Abstract

Curcumin is a compound which potential to be developed. The curcumin analogues havebeen synthesized. The compound 3–chloro–4–hydroxybenzaldehyde was used as starting material inthe synthesis of curcumin analogues with Cl substituent on the aromatic ring. The compound of3–chloro–4–hydroxybenzaldehyde were synthesized by chlorination reaction betweenp–hydroxybenzaldehyde and Cl2 gas. The purity of 3–chloro–4–hydroxybenzaldehyde wasdetermined base on the melting range, thin layer chromatography and LC–MS. The structureidentification of the compound was observed done by spectrometry techniques, including massspectra (ESI–MS), infrared spectra (cm-1, KBr), Nuclear Magnetic resonance proton spectra (δ, ppm,DMSO-d6, 1H–NMR, 500 MHz) and UV–Vis spectra. Chlorination reaction ofp–hydroxybenzaldehyde produced yellow crystal of 3–chloro–4–hydroxybenzaldehyde, its meltingrange is 128.1 to 130.8ºC. The average yield obtained of this reseach is 51.37%.

Abstract

Psycho active substance abuse is a common problem throughout the world, and it is associated with the increase of mortality and morbidity. One of substitution therapy program is methadone maintenance therapy program. The main problem is most of patients stop following the maintenance program before they feel the therapeutic effects of it. Compliance in taking daily medication is the focus in achieving the health status of patients. The research objective is to determine the relationship between the knowledge level with methadone maintenance therapy program in the public health center of Parakan, District of Temanggung. The research method was cross sectional approach. The population in this study were all patients of methadone maintenance therapy program in the public health centers of Parakan, District of Temanggung. Samples on this research were 31 patients and they are all active on methadone maintenance therapy program in the public health center of Parakan, District of Temanggung. Data analysis was performed using Chi Square (x2) formula correlation test. The results show there is a relationship between knowledge level with methadone maintenance therapy program in the public health center of Parakan, District of Temanggung with p value 0.001, with 13 respondents (42%) are in a good level of knowledge category and methadone treatment in obidiance catagory is 11 respondents (35.5%). It is concluded that there is a relationship between methadone maintenance therapy program in the public health center of Parakan, District of Temanggung.

Abstract

Antioxidant is compounds which have abilities to inhibit oxidation rate. Anredera cordifoliafolium consist of flavonoid which have activities as antioxidant. The purpose of this research was toknow antioxidant activity at ether fraction of hydrolysed aquous extract of Binahong folium (Anrederacordifolia). Ether fraction was obtained by hydrolysing the infuse Binahong folium (Anrederacordifolia) with HCl 2N then fractionated with ether and then evaporated. The antioxidant activity wasmeasured by DPPH method using spectrophotometer. The maximum wavelength and operating timequercetin and samples were 515,0 nm (quersetin) and 514,0 nm (ether fraction), 19-30 minute(quersetin) and 19-29 minute (ether fraction). Scavenging activity test done with the DPPH method(1,1- dyphenyl-2-picrylhydrazyl). The decreasing of DPPH radical solution decreased the absorbantintensity. The percentage of radical scavenging activity was performed as EC50 (EffectiveConcentration). The results indicated that both samples showed free radical scavenging activity withEC50 value for quercetin (2,702±0,15) µg/ml, and ether fraction of (249,31±9,26) µg/ml. Statisticalanalysis using Kruskall Wallis method with 95% of confident level followed by Mann Whitney testgiving evidence the results between samples and positive control (quercetin) are significantlydifference.

