Abstract

The purpose of this research was to evaluate the use of onion extract (Allium cepa L) diluted in Ringer’s Dextrose base extender on the semen quality of Peranakan Etawah (PE) buck. Fresh onion was grinded and filtered with filter paper. The filtrate was then inactivated by heating it at 56oC for 10 min to avoid the negative or detrimental effect of its biological activity. The onion filtrate was added to the tube containing Ringer’s–dextrose solution with the concentration of 0%, 1%, 2% and 3% (v/v). Semen was collected with artificial vagina from a healthy selected PE buck reared at the Teaching Farm Laboratory of the Animal Husbandry Faculty Brawijaya University. The semen was diluted with prepared extender to rich the sperm concentration of 75x10 millions/ml. The semen parameters were observed after 1 and 2 hours during storage at room temperature. The results showed that concentration of onion extract (OE) affected the individual sperm motility (P<0.05), sperm viability (P>0.05) and sperm abnormality (P<0.05) during semen evaluation at 1 and 2 hours following the semen dilution time. The highest quality of semen following the dilution was shown by the dilution containing 2% of OE (2 ml/100 ml extender). It was concluded that it was possible to use the OE to improve the semen quality during storage at room temperature. A further study of the use of OE should be conducted to explore more positive effect of OE when it is used in combination with standard semen extender and other technique semen storage such as cold storage or freezing process.   Keywords: semen quality, onion extract, Ringer’s dextrose

Abstract

The purpose of this study was to evaluate the nutritional value of cassava and tofu mixture fermented by Aspergillus niger, Rizhopus oligosporus and the combination among them (50% of Aspergillus niger and 50% of Oligosporus Rizhopus) as a substitute feed ingredients corn meal. The research method was laboratory experiment using the analysis of completely randomized design (CRD) factorial (3 x 4). The first factor was the 3 types of microbes (M) that consisted of M0 = fermentation by Aspergillus niger, M1 = fermentation with Rhizopus oligosporus and M2 = fermentation with the combination 50% of Aspergillus niger and 50% of Rhizopus oligosporus. The second factor was 4 incubation time (T) that consisted of T0 = non-fermented, T1 = 2 days of incubation time, T2 = 4 days of incubation time and T3 = 6 days of incubation time. Each treatment was repeated 3 times. Variables measured were proximate nutrient content, fiber component content (NDF and ADF, silica, lignin), soluble protein content and mineral content (calcium and phosphorus). Data were analyzed by ANOVA, if there were differences among the treatments then it woukd be followed by Duncan's Multiple Range Test. Results showed that microbes provided a significant influence (P <0.01) on ash (%), crude protein (%), crude fiber (%), crude fat (%), gross energy (Kcal / Kg), soluble protein (%), NDF (%), ADF (%), cellulose (%), hemicelluloses (%) and silicate (%) but they did not provide significant effect on lignin (%). The incubation period had highly significant effect (P <0.01) on ash (%), crude protein (%), crude fiber (%), crude fat (%), gross energy (Kcal / Kg), soluble protein (%), NDF (%), ADF (%), cellulose (%), hemicelluloses (%), lignin (%) and silicate (%). It was concluded that fermentation was best treated using a combination between 50% of Aspergillus niger and 50% of Rhizopus oligosporus with 2 days of incubation time. Abstract

Copra meal is by-product of oil extraction that is produced in large quantity and relatively cheap in Indonesia. However, its high cellulose becomes obstacle for poultry diet. Two studies was conducted to optimize the utilization of copra meal by producing crude enzyme that matched with copra meal using solid state fermentation method with various doses and incubation time that mixed with Pleurotus ostreatus (PO) and Trichoderma viridae (TV). In the first study, copra meal was fermented with 4 levels of inoculum (L0 = no inoculum; L1 = 17.7 CFU/g of TV and 175.00 CFU/g of PO per kg of copra meal; L2 = 35.4 CFU/g of TV and 218.75 CFU/g of PO per kg of copra meal; L3 = 53.1 CFU/g of TV and 262.50 CFU/g of PO per kg of copra meal and 4 incubation time (W1= 4 days; W2 = 6 days; W3 = 8 days; and W4 = 10 days). Parameters measured were crude protein, crude lipid, crude fibre and gross energy. A completely randomized factorial design was used in the study. In the second study, crude enzyme was produced from the best results found in the first study. A method of Jacob and Prema (2006) was used to produce enzyme. Meanwhile, activity of cellulase was measured based on the method of Omojosola (2008). The results showed that factor of inoculum level was found significantly increased protein content and gross energy but decreased crude lipid and crude fibre of the mixed fungi-fermented copra meal. Incubation time did not affect protein content but significanly affected crude lipid, crude fibre and gross energy. Interactions between inoculum level and incubation time was found in crude lipid, crude fibre and gross energy contents of mixed fungi-fermented copra meal. Activity of cellulase was 0.71 g glucose/l.   Keywords : Fermentation, Pleurotus ostreatus, Trichoderma viridae, inoculum level, incubation time, cellulase activity
