Abstract

Background:Ultraviolet B (UVB) exposure is one of the external factors which can cause reduction of cell viabillity through photochemistry reaction and can be managed using antioxydant. Failures of DNA repair and apoptosis have some important roles in photocarcinogenesis. Objective: This research is aimed to know the effect of vitamin C on the viability of UVB irradiated keratinocytes and HeLa cells.Method: This research employed a simple experimental method. Two groups of cells used in this research were: Group I normal foreskin keratinocytes passage III, and Group II HeLa cells. Each group was divided into 25 sub-groups consisting of 2x104 cells each, treated with vitamin C at 0, 6, 12, 40, 200 µg/ml concentrations, and UVB irradiated (Phillips UVB TL4OW/12RS) at 0, 200, 400, 800, 1600 mJ/cm2 intensities. Each treatment was done in quintet. Viable cells were determined based on formazan blue reaction using ELISA-reader 550 nm 24 hours after treatment. The results were analyzed by independent t-test, one-way ANOVA, and multivariate analysis using three-way analysis of variance,Result: Vitamin C at 6, 12, 40, and 200 ug/m1 concentrations significantly increased normal keratinocyte and HeLa cells viability which were not irradiated by UVB, statistically significant, compared to normal keratinocyte and HeLa cells which were not treated with vitamin C. HeLa cells, which were irradiated by UVB at 200, 400, and 800, 1600 mJ/cm2 intensities and treated with vitamin C at 6, 12, 40 and 200 ug/ ml concentrations, statistically significant in decreasing cell viability. Conclusion: Vitamin C at 200 µg/ml concentration, which was given to normal keratinocytes irradiated by UVB at 200, 400, 800, and 1600 mj/cm2 intensities used in this research showed its protection effect, thus enhancing the viability of the cell. Vitamin C 6, 12, 40 ug/ml, which were irradiated with UVB at 200, 400, 800, and 1600 mJ/cm2 intensities, did not show protection effect. HeLa cells were less resistant than normal keratinocytes to UVB irradiation at 200, 400, 800, and 1600 mj/cm2 intensities.Key words: vitamin-C, HeLa cell, keratinocyte, cell viability, UVB

Abstract

A randomized controlled study on the insertion of Norplant without prophylactic antibiotics had been carried out. The aim of the study was to compare the risk of complication happening shortly after insertion. A total of 500 acceptors completed their follow-up, 244 belonged to the antibiotic group (tetracycline), while 256 belonged to the control group. They come from suburban areas stuTounding the city of Yogyakarta.Insertion was carried out outside the hospital during the Safari Mass Campaign, with hospital standard asepsis. Follow-up was carried out one week after insertion to detect the presence of such complication or side effects as pain, inflammation, abscess and sense of itching. Data were processed with version 2.0 SPSS program using Nikon PC Computer. t-test, chi-square test and relative risk were statistical analysis used in the study. Results showed that both groups were comparable in. terms of age, body weight, parity, duration of insertion and level of education. The overall incidence of complication in the non-an tibiotic group was 19,9% compared to 24.2% in the antibiotic group (RRO.82, 95% CL 0.57– L19) . More detailed complications such as pain, inflammation, abscess and sense of itching show neither clinical nor statistical significant differences. The risk of complication in all, cases was influenced neither by duration of insertion nor by acceptors level of education. The study concluded that the insertion of Norplant without prophylactic antibiotics was as safe as with antibiotics.Key Words: Norplant - contraceptive mass campaign - antibiotics - obstetrics - contraceptive complication

Abstract

Total iodine contents in the urine samples were directly tested with and without destruction using Ion Selective Electrode (ISE) method. The dry destruction was carried out with volume variation of 0.5 M NaOH solution and wet destruction was done with 4 ml of concentrated H2504 and 1 ml of concentrated HNO3 mixture, followed by volume variation of 1% p-aminophenol solution and temperature to reduce 103- form to form. The Chloride ion contents were also determined using technique. It was found that the optimum of NaOH amount, the optimum of 1% p-aminophenol and temperature (wet digestion) was 2 ml, 12 ml, and 60°C respectively. The chloride ion contents in 6 urine samples were in the range of 2373-4495 ppm and didnt interfere on r determination. The total r contents in the six samples without destruction, using wet and dry destructions were in the range of 280-392 ppb, 291-505 ppb and 306-516 ppb respectively. The t-test showed that the total Iodine content in urine samples with and without destructions was significantly different.Key words: Iodine - urine - ISE method - wet and dry destruction - chloride

Abstract

Key words : inflammatory bowel disease -- TNBS colitis -- budesonide-lymphoid and non-lymphoid cell populations -- MHC class II expression -- adhesion molecules.

Abstract

Background: Almost all medical treatment may pose risk. The risk of adverse event in patients undergoing surgery is higher than those who undergo non surgical intervention. The incidence of adverse event varies from 3.7% in the US to 16.7% in Australia. Basically, most of the adverse events are preventable. It is therefore recommended to implement clinical risk management in health care services. Objective: To identify risk of adverse event among patients undergo elective urologic surgeryMethod: A cross sectional study was carried out to collect prospective data on patients underwent elective urologic surgery at Tabanan General Hospital between August-September 2005. Data on the risk of adverse event were identified since patients were admitted to Hospital, underwent pre-op preparation and surgical procedure. Follow up was carried out until 30 days after surgical procedureResult: Seven major potential risks were identified, i.e.(1) pre-operative stay> 3 days (28.85%);(2) patients risks for surgery (44.23%); (3) prophylactic antibiotic were given > 2 hours before surgery (88.46%); (4) ASA > (82.69%); (5) preoperative hair removal > 2 hours before surgery (86.53%); (6) use of intravenous line more than 24 hours (26.92%); and (7) use of urine cathether > 6 days (25%). It is found that pre-operative stay > 3 days was significantly related to the occurrence of adverse event. Conclusion: Pre-operative stay > 3 days contributed significantly in the occurrence of adverse event among patients underwent elective urologic surgery. However, other risk factors were not significantly related to adverse event due to small sample size.Key words: elective urologic surgery - risk - clinical risk management - clinical governance
