Mujiyati, Supriyadi. 2009. Effect of manure and NPK to increase soil bacterial population of Azotobacter and Azospirillum in chili (Capsicum annum) cultivation. Nusantara Bioscience 1: 59-64. The objectives of this research were to find out the increase number of two bacterials populations, Azotobacter and Azospirillum, due to the use of manure fertilizer. The exsperiment was conducted using group randomly designed with two treatments. The plant populations were treated (i) whithout fertilizer as the control, (ii) with manure fertilizer, and (iii) with NPK fertilizer. Data was experimentally collected by planting chili in several plots treated by manure, with three replications. The field experiment was conducted in Gathak Village, Karangnongko Sub-district, Klaten District, Central Java. The data collected consist of the total population of Azotobacter and Azospirillum, nitrogen content in soil and the chili yield. The primary data of research were analyzed using ANOVA test and followed by LSD test, with the degree of significance by 95% .The results showed that the manure fertilizer can increase the population of bacteria as many as 0.02% (Azotobacter) and 0.46% (Azospirillum) when they were compared to the control one. So that it can increase the soil fertility when they were used in long time. Therefore increasing the nutrient availability in the soil was occurred. Application of manure fertilizer could increase the total nitrogen content in the soil and it is very useful for the fertilizing of plants.

Abstract. Maryati KT, Sugiyarto. 2010. Characterization of white grub (Melolonthidae; Coleoptera) in salak plantation based on morphology and protein banding pattern. Nusantara Bioscience 1: 72-77. This research aims to find out the white grub (Melolonthidae; Coleoptera) variability based on the morphological characteristic and protein banding pattern found in "salak pondoh" farm in Regencies of Sleman, Yogyakarta and Magelang, Central Java. Each area has five sampling points. Morphological analysis on white grub was conducted using descriptive method and analysis on protein banding pattern was conducted using qualitative analysis based on the presence or absence of band pattern on the gel, and qualitatively based on the relative mobility value (Rf) of protein. The result indicated that the white grub in Sleman and Magelang, based on morphology characteristic is only one species, namely Holothricia sp. Based on the protein banding pattern, the white grub sample has differences in protein band number and molecular protein weight.

Hastuti D, Suranto, Setyono P. 2009. Variation of morphology, karyotype and protein band pattern of adenium (Adenium obesum) varieties. Nusantara Bioscience 1: 78-83. The aim of this research to find out the Adenium obesum variation from six varieties, namely: obesum, cery, red lucas, red fanta , white bigben and harry potter based on morphology, karyotype, as well as protein banding pattern. The chromosome preparation was made using semi-permanent squash method from the tip of root plant; while protein banding pattern was made using SDS-PAGE method. Qualitative data included shape and color of the leave and flower described from each variety. Data were presented in morphometry and analyzed using ANOVA and then followed by DMRT with 5% of confidence levels, indicated significance difference. Protein banding pattern, the root, stem, leave and all organs were analyzed using Hierarchical Cluster Analysis method with Average Linkage (between Groups) using SPSS 10.0. The result of research shows that the six A. obesum varieties have morphological character with no variation of light green to dark green leave, not hairy, smooth leave bone, meanwhile for light red to dark red flower crown color although some of them are white and the same funnel color, yellow. All varieties of A. obesum have same number of chromosome, 2n = 22 and shows the difference ranging from 2.56 to 5.13 μm. In the banding pattern formed qualitatively, there is variation among the six varieties.

