50) for H. edulis was 127.84g, P. graeffei 357.61g, and B. vitiensis 279.34g. The gonad somatic index (GSI) range 13.60-15.60% for H. edulis, 14.75-12.25% for P. graeffei and 10.83-14.57% for B. vitiensis. The result of gonad somatic index with a value above 10% means that it has entered the spawning phase. The above data indicated that the three commercial species spawned monthly, during new moon. It is likely that the spawning peak occurred when the temperature was increased. However, the pattern of reproductive activity in this study was limited at Nyamuk Island. Future studies are needed to observe whether there is a correlation between the increasing water temperature and the reproductive activity of the three sea cucumbers.

Abstract. Prianto E, Purwoko RM, Kasim K. 2021. Stock status of Nile tilapia (Oreochromis niloticus) in Aneuk Laot Lake, Sabang District, Aceh Province, Indonesia. Biodiversitas 22: 3364-3370. The Nile tilapia (Oreochromis niloticus) fishery in Aneuk Laot Lake plays a vital role in the local livelihoods. There is an indication of the massive exploitation of the species that potentially harm the stock and threaten its sustainability. This research aims to evaluate and assess the Nile Tilapia's stock status and use the results as baseline information to the management authorities. Surveys were conducted from May 2018 to April 2019 by collecting the length-weight data, sex, and gonadal maturity stage of 600 individuals. The Length-Based Spawning Potential Ratio (LB-SPR) was used to determine the Spawning Potential Ratio (SPR) and compared the results to the reference point. The results show that 50% of the females have matured at 15.1 cm, while the males at 17.8 cm. The individual fish length varies from 12.15 to 20.5 cm, whereas 50% of them were caught at size 17.8 cm (Lc50), meaning that predominantly of the fish had experienced first spawning before they were captured. The current SPR is about 5%, confirming the Nile tilapia's stock status is at an unhealthy state. Consequently, the precautionary approach, such as increasing the mesh size and limiting effort, should be considered to ensure the sustainability of the recruitment.

Abstract. Rezeki MS, Harahap IS, Sartiami D, Irmansyah, Watson GW. 2021. Identification key to nymphal and adult mealybugs (Hemiptera: Pseudococcidae) associated with dragon fruits in Indonesia. Biodiversitas 22: 3113-3118. Dragon fruit (Hylocereus spp.), belonging to Cactaceae family, is one of the horticultural commodities being developed in Indonesia for export. Phytosanitary regulations require that fruits for export must be free from diseases and pests, including mealybugs (Hemiptera: Pseudococcidae). Mealybug species commonly found on dragon fruits in Indonesia are Ferrisia virgata (Cockerell), Phenacoccus solenopsis Tinsley, Planococcus minor (Maskell) and Pseudococcus jackbeardsleyi Gimpel & Miller. Species-level identification of these insects in the nymphal phase is difficult due to the limited number of identification keys available, so it has been necessary to create an identification key to nymphs of mealybug species found on dragon fruit. A dichotomous identification key was constructed based on microscopic morphological characteristics of the cuticle. Thus, the four species can be differentiated based on general and unique characteristics of each species in the nymphal and adult stages. The diagnostic features that can be used to separate the four species in the nymphal phase are the presence or absence of: discoidal pores around the eyes; anal lobe bars; tubular ducts on the dorsum; and a denticle on each tarsal claw.
