P.Beauv.) is an invasive species commonly found in tropical and subtropical regions worldwide, consequently threatening global plant biodiversity, and sustainable agriculture. Nevertheless, some of its potential as a medicinal herb, phytoremediation agent, and alternative energy have been reported. genetic variation of cogongrass has been studied in certain areas, however genetic variation in Java, Indonesia has not been reported. Therefore, this study aims to assess the genetic variation of cogongrass in Java, Indonesia using atpB-rbcLand trnL-F intergenic spacer (IGS). Twenty-one plant samples were collected randomly from nine different sites and two pairs of universal primers were employed to amplify the markers. The results showed a much lower genetic difference among subpopulations in comparison to genetic variation within individual subpopulations. This result indicates that the subpopulations have high connectivity, meaning the rapid change in terrestrial ecosystems on the island does not affect the cogongrass population.

Abstract. Syamsuardi, Nurainas, Taufiq A, Harmawan T, Suwardi AB. 2021. Aneuk Jamee traditional foods in the South Aceh District, Indonesia. Biodiversitas 23: 443-454. The traditional foods of the Aneuk Jamee Tribe have a long history and unique traditions passed down for thousands of years. However, a lack of information about indigenous and traditional foods of the Aneuk Jamee Tribe will lead to a decline in local knowledge of traditional foods, particularly among the younger generation. Therefore, this study aimed to investigate the variety of Aneuk Jamee traditional foods in the South Aceh District of Indonesia. The study was conducted in South Aceh, Indonesia, Labuhan Haji Timur, Kota Bahagia, and Kluet Selatan Sub-districts. The study involved 90 respondents (30 from each sub-district) chosen at random. The interview was conducted using a questionnaire covering the respondents' backgrounds, traditional food names, food composition, mode of preparation, and uses. As a result, 52 Aneuk Jamee's traditional foods were identified from three sub-districts of South Aceh District, including 41 types of traditional cuisine and 11 types of traditional snacks. All respondents recognised Gulai ciik minyak, Gulai taleh, Gulai paku, Gulai cabadak, Gulai kabau, Gulai kambieng, Gulai ikan panjang, Gulai kitang, Kue kakareh, Limpieng sagu, and Limpieng ubi as Aneuk Jamee traditional foods. On the other hand, less than 20% of respondents were familiar with other traditional foods, such as Lamang ubi. The Aneuk Jamee Tribe has an excellent system for passing down traditional knowledge from generation to generation. Fifty-six plant species consisting of 47 genera and 28 families were used as Aneuk Jamee traditional food. The Aneuk Jamee Tribe prepares various foods from various plant species to maintain their health and contribute to household income and conservation efforts.

The health status of coral reef ecosystem in Taka Bonerate, Kepulauan Selayar Biosphere Reserve, Indonesia. Biodiversitas 23: 721-732. This study assessed the coral reef conditions in a tropical marine biodiversity hotspot, Tambolongan and Polassi islands, located within the transition zone of UNESCO's Taka Bonerate Kepulauan Selayar Biosphere Reserve in Indonesia. The islands' coral reefs receive a multitude of pressures from anthropogenic activities, risking the livelihood of local communities which rely on resources, such as fish stocks, from this ecosystem. This study measured the coral and reef fish characteristics and current biodiversity status in the coastal waters surrounding the two islands. The evaluation of the coral reefs condition used the Coral Health Index (CHI), which was determined from the two main components of benthic coverage (using underwater photo transect) and reef fish assemblages. The results showed that, overall, coral reef health status in both islands could be classified as very poor (with scores ranging from 1 to 3, out of 10 for CHI), with medium to nearly low diversity for coral (H' index of 1.5 to 2.24), and medium to high diversity for reef fish (H' index of 2.90 to 3.37). This study observed the ecological responses of both habitat and fish within each environment towards the existing damages, which indicated a degree of habitat resiliency. The results obtained contribute to further understanding the extent of pressures towards the quality of marine habitats in the area. They can also be used as baseline data to devise management measures, such as rehabilitation, restoration, or monitoring programs, to ensure that sustainable development can be pursued without neglecting the necessity for conservation.

Diversity report of freshwater gastropods in Buton Island, Indonesia. Biodiversitas 23: 1938-1949. This study was located in Buton Island consisted of 5 administrative districts/cities (Baubau City, Buton District, South Buton District, Muna District and North Buton District) and was conducted for 1 year (August 2019-August 2020). The output is to reveal the potential richness of the biodiversity of freshwater gastropods as a first step to optimize the utilization of freshwater gastropod resources in the Buton Islands. Determination of sampling location for gastropods was carried out by purposive sampling method and the selection of gastropod samples using simple random sampling method at a predetermined location based on the presence of gastropods. Gastropod samples (epifauna/infauna) were collected manually without special fishing gear with gloves, paralone pipes (3.5 inches), and filter (1 mm). The gastropods found in Buton Island consist of 18 genera and 40 species. Freshwater gastropods spread over 8 families, namely Cyclophoridae, Planorbidae, Lymnaeidae and Thiaridae. Among the species, there are several invasive alien species (IAS) such as Tarebia granifera, Melanoides tuberculata and Achatina fulica. These three species always dominate the habitat space where they are found. Most gastropod communities in 5 districts/cities live in fast-flowing rivers with rock, gravel and sand substrates. Only a few of them are found in artificial inland waters such as rice fields, embankments, dams and drainage. This study became a first step to optimize the utilization of gastropod resources and sustainable management, especially related to the conservation of native species from the threat of IAS.