Abstract. Ruwaida IP, Supriyadi, Parjanto. 2009. Variability analysis of Sukun durian plant (Durio zibethinus) based on RAPD marker. Nusantara Bioscience 1: 84-91. The purpose of the study is to assess the diversity of the durian varieties of Sukun, Sunan, Kani, Monthong, and Petruk; and Sukun durian variety is grown in different regions based on RAPD markers. Materials research is durian leaves of Sukun, Sunan, Kani, Monthong and Petruk from Ranukutri Garden Seeds, Karanganyar, and also Sukun durian leaf from Gempolan Karanganyar, Jepara and Salatiga. Then performed DNA analysis on samples of leaves that began the isolation of DNA, test of DNA quantity and quality, primer selection, and amplification with PCR. Visualization of PCR performed by electrophoresis using EtBr that generate DNA bands. DNA bands were then analyzed with NTSYS program to get durian diversity among varieties of durian and between Sukun durian at different planting areas. Six RAPD polymorphic primer was used, namely OPA-01, OPA-02, OPA-07, OPA-16, OPA-18, and OPA-19. Banding pattern of DNA amplification show the existence of diversity among varieties of Sukun durian, Sunan, Kani, Monthong and Petruk and show the diversity of Sukun durian grown in different areas. Abstract. Setyawan AD. 2009. Diversity of Sonneratia alba in coastal area of Central Java based on isozymic patterns of esterase and peroxidase. Nusantara Bioscience 1: 92-103. The study was intended to observe the diversity and the relationship of Sonneratia alba in southern and northern coast of Central Java Province based on isozymic patterns of esterase and peroxidase. This research was conducted in July until December 2003, at six sites, i.e. Wulan (WUL), Juwana (JUW), Pasar Bangi (PAS), in the northern coast, and Bogowonto (BOG), Motean (MOT), and Muara Dua (MUA) in the southern coast. The laboratory assay was conducted in Central Laboratory of Mathematics and Natural Sciences, Sebelas Maret University (UNS) Surakarta. The seedling plant samples that were collected from six mangrove habitats transplanted in greenhouse in the laboratory. There were 20 individual samples each site. The result indicated that the individual of S. alba of the same or near location has the same genetic diversity in common, because the genetic exchange on the same population was higher than on a different population. Therefore, the populations of S. alba from northern coast had higher similarity each other than southern coast one, on the other way the populations from southern coast had higher similarity each other than northern coast one.

Abstract. Latifa IC, Anggarwulan E. 2009. Nitrogen content, nitrate reductase activity, and biomass of kimpul (Xanthosoma sagittifolium) on shade and nitrogen fertilizer variation. Nusantara Bioscience 1: 65-71. The aims of this research were to study the effect of shade and nitrogen fertilizer variation and also their interaction to nitrogen content, nitrate reductase activity, and biomass of kimpul (Xanthosoma sagittifolium (L.) Schott.). This research was arranged in randomized complete block design (RCBD) with two factor, they were shade variation which contained 3 levels (0%, 50%, and 75% of shade) and nitrogen fertilizer dosage variation which contained 4 levels (0; 0.0625; 0.125; dan 0.25 g ZA/kg of soil of the dosage of ZA fertilizer). The variables of the research were nitrogen content, nitrate reductase activity, fresh weight, dry weight, and shoot/root ratio. The collected data were analyzed with General Linear Model Univariate, and continued with Duncan's Multiple Range Test (DMRT) on 5% significantly levels. The result showed that the shade treatment affected nitrogen content, fresh weight, and shoot/root ratio. The fertilizer application and the interaction between shade and fertilizer application were affected to all variables. The treatment of 75% shade and 0.25 g ZA/kg of soil of ZA fertilizer dosage increased nitrogen content (5.32%) and fresh weight (420.88 g). The treatment without shade and 0.25 g ZA/kg of soil, dosage of ZA fertilizer increased nitrate reductase activity (260.58 µ mol NO2-/g/hour) and dry weight (53.92 g). The treatment of 50% shade and 0 g ZA/kg of soil of ZA fertilizer dosage increased shoot/root ratio (0.98).

Abstract. Handajani NS, Dharmawan R. 2009. Effect of VCO to leucocyte differential count, glucose levels and blood creatinine of hyperglycemic and ovalbumin sensitized Mus musculus Balb/c. Nusantara Bioscience 1: 1-8. Chemical medicines and insulin can decrease glucose blood levels in hyperglycemic patients with macrovascular side effects. The quality and quantity of leucocytes influence diabetes and allergy incidences. Lauric acid within VCO reports decreased glucose blood level of diabetes and some allergy incidents. The purpose of the study is to know the effect of VCO on the glucose blood level, differential leucocyte count, and creatinine blood level on hyperglycemic and normoglycemic ovalbumin-sensitized mice. Forty-five (45) males (mice) of Mus musculus Balb/c with an average weight of 35 g are divided into nine groups with five repetitions; those are four non-alloxan groups and five alloxan-induced hyperglycemic groups. From the 22nd day to the 36th day, they are sensitized to ovalbumin as an allergen. A blood sample was obtained by orbital vena using heparin as an anticoagulant to measure glucose blood level by GOD method to 6 times, on 1st, 4th, 18th, 22nd, 32nd, and 37th days, then are tested by ANOVA followed by DMRT 0.05. On the 37th day, differential leucocytes are determined, the blood level is counted, and then compared to the normal value. This study showed that within differential leucocytes count of hyperglycemic mice, neutrophile percentage was much lower than the normal value (3.22%), and lymphocyte percentage was much higher than the normal value (94.54%). Consumed 0.003 mL/35 g VCO more 18 days decreased glucose blood level on hyperglycemic mice, decreased basophile percentage of ovalbumin-sensitized mice, normalized neutrophile percentage no increased creatinine blood level.

