Abstract

Research has been carried out 2,2-diisopropoksipropan catalytic synthesis of isopropanol to increase the octane number of renewable fuels. Catalytic Synthesis using Cu catalyst that falls on activated carbon (Cu / AC) for use as a process of dehydration of isopropanol to 2.2-diisopropoksipropan. This study also aims to obtain the optimum time and temperature for the dehydration of isopropanol. Preparation of carbon carried by softening coconut shell charcoal to 60-80 mesh size, then heated at a temperature of 800 ° C, washed, dried, impregnated with CuCl2 and reduced at 400 ° C with flowing hydrogen gas. The amount of metal content of Fe, Ca, and K in the carbon after the process of washing / soaking determined using Atomic Absorption Spectroscopy (AAS). Activated carbon and catalyst acidity was determined by adsorption of ammonia, isopropanol dehydration process with catalyst Cu / activated carbon (Cu / AC) carried out in an electric furnace with temperature variation: 250; 300; 350; 400; and 450 ° C and the variation in the amount of catalyst amount: 6; 8; and 10 grams, with N₂ flowed at a rate of 20 mL / min as the carrier gas, and no water vapor. Dehydration results in vapor cooled condenser and then collected for analysis by gas liquid chromatography (GC) and gas chromatography mass spectrometry (GC-MS). The results of the analysis using AAS showed that the presence of washing, soaking using 3 M HCl, the metal Fe, Ca, and K in the activated carbon is significantly reduced. The results of ammonia adsorption analysis showed that activated carbon and Cu / AC has acidity, respectively for 2.962 and 4.911 mmol / g. Abstract

Tomcat (Paederus sp.) venom can bring a condition called dermaconecrosis. Initially, paederin toxins was thought that come from the body of Tomcat (Paederus sp.). However, based on Piel’s research, the genes come from biosynthesis of tomcat (Paederus sp.) venom is more approach to Pseudomonas aeruginosa genes than body itself. In other fact, Pseudomonas aeruginosa can secrete enzymes metalloprotease (MMP), especially extracellular MMP-2 and MMP-9. MMP-2 and MMP-9 are gelatinase that can destroy the gelatin on human skin. There has been no previous report about the treatment of dermatitis paederus based on the mechanism of action except for the use of corticosteroids as symptomatic drugs. Because of that, researcher hypotheses that MMP-2 and MMP-9 have a role on dermatitis paederus and doxycycline can inhibit that process as MMP inhibitor and antibiotics component. The research design is quasi-experimental research, in vitro with a post-test only control group design. The research steps are Tomcat (Paederus sp.) venom extraction, doxycycline solution preparation, and electrophoresis of gelatin zymografi. The independent variables are induction of Tomcat (paederin sp.) venom and the doses of doxycycline, while the dependent variable is the density of the gel. Gel density was measured using densitometry software ImageJ and calibrated as optical density. This study describes the first time the mechanism Paederus dermatitis caused by MMP-2 and MMP-9, so that the first line of therapy that can be used is doxycycline with an effective dose of 10 mM in vitro. Keywords:  paederus dermatitis, doxycycline, gelatin zimografi, MMP

Abstract

Poverty is a major cause of a number of social problems, political, economic and particularly in developing countries. Poverty reduction is the key factor that must be done to achieve the targets in the Millennium Development Goals (MDGs) by 2015 government assistance program for the reduction of poverty that exist today have not been able to reduce the number of poor households in Jombang, whereas Jombang have a positive economic growth rate from year to year. Thus, in order to support effective and efficient government then carried out a classification based on the household assistance package that is expected by the poor households in Jombang. Poor households as the object of observation is divided into two according to their poverty status, namely Poor Households (RTM) and Chronically Poor Households (RTSM). Classification is done by using the CART approach is then combined with the method of the Random Forest to obtain a more accurate and stable classification. Classification results using Random Forest CART stated that the expected assistance package by RTSM determined by income per month, place of dispocing of fecal water, type of floor building, and source of drink water. While, the most important characteristic of RTM classification income per month, place of dispocing of fecal water, source of drink water, and large of house building. The classification result from Random Forest CART method produces a total accuracy rate of 99.50 percent for RTSM and 98.33 percent for RTM. The mapping of households in Jombang stated that cash money, cattle, and modal were the most expexted assistance package. Keywords: CART, Chronically Poor Households, Poor Households, Random Forests, Total Accuracy Rate.

Abstract

Cervical cancer is the third most diagnosed cancer and the fourth leading cause of cancer death in females in the world. Zebrafish have been used as experimental animals in human tumors because of similar gene expression (70%) than other experimental animals. Yogurt LBA-ST produces SCFA metabolites (acetate, lactate, and butyrate) that can provide anticarcinogenic effects. There has been a vast growth of related research focusing on proliferation and apoptosis without clear mechanism. However, Important concern need to considered in the development of tumors is protein-coding invasion, migration, and adhesion (IMA) of tumor cells. This study aims at investigating (1) Antiproliferation activities of supernatan yogurt LBA-ST (SY LBA-ST) on zebrafish embryos and HeLa cells, and (2) anti-IMA’s pathway on HeLa cells. The anti-cancer property of SY LBA-ST was evaluated by MTT assay for viability and imunositochemistry of MMP-2, Laminin5-y2, Hsp27, and TGF-β for anti-IMA mechanism. The data is collected and analyzed using One-Way ANOVA.This study could be concluded that (1) SY LBA-ST increase mortality rate and decreased the hatching rate in zebrafish embryos, LC50 ≈ 20% (v/v),  (2) supernatant, pellet, and whole yogurt shows antiproliferation activity on HeLa cells, (3) IC50 SY LBA-ST ≈ 30% (v/v) and the ED = 10% (v/v), and (4) the anti-IMA’s pathway was via decrease MMP-2, Laminin5-y2, Hsp27, and TGF-β expression (p<0.05 ± 0.005).Keywords:  lactobacillus bulgaricus-acidophilus, streptococcus thermophillus, cancer, HeLa, zebrafish
