Abstract

Despite the advancements in cancer therapy, there is still a need for new anticancer agents. Due to the tremendous range of chemical compounds that endophytic fungi create, they have become a crucial component of the current drug development process. The aim of this study was to evaluate the cytotoxic potential of endophytic fungi associated with Catharanthus roseus (L.) G.Don collected from Yogyakarta, Indonesia. All fungal EtOAc extracts were tested against three human cancer cell lines, HeLa, T47D, MCF-7, and Vero cell lines. The potential fungal isolate was identified using molecular techniques. The results showed that a total of seven endophytic fungi were obtained from the different parts of C. roseus. Only one fungus (SCR 3) showed the best cytotoxic activitiy towards all three cell lines used, especially the MCF-7 cell line (IC50 130.90 ?g/mL, Selectivity Index=11.42). The findings of using LC-HRMS also revealed several exciting compounds such as genistein and D-(-)-Mannitol. The results showed that EtOAc extract of SCR 3 (Aspergillus sp.) induced apoptosis but did not affect the cell cycle. This study suggests that EtOAc extract of SCR 3 may be a potential source of secondary metabolites that can function as a new source of anticancer agents.

Abstract

Identification of lactic acid bacteria isolated from developed dried coffee starter culture used as a fermentation agent to produce Robusta civet coffee. Biodiversitas 24: 3715-3722. Native civet coffee is a popular coffee bean, produced by the processes which undergo spontaneous fermentation in the civet digestive system. Since the native civet coffee has a higher selling value and its production depends on the uncontrolled civet life activities, the effort to produce a similar quality of this coffee product has been made by designing a dried starter culture isolated from civet caecum, and utilizing coffee cherry peels as the potential inducer and rice flour as the matrix for coating and immobilizing the isolates. The proportion of rice and coffee cherry peel flour mixtures was formulated to fabricate the dried starter culture by determining the viability and fermentability of lactic acid bacteria (LAB) survive in the starter culture. Furthermore, the starter culture was analyzed for the number of microflora and identification of LAB, water content, glucose fermentability, total titrated acid production rate, and pH value. The most viable starter cultures were obtained from the formula of 80% rice flour and 20% coffee cherry peel flour exhibiting the number of microflora of 5.6 log MPN/g and fermentation capacity of 245.73 mg glucose/gh. The total titrated acid production rate was 91.07 mg lactic acid/gh, and a pH value of 5.33. The LABs survived in the starter culture were identified as Lacticaseibacillus paracasei. The quality of hot brewed roasted Robusta coffee prepared by fermentation using this dried starter culture was higher than that of unfermented one, as observed from their organoleptic quality. These findings indicate that the LAB isolated from civet caecum is potential to be developed as a dried starter culture in coffee cherry fermentation processes.

Ethnobotanical knowledge of Dayak Ngaju in utilizing plant species to locate gold mining in Sei Riang Village, Central Kalimantan, Indonesia. Biodiversitas 24: 4145-4150. Indonesia is one of the richest countries in flora and cultural diversity. The strong relationship between the culture and plant diversity has developed ethnobotanical knowledge of the indigenous community, including the knowledge of the Dayak Ngaju Tribe in utilizing plant species as gold indicators. Therefore, this study aimed to inventory and identify the plant species used as gold indicators by Dayak Ngaju in Sei Riang Village, Gunung Mas District, Central Kalimantan, Indonesia using ethnobotanical study and then empirically assess the gold content in the identified plants using laboratory analysis. Ethnobotanical information was collected through in-depth interviews with 25 informants and analyzed with the Informant Agreement Ratio (IAR). Plants mentioned as gold indicators were then identified in the field. The samples of the plants were then examined by atomic absorption spectroscopy to determine gold (Aurum; Au) contents in their root, stem, and leave tissues. The data was analyzed using SPSS 25 with the Parametric Pearson Correlation test to determine the correlation and significance level of Au contents found in the plant tissues and soil. Based on the interview results, the gold miners believed that several plant species could be utilized to indicate gold’s presence in the mining area, as indicated by the value of IAR of 0.540, suggesting that 54% of the informants agreed to use the plants as the gold indicator. Two plant species were identified as gold indicators: Kasuhui (Dipterocarpus sp.) and Hara (Ficus racemosa L.). Significant positive correlations were also exhibited between Au contents in the Kasuhui stem and the Hara root and stem and the soils where they grow. This research is expected to promote the ethnobotanical knowledge of the gold miners from the Dayak Ngaju Tribe in selecting scientifically justifiable gold mining areas.

