Abstract

The study was conducted to investigate the clinico-pathological features of highly contagious disease occurred in chicken located in East and West Java during the outbreak in September-October 2003. Six farms located in Districts of Surabaya, Malang and Blitar of East Java had been visited. They were mainly commercial layer, breeder layer and breeder broiler, which the population was between 14.000, 80.000, and aged 17-70 weeks. Where as five farms in West Java (Districts of Bogor, Bekasi and their surrounding areas) were visited and consisted of commercial layer and breeder broiler, having population of 3000-16.000 and aged 11-53 weeks. Observation was made according to clinical, gross pathological and histopathological changes. Clinically, most of them had cyanotic wattle and comb and subcutaneous petechiation of non-feathered part of the legs. These were also seen in necropsy, accompanied by general circulatory disturbances in most organs: namely pectoral and thigh muscle, trachea, lungs, epicard, myocard, proventriculus, liver, kidney and ovary. In addition, the liver was congested, friable and necrotic in some parts. Histologically, hemorrhage and non suppurative inflammatory reaction were observed in the brain, skin (comb, wattle and non feathered leg), skeletal muscle, trachea, lung, heart, proventriculus, liver, kidney and ovary whereas vasculitis was found especially in the skin of the wattle and comb, brain and kidney. It is concluded that based on the clinicopathological findings the outbreak of poultry disease in East and West Java were attributed to highly pathogenic avian influenza.   Key words: Highly pathogenic avian influenza (HPAI), chicken, clinico-pathology, outbreak, East Java, West Java

Abstract

Susceptibility of ostriches (Struthio camelus) to the Indonesian velogenic strain of Newcastle disease virus (NDV) was evaluated by artificial infection . Twelve - 5 to 6 week old ostriches were divided into 3 groups each containing 4 birds . The first group was inoculated through respiratory system by dropping directly the virus solution into the nostrils, while the second group was inoculated through digestive system by dropping directly the virus solution into the oesophagus, with the dose of infection 106ELDSo (50%-embryo lethal dose) per bird . Meanwhile, the third group was treated as uninfected control . All infected birds developed antibody responses, but only two inoculated birds from the first group and two inoculated birds from the second group developed clinical signs of Newcastle disease (ND), with no specific pathological alterations . Infected birds, either sicks or healthy, excreted the challenge viruses through the respiratory system and still be detected up to the end of this experiment, ie . 15 days post-inoculation . The challenge viruses can be re-isolated from the brain, trachea, lungs, heart, liver, spleen, kidneys, small intestine, cecal-tonsil, and proventriculus of the infected birds . This study concludes that: (1) the ostriches are susceptible to the infection of the Indonesian velogenic strain ofNDV; (2) all infected birds developed immune responses, but only half of them develops el jtigi aj i disease ; (3) the infected birds excreted the challenge viruses for a considerable long time which may play role as the Mginiseti.ce ofinfectron the other healthy ostriches ; and (4) the challenge viruses can be re-isolated from various organs of the birds . .   Keywords : Newcastle disease vir4, 9strich, immune response, artificial infection

Abstract

The effect of supplementation of graded levels of Zn-bio-complex in concentrate on the growth of young sheep was studied. Fourty young Thin Tailled sheeps were devided into 4 groups of Zn-bio complex treatments. Zink-bio-complex was produced by IRIAP’s laboratory. King grass powder (CP = 10.4%) and concentrate (CP=15.5%) were given as a basal diet. King grass powder and drinking water were given ad libitum. The concentrate was given daily at 200 g h-1d-1 supplemented with 0 mg Zn kg-1 of DM (R1), 50 mg Zn kg-1 of DM (R2), 100 mg Zn kg-1 of DM (R3), and 200 mg Zn kg-1 of DM (R4). The initial live weight of sheep was 11.99 ± 0.17kg h-1. Live weight were measured fortnightly and feed consumption were measured daily. The experiment was carried out for 12 weeks with 4 weeks prior adaptation periods. Data obtained were analyzed statistically based on Completely Randomized Design. The results of experiment showed that the supplementation of Zn-bio-complex improved the live weight from 57.60 g h-1d-1 (R1) to 85.47, 72.14 and 67.86 g h-1d-1 with the feed conversion ratio improvement from 11.9 (R1) to 8.0, 9.6 and 10.5 for treatment R2, R3 and R4 respectively (P<0.05). The graded level of Zn-bio-complex did not affect daily feed intake which was 687, 686, 695 and 711 g h-1d-1 (P>0.05) for R1, R2, R3 and R4, respectively. It can be concluded that the supplementation of Zn-bio-complex at the level of 50 mg Zn kg-1 in daily concentrate resulted in the best response on live weight gain and feed conversion ratio of young sheep. Abstract