Abstract. Saensouk S, Saensouk P. 2021. Pollen morphology of some species in family Amaranthaceae from Thailand. Biodiversitas 23: 601-611. The aim of this study was to study the pollen morphology of 14 species, eight genera from the family Amaranthaceae in Thailand by the applied acetolysis method and it was taken under light and scanning electron microcopies. All pollen grains had monad, radial symmetry and apolar. The pollens in this study can be divided into several groups based on shape, size, aperture, mesoporia, the character of aperture, and exine sculpturing. The sizes of the pollen can be divided into three groups, i.e., very small pollen group, small pollen group, and medium pollen group. The shapes of the pollen can be divided into three groups, i.e., spheroidal, pentagon, and dodecahedric. Exine sculpturing can be divided into three groups, i.e., granular, macrogranular, and microspine. The pollen in this study can be divided into five groups based on the character of the aperture, i.e., aperture with convex circular-like shape, aperture with deeply collapsed circular-like shape, aperture with collapsed circular-like shape, aperture with collapsed pentagon-like shape, and 6-angular element or hexagon-like shape. The pollen in this study can be divided into three groups based on the mesoporia, i.e., mesoporia broadly flat, mesoporia moderately and broadly vaulted, and mesoporia narrow and highly vaulted. The number of apertures was found to be between 12-54 apertures per pollen. The aperture diameters of Amaranthaceae pollen in this study ranged from 0.5-5 µm. The exine thickness of the pollen in this study was 1.12±0.04 - 2.14±0.31 µm. The pollen character of this study can be divided into eight types, i.e., Achyranthes-type, Alternanthera-type,  Amaranthus-type, Celosia-type, Cyathula-type, Gomphrena-type, Ouret-type, and Psilotrichum-type. The pollen characteristic of Alternanthera pungens Kunth, A. Sessilis (L.) R. BR., Amaranthus blitum subsp. oleraceus (L.) Costea, Cyathula prostrate (L.) Blume., Ouret sanguinolenta (L.) Kuntze, and Psilotrichum ferrugineum (Roxb.) Voigt were reported for the first time.

The use of statistical models in identifying skipjack tuna habitat characteristics during the Southeast Monsoon in the Bone Gulf, Indonesia. Biodiversitas 23: 2231-2237. Skipjack tuna (Katsuwonus pelamis) is the main fishery product in the Bone Gulf, Indonesia, since it has high economic value and is widely accepted by the market. The migration of skipjack tuna can be described by oceanographic dynamics, influenced by seasonal changes. This study aimed to explore the relationship between the oceanographic factors and the distribution and abundance of skipjack tuna obtained by pole and line fishing in the Bone Gulf, Indonesia. The research was conducted during the southeast monsoon season (April-September) from 2015 until 2018. Three oceanographic parameters were used to understand this relationship, i.e., Sea Surface Temperature (SST), Sea Surface Chlorophyll-a (SSC), and water depth. Generalized Additive Models (GAMs) and Generalized Linear Models (GLMs) used to predict the spatial pattern of the skipjack tuna showed that all considered parameters significant influenced the distribution of skipjack tuna in the study area. The fish are mostly distributed from coastal to offshore areas, ranging between 50 to 2000 meters and even wider. The concentration tends to be high within the range of 29.5 to 30.5°C of SST, 0.25 to 0.35 mgm-3 of SSC, and 500 to 1500 meters of water depth. The research also found that the skipjack tuna migration to the Bone Gulf may have followed the current water interruption during the southeast monsoon, for preferred warmer temperatures and lower primary productivity waters in the offshore areas. This factor may represent the optimal habitat of skipjack tuna during the period and can be the basis for determining the skipjack fishing schedule in the Bone Gulf.

