Abstract

Solang M. 2017. Blood cockle (Anadara granosa) supplementation to increase serum calcium level and femur growth of lowprotein diet rat. Nusantara Bioscience 9: 62-67. Blood cockle (Anadara granosa) contains protein, zinc and calcium. The aim of this study was to evaluate the effects of blood cockle flour supplement on serum calcium level, rat femur length and weight of rat femur on low-protein diet. The design of research used The Separate Sample Pre-Post Test Design. Forty eight Wistar male rats were randomly grouped into 2 groups, 12 rats as normal control group and 36 rats as malnourished group. After eight weeks, 4 rats from each group were sacrificed. Furthermore, 8 rats as normal control and 32 rats as malnourished group were randomly grouped. Malnourished rats were treated with blood cockle flour of 12.5%/kg diet, 25%/kg diet, and 50%/kg diet ad libitum for 8 weeks. Data analysis was performed with One Way ANOVA, LSD, Kruskal-Wallis and Mann-Whitney tests. The results of study showed that the supplementation of blood cockle flour of 12.5%/kg diet was significantly increase the serum calcium level (p=0.003), rat femur length (p=0.000) and rat femur weight (p=0.002) on low-protein diet. This study demonstrated that blood cockle flour can be used as alternative food to improve the growth of rat femurs in a malnutrition condition.

Eriani K, Sari N, Ihdina M, Rosnizar. 2017. The effect of equilibration time on semen freezing of local swamp buffalo (Bubalus bubalis) with combination extender of lactose and glycerol. Nusantara Bioscience 9: 77-82. This study was to determine the different time of equilibration on semen of swamp buffalo (Bubalus bubalis) which was diluted by adding Tris-Egg Yolk extender of lactose and glycerol cryoprotectant combination. Fresh sperm of the swamp buffalo (B. bubalis) was diluted by using Tris-Egg Yolk extender of lactose cryoprotectants combination 0 mM (L0), 60 mM (L60), 120 mM (L120) and glycerol 3% (G3), 5% (G5), 7% (G7) with the equilibration of 2,5, 3, and 4 hours. The parameter used in this study was the percentage of sperm motility and live treatment. The results of the 2.5-hour equilibration with a combination of cryoprotectants L60G7 showed the best quality in all parameters. The results of the 3-hour equilibration with combination of cryoprotectants L60G7 showed the best quality in all parameters whereas the best quality in all parameters in 4-hour equilibration was the combination of cryoprotectants L120G7.

Nusantara Bioscience 9: 68-76. Micronutrient bioavailability from cereals is poor to monogastric animals due to presence of phytate as a chelating agent in seeds and an exogenous application of phytases is required to overcome antinutritional effect of phytate. In the present investigation, 40 fungal isolates including Aspergillus, Penicillium, Trichoderma, Toliposporium, Cladosporium, Talaromyces etc. from different habitats were screened positive for phytase activity on specific media. On the basis of phytase production potential in solid state fermentation conditions, plant epiphytic fungi Penicillium oxalicum strain EUFR-3 has been observed with maximum phytase activity of 12.8 U/g. The biochemical properties i.e. activity and stability under optimum pH and temperature, and effect of modulators on phytase activity from this isolate were studied for its prospective application in human food. The P. oxalicum EUFR-3 phytase (PhyP-EUFR3) had maximum activity at pH 7.0 and temperature 40 °C. It was stable in pH range of 3-8 with more than 60 % activity throughout investigation. The thermostability of PhyP-EUFR3 was about 30 % residual activities after 10 min at 80 °C. Further purification, characterization and study of catalytic mechanism will be useful in better utilization of this phytase in human food for increased bioavailability of important metal ions.

Hasan Z, Dhahiyat Y, Andriani Y, Zidni I. 2016. Water quality improvement of Nile tilapia and catfish polyculture in aquaponics system. Nusantara Bioscience 8: 83-85. Research on the improvement of polyculture water quality in aquaponics system was conducted from July 2014 to August 2014. This study aims to improve the water quality in Nile tilapia and catfish polyculture by applying the aquaponics system. The results show that several water quality parameters increase, such as dissolved oxygen, ammonia, and nitrate. Meanwhile, Orthophosphate tends to be similar both in aquaponics system and control. The study is conducted experimentally by using completely randomized design. The cultured fish are catfish and Nile tilapia while the aquatic plants are water spinach and lettuce. At the end of the study the concentration of dissolved oxygen in the aquaponics system ranges from 5.3 to 7.6 mg/L, while in the media control ranges from 4.2 to 4.3 mg/L. Average concentrations of ammonia, nitrate and orthophosphate in the aquaponics system range from 0.003 to 0.25 mg/L, 10.0 to 50.7 mg/L and 3.0 mg/L-5.0 mg/L respectively. Meanwhile, in media control, concentration of those three parameters are 0.003 to 0.35 mg/L, 10.0 to 60.0 mg/L and 3.0 to 5.0 mg/L respectively. Based on this study, it is concluded that the polyculture water quality can be improved through the application of the aquaponics system.