This study is to investigate the residue level of TCDDs/TCDFs in beef collected from Animal Slaughter Houses in Giwangan (Yogyakarta), Klaten (Central Java) and Kupang (East Nusatenggara). Residue of persistent organic pollutants (POPs) was also analysed with GC-ECD, and dioxins with GC-MSMS. Fifty samples were collected form Giwangan (20), Klaten (15) and Kupang (15). Results shows that POPs were detected in beef samples including aldrin, dieldrin, DDT, endrin and heptachlor. The highest total residues of POPs were detected in beef from Giwangan (Yogyakarta) at 93.11 hg/g, followed by Klaten (17.79 hg/g) and Kupang (12.87 hg/g). Residues of POPs were below the maximum residue limit stated by the SNI 7313: 2008. Average of total TEQ for TCDDs/TCDFs in beef from Giwangan was 13,624.38 rg/g with a range between 4,496.66 to 20,642.40 rg/g was higher than that from Kupang at 1,623.98 rg/g with a range between 0.83 to 6,471.07 rg/g. On the other hand, dioxins were not detected in beef from Klaten. The total TEQ of TCDDs/TCDFs in beef samples from Giwangan was 54,497.52 rg/g and Kupang was 6,495.9 rg/g. These results indicated that there was an association between the presence of endrin and heptachlor with the presence of TCDDs/TCDFs residues in beef that this is may be new information for further studied.

Abstract

An outbreak of avian influenza in Indonesia was reported at the first time at the beginning of September 2003 causing high mortality among poultry population especially commercial layer chicken farms in Java, Sumatra and Bali islands. From the outbreaks highly pathogenic avian infuenza viruses have been isolated and characterized by rapid, HA, HI and AGP tests. However, these isolates are still needed to be further molecularly characterized. The aim of this study is to identify by further subtyping the avian viruses by means of RT-PCR using Matrix, H7 and H5 primers. The study reveals that the RT-PCR using Matrix primer amplified a 200-300 basepairs (bp) Jawa Timur isolates were collected from East Java, while Jawa Barat isolates were from West Java. The RT-PCR using H7 primers did not amplify any product, while H5 primer amplified a 500-600 bp product from the isolates. It is concluded that the outbreak of poultry disease in East and West Java was caused by an avian influenza H5 subtype.   Key words: Identification, avian influenza virus, RT-PCR, H5 subtype

Abstract

A series of experiment was conducted to improve the utilization of coconut meal in poultry ration . A bioprocess (fermentation) approach was adopted. Coconut meal was fermented followed by an enzymatic process at room temperature or 50 OC.The nutritive value of the fermented products was determined chemically. Product with the highest nutritive value was then used for a feeding trial to study the interaction effect between dietary levels of fermented coconut meal and phosphorous level on the performances of laying ducks. Nine experimental diets were formulated, i.e ., the factorial of 3 levels of fermented coconut meal (0, 30 and 40%) and 3 levels of total phosphorous (0.6, 0.8 and 1 .0%) . The diet was fed to pullet ducks and the performances of the ducks were observed for 16 weeks of production period. The results showed that the nutritive value of coconut meal increased by fermentation and the highest yield was achieved when the enzymatic process was performed at 500C. There was no significant interaction effect between dietary fermented coconut meal levels and the phosphorous levels on all parameters observed, except on egg weight. Inclusion of fermented coconut meal in the ration up to 30% did not produce negative effects on the productivity oflaying ducks, however, higher levels (40%) ofinclusion reduced the egg size   Keywords: Coconut meal, fermentation, ducks, egg production