Abstract

Curcumin is a chemical compound that has low solubility and bioavailability and, for these reasons, limited biological effectiveness. For improvement, the solubility needs to be increased by nanotechnology and nanoparticles, among others. Nanostructured Lipid Carrier (NLC) is a new drug delivery system that offers several advantages, including a significant increase in drug solubility and entrapment efficiency. This study aimed to formulate curcumin into curcumin-loaded NLC preparation and determine its characteristics, absorption efficiency, and drug loading. The formulation used evaporation and solvent diffusion methods with three different concentrations, namely 5%, 10%, and 15%. Spectrophotometry and HPLC were employed to test the absorption efficiency and drug loading capacity.  The results showed that the curcumin-loaded NLC preparation containing 10% curcumin had stable characteristics and produced particles sized 17.4 nm with a polydispersity index of 0.574 and zeta potential of -63.43 mV. Based on the spectrophotometry results, the entrapment efficiency was 93.212%, and the drug loading capacity was 0.708%. Meanwhile, the HPLC showed that the entrapment efficiency was 93.007%, and the drug loading capacity was 0.795%.

Abstract

Growth inhibitory activity of combination of mango honey-probiotic milk produced by Lactobacillus acidophilus (L.a) and Lactobacillus casei (L.c) against E. coli and S. aureus has been studied. The fermented milk was prepared by inoculating each probioticin milk at 43ºC and then incubated at room temperature for 24 hours. Diffusion agar technique by using nutrient agar media was used to assess the minimum inhibition concentration (MIC). Result of the research showed that pH of fermentation broth of the probiotic milk reached 5. Biomass obtained that calculated by Total Plate Count of L.a and L.c was 1.3 x 107 cfu/mL and 20.7 x 1013 cfu/mL respectively. Inhibition zone diameter ofthe mangohoney-L.c showed an optimum antimicrobial effect against tested bacteria at 1:9 ratio. The MIC value against E. coli and S. Aureus was 15.7 ± 6,1 mm and 13.3 ± 3.0 mm respectively. The activity of mango honey-L. aexhibite dan optimum antimicrobial effect against tested bacteria at 2:8.The MIC value againstE. coli and S. Aureuswas14,7 ± 1,1mm and 13.3 ± 1.3mm respectively.

Abstract

The tendency to use herbal drug simultaneously with synthetic drug may result inan interaction. Based on research, aqueous extract of Centella asiatica (L) Urbancontains saponin (asiaticoside and madecassoside) compounds that inhibit CYP450and is a safe herbal alternative that is used for anti-inflammatory treatment. The aim ofthis study was to investigate the effect of aqueous extract of Centella asiatica (L) Urbanto the ibuprofen tablet bioavailability in male rabbits. Both compounds are metabolizedby CYP450 enzymes in the same way with different mechanism. The cross over designwith the same subject was used in this study. The samples consisted of 400 mg ibuprofenwith 0%b/v, 25%b/v, 50%b/v dan 100%b/v aqueous extract of Centella asiatica (L)Urban respectively. The blood were taken after 0; 0,5; 1; 1,5; 2; 2,5; 3; 3,5; 4; 6; 8; 10hours after samples treatment, then the ibuprofen plasma concentration weredetermined spectrophotometrically. The results then were used to evaluat thebioavailability parameters: tmaks, Cpmaks, and AUC and were analyzed using ANOVAtest in the same way as well as the 95% trust standart. The results showed, the use ofibuprofen tablet stimultaneously with aqueous extract of Centella asiatica (L) Urbanincreased Cpmaks dan AUC, but there were no influence to the tmaks value. The use ofibuprofen tablet stimultaneously with 50 %b/v and 100 %b/v aqueous extract ofCentella asiatica (L) Urban had a significant influence to the bioavailability ofibuprofen.

