Abstract

A total of 4,977 sera from a sentinel cattle in West and East Nusa Tenggara were tested for antibody against BHV-1, the causal agent of infectious bovine rhinotracheitis (IBR). These sera were collected between June 1990 and June 1993, and were tested by using serum neutralization test (SNT). Out of these sera, 3,713 were suitable for IBR SNT. A total of 349 sera (10.4%) reacted. IBR reactors were more prevalent in East Nusa Tenggara (NTT) than in West Nusa Tenggara (NTB) . Based on this survey, it is concluded that antibodies against IBR virus are present among cattle in East and West Nusa Tenggara .

Abstract

Immunological responses of sheep against adult worm extract antigen of Fasciola gigantica were evaluated in an effort to identify the protein antigen for the candidate of vaccine. In this study the protein antigen from adult worms was extracted, and the extract antigen was then intramuscularly injected into 4 groups of 5 sheep. Two groups received one injection, these included one group injected only with extract antigen and the other group injected with extract antigen emulsified in Quil A adjuvant. The other two groups received two injections with a two week interval, these included one group injected only with extract antigen and the other group injected with extract antigen emulsified in Quil A adjuvant. Three weeks later all of the sheep were challenged with 300 metacercariae of F. gigantica. The antibody titer was monitored every two weeks by using ELISA and the protein profile from each group was compared by using western blotting. Fifteen weeks after challenged all of the sheep were killed and the liver flukes were collected from the liver and counted. The results showed that the antibody titer was higher in the group which received two injections, and the additional of Quil A adjuvant gave much better protection from the infection of F. gigantica (57%) and could avoid the death of the sheep than twice injection of antigen without Quil A adjuvant (37%).   Key words : Fasciolosis, Fasciola gigantica, protein antigen, antibody responses, vaccine

Abstract

Prolactin gene plays crucial role in the reproduction and egg production of birds. The objectives of this study were to characterize single nucleotide polymorphism in partial intron and coding region of duck prolactin gene. Blood samples were collected from 168 ducks consisted of 19 Pekin, 36 Mojosari, and 113 of their crossbreds collected from Indonesian Research Institute for Animal Production (IRIAP). Primer pairs for the coding regions in prolactin gene were self designed based on the duck genomic sequence database (GeneBank: AB158611.1). PCR products based on DNA of prolactin gene exon two was amplified approximately 400 bp. There is one base insertion of Adenin at the position of 2001 bp intron two region of duck prolactin. Homology test based on BLAST method indicated 99% identity with duck refference (Code Access GeneBank: AB158611.1). Adenin composition in all of duck samples was higher than refference. Triplet hydrogen bonds between Guanine and Cytosin pairs was higher than those at duplet hydrogen bonds between Adenine and Thymine. All duck samples were homozigous and monomorphyc.

Abstract

An experiment was conducted to evaluate the optimum level of inclusion of crude palm oil (CPO) in ducks diet . The treatments consisted of 5 different levels ofCPO (0, 5, 10, 15 and 20%) with 5 replicates and 10 day-old ducks/replicate. The ducklings were allocated in litter system covered with rice hull . At 0-3 weeks old, ducklings were fed with starter commercial diet . The experiment diet was given to starter (3 - 5 weeks old) and to grower (5-8 weeks old) ducks containing crude protein 19 .5 and 17%, respectively and metabolizable energy 2,900 kcal/kg. Measurements done were on body weight, feed consumption, carcass weight, internal organ and abdominal fats . There were no significant difference among treatments on body weight gain, feed consumption, feed conversion, carcass or internal organ weight . There was an increase on abdominal fat with an increase level ofCPO in the diet. Male duck had the ability to accept up 20% CPO in the diet without any negative effect.   Key words: Performance, male duck, crude palm oil

Abstract

Morphological characterization of Madura cattle in Madura islands was done as an input for "action plans" of national animals genetic resources management according to the global system ofFAO. Assessments were done in Sumenep District and Pamekasan District, East Java. According to the body measurements, Madura cattle can be classified as a small to medium type with withers height of about 120 cm. Because of potential productivity in the limitation of environmental resources, Madura cattle can be classified as a "superior" cattle . Body measurements of Madura cattle in the present study were relatively the same with those of 50 years ago, indicating that there is no breeding improvement activities except natural selection . The variability of body measurements is relatively narrow . Improving productivity by outbreeding is needed . To conserve the unique germ plasm of the Indonesian genotype, such as Madura cattle and a possibility to improve their productivity by a complete prevention of cross breeding in the Madura islands needs further evaluation .   Keywords: Madura cattle, morphological characteristic, conservation