Abstract

In order to determine grazing characteristics in the dwarf Elephantgrass pasture under the rotational grazing system, 5 a of pasture established by rooted tillers of dwarf variety of late-heading type (DL) was grazed by 3 head of breeding beef cows in the following 2 years after establishment in 2003 and 2004. Grazing system was conducted at 4 and 5 times for about 1-week grazing with 3-weeks and 1-month rest period in 2003 and 2004, respectively. Tiller number increased seasonally from the first through the fourth or fifth grazing, and herbage dry matter weight reached the peak at the fourth grazing both years, while some characters such as plant height, mean tiller dry matter weight and percentage of leaf blade tended to decrease with the grazing. From the stratified clipping method, percentage leaf blades in the top two, third and in the fourth strata were consumed perfectly and almost perfectly, respectively both years. Herbage consumption increased with the grazing up to the third and fourth grazing and dropped at the last grazing due to the poor regrowth under the decreasing air temperature both years. Dry matter intake per head averaged 4.3–13.4 kg DM head-1 day-1 in 2003 and 6.7-11.4 kg DM head-1 day-1 (15.5-28.6 g DM/kg LW-1 day-1) in 2004. Grazing time on DL elephantgrass was stable at 50–60% in daytime at the first grazing day both years, while that at the last grazing day tended to increase with the grazing both years. Thus, live weights of 3 head of beef cows were maintained in the grazing from the second to the fourth time without concentrate feeding in 2004, when dry matter intake was above 20 g DM  LW-1 day-1. Abstract

In farm animals, vocal analysis is accepted as a non-invasive method for assessing animal welfare in comparison to most physiological measurements. The aim of this study was to investigate the influence of different degrees of social isolation on the acoustic characteristics of stress-induced bleats from three different breeds of sheep. The experiment consisted of subjecting the animals to three different levels of social isolation during which both locomotive and vocal data were recorded. The data on locomotive behaviour was subjected to descriptive analysis while specialized acoustic software was used to measure a total of thirty-six parameters of recorded bleats. Descriptive analysis showed that the animals displayed more locomotive activity during partial isolation compared to complete isolation. Number of bleats during partial isolation was also found to be higher. The application of two-way analysis of variance showed a significant effect of isolation level and breed on both temporal and structural acoustic properties. Amplitude, power and time acoustic properties were found to affect acoustic quality of vocal responses to isolation, whereas frequency related properties were also found to be different significantly (P < 0.05) between breeds. From spectrogram analysis, the patterns of energy distribution within the calls proved to offer the most evident that differ between isolation levels and breeds. It was concluded that acoustic analysis can be helpful in revealing the affective state of socially isolated rams. Key Words: Sheep, Social Isolation, Vocalization, Behaviour, Animal Welfare

Abstract

This study was aimed at determining the interaction between drought stress and defoliation interval on the productivity of Indigofera sp. This study was done based on Complete Randomized Design (CRD) 3x2 factorial, the first factor was without drought stress (100% FC), medium drought stress (50% FC), and severe drought stress (25% FC), and the second factor was the defoliation interval of 60 day and 90 days, each treatment was repeated 5 times. Observed variables were biomass production, and leaf/stem ratio. Results showed that there were significant interactions (P < 0.05) between drought stress and defoliations interval on the productivity of Indigofera sp. The highest biomass production of Indigofera sp. (424.47 g/plant) was obtained from treatment of wihtout dought stress (100% FC) combined with defoliations interval of 90 days, while the lowest (184.55 g/plant) was reached at treatment of severe drought stress (25% FC) and depoliations interval of 60 days. The highest leaf/stem ratio (1.47) was reahced at treatment severe drouht stress (25% FC) and depoliations interval of 60 days and the lowest (1.05) was reached at combination treatment of drought stress and depoliations interval of 90 days. Abstract

This paper represents an attempt to examine gender relations in goat-farm households in Central Java . Guided by theory of farm household in which labor for household activities is supplied and organized based on gender and age differences of the household members . The study was conducted at Klirong village, sub-district Kebumen, Central Java in May 1995 . Twenty-four families who owned goats were chosen and divided into two groups based on the ownership to land . Participant observation method were used to collect information on the time spent for household maintenance activities, agricultural activities and on/off farm activities by household members . The results indicated that gender appears to be the primary determinant of labor and time allocation in two types of farm households . Since goat production is secondary economic activities for both type of households, therefore, the responsibility to take care of was allocated to the children, especially male children, while adults -- man and woman -- engage in the primary economic activities such as agricultural activities and selling labor for on/off farm activities .
