Abstract

The improper and uncontrolled uses of antibiotics against pathogenic bacteria have resulted in the occurrence of MultiDrugs Resistant (MDR) strains. There is now an urgency to find alternative antibiotics to combat the MDR strains.Sponge associated microorganisms are among the most interesting and promising marine natural product sources whichproduce polyketide and non ribosomal peptide products with various biological activities. In this study, marine bacteriawere isolated from sponge Aaptos sp. collected from North Java Sea, and were screened for antibacterial activityagainst MDR strains. Three out of 64 bacterial isolates were successfully screened and were found to be active againstMDR strains, in which two isolates (SPA1 and SPA5) were active against resistant strain Escherichia coli and one isolate(SPA21) against resistant strain Proteus sp., respectively. These active isolates were also capable of amplifying NRPS(Non Ribosomal Peptide Synthetases) gene fragments necessary for the biosynthesis of non ribosomal peptides. Themolecular identification based on partial 16S DNA nukleotide sequences indicates that the active isolates areHalomonas aquamarina, Alpha proteobacterium, and Pseudoalteromonas luteviolacea.

Abstract

Astrometric study of galactic clusters using UCAC2 catalog containing proper motion data is described. Kinematicalanalysis is applied to determine cluster membership. The near infrared photometric data of the same catalog is used toderive physical parameters of the cluster. In the present study the case for NGC 3532 is given. An isochrones fitting tothe color-magnitude diagram gives the age of cluster in log t = 8.85 and the distance of 515 ± 41 pc with infraredreddening in J filter of 0.043 ± 0.017 mag. Our results are in good agreement with results of previous studies.

Abstract

3-(1-Hydroxy-4-chlorobenzyl)-1-methylindole (3) has been synthesised in three steps. The first step involved methylation of indole to give N-methylindole (4) in 87% yield which on Vilsmeier-Haack formylation in the next step gave new ketone (5) in 67% yield. Sodium borohydride reduction of the new carbonyl compound (5) in the last step afforded the new indole alcohol (3) in 99% yield. Treatment of the secondary alcohol (3) with p-toluene sulfonic acid has been examined which afford di-(1-methylindol-3-yl)-(p-chlorophenyl)methane (7) in 71% yield.

Abstract

This study aims to find out a method of spawn characterization for wood ear mushroom (A. polytricha [Mont.] Sacc.)based on microsatellite DNA marker. Four strains of wood ear mushroom cultivated in several regions of Java i.e AUC(West Java), AUCN (Central Java), AUP (West Java) and AUT (West Java) were evaluated. A pair of designed primers(5’ -GGGAAAGTGATCCCATCTT-3’ and 3’-AGTTGTGGGAACATCGAACT-5’) was able to amplify microsatelliteDNA from those four strains of wood ear mushroom. Strains AUC, AUCN and AUP share the same positions (2 loci) ofmicrosatellite motive (TC)n with PCR products of 212 and 201 bp, while the same motive for strain AUT was found atdifferent loci with PCR products of 127, 108, and 96 bp. The same motive was also found at another locus of strainAUC and AUP with a PCR product of 228 bp. Another different locus with a PCR product of 85 bp also gave positiveamplification result for strain AUC. The data on macroscopic characters (mycelial growth rate and quality, fruitingpercentage and mushroom yield) revealed that AUC, AUCN and AUP are good strains, while AUT is bad strain.Overall, there is a good correlation between molecular (microsatellite DNA) patterns and macroscopic data on mycelialgrowth rate and quality, as well as flushing percentage and mushroom yield. Therefore, microsatellite DNA motive of(TC)n can be adopted for spawn characterization of wood ear mushroom.

Abstract

Optimization of overproduction, purification and characterization of Streptococcus pyogenes’s Mga recombinant protein from Escherichia coli pMALMga carrying mga49 gene had been carried out. The overproduction optimization was conducted by culturing the Escherichia coli pMALMga of OD600 = 0,7 in LB medium with or without glucose, and induced by 0,3 mM IPTG for a certain period of time. The protein produced was MBP-Mga fusion protein with a molecular weight of about 104 kDa. The purification was conducted by affinity chromatography using amylose ligands at pH 7,4 and continued by SDS-PAGE. One band identified at 104 kDa showed that the MBP-Mga fusion protein was pure enough for further analysis. The MBP-Mga protein was then injected into rabbit to produce polyclonal antibodies. The antibody development was monitored by dot blot and Western blot. The antibodies produced gave a positive reaction to MBP-Mga fusion protein. Adsorption and precipitation of the polyclonal antibodies which gave a positive reaction to MBP protein was done by adding pure MBP protein (56 kDa) produced by Escherichia coli pMALc2, and purified by affinity chromatography using amylose ligands at pH 7,4. The results showed that the antibodies gave a positive reaction to Mga recombinant protein and a negative reaction to MBP recombinant protein.

