Mushrooms play an important role in ecosystems and human life. They can be found in various places including in oil palm plantations. The aim of this study was to survey and identify the population of macroscopic mushrooms or macrofungi in small-holder oil palm plantations on tropical peatlands in Kampung Baru Village, Kubu Raya District, West Kalimantan, Indonesia using the field survey method. In the research area, a dam has been constructed to regulate the water level at 30 cm, 40 cm, 50 cm, and 60 cm from the surface. Sampling was carried out in these four conditions. The study revealed the presence of 34 species of macrofungi belonging to the Basidiomycota. A groundwater level ranging from 40-60 cm from the surface was identified as favorable for macrofungal growth. Meanwhile, a groundwater level of 30 cm from the surface was found to be unsuitable for macrofungal growth in the peatlands. The level of macrofungal species diversity in small-holder oil palm plantations on the peatlands was determined to be moderate.

Barcoding of ornamental freshwater shrimp, Neocaridina denticulata (De Haan, 1844) from the aquatic ornamental market in Purbalingga, Central Java, Indonesia. Biodiversitas 24: 3601-3608. Indonesia is one of the world’s leading aquatic ornamental shrimps producers and exports. One of the aquatic ornament shrimp species is a Neocaridina denticulate (De Haan, 1844). There is a wide range of colors including red, bright red, yellow, orange, green, blue, violet, black, and color combinations. It is a landlocked Atydiae family species comprising31 species and subspecies. Due to taxonomic difficulties, this species is continually under revision, and the validation of a few species is currently questionable and uncertain for the relationships among the various colors of N. denticulata. The Cytochrome Oxidase gene I (CO1) gene sequences of mitochondrial DNA were used for species DNA barcoding studies. This study aimed to validate the taxonomic status of different colors of N. denticulata species using the mitochondrial DNA or mtDNA Cytochrome oxidase (CO1) gene sequences. Neighbour Joining trees were constructed based on 545 bp of CO1 gene from white, black, red, yellow, blue colors, and their combination. Several sequences were derived from GenBank for relevant species from different countries and the outgroup. The result showed that all samples are homolog, even to N. denticulata from China NC_023823.1.

Abstract. Tenriware, Nur M, Nasyrah AFA. 2023. Biological reproduction of mackerel scad, Decapterus macarellus (Cuvier, 1833) caught by purse-seine net in Majene Waters, West Sulawesi, Indonesia. Biodiversitas 24: 3012-3018. Mackerel scad, Decapterus macarellus Cuvier, 1833 is an economically important fish resource as an income source for fishermen in Majene waters, West Sulawesi. This study aims to determine some aspects of reproductive biology, including sex ratio, gonadal somatic index (GSI), gonad maturity stage (GMS), and the size at the first maturity. The sampling was carried out at the Fish Landing Sites in the Coastal Waters of Pangali-Ali Village, East Banggae District, Majene District, West Sulawesi Province, Indonesia. Each specimen was measured to the total length with digital calipers and body weight using a digital weighing scale. Initial sex assessment was noticed at the time of dissection, and gonadal maturity was also assessed. A total of 275 specimens consisting of 177 males and 98 females were obtained from April to July 2017. The result indicates that the sex ratio between males and females deviated from an expected 1:1: and was not balanced. This species spawns in April at the size of 173.4 mm and 168.6-178.3 mm for males and 189.1 mm or the range of 182.6-195.8 mm for females.

Short Communication: The isolation of Klebsiella variicola’s cellulase from Macrotermes gilvus gut in Indralaya Peatlands, Indonesia. Biodiversitas 24: 4218-4222. Termites are organisms that can inflict damage on agricultural, forest, and furniture materials. Furthermore, they can positively affect the ecosystem due to their ability to break down lignocellulose-containing materials. According to previous findings, the cellulolytic bacteria from the termite Macrotermes gilvus gut obtained in Indralaya Peatlands showed activity and hydrolyzed cellulose in a CMC agar medium. In this study, the cellulase enzymes of Klebsiella variicola isolated from Macrotermes gilvus gut showed higher cellulolytic indexes after staining with Congo red. Furthermore, the enzyme was purified and characterized using ammonium sulfate precipitation and dialysis. SDS PAGE and zymogram analysis reported a molecular mass of 20 kDa, with optimal activity at pH of 6 and temperature of 50°C.