Abstract

Scope of pharmacy practices as well as the role of pharmacist have been improving recently. Pharmacists have responsibility for doing the pharmacy practices, including pharmacist it self and also the technicians. According to PP 51 of the year 2009 it was known that pharmaceutical practices have vary of choice of carreer-field. This present study was a descriptive study which was conducted by survey method using stratified random sampling. The respondents of this study were 1st, 2nd, 3rd and 4th grade students from Faculty of Pharmacy, Andalas University. The result showed that, in term of the profile of respondent, it was known that most of the students chose to work at industry of pharmacy of 48.2%, followed by hospital of  35.3%, University/Research Institute of 4.7%, pharmacy of 3.5%, pharmacy retail of 2.4% and others career-field (such as BPOM, civil servant at ministry of health and enterpreneur) up to 5.9%. Based on descriptive analysis, it was described that field of work was the most important criteria of affecting factors for pharmacy student in choosing their future career. The others important criterias those also afforded as the affecting factors were geographis, payment/income, flexibility of work-schedule, possibility of career-development and benefit, successively. As for result of independent sample t-test in term of gender, both of men and women chose same three main affecting factors in choosing career-field (including field of work, geographis as well as payment/income). The different result showed for 4th, 5th and 6th factors.

Abstract

Fries Rearrangement of eugenyl acetate in order to study allilic substituentgroup influence has been done. Fries rearrangement of eugenyl acetate was done byheating at 120°C for 3 hours under AlCl3, dichloromethane as the catalyst and solvent,respectively. The structure of the compound was identified using Infra Redspectrometry (IR) and GC-MS. The result of te research showed that rearrangementfries of eugenyl acetate are not formed, but yielded lightly brown oil of dimmer.

Abstract

Obesity may cause the lipid metabolisms disorder so that a risk factor for hypercholesterolemic. Catechin and epigallocatechin gallate in white tea potentially inhibit lipase activity. Caffeine compounds and catechin derivatives have a role as antihypercholesterolemic. The objective of this study was to explore antiobesity and antihypercholesterolemic activity from white tea infusions to lipase and total cholesterol in rats model fed a high-fat-diet. Twenty-five rats were divided into five groups. Negative control group was administered with distilled water. The positive control group was administered with orlistat dose 14.2 mg/kgBW. Group 3rd, 4th, and 5th were treated with white tea infussion doses of 125, 250 and 500 mg/kgBW/day, respectivelly. period and feed intake was calculated daily during the treatment. White tea doses of 500 mg/kgBW decrease the total cholesterol by 37.16±11 mg/dL, lipase level of 23.70±0.64 U/L, weight up to 3.53±1.21 g/day and decrease feed intake by 12.71±1.76 g/day. The infusion of white tea can decrease total cholesterol level, lipase level, body weight and feed intake. This study concluded that infusion of white tea has antiobesity and antihypercholesterolemic effect on rats fed high-fat-diet.

Abstract

Liver is one of the most important organ for drug metabolism in the body. The damage to the liver can be marked by increased activity of the enzyme alanine aminotransferase (ALT) in the blood. Lycopene is an antioxidant that has the ability to eliminate free radicals and detoxifies electrophilic compounds that damage can be prevented. The purpose of this research is to know whether lycopene can inhibit the increase alanine aminotransferase activity of SD (Sprague Dawley) rats induced by parasetamol. This experiment using 20 male rats Sprague Dawley. They were divided into 4 groups. Group I is a healthy control (baseline) was only given aquadest, group II were given paracetamol as pain control, group III and IV are treatment group were each given lycopene with a dosage 15 µg/kg BW and 30 µg/kg BW for 21 days. On 19th day group II, as well as all treatment groups induced paracetamol. The blood sampling was performed on orbital sinus on 19th day (before induced by paracetamol) and on 21th day or two days after induced paracetamol to know the difference alanine aminotransferase activity. The data were analyzed using non-parametric statistical methods with Kruskal-Wallis test followed by Mann-Whitney test with 95% confidence level. The result has showed that the lycopene doses 15 µg/kgBW had alanine aminotransferase activity of 130.10 ± 11.83 U/I and lycopene doses 30 µg/kgBW had alanine aminotransferase activity of 106.46 ± 2.36 U/I. If compared with paracetamol control had alanine aminotransferase activity of 92.53 ± 2.57 U/I, should take a summary that the lycopene doses 15 ìg/kg BW and 30 ìg/kg BW couldn’t decrease alanine aminotransferase activity on SD (Sprague Dawley) male rats.
