Abstract

A common approach for improving the nutritive value of low quality roughages and crop by-products is by pre-treatment or processing either physical, chemical, or biological treatments. Microwave radiation is one type of physical treatment that could be used to treat low quality roughages. Research was carried out to investigate  the effects of microwave radiation on the rumen degradation characteristics of barley straw obtained from two different stages of maturity. The experiment was run factorially based on completely randomized block design. The first factor was stage of maturity, straw cut during the soft elongation time (C1) and during the harvest time (C2). The second factor was levels of microwave radiation times (MWRT) (T0 = control, without MWR; T1 = MWR for 1 minute, T2 = MWR for 2 minutes). The results indicated nutritive values of barley straw obtained from C1 sampling time were significantly better than that obtained from the C2 sampling time in terms of a higher rumen degradation rate and a much greater total potential rumen degradability. In contrast, MWR did not have significant effects on the rate of degradation and total potential degradability of straw in the rumen. Key Words: Microwave Radiation, Barley Straw, Rumen Degradability

Abstract

An experiment had been conducted to evaluate the effect of marine yeast and Saccharomyces cerevisiae (Sc) as probiotic supplement on poultry performance. Marine yeast isolated from rotten sea-weed and commercial Saccharomyces cerevisiae were used. Evaluation was conducted by comparing performance of broiler chicken supplemented with marine yeast or Sc, which were given through drinking water (5 ml/l) to negative control (feed without antibiotic growth promotor/GPA), positive control (feed with GPA), and reference commercial probiotic. Forty DOC broiler birds were used for each treatment, divided into 4 replicates (10 birds/replicate) and raised in wire cages for 5 weeks. Body weight and feed consumption were measured weekly and mortality was recorded during the trial. The results showed that there were no significant difference on the birds performance among marine yeast, Sc, positive control and probiotic reference control treatments. However their effects on bird performance were better (P<0.05) than treatment of negative control. It is concluded that marine yeast or Saccharomyces cerevisiae could replace the function of antibiotic as a growth promotant.   Key words : Marine yeast, Saccharomyces cerevisiae, probiotic, GPA, poultry

Abstract

Bluetongue virus is one of arbovirus diseases which is transmitted by insects, Culicoides spp. Insect collection wasconducted weekly from 1991 to 1993 and forthnightly from 1993 to 1997 in West Java using Pirbright-type miniature light trapin to phosphate buffered saline for identification and viral isolation. A total of 1155 pools of insects were inoculated intoembryonated chicken eggs before passaging in a mosquito cell line and three times blind passages in BHK-21 cells. Fourteenpools of insects produced cytopathic effect in BHK-21 cells. Four of the infected BHK-21 cells reacted in the antigen captureELISA test using a specific monoclonal antibody to bluetongue (BTV) virus. Further identification into serotype in ReferenceLaboratory, indicated that BTV serotype 1 was isolated from C. fulvus, BTV serotype 6 was isolated from C. peregrinus andBTV serotype 21 from pools of C. shortii and C. orientalis.Key words: Bluetongue viruses, isolation, insects

Abstract

The effects of planting density and cutting frequency on dry matter productivity were compared in two years following establishment among dwarf varieties (early-heading, DE and late-heading, DL) and normal varieties, Wruk wona (Wr) and Merkeron (Me), in the southern part of Kyushu, Japan. The planting densities examined for Wr, DE and DL were high (16 plants/m2, 25 cm × 25 cm of spacing), medium (8 plants/m2, 50 × 25 cm), and low (4 plants/m2, 50 cm × 50 cm), while for Me was only medium. The cutting frequency was three times with 60-day intervals in 2002 and two times with 90-day intervals in 2003. Irrespective of the planting densities, dwarf varieties were higher in tiller number, leaf area index and percentage of leaf blade (PLB) than those of normal varieties, but lower in plant height and total dry matter weight at all planting densities in both years. With the increase in planting density, annual herbage dry matter yield (HDMY) increased. The annual HDMY was higher in 2002 (cut twice) than that in 2003 (cut three times), and the difference in annual HDMY between the dwarf and normal varieties decreased by planting at high density and cut twice. Even though, the dry matter productivity was higher in the normal varieties than in the dwarf varieties at any planting density and cutting frequency, DL tended to show a stable productivity with high PLB irrespective of planting density and cutting frequency. Abstract

Most of duck husbandry in Indonesia is still run traditionally, herded in rice field or in the swampy area. This kind of husbandry seemed to be much preferred by farmers as they thought it was a simple and did not need high skill and high capital "IP padi 300" was a term of rice planting system tree times instead of twice in a year. This kind of changing might have significantly affected duck faming. The objective of the study was to observe the interactively effect of "IP padi 300" to duck husbandry at the same area. Two locations were choosen (Subang, West Jawa and Pemalang, Central Java) with 5 farmers at each location to be involved in the study. As many as 1200 laying pullet ducks were distributed to 10 farmers at two locations. The farmers were suggested to raise laying ducks with their own systems (fully intensive, semi intensive and fully herded) and were observed for 6 months. Biota was observed on both field and in the crop of the laying ducks. There was an interactive effect of "IP padi 300" and the duck farming on the same area. The availability of feed was increased on the "IP padi 300", which gave benefit to duck farming especially fue herded system, not to go far from owner's home base. Whilst the benefit to "IP padi 300" was assumed to the reduction of pest and desease, which was frequently attacked the rice field. The production of egg from herded duck was very fluctuative due to the movement and feed availability in the rice field. Field biota in Subang and Pemalang was very much the same in profile, although "golden snail" was only found in Subang. Ducks' crop content seemed to be very much similar with the profile of field biota, although rice grain was the most in the crop. Thus, it was found that field biota was not reduced by "IP padi 300", in fact it was rather increased.   Key words: IP 300, herded ducks, duck farming