Diversity and ecology of molluscs (Gastropods) in mountain streams, Nurota mountain range, Uzbekistan. Biodiversitas 24: 2402-2408. Characterization of the faunal biodiversity of the regions is one of the important tasks in today's era when the influence of the anthropogenic factor has increased, and the natural ecosystems are undergoing drastic changes. Therefore, the faunal composition analysis of molluscs as a part of the natural food chain, the development of measures related to their protection, the disclosure of bioecological features, and the substantiation of scientific solutions are of great scientific and practical importance. This study provides information on the species composition of gastropod molluscs in the streams of the Nurota Mountains of Uzbekistan. Sampling was carried out at 11 stations from April to November 2019. Molluscs include 4 families (Belgrandiellidae, Lymnaeidae, Physidae and Planorbidae), 5 genera, and 15 species. The dominant family in the malacofauna of the study area is Lymnaeidae, one genus and eight species belonging to this family are found in the area. Most molluscs are found on rocky, sandy substrates around plants. This article describes the biological diversity of animals in streams in the Nurota Mountains of Uzbekistan.

Floristic richness and diversity of Bintuni mangrove, Bird's Head Peninsula, West Papua, Indonesia. Biodiversitas 24: 2887-2897. Mangrove ecosystems play an important role in the functions of coastal protection, fish and other living-organism habitats, carbon storage, and the livelihood of local people. Yet, mangrove forests generally face ecological threats like anthropogenic pressures. However, fewer studies were carried out in this area, so this research aimed to reveal the vegetation diversity and richness in Bintuni, not only the true mangroves but also the mangroves associates with three different conditions, namely sandy areas called protection forests, disturbed forests called production forests, and delta called nature reserve. The results showed that species vegetation in the Bintuni Mangrove could be differentiated as true mangroves (29.82%) and mangrove associates (70.18%) of the total species richness of mangrove vegetation where in terms of taxonomic composition, the three forests encompassed 25 families, 47 genera, and 57 species clustered into 9 lifeforms. Then, the vegetation diversity and richness were highest in the nature reserve, and 34 (59.65% of total species number) species of vegetation in common share in three forest types. Consequently, frequent checking of vegetation richness is important since the Bintuni mangrove does not seem to have a significant threat like massive conversion of mangrove.

Biodiversitas 24: 3481-3487. Etlingera elatior (Jack) R.M.Sm. has been well-documented to have antioxidant and antidiabetic properties. Among all medical conditions worldwide, diabetes mellitus is one of the leading causes of death. Therefore, research on the discovery of antidiabetic medicines inhibitory to ?-glucosidase (an enzyme for degrading complex dietary carbohydrates into sugar in the digestive process) is urgently needed. This study aims to isolate and evaluate the potential of the fermentation culture of endophytic fungi associated with E. elatior to inhibit the activity of ?-glucosidase. The endophytic fungi were isolated from various parts of E. elatior. A total of 29 isolates were successfully isolated from the plant's roots, stems, leaves, and flowers. The endophytic fungi were tested for their ability to inhibit ?-glucosidase activity in bioassays. Among those isolates, 8 were inhibitory to such an enzyme, and 2 showed the highest inhibitory activities. Morphotype and molecular identification of those isolates, using their Internal Transcribed Spacer (ITS) regions of ribosomal DNA (rDNA), identified them as Daldinia eschscholtzii isolate CFL 7 and Hypoxylon trugodes voucher YMJ 57, with IC50 values of 738 µg/mL and 825 µg/mL, respectively. That indicates that they are potentially used in preventing or treating diabetic mellitus.

