Abstract

Propanolol is a non-selective beta-blocker that is used widely to overcome cardiovascular disorder.Development of propranolol in transdermal delivery is necessary to avoid the first pass metabolism that reducesthe active metabolite up to 15-23% left. This study was objected to determine the effect of VCO (Virgin CoconutOil) concentration as a base and penetration enhancer of propranolol in the cold cream preparation through rat skinmembrane in vitro. Variation concentration of VCO (0%, 14%, 28%, and 42%) was added to propanolol coldcream. Transdermal in vitro study was performed using vertical type diffusion cell with PBS pH 7,4 as receptormedia. The temperature was maintained at 35ºC with a constant stirring rate at 300 rpm. The transport wasconducted for 8 hours. Flux, efficiency, and lag time were calculated as responses. The results showed that flux atvarious concentration of VCO (0%, 14%, 28%, 42%) were 12,30; 14,13; 14,52; and 23,06 -1 cm-2respectively. The transport efficiency were 6,5x10-4; 7,5x10-4; 8,1x10-4; and 1,22x10-3 % cm-2 respectively. Thelag time were 1,13; 1,26; 1,11; and 0,92 hours respectively. It can be concluded that the VCO can be used mainlyas a base in the preparation of cold cream and can increase percutaneous permeation of propranolol significantly(p <0.05). VCO concentration of 42% has the highest performance.

Abstract

The study of effect ethanolic extract Curcuma heyneana rhizoma as antihypertrigliserida examine of rats after given by dietary of high fat has been carried out. This experiment use 28 white male Wistar grooved rats with weight about 150-200 gram body weight, at about 2 -3 months old. They were divided into seven group randomly and each group 4 rats. The first group was given by standart treatment of dietary (BR II) and aquadest, the second group was given by dietary of high fatty and aquadest, the third group was given by CMC Na solution, the fourth was given by gemfibrosil in CMC Na suspension, the fifth group, the sixth group and the seventh were orally given by of ethanolic extract with dose 10g/kgBW; 20 g/kgBW; and 40 g/kgBW. Hyperlipidemia was treated with higth dietary fatty (5 gram fat cows and 10 gram egg yolk) at the second period (two weeks). The treatment of ethanolic extract Curcuma heyneana rhizoma was given after hyperlipidemia. The trigliserida content determined with GPO (Glycerol-3-Phosphate Oxidase) test. The trigliserida level in the serum analyzed on the before treatment, during the treatment and after treatment. The result of the research showed that ethanolic extract Curcuma heyneana rhizoma 2 weeks can decrease trigliserida level.

Abstract

Apigenin is one of the flavonoid compounds contained in celery (Apium graveolens L.) and has been used for the treatment of various diseases. This study aims were to ensure the validity and accuracy of analytical method for apigenin assay in celery extract using UV-Vis spectrophotometry. The flavonoids test of celery extract give a positive result which produces a yellow color that can be drawn by amyl alcohol. The result of TLC test using a mobile phase of chloroform and methanol (9.5 : 0.5) produced Rf value of 0.75. Validation of analytical methods were sensitivity, linearity, limit of detection as well as limit of quantification, accuracy, and precision. Validation of analytical methods showed that sensitivity results as represented by molar extinction (ε) was 77004.35 M-1cm-1. Linearity from calibration curve at concentration of 2, 4, 6, 8, and 10 mgL-1 resulted an equation of y = 0.074x – 0.0011 with coefficient correlation (r) of 0.9996. The limit of detection and limit of quantification were 2.99 mgL-1 and 9.92 mgL-1 respectively with recovery percentage of 97.59% and the coefficient variation values were 1.53%; 2.43% and 1.68%. The average contents of apigenin in the celery extract were of 3.74 ± 0.09%.

Abstract

Mucoadhesive is a topic of current interest in the design of drug delivery systemand treatment of peptic ulcers. This concept is based on the strength of adhesion of aprotective material to the mucosa. King Banana (Musa xparadisiaca AAB) is oftenused in the treatment of abdominal pain. It has been suspected having muchoadhesivestrength to cover peptic ulcer. The study was armed to formulate king banana tabletwhich has muchoadhesive ability to protect gastric mucosa become ulcer. The aim ofpeptic ulcer cover test was to see the ability of king banana tablet to covered ulcer at thegroups test that received different treatment of the negative control group (solvent), thebanana tablet suspension group, and sucralfate group. The ulcer severity wasdetermined by scoring method. All experimental data were tested by SPSS analyticalstatistics. The result showed that king banana tablet has similar strength to peptic ulcercover of sucralfat and not significantly different.

Abstract

Endophytic fungi are microorganisms reside in the living tissues of the host plant. This fungi can contribute to provide protection to the host from the infection caused by another microorganism. The one reasonable strategy to discover a new antibacterial agent from endophytic fungi are from the plant which lived in special condition such Rhizophora mucronata. The aim of this research is to determine antibacterial activity of the endophytic fungi extracts assosiated with Rhizophora mucronata from Sagara anakan. The isolated endophytic fungi was identified as Neopestalotiopsis sp. and Peniophora lycii using molecular analysis method. The antibacterial activity was carried out by using microdillution method. The antibacterial properties from mycelium extracts showed moderate to low antibacterial activity with Minimum Inhibitory Consentration (MIC) values of 125-500 µg mL-1 against Eshcerichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923 bacteria. The n-hexane extract of both endophytic fungi shows the strongest antibacterial activity against Escherichia coli with MIC values of 125 µg mL-1

Abstract

The compound N-(2-nitrobenzyl)-1,10-phenantrolinium chloride has been synthesized bynucleophilic substitution reaction bimolecular (SN2) between the 2-nitrobenzyl chloride with1,10-fenantrolin monohydrate. This study is an experimental research of Organic Synthesis.Synthesis is done by varying the compound 2-nitrobenzyl chloride to 1.10 Phenantrolinemonohydrate ie 1:1, 3:1, 5:1 and 7:1. The purity of the compounds synthesized are determinedbased on the melting range data and TLC. Identification of the structure of the compoundssynthesized done by spectrometry UV/Visible, IR and GC-MS. The results of this research is acolorless amorphous solid from N-(2-nitrobenzyl)-1,10-phenantrolinium chloride with anoptimal yield 37%+5% and the variation of mole 2-nitrobenzyl chloride effect on the amount ofyield product, there is an optimal yield in the mole ratio of 1:1.

