Abstract

Ruminal bypass protein’ is one of the strategies to increase the amount of protein, which enters abomasum and hence, increases ruminant productivity. One of the strategies to obtain “ruminal bypass protein” is by complexing tannin or formaldehyde with protein. Two experiments were conducted. The first experiment aimed to obtain the optimum level of added tannins to decrease the dry matter digestibility of protein sources. Tannins were isolated from Calliandra calothyrsus leaves. Tannins were added at the level of 0, 10, 20, 30, 40 and 50 mg to each in vitro tube containing 0.5 g of protein source (gliricidia leaves, soybean meal or casein) and the tubes were incubated for 48 hours. The result showed that the optimum level of tannin was 60 mg/g of gliricidia leaves or casein and 80 mg/g of soybean meal. The second experiment aimed to compare the ability of tannin to formaldehyde to form complex with protein. One set of tubes containing tannin-protein or formaldehyde-protein complex was incubated with rumen liquor for 48 h and another set was incubated with rumen liquor (48h) and followed by pepsin-HCl for 24 h (total incubation time: 72 h). After incubation at 48 h or 72 h, the dry matter or protein digestibility of tannin-protein complex was much higher than those of formaldehyde-protein complex. The amount of ruminal bypass protein was almost similar between the two agents to complex gliricidia leaves (34.4 and 32.1 g/100g for tannin-gliricidia and formaldehyde-gliricidia, respectively). Tannin-soybean meal interaction produced 27.9 g/100 g of ‘ruminal bypass protein’, which was half of that from reaction between formaldehyde and soybean meal (54.1 g/100 g). Tannin-casein complex was easily broken down in the rumen so that ‘ruminal bypass protein’ from this complex was very small. It can be concluded that the ability of tannin to form ‘ruminal bypass protein’ depends on the type of protein source but is the same with the ability of formaldehyde to bind forage protein (Gliricidia sepium). Abstract

Aspergillosis detection by using serological method has not been reported in Indonesia. In this case, a study was conducted, by using mycelium extract of A. fumigatus as the antigen. Rabbits and chickens were injected with the antigen to produce positive serum (antiserum). The antigen and antiserum were tested serologicaly by Immunodiffusion/Agar Gel Precipitation (AGP), ELISA and Immunoblot. Chicken serum of broiler and layer collected from field were also included in the test. All positive serum of the experimentally animals gave positive results with all methods of serological tests. No bands of precipitation reaction in AGP test with chicken serum from the field. Both chicken and rabbit positive serum with ELISA test showed high Optical Density (OD), while field chicken serum from broiler commonly gave lower OD compared to layer. Immunoblot test of chicken positive serum showed bands of reaction with the antigen in nitrocellulose membrane, approximately on 33, 38, 44, 52, 70, 77, 97, and 110 kDa, meanwhile field chicken serum with high OD in ELISA test, showed bands approximately on 16, 18, 33, 38, 44, 47, 52, 70, 77, 84, 97, and 110 kDa. It means that the field chicken serum contain immunoglobulin molecules has spesific antibody of aspergillus antigen. It is concluded that the ELISA test can be used for screening on chicken aspergillosis in serological methode.     Key Words: Antigen, Aspergillus fumigatus, Serology, Chicken

Abstract

Inclusion limit of palm oil sludge (POS) in poultry diet varies according to processing, and species or strain of the animal. Therefore, an experiment was conducted to study the utilization of fermented (FPOS) and non-fermented palm oil sludge (POS) for growing drakes. A number of 224 one week-old male ducklings were randomly distributed into 7 dietary treatments, with 4 replicates and 8 ducklings for each replicate. The dietary treatment consists of 2 kinds of feedstuffs (POS and FPOS) with 3 levels (5, 10, and 15%) and one control diet consisting neither POS nor FPOS. All diets were formulated with similar nutrient contents and meet the requirement of growing duckling and fed to 8 weeks old. The results showed that at the first week of the trial, feeding of POS or FPOS significantly (P<0.05) depressed growth of the ducklings, although the feed consumption was higher than the control. However, overall performances (body weight, feed consumption and feed conversion) of the ducklings, carcass yield, liver weight, gizzard weight and abdominal fat weight measured at the end of the trial were not significantly (P>0.05) affected by the dietary treatments. Therefore, it is concluded that it is safe to include POS or FPOS up to 15% in the diet of growing ducklings.   Key words: Palm oil sludge, fermentation, ducks, growth