Abstract. Zubaidah S, Mujtahida I, Kuswantoro H. 2023. Response of morphological, anatomical and agronomic characteristics of soybean genotypes to Cowpea Mild Mottle Virus. Biodoversitas 24: 4017-4026. Cowpea Mild Mottle Virus (CpMMV) is a significant disease in soybeans. This disease can cause plant malformations and reduce seed yields. The study used a randomized block design with three replications. Plant materials were five resistant genotypes (MLGG 0006, MLGG 0106, MLGG 0297, MLGG 0315, and MLGG 0599) and five susceptible genotypes (MLGG 0123, MLGG 0379, MLGG 0603, MLGG 0695, and MLGG 0796). The results showed a significant difference between resistant and susceptible genotypes to CpMMV attack. Resistant soybean genotypes had relatively tall stems, many branches, and larger leaf widths. The number of stomata and trichomes of the resistant genotype was less than that of the susceptible genotype. The number of filled pods, the total number of pods per plant, and the number of seeds of the resistant genotype had more numbers than the susceptible genotype. MLGG 0695 was the most responsive genotype because it had a relatively low stem height, few branches, small leaf size, many stomata and trichomes on the leaf surface, the thickest leaf size, and a small number of pods and seeds. The most resistant soybean genotype was MLGG 0297, having a relatively high stem, many branches, large leaf size, few stomata and trichomes on the leaf surface, thin leaf size, and many more pods and seeds. MLGG 0297 can be used as a gene source to develop a superior variety resistant to CpMMV.

Abstract. Rinaldy H, Iskandar J, Cahyandito MF, Sunardi. 2023. The potential feasibility of mangrove crab (Scylla serrata) silvofishery aquaculture. Biodiversitas 24: 3344-3354. The degradation of mangrove forests in Indonesia has reached more than 50%, and increases continuously. Sei Beduk is one of the Sub-districts in Batam City, Indonesia, that has mangrove forest damage where the density of trees per hectare is below the normal value of <1,000 trees per hectare. One approach that can be applied to overcome forest degradation is fisheries management with an ecosystem approach. This study aims to determine the suitable location for cultivating mangrove crab (Scylla serrata Forskål, 1775) and analyze the potential feasibility of S. serrata silvofishery cultivation using a quantitative approach with the Ecosystem Approach to Fisheries Management (EAFM) method. According to Slovin's calculations, a minimum of 100 people samples were obtained, and 147 samples were obtained at the time of data collection. The results showed that the location of the Perahu was highly potential for silvofishery S. serrata cultivation. The overall potential feasibility of silvofishery S. serrata cultivation in Sei Beduk, Batam was 53.10% or moderate/relatively good. The potential feasibility of S. serrata cultivation management planning is sufficient to support the sustainability of mangrove forests and allow it to be implemented. The feasible plan for managing S. serrata cultivation is to preserve ecology while improving and considering aquaculture policies and regulations. The relatively appropriate mangrove silvofishery cultivation in Sei Beduk, Batam, can provide community solutions to carry out silvofishery cultivation as a measure to overcome the problem of mangrove forest degradation, particularly in Sei Beduk, Batam.

Abstract. Aditya IGMRA, Purwanto H. 2023. Molecular detection of the pathogen of Apis mellifera (Hymenoptera: Apidae) in honey in Indonesia. Biodiversitas 24: 2612-2622. Apis honey bees play a major role in global crop pollination. The honey bee population faces various threats, one being entomopathogens, responsible for 13% of honey bees' declining population cases. Some major pathogens are Nosema sp., Melissococcus plutonius, and Paenibacillus larvae. Data on the prevalence of these pathogens is not available in Indonesia. Therefore, this study aimed to detect entomopathogen in Java, Bali, and Sumatra honey. We extracted 30 honey samples from Java, Bali, and Sumatra in this study. DNA from honey amplified by conventional Polymerase Chain Reaction (PCR) to detect Nosema apis, Nosema ceranae, M. plutonius, and P. larvae. Amplicons were sequenced to confirm the species of pathogens detected. Results showed that N. ceranae has a 20% prevalence, found in Java and Bali; M. plutonius was, for the first time, found in Indonesia and only in Java, with a 3.33% prevalence. In comparison, this study didn't find N. apis and P. larvae. Environmental factors were studied for their correlation with the incidence of pathogens. From several factors, only precipitation significantly correlates with the occurrence of N. ceranae and M. plutonius, while other factors do not. This research concludes that honey can be used as source of DNA for pathogens detection and monitoring in a certain area globally.