Abstract. Suriani, Patandjengi B, Muis A, Junaid M, Mirsam H, Azrai M, Efendi R, Sebayang A. 2023. New corn resistant lines to stalk rot disease (Dickeya zeae) in Indonesia. Biodiveritas 24: 3190-3200. Stalk rot disease caused by Dickeya zeae is one of the important diseases of corn in Indonesia. Host resistance cultivars are an effective and sustainable control measure of the disease. Therefore, the present study aimed to evaluate the resistance of 15 S1 hybrid maize lines to stalk rot disease. The research was conducted in two seasons (DS and WS) using a randomized block design with 3 replications. The D. zeae suspension with 108cfu/mL concentration was inoculated into the plant test 45 Days After Planting (DAP). Disease incidence and severity were observed during the two seasons. The results showed that all tested lines were infected with stalk rot disease but had various resistance reactions. Disease incidence and severity in the dry season were higher than in the rainy season. In the rainy season all test lines followed the 3 models of disease development, but in the dry season, all lines followed the monomolecular model. Further analysis showed that 3 lines of hybrid maize had the lowest AUDPC value with a protection index of more than 50% in two growing seasons. Stalk lignin content had negative correlation with a disease incidence of -0.60877, so it can be used as a parameter of plant resistance to disease. Tested lines that show resistance to the disease could potentially be useful as new varieties of maize.

Abstract. Sintia M, Ardie SW, Suwarno WB. 2023. Genetic variability of F2 foxtail millet population derived from ICERI-5 and Botok-10 cross. Biodiversitas 24: 3559-3567. Foxtail millet (Setaria italica (L.) P. Beauv.) is a carbohydrate source with good tolerance to abiotic stress. High productivity, short stature, and early flowering time are the breeding targets of this species. This study aimed to obtain genetic variability of F2 foxtail millet population derived from ICERI-5 and Botok-10 crossand to predict selection response based onweighted selection index. This experiment was conducted in Bogor and consisted of 522 F2 individuals and two parental genotypes. The results showed that the F2 population had shorter plant height and earlier flowering time compared to Botok-10, and higher grainweight per plant compared to ICERI-5. Plant height, flowering time, and grainweight per plant showed moderate to high genotypic coefficient of variation with moderate to high broad-sense heritability. Weighted selection index using the three target traits resulted in ten F2 individuals with higher selection index compared to both parents, with individual number I5B10-4-96 having the highest selection index. Selection responses based on a weighted selection index indicate an expected decrease in flowering time and an increase in grain weight per plant in the next generation.

Abstract. Wahidah, Omar SBA, Trijuno DD, Nugroho E, Amrullah, Khatimah K. 2023. Genetic variation of giant freshwater prawns Macrobrachium rosenbergii wild population of South Sulawesi, Indonesia. Biodiversitas 24: 3081-3090. Quality giant freshwater prawn broodstock can be provided using broodstock sources from wild populations with good morphological and genetic performance. This study aimed to analyze the genotypes of the wild population of giant freshwater prawns in South Sulawesi, hence it can be used as basic information for quality broodstock. Evaluation of giant prawn haplotypes, Macrobrachium rosenbergii, from 3 rivers in South Sulawesi: 26 samples from the Waelawi River (11 males, 15 females); 25 samples from the Kalibone River (10 males, 15 females); and 26 samples from the Kariango River (12 males, 14 females), so there are 6 population groups. Analysis used mt-DNA and obtained an amplification of 700 bp. The amplified restriction uses four enzymes (Hae III, Mbo I, Rsa I, and Taq I). The identified male and female Kariango populations each had 3 haplotypes: female Kalibone had 2 haplotypes; male and female Waelawi had 1 haplotype; and male Kalibone had 1 haplotype. Female Kariango had the highest haplotype diversity (0.3651), followed by male Kariango (0.1449) and female Kalibone (0.1011). Kariango females have the farthest genetic distance from Kalibone males, Waelawi males, and Waelawi females. The male Waelawi, female Waelawi, and male Kalibone are in one cluster and form a new cluster with the male Kariango, then with the female Kalibone, and the last cluster is formed with the female Kariango. The Kariango female population can be used as potential broodstock, which can be mated with Kalibone males, Waelawi males, or other populations.