Abstract

The mutation and genetic variation of avian influenza virus ussually associated with Hemmaglutinin (HA) and Neuraminidase (NA). The HA and NA protein are surface glycoproteins which have role for receptor binding site of the virus, determine virus subtype and genetic variation occured in those proteins. On the other site, the virus have the internal protein that posses  function for virus replication. This study analyzed the mutation on the the internal protein virus especially the Matrix (M1) and non structural (NS1) protein and its three dimensioanal structure of proteins. The methods used in this study were virus propagation, (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR sequencing of M1 and NS1 and using DS Modeller dan  DS Standalone from Discovery Studio for Modeling and Simulation to predict the three dimentional structure of the proteins. The result of this study showed that Indonesian AI H5N1 subtype had genetic variation internal protein and have no change on conserved of condition such as putative zinc finger and nuclear localization signal (NLS). Genetic variation that occured on PDZ motif of NS1 especially have human origin motif that might be correlated with virus adaptation on human. Abstract

This study was conducted at Laboratory Reproduction of Physiology, Research Institute of Animal Production. The experiment was factorial design with two kinds of cryoprotectant (DMF and DMA) as the first factor and two levels of them (7 and 9%) as the second factor. This study was invented to determine the effect of cryoprotectant and its level to survivability of drake semen. Sperm was collected from fifteen drakes two times per week using artivicial vagina. Only the best quality of sperm was used in this study. Collected sperm was diluted in medium to get concentration of 400 x 106 per ml. Equilibrated at 50C in mini straw (0.25 ml) for 60 minutes, then kept them up 8 cm above the LN2 for 4 minutes before plunged in LN2. Parameters measured of this study were survability of drake semen after diluted, at 50C and after freezing-thawing at 350C for 30 seconds. Result of this study showed that percentage of motility and percentage of live sperm were significant different (P<0.05) between DMA (33.24 and 42.03) and DMF (25.82 and 34.30). Level of cryoprotectant (7 and 9%) were not significant different. Based on this study, using DMA as cryoprotectan with 7% in medium was better than that of DMF.   Key words: Cryoprotectan, survivability, drake sperm

Abstract

Diacylglycerol acyltransferase 1 (DGAT1) gene is one of the major genes that has an important role in milk fat synthesis. This research was aimed at to identifying genetic polymorphism of the DGAT1 gene by PCR-RFLP method and its association to milk fatty acid components.  Animals studied were Holstein Friesian (HF) cattle from BBPTU Baturraden (123 cows) and BPPT SP Cikole (36 cows). The length of PCR product of the DGAT1gene was 411 bp. Genotyping resulted in two types of alleles, namely K (411 bp) and A (203 and 208 bp); and two genotypes, namely KK (411 bp) and AK (203, 208 and 411 bp). For both locations, genotype frequency of AK (0.75) was higher than KK (0.25). The allele frequency of K (0.64) was higher than A (0.36). Heterozygosity of HF cattles at both locations was relatively high (Ho>He). The DGAT1 gene of the observed HF cattle was polymorphic. Result showed that there was an association between the DGAT1 polymorphism with unsaturated fatty acids especially in nervonat acid. The AK cows had a significant effect on unsaturated fatty acid content of which having a higher nervonat content (0.05%) (P<0.05) than that of the KK cows (0.03%). From the results, it is concluded that the DGAT1 gene can be functioned as a marker of selection for milk fatty acids.

Abstract

The objectives of research were to find out physico-chemical characters and to detect flavor volatile compound of kefir-like. Material used was skim milk TS 9.5% which was heated at 85oC for 30 minutes and cooled at 22oC before innoculation of the starter. Microorganisms used were (a) Lactobacillus acidophilus P155110, (b) Lactobacillus delbrueckii subsp. Bulgaricus NCIMB 11778, (c) Lactococcus lactis P155610, (d) Leuconostoc mesenteroides subsp. dextranicum NCIMB 3350, (e) Acetobacter aceti P154810, (f) Bifidobacterium longum BF1, and (g) Saccharomyces cerevisiae P156252. The treatments consist of P1 = without (b); P2 = without (a); and P3= used (a) until (g). The physico-chemical characters identified were lactic acid and lactose percentages, pH, viscosity, organoleptic test for intensity of kefir-like sensory attributes. Results indicated that B. longum was potential bacterium use for starter combination on kefir-like making. The use starter P1 combination has high acidity and viscosity, low pH and lactose percentage, and high intensity on attribute creamy-white color, soft and curdle consistency, and kefir specific aroma on kefir-like. Volatile compound acid group were dominate by high acidity character on kefir-like resulted from starter P1 combination. Compound of 3-hydroxi-2-butanone (acetoin) was affecting butter-like of P3 character. This compound resulted from which is a character of fermented milk flavor was not detected on P1 kefir-like.     Key Words: Fermented Milk Kefir-Like, Volatile Compound, Physicochemical Character, Bifidobacterium longum