Assessing the impact on growth and yield in different varieties of chili pepper (Capsicum frutescens) intercropped with chaya (Cnidoscolus aconitifolius). Biodiversitas 24: 2639-2646. Intercropping chaya and chili plants is advantageous as it can boost land productivity and increase farmer income. However, the growth of the chili plant is more likely to be hampered by the shade formed by the canopy of the chaya plant. Therefore, this study aimed to investigate the response of chili to the shade of the chaya plant canopy and identify varieties that can thrive in intercropping systems. The experiment was arranged based on a split-plot design with a planting pattern as the main plot, consisting of chili monoculture, intercropping of chaya-chili, and intercropping of pruned-canopy chaya with chili. Three chili varieties, i.e., Rajo, Taruna, and Bara, were the subplots with four replications. Chaya plants were planted at a distance of 200 x 200 cm. Chaya-chili intercropping was done by inserting two rows of chili plants with a distance of 50 x 50 cm between rows of chaya plants. The results showed that chili pepper planted in an intercropping system exhibit an etiolation effect as indicated by a taller stem but with fewer leaves than the chili plants cultivated with a monoculture system. The shading effect of the chaya plant resulted in lower leaf temperature of the chili peppers in intercropping system. Pruning treatment on the shoots of chaya plants positively impacted the growth and yield of chili plants, which was indicated by an increase in biomass weight and fruit yield. The Bara variety was harvested two weeks earlier and produced higher fruit yields compared to Rajo and Taruna varieties. The growth and yield of chili plants were relatively comparable between the monoculture and intercropping system if the canopy pruning in chaya plants was carried out, especially in the Bara variety.

Abstract. Asrul. 2023. Potential of local Bacillus spp. isolates as wilt disease biocontrol agents for Fusarium oxysporum f. sp. cepae on Allium x wakegi. Biodiversitas 24: 4989-4997. Implementation of biological agents for eradication and inhibition of plant pathogens is often ineffective due to its protracted consequences. The objective of this research was to examine the potential of local isolates of Bacillus spp. as a biocontrol agent in suppressing Fusarium wilt disease (Fusarium oxysporum f. sp. cepae) on wakegi onions (Allium × wakegi Araki). The research was conducted in a completely randomized design (CRD) with the treatment of rhizospheric bacterial isolates. The treatments included control (without isolate application) and seven bacterial isolates, namely KP17, KP5, DB9, DB12, DB18, DG4 and DG11. Each treatment was repeated five times and each replication consisted of 10 wakegi onion plants. The results showed that a total of 46 isolates were isolated from the rhizosphere of wakegi onion and seven isolates among them were selected as biocontrol agents. These isolates had similarities with Bacillus spp. in accordance with the colony morphology, physiology and biochemical characteristics. Among the isolates found, DB12 emerged as the isolate that had the potential to be developed as a biocontrol agent.

Sampling was conducted to record clump growth and habitat properties of 38 sites grouped based on the clump density using PAST 4.11 and ArcGIS 10.8.2 software. Antibacterial activity of lactic acid bacteria from various freshwater fish species against pathogenic bacteria in caged red tilapia (Oreochromis sp.). Biodiversitas 24: 3373-3383. Antibiotic-resistant bacteria are a big challenge for many aquaculture countries around the world. The search for new solutions, such as probiotics to replace antibiotics in aquaculture, attracts the interest of many scientists. The investigation was accomplished by isolating Lactic Acid Bacteria (LAB) inhibiting Streptococcus agalactiae and Aeromonas hydrophila, which cause swollen eyes and hemorrhagic skin disease in caged red tilapia (Oreochromis sp.) in Vinh Long province. The findings isolated 37 isolates of LAB from the intestines of catfish, red tilapia, and tilapia. Using the diffusion well method, the results showed that 13/37 (35.14%) and 18/37 (48.65%) isolates were inhibitory against S. agalactiae and A. hydrophila, respectively. The research found that eight isolates were inhibitory to two species of S. agalactiae and A. hydrophila, especially isolate CT6.2, which has the strongest inhibition (10.5 mm inhibition zone diameter). Our finding revealed that the isolates CT6.2 and RP6 demonstrate the highest antagonistic activity against S. agalactiae and A. hydrophila at a density of 108 CFU/mL after incubation at 35°C, pH = 5-6 for 48-60 h. Using PCR techniques and 16S rRNA gene sequencing combined with morphological and biochemical characteristics, the LAB isolates in the findings belong to the genera Lactobacillus and Pediococcus, being 99.35% and 99.02% similar to Lactobacillus plantarum and Pediococcus pentosaceus, respectively. This is the first report on the antimicrobial activity of LAB against S. agalactiae and A. hydrophila in caged red tilapia in the Mekong Delta.
