Abstract

The aim of this study was to evaluate genetic analysis including heritability and further phenotypicand genetic trends of reproductive traits in Bali cattle. Reproductive traits studied were age at firstcalving (AFC), calving interval (CI) and pregnancy rate (PR). Data of reproductive traits were collectedfrom Breeding Centre of Bali Cattle, Denpasar-Bali at the year period of 2000-2007. To evaluate thegenetic analysis, heritability were estimated using the Mixed Model Least Squares and MaximumLikelihood procedure. The phenotypic and genetic trends were calculated using regression equation.Estimation of heritability for AFC, CI and PR were 0.22, 0.41 and 0.40, respectively. The phenotypictrend of AFC, CI and PR decreased at an average rate of 1.70 month, 10.4 days and 0.75% per year,respectively. The same pattern was showed for genetic trends for AFC, CI and PR decreased at 0.38month, 4.25 days and 0.30% per year respectively in the same period. The heritability of reproductiontraits in Bali cattle were considered as moderate to high (0.22-0.41) which means that the selectionprogram will be more effective and efficient in improving the genetic merits in Bali cattle.

Abstract

This study was aimed to analyze factors and models of business sustainability of smallholder layer farms in Kendal Regency, Indonesia. Purposive sampling method was used to select 120 active layer chicken farmers in Sukorejo, Pageruyung, Patean, Limbangan, and Boja Districts as research respondents with the ownership of 300 to 10,000 birds. Research variables consisted of three external variables (social, economic, and institutional) and two internal variables (income and business sustainability). Closed questionnaires were used to collect data. The data were analyzed descriptively using Structural Equation Modeling (SEM) by AMOS 21 software. The results showed that the exogenous and endogenous variables met the modeling criteria with Chi square value = 160.764; probability = 0.380; CMIN / DF = 1.031; GFI = 0.892; AGFI = 0.841; TLI = 0.998; CFI = 0.998; RMSEA = 0.016. Social, economic, institutional and income variables had a significant and positive influence (P≤0.05) on business sustainability. The constructed model had a strong and positive relationship, so it could illustrate the sustainability model of layer business in Kendal Regency.

Abstract

This study aimed to investigate the effect of decocted turmeric on performance, haematological parameters and carcass traits of broilers. Broiler chicks at 11 days of age were allotted to five groups, including T0 (birds drinking 100% water), T1 (25% decocted turmeric + 75% water), T2 (50% decocted turmeric + 50% water), T3 (75% decocted turmeric + 25% water) and T4 (100% decocted turmeric). The study was arranged based on a completely randomized design (CRD), parameters investigated was performance and blood chemistry (leukocyte, total protein, albumin, globulin, uric acid, glucose). The data were subjected to analysis of variance at 5% probability. Decocted turmeric did not affect weight gain, feed and water consumption and feed conversion ratio of broilers. Lymphocytes proportion was higher (P<0.05) in T1, T2 and T4 than in T0 and T3 birds. The heterophils to lymphocytes ratio was lower (P<0.05) in birds receiving decocted turmeric than control. Globulin concentration and albumin to globulin ratio were higher and lower (P<0.05), respectively, in serum of T2, T3 and T4 than T0 and T1 birds. The proportion of heterophils and concentration of serum total protein, albumin, uric acid and glucose were not different among treatments. The weight of breast meat was higher and lower (P<0.05) in T3 and T4 birds, respectively, compared to the control. In conclusion, providing decocted turmeric through drinking water helped to improve stress responses and increased the weight of breast meat of broiler chickens.