Effect of katuk (Sauropus androgynus) powder supplementation on the levels of progesterone (P4) and estradiol-17β (E2) hormones in kacang goat (Capra aegragus). Nusantara Bioscience 9: 86-91. Kacang goat (Capra aegragus) is one of Indonesian endemic ruminant species which commonly retained by the community in Indonesia. This species is classified as a small ruminant which has many benefits i.e easy to be domesticated and to adapt, and has an economical value especially for saving purposes to farmers in marginal areas. This study aimed to observe the effects of the katuk powder supplementation (Sauropus androgynus) in the feed of kacang goats on the level of reproductive hormones (progesterone (P4) and estradiol-17β (E2)) in female kacang goats. A total of 9 female kacang goats which meet the prerequisite samples i.e reaching the age of a sexual maturity, having a homogenous weight (12.00 ± 1.00 kg) and in healthy condition were then devided into three groups of treatments using a Completely Randomized Design method. Each treatments consisted of 3 kacang goats as a replication. The treatments consisted of: (i) feed without katuk powder supplementation (control),(ii) feed with the concentration of katuk powder supplementation was 3% of kacang goat’s body weight, and (iii) feed with the concentration of katuk powder supplementation was 6% of kacang goat’s body weight. After 60 days of treatments, the results showed that the supplementation of katuk powder on female kacang goats had no significant effect on the level of progesterone and estradiol-17β in the first sampling (day-0, P>0.05), however the treatment had a significant effect to the level of progesterone and estradiol-17β on the second sampling (day-28) and the third sampling (day-56) with a significance level of P<0.05. It was concluded that the supplementation of katuk powder could increase the level of progesteron and estradiol-17β hormones in kacang goats. The katuk powder supplementation about 3% of goat weight has sufficiently increased the level of reproduction hormones in female kacang goats.

Abstract. Indrawati I, Miranti M, Mayfi IR. 2017. Antibacterial activity of rhizome extracts of three ginger varieties against acne-isolated bacteria. Nusantara Bioscience 9: 92-96. Antibacterial activity of rhizome extracts of three ginger (Zingiber officinale Rosc.) varieties against bacteria isolated from acne was done. Three ginger varieties, namely emprit, gajah and red ginger, were tested to obtain the best concentration of ethanolic extracts among them in inhibiting the growth of the acne-isolated bacteria. This research used experimental approach by Completely Randomized Factorial Design with two factors of treatment, A x B. The first factor (A) were bacteria strain isolated from acne that consists of 3 levels. The second factor (B) was a varied concentration of rhizome ethanolic extract of three ginger varieties which consist of 7.5, 10, 12.5, 15, 17.5 and 20% for emprit and gajah, while for the concentration used for red were 2, 2.5, 5, 7.5, 10 and 12.5%. Moreover, a synthetic antibiotic clindamycin 30 mg mL-1 was used as a comparison. The obtained data were statistically analyzed using ANOVA with 95% of confidence and then followed by Tukey test when the significance was detected. The results showed that the acne-isolated bacteria were identified as Propionibacterium sp 1 and 2 and Staphylococcus. The rhizome ethanolic extract of red ginger had the highest antibacterial activity against the tested bacteria. This finding was supported by the fact that the red ginger variety had a small value of Minimum Inhibitory Concentration (MIC). Additionally, the measurement of clear zone revealed that the interaction between ethanolic extract of red ginger with a concentration of 12.5% ??producing a clear zone of 13.5, 25.0 and 17.5 mm against Propionibacterium sp.1, Staphylococcus sp and Propionibacterium sp.2, respectively.

Overproduction, characterization and antiproliferative activity determination of non-tagged recombinant human interferon alpha-2a produced in Pichia pastoris. Nusantara Bioscience 9: 97-101. Recombinant human interferon alpha-2a (rhIFNα-2a) has been widely used for antiviral, anticancer as well as immunomodulatory clinical therapy. In our previous research, we constructed the Open Reading Frame (ORF) encoding synthetic rhIFNα-2a to be in framed with N-terminal alpha factor secretion system in pPICZαB shuttle vector. This research covered overproduction, characterization of the protein and preliminary determination of biological potency assay by using the estrogen positive cell line MCF-7. To overproduce the protein, cultivation was performed in Buffered complex medium containing glycerol (BMGY) for 24 h and production was performed in Buffered complex medium containing methanol (BMMY) during 48 hours at 30°C. The recombinant protein was purified by affinity chromatography using blue sepharose resin. Analysis of amino acid sequence by using MS/MS2 mass spectrometry covered 21% of the total amino acid residues. To determine the initial biological activity, the effect of rhIFNα-2a on MCF-7 breast cancer cell line was studied in vitro. The anti-proliferative activity of hIFNα-2a was performed by 3-[4.5-dimethyltiazol-2il]-2.5 diphenylltetrazolium bromide (MTT) assay. The result showed that the protein is able to inhibit MCF-7 proliferation and the reduction of cell growth was dose-dependently. To monitor the protein production, we performed stability expression assay as well. The result informed that the expression of ORF still stable until 60th generation

