Abstract

This research is conducted to study the effect of anaerobic digestion on the reduction of BOD, COD, TSS, VFA, and Cr(VI) content and biogas production of industrial wastewater bearing chromium. Material used in this experiment are sludge of tannery waste and dairy cattle manure.  Splitplot design was used in this research, as mainplot is level of sludge i.e. 0, 10, 20 and 30%, and the subplot is days of digestion i.e. 0, 10,20, and 30 days, respectively.  The results of this research show that increasing level of chromium in the waste inhibit biological activity due to the toxicity of the chromium. Anaerobic digestion decreased the value of BOD, COD, TSS and Cr(VI) concentration, the value is 75.54%, 44.62%, 41.30% and 22.40%, respectively. Biogas production and VFA concentration decreased significanly after 20 days of digestion.  The conclusion is biometanogen can be used to reduce the level of pollution of organic matter and Cr(VI).  Anaerobic digestion can be used as alteranative of bioremediation of industrial wastewater bearing chromium.

Abstract

This study aimed to obtain binary vectors carrying  cryIB-cryIAa hybrid gene suitable for Agrobacterium-mediated transformation of two Indonesian rice cultivars, Rajalele (Javanica) and Cisadane (Indica).  Introduction of hybrid Bt toxin, encoded by cryIB-cryIAa hybrid gene with different binding receptor in the insect gut driven by maize ubiquitin promoter, was intended to reduce the possibility of the appearance of stem borer resistant to Bt toxin. A HindIII fragment containing the ubi cryIB-cryIAa hybrid gene was excised from pBKS ubi cryIB-cryIAa and cloned into the HindIII site of binary vector pCAMBIA 1301, 1303, or 1304.  Restriction analysis of recombinant plasmids confirmed the insertion of the gene and demonstrated that the gene inserted in two orientations. The recombinant plasmids were transformed into electrocompetent Agrobacterium tumefaciens EHA105 by electroporation.  Embryogenic calli of Rajalele and Cisadane were co-cultivated with A. tumefaciens EHA 105 harbouring binary vector pCAMBIA 1301, 1303, or 1304 containing ubi cryIB-cryIAa. GUS histochemical assay revealed that high transformation efficiency (75-100%) on both cultivars were obtained from embryogenic calli transformed with pCAMBIA 1303 or 1304.

Abstract

Rambutan fruit have a great potential to be developed as an exported commodity. The constraint is the existence of biannual bearing characteristic of the plant, which is an alternation of fruit yield between on year and off year. This condition resulted in a problem in product availability continuity. The aim of this research was to induce rambutan plant flowering in the off year with the application of paclobutrazol. There were three doses of paclobutrazol treatment including 0 g/tree, 1.5 g/tree, and 3 g/tree. Bud dormancy as an implication of paclobutrazol application were broken by the application of KNO3 at 20 g/l concentration. The experiment was arranged in completely randomized factorial design 3X2 in triplicate. The result showed that paclobutrazol treatment was able to induce rambutan flowering in the off year. Paclobutrazol treatment significantly increased the number of generative buds. Based on the generative bud percentage, the higher doses of paclobutrazol application the stronger stimulus of flowering. Paclobutrazol treatment also reduced the inflorescences length and increased the chlorophyll content of the flowering support leaves. It seems that application at dose 3 g/tree caused bud dormancy but dose 1.5 g/tree did not. Bud dormancy as a result of paclobutrazol application could be broken by the KNO3 application. The optimum dose to induced rambutan flowering was 1.5 g/tree because this dose seems did not caused bud dormancy and gave a higher number of generative buds compared to 3 g/tree dose.

Abstract

During the 2002-2003 period, the research was done to study 11 Rainbow Lorikeets reared in an animal house laboratory. The aim of this research was to find the food rations formule of the Rainbow Lorikeets so that those birds can be able to grow and breed well. The food rations were composed of 26.3% local bird foods (521), 35.09% lampung bananas, 8.77% slice corns, 10.5% boiled quails eggs, 1.75% white bread, bean sprouts and red sugar are 8.77%, respectively. All of food materials were mixed on the plastic cup and mixed with 450 ml of water, then pulverized like sweet porridge. That porridge was given to birds in cafeteria and the water was made ready “ad libitum” everyday. The results have shown that giving food rations formula can stimulate two pairs of the Rainbow Lorikeets breeding and during the 2002-2003 period they produced three young birds.

Abstract

There has been a revolutioner development in our understanding about coral reproduction in the middle of 1980s. All publication, before this period, therefore provides incomplete facts and wrong conclusions about the modes of coral reproduction. Most corals are hermaphodite and broadcast spawners, in which fertilization takes place externally. Temperature, photoperiod, lunar and tidal cycles are thought to be major environmental factors influencing reproductive cycle in corals. In Indonesia, sexual pattern of scleractinian corals consistent with previously reported data from other regions. Available data show that multispecific spawning may occurs in Indonesia. There are also some evidences that there is variation in spawning season between corals in Lombok and Karimunjawa Islands.