Abstract

Variation of DNA sequences of the internal transcribed spacer (ITS) region has been used as a molecular character tounderstand the phylogenetic relationships within different strains of Phyllanthus niruri and with other species belonging tofamily Euphorbiaceae. Phylogenetic analysis of 19 plant samples using parsimony method revealed several findings: as awhole family, Euphorbiaceae is a monophyletic group and the family is divided into two major clades based upon its singleleaf arrangement; genus Phyllanthus is a non-monophyletic group, though species Phyllanthus niruri is a monophyleticgroup; major classification system of Phyllanthus niruri at the molecular level does not support previous majorclassification; Saoropus androgynus is closely related with Phyllanthus niruri.

Abstract

Two dimers of stilbenoids, (-)-balanocarpol (4), and (-)-ampelopsin A (5), along with a tetramer (-)-hopeaphenol (6) were isolated from the stem bark of Shorea multiflora Burck (Dipterocarpaceae), locally known as Meranti. Their structures were determined by physical and UV, IR, 1H-NMR, and 13C-NMR spectroscopic data.

Abstract

Copolymers of δ-valerolactone and 2,2-dimethyl-1,3-propandiol monomers P(VL-co-DP) as raw materials in synthesisof poly(urethane-ester) were prepared by a ring-opening polymerization of 2,2-dimethyl-1,3-propandiol and δ-valerolactone monomers in various composition using 1-hydroxy-3-chloro-tetraisobuthyldistanoxane as a catalyst.Characterizations of polymers were carried out by analysis of functional groups, microstructure, viscosity, hydroxylnumber, and also thermal properties of copolymers. The copolymers of 2,2-dimethyl-1,3-propandiol and δ-valerolactonemonomers have a triblock chain structure, that is P(VL-block-DP-block-VL) with various chain lengths, which dependon VL/DP ratio used in copolymerization. The increase of δ-valerolactone composition used in polymerization leads toan increase of molecular weight and melting point as well as a decrease of hydroxyl number of copolymers.

Abstract

In actuarial science, a multistate process deals with the joint distribution of two random variables: time until termination and cause of the termination. In a three state process (active, retired, death), an insured in active state canbecome retired according to a force of decrement of retired, insured in retired state can move to death state accordingto a force of decrement of mortality. The main issues are the development of the estimation methods for transitionprobabilities and its implementation on a real data set. Analytical studies of multistate model are well developed in theliterature. However, the empirical studies based on real data set rarely reported. Furthermore, published works focusedon modeling the transition probabilities as function of period of transition. This work presents an extension of transitionprobabilities as function of age of insured and period of transition. A case study of modeling three state process andmethodology for estimating the model parameters from an insurance database under constant forces of decrement ispresented. The forces of decrement was estimated using single decrement moment estimation procedure, and yields atransition probability for a three state models. This paper aims to contribute to the development of empirical transitionprobabilities of multistate model based on real data set as an alternative to the traditional approach to doubledecrement modelling.

Abstract

In a drug discovery program to combat various diseases, many natural chemicals originating from Indonesian tropicalplants including Moraceae and Dipterocarpaceae have been investigated in our laboratory. These family of plants areknown as rich sources of stilbenoid type of compounds which have been shown to have various bioactivities, that arepotentially important in cosmetic bioindustry. In the course of our search for bioactive natural products from plants, wewere interested in the stilbenoid constituents of these plants. Thus, as part of this program, several stilbene derivativeswere successfully isolated from extracts of Morus macroura, a Moraceous plant locally named andalas, includingoxyresveratrol (1), lunularin (2) and new resveratrol dimers, namely andalasin A (3) and andalasin B (4). While,biotransformation of oxyresveratrol (1) using cell suspension culture of Solanum mammosum affordedtrans-oxyresveratrol-4-O-β-D-glucopyranoside (5). Similarly, some resveratrol derivatives have been isolated fromDipterocarpaceous plants, such as Shorea assamica, S. seminis, Vatica umbonata, and Dryobalanops oblongifolia.These species gave, among others, resveratrol (6), resveratrol-12-C-β-glucopyranoside (7), the resveratrol dimers(-)-ε-viniferin (8) and the new compound diptoindonesin A (9), in addition to resveratrol trimers (-)-α-viniferin (10), thenew compounds cis-diptoindonesin B (11) and trans-diptoindonesin B (12). Among these compounds oxyresveratrol (1),andalasin B (4), trans-oxyresveratrol-4-O-β-D-glucopyranoside (5), resveratrol (6), cis-diptoindonesin B (11) andtrans-diptoindonesin B (12) exhibited the most potent antioxidant activity [1,1-diphenyl-2-picrylhydrazil (DPPH)radical-scavenging activity]; moreover, andalasin A (3) have been reported to show a very strong tirosinase inhibitoractivity, thus valuable in cosmetic industry.