Abstract

Verotoxigenic Escherichia coli (VTEC) strains are responsible for serious human illnesses. These strains are commonly found in milk. The aim of this study was to determine the occurrence of verotoxigenic E. coli in milk. A total of 351 milk samples, were collected from dairy farms in Bogor, Sukabumi and Cianjur. These samples were analyzed for VTEC using biochemical, serological and vero cell cytotoxicity assays. VTEC O157:H7 isolates were found in milk collected from dairy herds in Bogor and Sukabumi at rates 0.47% of 214 samples, 1.10% of 91 samples respectively, and none in Cianjur. Hemolytic E. coli isolates were found in 0.94% of 214 milk samples from Bogor, 2.2% of 91 milk samples from Sukabumi and none from Cianjur. From E. coli isolates, 53 isolates (67.95%) were verotoxigenic, consisted of: two E. coli O157:H7 isolates and 51 non O157:H7 isolates.Therefore this study showed the occurrence of VTEC in milk samples from dairy farms in Bogor, Sukabumi and Cianjur. Abstract

This research aimed to find out the genetic progress for Aceh cattle by several growth traits selection such as birth weight (BW), weaning weight (WW), yearling weight (YW), mature weight (MW) of does to identify the best selection response from bulls and cows breeding patterns. The recorded data of production and reproduction of Aceh cattle from 2010 to 2014 at breeding centre were analyzed and used as a technical coefficient on estimate heritability (h2), genetic correlation (rG), direct selection response (RY) and correlated selection response (CRY) on several breeding patterns. Most of  h2 and rG value among growth traits were positive and high. The highest value of Ry found on BW (0.04), WW (1.21), YW (2.05), MW (3.28) and this was obtained on breeding pattern for 3 years and cow for 6 years. The indirect selection or CRY value based on BW were lower than WW. It is concluded that WW might be used as selection criterion in order to increase WW, YW and MW.

Abstract

Eleven male buffalo calves were divided into infected group of 7 animals and non-infected control group of 4 animals. The infected group was then inoculated with trickle doses of 15 Inetacercariae of Fasciola gigantica twice weekly for 32 weeks and killed 36 weeks after first infection. There were no clinical symptoms observed . Infected and non-infected buffaloes, had similar values of packed cell volume, haemoglobin and red blood cell counts . In infected buffaloes, plasma glutamate dehydrogenase enzyme activity increased in proportion to the dregree of hepatocyte destruction level and evidence of necrosis caused by the migrating of immature flukes through the parenchyma prior to their entry into the bile ducts. While the values of plasma glutamyl transpeptidase showed only a minimal rise with a small peak in week 20 as evidenced by histological observation that infected caused limited damage to epithelial surface of the bile duct . These results indicates that, the resistance mechaninisms of buffalo against fasciolosis infection occurred in the liver or before flukes entering into the bile ducts. These results seem to indicate that, in buffalo, resistance mechanisms to fasciolosis infection occured in the liver or before flukes entry into the bile ducts.   Key words: Fasciola gigantica, clinicopathology, buffalo calves, trickle infection, resistance

Abstract

An experiment has been conducted to determine the effect of type of packaging and time of storage on fermented palm oil sludge. The palm oil sludge was fermented with Aspergillus niger, dried, kept in differentpackaging materials (plastic bags, feed bag, and paper bag) and stored under room temperatures for 12 weeks. The experiment was assigned in a split plot design (3x7). The parameters measured were water content, crude protein, soluble nitrogen, true protein, in vitro dry matter (IVDMD) and true protein digestibilities (IVTPD), total in vitro digestible protein, and the activities of mannanase and cellulase. Results showed that fermented palm oil sludge stored for 12 weeks increased the water content, decreased the true protein and fiber contents, and also reduced the activity of mannanase and cellulase and in vitro dry matter digestibility but no changes on the crude protein content, protein digestibility, and total digestible protein. It was concluded that fermented palm oil sludge should be kept in feed bag under room temperature if to be stored for 12 weeks.   Key words : Palm oil sludge, fermentation, nutritive value, storage