Fajriaty I, Hariyanto I.H, Haryanto Y. 2017. Anti-fertility effect of ethanol extract of lerak (Sapindus rarak DC) fruits in female Sprague Dawley Rats. Nusantara Bioscience 9: 102-106. This research aims to observe the antifertility effect of ethanol extract of Lerak fruits (SFEE) in female sprague dawley rats. Group of rats are divided into four groups, the first group is given carboxymethyl cellulose (CMC) as negative control (KN), the second group is given ethinylestradiol as positive control (KP), the third group is given SFEE with dose 50 mg/kg BW, and the fourth group is given SFEE with dose 100 mg/kg BW. The female rats in estrus phase is mated with male rats. In plug vaginal checking and smear vaginal there is sperm that signify if female rats have experienced copulation and currently on the day of the zero pregnancy. All treatment are given orally from 0 day of pregnancy to 10th day of pregnancy. Antifertility effect on treatment in rats is shown from laparotomy in the 20th day of pregnancy. Determining of the antifertility effect that to do include pre-implantation loss effect and post implementation loss effect. The result of this research shows that SFEE with dose 50 mg/kg BW has antifertility effect that is pre-implantation loss as big as 0% and post-implantation loss as big as 10.53% and SFEE with dose 100 mg/kg BW shows antifertility effect is pre-implantation loss and post implantation loss in each of 47.50% and 28.57%. This observation shows that SFEE has antifertility effect.

Abstract

Purwantoro RS. 2017. Seed germination and seedling growth of Chiococca javanica (Rubiaceae) in relation to fruit maturation and media growth. Nusantara Bioscience 9: 111-119. Chiococca javanica is a plant having ornamental potential. The seeds extracted from the fruits were classified according to maturity stage: Unripe - yellow; ripe - reddish; and over ripe - dark-red and seed germination were done in two germination media: sand and rice husk. The statistical approach was a randomized complete block design with three replications. Analysis of variance indicated that the seeds of C. javanica from the ripe and over ripe fruit had higher significances (P<0.05) on germination capacity, speed of germination, mean germination time, mean daily germination, peak value and germination value than the seeds from yellow fruits. The effect of growing media on germination process showed higher significance in sand than that in rice husk. Similarly, mean daily germination in sand was higher than that in rice husk. All seedling growth parameters of the seeds from ripe and over ripe fruit showed significant different on number of leaves, shoot length, diameter of seedlings, number of lateral roots, roots length, fresh weight and dry weight of plants than the seeds from unripe fruit.

Nugrahaeni N, Rahayu M. 2017. Short Communication: Peanut introduced germplasm response against Ralstonia bacterial wilt disease. Nusantara Bioscience 9: 138-140. Peanut germplasm introduction is a means to increase genetic variability within the germplasm collection. However, its utilization is frequently hampered by its susceptibility to bacterial wilt disease, the main disease in peanut cultivation in Indonesia. The aim of the present study was to assess peanut introduced germplasm response against Ralstonia bacterial wilt disease. One hundred and fifty peanuts introduced germplasm were planted on the bacterial wilt endemic area in Pati, Central Java, Indonesia. Pre-planted isolation revealed that the bacterial population in the soil was considered high, i.e. 2.6x106 cfu.g-1. The trial was laid out in RCBD, replicated two times. Each accession was planted in two rows of 4-m length with 40 cm x 10 cm plant spacing. The crop received a basal dose of Phonska at 300 kg.ha-1 and SP36 at 100 kg. ha-1. Observations were made on bacterial wilt disease incidence and agronomic traits. Research result showed that the introduced germplasm was highly variable in their responses to the wilt disease with wilt incidence ranging from 0% to 100%. Twenty-three accessions, out of 150, were resistant to the disease. Those resistant accessions showed good agronomic characters and gave dry pod yield in the range of 1.56-4.5 t.ha-1. This results indicated that introduced germplasm could be used in increasing variability of bacterial wilt resistant gene source as well as in increasing yield potential in developing new peanut improved cultivars.
