Abstract

Seagrasses in Bali are distributed on coastal areas of south east Bali coastal waters of Nusa Dua, Serangan Island,Sanur Beach and beaches in Nusa Lembongan. In Bali, it has been reported that there are eight species of seagrasses. Thisresearch aimed to identify seagrass species on Lembongan Beach based on morphological characters and optimize PCRcondition for molecular analysis. This research is a preliminary research on molecular method for seagrass analysis. Seagrasssampling was conducted in Lembongan Beach (in front of Ketut’s Losmen) in Nusa Lembongan, Nusa Penida Subdistrict,Bali Province. DNA extraction method used was the method of Doyle and Doyle with modifications. Result showed thatthere are five species found in Lembongan Beach, Nusa Lembongan. These seagrass are Cymodocea rotundata, Enhalusacoroides, Halodule pinifolia, Thalassodendron ciliatum and Thalassia hemprichii. DNA extraction resulted in high size ofDNA and smear DNA. Optimation of PCR reaction of rbcL fragment was done at DNA concentration of 30 ng and 50 ng.The electrophoresisof PCR products showed that DNA concentration of 50 ng had thicker band than concentration of 30 ng.Keywords: DNA extraction, Morphology, PCR rbcL, Seagrass

Abstract

This research aims to find out the quality of curled satay in terms of Staphylococcus aureusthat are spread around in the traditional markets of Karangasem, Bali. Samples of the curled satay were taken from the 4 regions in Karangasem (Selat, Rendang, Sidemen, and Seraya), every region takes 1 traditional market and every market are owned by 2 salesman. The method used in this study is the method of Platting Method (Pelczar and Chan, 2005).  The results shows that the highest of curled satays in traditional markets of Karangasem is found in S. aureusbacteria was obtained for up to 73 x 102 CFU/g found in Sinduwati market, Sidemen on salesman 1 and 57 x 102 in Seraya Tengah market, on salesman 1. Based on the tests that has been done shows that a number of curled satay sold in traditional markets of Karangasem, Bali has exceeded the threshold amount Indonesian National Standard. The maximum lilit of microbial impurities in satay ready consumption for S. aureusbased on Indonesian National Standard is 102CFU/g. Abstract

The research aimed to analyze genetic variation of Rangda coconut in Bali based on microsatellite DNA markers. The young leaf (busung) were collected from Gunaksa village at Klungkung regency, Tulikup village at Gianyar regency, Kubu village at Bangli regency, Sanghyang village, Jembrana regency one sample each, and three samples  are from Ngis village at Karangasem regency. Doyle and Doyle DNA extraction method was used to extract DNA from the samples. Three primer pairs of CnCirC3, CNZ05 and CNZ51 were used to amplify DNA samples. Amplicons were electrophoresed on PAGE 10% and stained with silver nitrate. The allele size was determined by plotting the amplicon migration distance on semilog paper. The results showed there were 11 alleles with an average of 13.33 alleles per primer. There were 3 alleles on primer CNZ05, 4 alleles on primer CnCirC3 and CNZ51 each. Heterozigosity expectation (He) value on primer CnCirC3, CNZ05, and CNZ51   were   0,49±0,04, 0,27±0,03 and 0,71±0,06 consecutively. The result showed that Genetic relationship among 7 coconut samples were divided into two big groups. Key word : Rangda coconut, Microsatellite marker, Number allele, Genetic relationship

Abstract

Breadfruit (Artocarpus communis Forst.) leaves have a high antioxidant content. This research aims to know the effect of breadfruit leaf extracts on blood sugar levels and the amount of sperm in rat. This study used a Completely Randomized Design (CRD) using 30 rats aged 2-3 months and weighing 150-200 grams. Male rats were divided into six groups, i.e. negative control group (K1) without treatment, groups K2, P1, P2, P3 and P4 are given then NaCl 0.9%, breadfruit leaf extracts 50 mg/kg, 100 mg/kg, 200 mg/kg body weight, and glibenclamide 1 mg/kg body weight respectively. The treatments were given every day for 21 days. Blood sugar levels were measured before and after aloksan administration, day 7, 14, and 21 after treatment as well as counting the number of sperm after 21 days. The data obtained were analyzed with ANOVA and Duncan Multiple Range Test (DMRT), using the program of SPSS For Windows version 22. Results of the study showed that breadfruit (Artocarpus communis Forst.) leaf extracts could lowered blood glucose levels and the most effective concentration was 100 mg/kg body weight of breadfruit leaf extracts given for 21 days with the effectiveness of 66,77%. Breadfruit leaf extracts did not inhibit spermatogenesis. So the amount of rat sperm did not different significantly among treatment. Keywords: blood sugar levels, breadfruit, rat, sperm count

Abstract

This research aimed to determine the effect of colchicineby seed immersion ondessert rose (Adenium sp.) seedling. Observation were done on morphological and anatomical characters. This research was conducted using colchicine concentration of 0%, 0.05%, 0.1%, and 0.15%. Each treatment had 10 replications. The parameters observed included seedling emergence, seedling height, number of leaves, leaf length, leaf width, and stomatal density of cotiledone. The results showed differences in morphological characters, led to the emergence of seeds on the ground inhibited by immersion in colchicine. Anatomically giving of colchicines cause a reduction instomatal density of cotiledone. Keywords: anatomy, colchicine, dessert rose, morphology

Abstract

This research aimed to determine the concentration giving Carbon (C) the right to know the waste liquid as solvent Bacterial growth media Phosphate (BPF) performed in the Laboratory of Microbiology, University of Udayana. Experimental research method by adding Dextrosa concentrations varying the liquid waste that has been given out bacterial isolates phosphate solvent the concentration of 0 g (as a control), 0,54 g, 0,77 g and 1,00 g Dextrosa. The results showed the growth of bacteria Solvent Phosphate (BPF) E. gergoviae no significant differences (P <0.05) between the control and treatment of 0,54 g and 1,00 g, 0,77 g where as treatment showed significant differences (P> 0.05) are marked with the number of total number of bacterial colonies of E. gergoviae as many as 2,45 CFU / mL.