This research is aimed to observe the body weight and vital statistic measurement of 50 Texel sheep. Sheep are classified into five treatments of giving woof P0 (giving greenish tree woof without concentrate), P1 (giving greenish woof and concentrate without adding the ramie hay/0%) concentrate), P2 (giving greenish woof and concentrate by adding 10%) ramie hay), P3 (giving greenish woof and concentrate by adding 20%) ramie hay), P4 (giving greenish woof and concentrate by adding 30%) ramie hay), every treatment was repeated 10 times. The result shows that even though it can’t replace the concentrate function, adding ramie hay as much as 10%), 20%) and 30%) on sheep woof can increase the body weight’s growth, respectively 186.67 g/day, 153.34 g/day dan 103.34 g/day. The addition of ramie hay 10%), 20%) and 30%) can increase the addition of statistic vital’s measurement on breast of sheep livestock 1.20 cm); 0.95 cm) and 0.90 cm); the addition of statistic vital measurement on the body length of sheep livestock 0.05 cm); 1.00 cm) and 0.75 cm) and also the addition of breast width is 1.50 cm); 0.15 cm) and 0.3 cm). Meanwhile, the addition of ramie hay on livestock woofs can only increase the addition of vital statistical measurement on the breast at giving 30%) as big as 0.15 cm). It is needed to know further about giving ramie hay by comparing hay of different concentrations of leaf and stems.

Abstract. Wardani S, Sugiyarto. 2009. Characterization of white grubs (Melolonthidae: Coleoptera) at salak pondoh agroecosystem in Mount Merapi based on isozymic banding patterns. Nusantara Bioscience 1: 38-42. The aim of this research was to know the characteristics of white grubs (Melolonthidae: Coleoptera) based on isozyme banding patterns. This research was conducted at Sleman, Yogyakarta, and Magelang-Central Java for morphological purposes. The sample was taken from 5 places with different heights in which five samples were taken from each location. The method used in this research was polyacrylamide gel electrophoresis (PAGE) using the vertical type. The enzyme system used in this research were peroxidase and esterase to detect the isozyme banding patterns. The results showed a variation in isozyme banding patterns of white grubs (Melolonthidae: Coleoptera) at salak pondoh agroecosystem in Mount Merapi’s slope (peroxidase in station II and IV while esterase in station III and V). It means that genetic variation on the white grubs population at salak pondoh agroecosystem in Mount Merapi’s slope was found. The environmental condition also influenced the isozyme banding pattern’s appearance because each location had a different state.

Abstract. Setyawan AD. 2009. Effect of global warming on plant evolution and diversity; lessons from the past and its potential recurrence in the future. Nusantara Bioscience 1: 43-52. Lessons from the past show that global warming and glaciation are repeated natural cycles; the trigger factor is not always the same. Still, global warming is always accompanied by elevated levels of CO2 and greenhouse gases in the atmosphere, which causes the other rising global temperatures. Present and destruction of various plants and other living things continue to happen from time to time. Every era has its own life form, as a mirror of global environmental conditions at the time. Biodiversity is not always the same between one period of global warming and the subsequent global warming or one period of glaciation with the next glaciation. However, new breeds always show traces of the evolution of their ancestors. Man is one of the agents of global warming that began with developing agricultural systems since 8,000 years ago. The impact of climate change due to global warming should continue to be wary of. Based on past experience, global warming is always followed by mass extinctions, but various life forms will still survive even though their shape is almost certainly not the same as before. Living organisms that can survive will evolve into new taxa different from their parental taxa. Humans who were present at that time probably were not men present at this time, given Homo sapiens may have been extinct for not being able to adapt or otherwise has evolved into a new man who may no longer show characteristics of human wisdom.