Abstract

The aims of this study were to estimate the heritabilities, examine the effects of dairy femaleselection and calculate the genetic gains on milk yield and milk composition in Baturraden Dairy CattleBreeding and Forage Centre (Balai Besar Perbibitan Ternak Unggul dan Hijauan Pakan Ternak /BBPTU–HPT Baturraden), Indonesia. The first lactation records of 221 dairy cows from 2006 to 2014were used. Heritabilities were estimated by paternal half-sib correlation. Comparison of averageperformances between daughter population (Ā) and initial dam population before selection ( ) wereconducted by Z-test. Annual genetic gain was calculated as genetic gain per generation (the differencesbetween Ā dan ) divided by generation interval. Heritabilities for milk fat percentage (FP), milk fatyield (FY), milk protein percentage (PP) and milk protein yield (PY) were 0.46, 0.30, 0.28 and 0.17,respectively. A significant increase (P=0.025) in the total milk yield (TMY) from the first generation(G1) to the second generation (G2) resulted in a high significant decrease in the FP (P=0.004). Geneticgains of TMY, FP and PP were 9.76 kg, -0.04% and -0.01% per year, respectively. It is concluded thatselection for higher TMY only negatively affect FP and PP. Selection can be applied based on FY to avoid the decrease of FP. Negative effects of genetic-environmental interaction resulted in slowergenetic gain because the imported cows needed time to adapt to the local environment.

Abstract

Fish waste (“ikan rucah”) is part of discarded fishing product, which is composed by non-foodcategorized fish (NFC-fish). Quality of NFC-fish meal that was made by dipping in extract of fermentedvegetable’s waste has been reported better than commercial fish meal, but the effect after storingremained in question. Experiment was conducted to study the effect of different time of storing onphysic-organoleptical, chemical and microbiological characteristics of the acidified NFC-fish meal thatwas made by dipped in extract of fermented vegetable’s waste (FVW-exctract). The NFC-Fish wassoaked in the FVW-exctract by the ratio 1:1 (w/v) for 4 hours, and then it was dripped out and dried. Thedried fish was ground passed through 20 mesh, kept into plastic bags and stored at the room temperature(23-25 °C and 70-80% RH) for 0, 1, 2 and 4 months. Physic-organoleptical, chemical and biologycalcharacteristics were parameters observed. Experiment was conducted by completely randomized design(CRD). Data were analysed by the GLM of SAS. Actual number of moisture, crude protein, extractether, and proteolytic bacteria of fish meal were significantly influenced by time of storing (P<0.05).Dipping NFC-fish in the FVW-exctract effectively maintain the physical characteristics, pH, moisture,crude protein, extract ether and the number of proteolytic bacteria of fish meal up to 2 months ofstorage. Dipping NFC-fish in the FVW-exctract provide better characteristics on physical, chemical andmicrobiologycal than the commercial fish meal at the same condition of storage.

Abstract

This study aimed to examine the effect of turmeric and garlic inclusion to fermented Sauropus androgynus-bay leaves (FSBL) containing diet on fat deposition, and chemical composition of broiler meats. Two hundred 15-day-old female broilers were distributed into 5 treatment groups with 4 replications as follows. Control feeds with commercial feed additive (T0); Feeds with 1.25% FSBL (T1); Feed with FSBL plus 1 g turmeric (T2); Feed with FSBL plus 2 g garlic (T3); Feed with FSBL plus 1 g of turmeric and 2 g garlic (T4). It was shown that the fat deposition in the abdomen was significantly affected (P<0.01). Furthermore, the inclusion of turmeric and garlic to FSBL containing diet did not affect fat, protein, moisture and ash contents, but increased arginine, methionine and histidine (P<0.01). The addition of turmeric and/or garlic to the FSBL containing diet significantly influenced lauric acid, myristic acid, linolenic acid, cis-11-eicosenoic acid, cis-4,7,10,13,16,19-docosahexaenoic acid (P<0.05), n-3 fatty acid and n-6/n-3 fatty acid ratio (P<0.01). In conclusion, turmeric plus garlic supplementation improved amino acid profiles and changed fatty acid profiles. The possible interaction should be considered when we formulated feed additive from medicinal plant mixtures

Abstract

Stapphylococcus sp., is bacteria that caused subclinical mastitis in Ettawa Grade (EG) goat. Thepurpose of this study was to determine virulance factor Stapphylococcus sp., which was isolated fromsubclinical mastitis EG goat’s milk in Sleman regency, Yogyakarta. A total of 7 isolate Stapphylococcussp., were isolated from subclinical mastitis EG goat’s milk were determinated by several virulancefactors such as haemolysin, clumping factor, and coagulase. Haemolysin was determinated by culture inblood agar plate and incubated in the temperature of 37°C for 24 hours. Clumping factor wasdeterminated by mixing the rabbit plasma with Stapphylococcus sp., in the glass objects. Coagulase wasdeterminated by mixing the rabbit plasma and broth culture of Stapphylococcus sp. After incubated inthe temperature of 37°C for 24 hours in tube, then the gel formation was observed. Haemolytic type ßwas yielded from 5 isolate Stapphylococcus sp., whereas 2 isolates were not haemolytic. Clumpingfactor and coagulase were produced from 2 isolate Stapphylococcus sp. This study showed that not all ofStapphylococcus sp., isolate causing subclinical mastitis in EG goat have virulance factor.