Abstract

Blocks containing 3% phenothiazine in solidified molasses (Wonnolas, Animeal Australia Ltd .) were tested to control gastrointestinal nematode infections and the effect on mineral status in sheep in villages in Cirebon, Indonesia. Two hundred and thirteen Javanese Thin Tail sheep which were grazed during the day and housed at night were divided into two groups on the basis of location . One group was allowed in an access to Wonnolas blocks when penned for 24 weeks while the other group remained untreated. The bodyweight was recorded and faecal samples were collected for egg counts and larval culture every four weeks. Sera and saliva samples were collected from 20 adult sheep in each group at weeks 0, 12 and 24 for mineral analysis by AAS and colorimetry. The results indicated that the mean egg counts of the treated group decreased from 576 epg to 123 epg and the percentage of sheep producing viable larvae decreased from 50% to 24%. In contrast egg counts of the control group increased from 768 epg to 4,840 epg and the percentage of sheep producing viable larvae increased from 65% to 84% over the same period . In the treated group the number of Haenronchus larvae declined significantly (36% to <6 %) and at the end of the trial Tiichostrongylus larvae predominanted in larval cultures (>80%) . Mineral analysis revealed deficiencies in sodium and copper, low levels of zinc and normal levels of potassium, calcium, magnesium and phosphorus . Wormolas had a significant effect on sodium and zinc status but not on copper although sufficient levels of this element were available. Comparison of bodyweight gains showed a significantly (P<0.05) higher rate of increase in the treated animals.

Abstract

The hemagglutinin and neuraminidase are important immunogen of avian influenza virus that are suitable for recombinant experimentation. However, both genes have been experienced rapid mutation resulting in diverse variety of genotypes. Hence, gene expression in recombinant systems will be difficult to predict. The objective of the study was to examine expression level of H5 and N1 genes from a field isolate by cloning the genes into expression vector pEGFP-C1. Two clones respresenting fulllength of H5 and N1 gene in plasmid pEGFP-C1 were transfected into chicken embryo fibroblasts (CEF), rabbit kidney (RK13) and African green monkey kidney (VERO) cells using Lipofectamine ‘Plus’ reagent. The experiment showed level of gene expression in the VERO cell was higher than in the RK13 and CEF cells. Observations using fluorescent microscopy and Western blotting revealed that the N1 gene was expressed better in all cells compared to the H5 gene. Abstract

This study has been conducted at the Laboratory of Physiology Reproduction, Research Institute for Animal Production (RIAP), Ciawi-Bogor, West Java. Sperms were collected from FH bulls with body weight 613 kg (FH-1) and 480 kg (FH-2) twice a week. Briefly after quality evaluation, semen was diluted in Tris-Citrate buffer medium, containing egg yolk (20% v/v) and (4% v/v) glycerol to get spermatozoa concentration of 50 x 106 per ml. Sperm diluents were added with glutathione (GSH) with doses of 0.0; 0.5; 1.0 and 1.5 mM as treatments A, B, C and D respectively. The diluted semen was then cooled from 35 to 5°C using a cooling machine for 60 minutes then stored in the refrigerator (5°C). Recorded parameters were the survivability of spermatozoa by evaluating the percentage of motile and live, the condition of acrosome and plasma membrane. Data were analysed by completely randomised design with the general linear model (GLM) procedure. The characteristics of collected semen were normal. Viability of spermatozoa stored at 5°C for 0, 1, 4 and 8 days shown by intact acrosomal were 74.42; 69.27; 57.80 and 42.58% for A, B, C and D respectively. Those data were significantly different (P<0.01). Motility, live and intact plasma membrane were 46.72; 52.34; 53.44 and 51.09%; 63.59; 69.11; 68.64; and 66.89%, and 66.01; 69.75; 68.38 and 68.44% for treatment A, B, C and D respectively. Additional 0.5 mM GSH gave the highest (P<0.01) motility, live and intact plasma membrane of sperm. Therefore, it is concluded that the effect of addition 0.5 mM of GSH to the sperm diluents can improve the viability of spermatozoa and possibly protect the spermatozoa from free radical damage.   Key words: Glutathione, viability, spermatozoa