Abstract. Isnansetyo A, Handayani DP, Istiqomah I, Arif A, Kaneko T. 2021. An antibacterial compound purified from a tropical coastal plant, Diospyros maritima. Biodiversitas 23: 135-142. This study aimed to isolate, purify, and characterize an antibacterial compound from Diospyros maritima and evaluate its antibacterial activity. Diospyros maritima bark was collected from Kei Kecil Island, extracted with ethanol, separated with hexane and chloroform, and purified by column chromatography. The isolated compound was tested for antibacterial activity by paper disk diffusion on the double-layer agar medium. The minimum inhibitory concentration (MIC) was determined by the microdilution method in a 96-well microplate. The pure compound inhibited Staphylococcus aureus and Aeromonas hydrophila at MICs of 0.625 and 5 µg/mL, respectively. The molecular weight of the compound was 188 m/z based on GC-MS analysis. The 1H-NMR spectrum indicated two protons which appeared as a multiplet at 7,687.59 ppm, one proton appeared as a singlet at 6.87 ppm and one proton appeared as a doublet at 7.26 ppm. The 13C-NMR spectrum showed eleven carbon signals, eight signals for the aromatic ring (186.06, 162.38, 151.17, 137.41, 134.49, 133.76, 17.75, and 16.51 ppm) and two signals at 191.91 and 191.05 ppm. The spectrometric data showed that the isolated compound was plumbagin (1,4-naphthalenedione-5-hydroxy-2-methyl). This finding opens up a new perspective for D. maritima as a source of lead compounds for the development of antibacterial substances.

Biodiversitas 23: 1852-1856. Nipa (Nypa fruticans) is one of the most common, widely distributed, and useful trees in the mangrove forest of Southeast Asia. It is being utilized in the production of roof thatching, wine and vinegar, and recent scientific studies have been done for the utilization of nipa sap in the production of bio-ethanol. Moreover, nipa has also contributed ecologically as it acts as a barrier during typhoons and storm surges helps prevent soil erosion along the banks of villages near river areas. The study area is located in barangay Bisagu, Aparri, Cagayan, Philippines where nipa plantation covers 550.51 ha and is utilized by the community. As we all know, mangroves areas as home to aquatic animals thus the study conducted takes on the diversity of macrofaunal species in a nipa-dominated mangrove area. Sampling areas were constructed, the proximal, middle and distal quadrat with 10 m by 10 m dimensions. Species collected in the area yield 11, where 10 is successfully identified and 1 species is not identified. Results showed that macrofaunal species in a nipa-dominated mangrove area show very low diversity. Shannon-wiener diversity index of the community showed a result of only 0.97457 and its equitability at 0.04, which concluded as low species evenness distribution.

Antimicrobial activity of actinomycetes isolated from mangrove soil in Tuban, Indonesia. Biodiversitas 23: 2957-2965. This study aims to isolate, identify, and investigate the antimicrobial activity of actinomycetes isolated from mangrove soil of the Mangrove Center Tuban in Jenu Village, Tuban District, Indonesia. Soil sampling was carried out with a purposive sampling technique. The dry-heating and pour plate methods were used to isolate actinomycetes. Four isolates (LIB.2; LIA.4; LIA.9; and LIA.10) were selected and tested for their antimicrobial activities against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, and Candida albicans ATCC 10231 using disc diffusion method. Colony morphology characterization and Gram staining were then conducted on the 4 isolates. The identification of LIB.2 was performed by biochemical assay and 16S rRNA gene sequence analysis. The results showed that crude antimicrobial extracts (CACs) of all isolates inhibited the growth of E. coli but not S. aureus and C. albicans. Isolate LIB.2 showed the highest inhibition zone with a diameter of 15,65 ± 2,33 mm, followed by LIA.9 (10,19 ± 2,78 mm), LIA.4 (9,80 ± 0,47), and LIA.10 (8,75 ± 0,42 mm). The most potential isolate was identified as a member of Streptomyces genera. Based on biochemical assay and 16S rRNA gene sequence analysis, LIB.2 was identified as Streptomyces vellosus strain LIB.2.

Identification and characterization of strawberry FvGELP1 (Fragaria vesca GDSL esterase/lipase).Biodiversitas 23: 907-915. Fruit softening is primarily the result of the hydrolytic enzymes activity. Esterases are reasonable candidates due to their putative role in cell wall components degradation. This study aimed to identify and characterize a new enzyme related to strawberry fruit ripening. Esterase activity was detected in the native PAGE of protein extract isolated from Fragaria ×ananassared fruit. Amino acid sequence analysis of the protein band revealed several esterases as possible candidates. GDSL esterase/lipase from Fragaria vesca(gene27964; FvGELP1) was chosen for cloning purposes and further analysis. Quantitative RT-PCR of FvGELP1in plant tissues of F. vescaindicated high expression levels in fruit, in particular in early developmental stages. The gene FvGELP1(1,161 bp) was amplified from F. vescafruit cDNA and expressed as a 43 kDa HisTag fusion protein in Saccharomyces cerevisiae.FvGELP1 possesses four conserved residues Gly43-Asp44-Ser45-Asn46. FvGELP1 contains Ser45, Asp364, and His367 as the catalytic triad. Esterase assays of FvGELP1 resulted in high levels of the aromatic alcohol products by applying ?-naphthyl acetate (?NA), p-nitrophenyl acetate (pNPA), phenyl acetate, and benzyl acetate as substrates. It could be suspected that FvGELP1 plays a role in strawberry fruit ripening and might be involved in the hemicellulose degradation, presumably by deacetylation of the polysaccharide.