Abstract

The degenerative diseases is caused by endogenous antioxidant cannot neutraliced from increased concentration of free radicals, therefore it is necessary to consume antioxidant from the outside ofbody for neutralizing and destroying free radicals. The aim of this research is to evaluate the antioxidant activity of methanolic extract of longan leaves. The experiment of antioxidant activity from extract longan leaves uses the scavenging assay using 2,2’-diphenyl-1-pycrylhydrazyl (DPPH) method and quersetin as positive control. The analytical result is processed using linear regression methode in order to get ES50 value. The results of research showed that theincreasing concentration of methanolic extract, and quercetin causes the higher percentage of DPPH radical scavenging. The ES50 values of methanolic extract and quercetin obtained are 40.32 µg/ml and 2.48 µg/ml respectively.

Abstract

Candidiasis is an infection caused by Candida albicans. Synthetic treatment against Candida albicans has been developed, but reports of resistance to antifungal agents that there continue to emerge, and cause a  lot of  side effects. Pacar kuku (Lawsonia inermis L.) is one of  the  plants  that has antifungal activity. Chemical content of polyphenols inhibit candidiasis is a form of naphthoquinone. This  study  aimed  to test the antifungal activity of ethanolic extract  of  insoluble ethyl acetate fraction of  pacar kuku leaves  against Candida albicans. The ethanol extract was obtained  from the ethyl acetate extraction of previously fractionated with ethyl acetate. Identification of chemical compounds made with phytochemical  screening. Test antifungal activity against Candida albicans using microdilution method. Amphoterisin B infusion in doses 250 µg/ml with the  concentration  of 1,50 µg/ml, 0,75 µg/ml, 0,38 µg/ml, 0,19 µg/ml, 0,09 µg/ml, dan 0,05 µg/ml were used as standard solution. The parameters in this study are the Minimum Inhibitory Concentration (MIC) and Minimum Fungicidal Concentration (MFC). The concentration of the extract used in the MIC and MFC determination  were 80%, 40%, 20%, 10%, 5%, 2,5% (w/v). Identification of chemical compounds was performed using test tubes and assay on anthraquinone compounds. The results showed that the MFC of the ethanol extract is 40% w/v. The MIC and MFC of amphoterisin B were 0,09 µg/ml and 0,19 µg/ml respectively. The test tubes showed that the ethanol extract contains  polyphenols, saponins, and quinone. The naphtoquinone total content in the ethanol extract consecutively 7,97±0,14%.

Abstract

Pneumonia is a lung infection caused by bacteria with high fever symptoms accompanied by cough with phlegm, shortness of breath. According to Riskesdas 2013 results, the prevalence of pneumonia based on diagnosis at Yogyakarta Special Region is 1.7% and 4.6%. The researcher has founds 172 adult patients diagnosed pneumonia during 2011-2016 in PKU Muhammadiyah Yogyakarta hospital. This study aims to determine the level of effectiveness between 3rd generation cephalosporins with 3rd generation cephalosporins and macrolides on the outcome of adult patients with pneumonia in PKU Muhammadiyah Yogyakarta hospital period 2015-2017. It was a retrospective cohort study with an affordable population. There were two groups of 3rd generation cephalosporins (n = 24) compared to the 3rd generation cephalosporins and macrolide (n = 13) measured results in the form of body temperature, shortness of breath, leukocyte count, and length of hospitalization using univariate and bivariate analyzes. Based on the results of the study, 70.3% was dominated by men, and 51.4% was dominated by the age group > 59 years. Both of groups showed a significant difference to the temperature where p=0.001 with difference of temperature decrease 0.79 ± 1.06 oC for single group while p=0.049 with difference of temperature decrease 0.65 ± 1.07 oC for combination group; as well as against leukocyte, both of groups had a value of p=0.001 where difference of leucocyte decrease was 4.80 ± 3.92 103/mm3 for single group and 5.43 ± 4.27 103 / mm3 combination group. While both of group showed no significant difference for shortness of breath p=0.638 with OR (95% CI) 2.4 (0.23 – 24.06) and p=0.435 for hospitalization, in single group was 5.88 ± 3.261 days while in combination group was 5.00 ± 1.826, this shows that the combination group had an average length of hospitalization of 0.88 days faster than the single group. The effectiveness between two groups showed a significant difference with p<0.05 on body temperature and leukocyte count. While both groups did not show a significant difference with p>0.05 to shortness of breath and length of stay.

Abstract

The synthesis and kinetics studies of Al-MCM 41 of the methylene blue adsorption have been done. The observations of the adsorption kinetics are very important parameter in the adsorption process. This is the higher of adsorption kinetics tend to faster adsorption process. The results of characterization of Al-MCM 41 have a wall thickness is 15.173 Å, Pore Volume Pore is 0.584 cm3/g, Pore Diameter is 30.446 Å, Surface Area is 995.513 m2/g. The results of the adsorption kinetics of Al-MCM 41 to  methylene blue adsorption follows pseudo second order with coefficient of determination (r2) value of 0.999 and the value of adsorption rate constant of Al-MCM 41 is 10-2 g.mg-1.min-1.