Abstract

MC4R gene is known as an important candidate gene for the growth trait. The purpose of this research was to identify the MC4R gene in Kebumen Ongole grade cattle and examine its association with growth traits. Data of birth weight (BW), weaning weight (WW), birth body length (BBL), birth chest circumference (BCC), birth shoulder height (BSH), weaning body length (WBL), weaning chest circumference(WCC), weaning shoulder height (WSH) and average daily gain (ADG) were collected and used for analysis of MC4R gene. Sixty blood samples were collected for DNA isolation and PCR amplification. The single nucleotide polymorphisms (SNP) g.1133 C>G was used for genotyping by using PCR-RFLP methods. The frequenciy of G allele (0.59) was greater than C allele (0.41). The highest genotype frequencies have been detected in CG heterozygote animals (0.52) followed by GG (0.33) and CC (0.15) in homozygote animals. The results of Pearson ‘s Chi-square test indicated that the population was not deviate (P>0.05) from the Hardy-Weinberg equilibrium (HWE). The SNP g. 1133 C>G of MC4R gene indicated affecting high birth body length with GG genotype (P<0.05). In conclusion, the SNP g. 1133 C>G may can be a marker for birth body length of calf selection.

Abstract

This research was conducted to study the chemical and microbiogical properties of goat milk kefir stored under different temperatures and storage time. A completely randomized design, factorial pattern 3 x 3 was used in this study. The first factor was storage temperature (-1 to -5; 1 to 5 and 6 to 10oC) and the second factor was storage time (10; 20 and 30 days). Each treatment has three replicates. Variables observed included pH, water activity (aw), total lactic acid bacteria (LAB), and total yeast. Data were subject to analysis of variance and Duncan’s multiple range test. Results showed that storage time and temperature had significant effects on pH. The lowest pH of Kefir was obtained by storing it for 10 days at 6 to 10oC. Titratable acidity was significantly affected by temperature, and kefir stored at 6 to 10oC has the highest titratable acidity. Storage time and temperature had no significant effects on water activity, and the average water activity of kefir was 0.875±0.028. Total LAB and total yeast were significantly affected by temperature, but not by storage time. In average, total LAB and total yeast in kefir were 7.17± 0.92 log cfu/ml and 6.76± 0.39 log cfu/ml, respectively. In conclusion, this study confirmed that temperature of storage has a major contribution to the characteristics of kefir made from goat milk; hence it has to be considered when handling kefir for a longer period of time.

Abstract

Growth hormone (GH) is an anabolic hormone which sintesized and secreted by somatrotop cell inpituitary anterior lobe. GH exert its effect on growth and metabolism by interacting with a specificreceptor on the surface of the target cells. Growth hormone receptor (GHR) has been suggested ascandidate gene for traits related to meat production in Bovidae. The objectives of this study were toidentify polymorphism of GH and GHR genes in buffalo. The 452 DNA samples buffalo were collectedfrom five populations in Indonesia (Siborong-Borong-Medan (65), Lebak-Banten (29), Pandeglang-Banten (180), Semarang-Central Java, and Mataram-West Nusa Tenggara (103)). A gene fragment of theGH|AluI gene at 432 bp located on exon 3 and GHR|AluI gene at 298 bp on exon 10 were successfullyamplified by using the techniques of a PCR (polymerase chain reaction) and genotyped by PCR-RFLP(restriction fragment length polymorphism) then -SSCP (single strand conformation polymorphism). Theresults showed no polymorphisms were detected in these genes. All buffaloes tested had LL genotype forlocus GH|AluI and AA genotype for locus GHR|AluI.