Abstract

Organic phosphorous in the form of phytate salts are found in feed component originated from cereals which can not be digested by chicken unless after the addition of an enzyme, i.e. phytase. A research to investigate phytase-producing moulds had been carried out. A total of 60 isolates from various collection units and isolation from samples had been collected then screened in Czapek’s medium with Ca-phytate as the source of phosphorous. From mould growth measurements and clearing zones formation there were 4 mould isolates, i.e. Aspergillus ficuum NRRL 3135 and NRRL 320, Aspergillus niger M94/18/D2 (Balitvet isolate), and Aspergillus niger no. 58 (IPB isolate), which were further observed. Enzyme was produced from medium containing 8% of corn’s starch, 3% of glucose, 0.0005% of KCl and MgSO4.7H2O, 0.0002% of K2HPO4, and 0.00018% of FeSO4.7H2O. The enzyme activity, protein and biomass contents were observed at 16, 24, 32, 48 and 72 hours incubations for Aspergillus niger and at 1, 2, 3, 4 and 5 days incubations for Aspergillus ficuum. The highest protein content was found at 5 days incubation for Aspergillus ficuum NRRL 3135 and NRRL 320, (165 and 169 μg/ml respectively), while for Aspergillus niger (Balitvet and IPB isolates) at 16 hours incubation (102 and 112 μg/ml respectively). Meanwhile, the biomass contents were decreased in accordance with the duration of incubations. The maximum enzyme productions were reached at 24 hours incubation for Aspergillus niger and at 4 days incubation for Aspergillus ficuum. The highest phytase activities were reached at 2.808 U/ml for Aspergillus ficuum NRRL 3135, followed by 1.520 U/ml for Aspergillus ficuum NRRL 320. Aspergillus niger produced enzyme quicker than the other moulds, with lower activity level and decreases quicker at the following hours.   Key words: Phytate, phytase, Aspergillus ficuum, Aspergillus niger

Abstract

Incorporating high amount of fats into the ration for ruminants will affect the rumen microbes adversely and will reducefiber digestion potential. To correct such negative effects, the free fatty acids used for feed should be bond with Ca++, so it canpassing through the rumen savely (rumen by-pass fat). To test the Ca-fat utilization biologically, 20 growing male Garut shee pwere used and fed with 4 type of isocaloric-isoprotein concentrate feed which were allotted based on a randomized block desig nwith 5 replications. The concentrate (C)-A was a positive control diet, while C-B was substituted with 10% free fatty acids (negative control), C-C was substituted with 10% Ca-fat, and C-D with 15% Ca-fat. The concentrate feed was fed at 500 g/d, while forage (King grass) was 4 kg/d. Results of the experiment showed that the negative effect of free fatty acids can be corrected if it was given in the form of Ca-fat. Growth rate curve indicating a good growing pattern, with average daily gain was 100.18, 87.68, 112.86, and 115.00 g/d (P<0.05) for C-A, C-B, C-C, and C-D treatments, respectively. Total dry matter intake for the respective 4 treatments were 875.9, 855.2, 866.7, and 847.4 g/d (P>0.05). Carcass production was relatively good, where for C-A, C-B, C-C, and C-D were 14.84, 14.68, 16.34, and 15.72 kg (P<0.05) respectively, with final live weights of 34.00, 31.74, 34.58, and 34.30 kg (P<0.05). It can be concluded that Ca-fat (rumen by-pass fat) can be used as an energy source component for growing sheep diet, and give the best result at 10% substitution rate in concentrate feed.   Key words : Rumen by-pass fat, growth rate, carcass, sheep

Abstract

Thirty rabbits were used in this study: 22 were infected and eight were used as controls . Fourteen rabbits were inoculated with blood taken from Ongole (Bos indices) cattle or buffalo naturally infected with malignant catarrhal fever (MCF). Another eight rabbits were inoculated with lymphoblastoid cell liras which contain Ovine Herpesvirus-2 (OHV-2), the causal agent of sheep-associated MCF. Various degrees and distributions of histological changes were observed and resembling the lesions developed in MCF. However 17 infected rabbits (77.3%) with non-suppurative meningoencephalitis, eight of them (47.196) were associated with the detection of Encephalhozoon cuniculi histologically .

Abstract

The objective of the study is to determine that sheep farming in the villages to increase farmers welfare could be developed taken into consideration of farmers perception and attitude which were included in the planning of development programme. In order to achieve the objective more effectively, the institutional system of sheep farming needs to be transformed into a corporate institution. Field study was conducted during the period of 1994-1997 in two villages of Jambu sub district, District of Semarang. Two approaches were used simultaneously, i.e. ideografis and nomotetis to analyze the data and information. Result s showed that, first, sheep farming in the villages still had big development opportunity .second, the threat of stagnation in the sheep farming may occur due to high dependency on family labor and lack of capital. Third, due to top down policy and lack of vision on the agrobusiness activities, the development of sheep farming could be hampered. Fourth, transforming into a corporate institution may increase economic profitability of sheep farming as well as performance of the farms. Fifth, in order to realize the development of corporate business systems, need to establish a pilot project. To implement the concept, support from banking institution especially for discount rate was necessary, along with the involvement of ATAI (Agricultural Technology Assessment Institutes), local livestock services, credit system and local government leader that has people oriented vision.

Abstract

Gumboro (infectious bursal disease, IBD) virus was isolated from darkling beetles (Carrinaps pumilin) and their larvae in a commercial pulletchicken farm with repeated outbreaks incidence of Gumboro disease in Tangertng, West Java. In addition, these over populated beetles and their larvae were suspected to be infected and then shed the virus or acted as vectors. Isolation was done by repeated passages of virus using chicken embryo fibroblast cells as prime media, which then showed the evidence of cylop: ihic effecis. The isolation was followed by antigen detection by means of ELISA test.   Key words: Gumboro disease, infectious bursal disease, darkling beetle, Carcinops punulin
