Abstract

Gibberellin is plant growth regulator that stimulates cell elongation, seed germination, flowering, and fruit ripening. This study was conducted to isolate, identify, and optimize growth media for gibberellins producing- rhizobacteria isolated from rhizosphere soil of “keruing“ (Dipterocarpus sp.) tree in forest research Carita, Pandeglang, Banten. Eight bacterial isolates were obtained and all produced gibberellin. The BC2 isolate produced the highest of gibberellin (0.897 mg mL-1) and then selected for identification based on physiology, molecular character, and effects of growth media with variation of temperature, pH, and light. The result of physiological test indicated that BC2 isolate does not produce indole, positive on urease and oxidative carbohydrate. The phylogenetic analysis showed that BC2 isolate is belonged to Stenotrophomonas maltophilia with 98% similarity level. The optimation of growth media indicated that the growth of BC2 isolate was optimum at 30°C, pH 7, and dark condition.

Abstract

An extensive study about egg parasitoid biodiversity in different plant growth stages is an important need. This study was aimed to determine the diversity and abundance of egg parasitoids Nilaparvata lugens on different rice growth stages in Saketi, Pandeglang, Banten. The parasitoids sample was collected three times with two days interval sampling in three villages, i.e. Sodong (location I), Sindanghayu (location II), and Sukalangu (location III) from February to September 2018. Ten plants (as bait: rice that infected by N. lugens eggs) were put during two days along the transect line (100 meters) with 10 m space in between. The findings revealed that the abundance richness (S), the diversity index (H'), and similarity (J') of egg parasitoids of N. lugens were higher in the location I than those in the locations II and III. The study indicated that the diversity and abundance of egg parasitoids of N. lugens decreased in the generative phase of rice growth.

Abstract

Pseudomonas has the potential ability for production of citrate synthase synthesis. Pseudomonas aeruginosa could synthesize the enzyme of citrate synthase which is most likely compatible with microalgae cell. Pseudomonas aerugenosa can be found in the rhizosphere of Kruing (Dipterocarpus sp., Dipterocarpaceae). This bacteria is commonly used in agriculture purposes because it is able to synthesize organic acid such as citric acid. These organic acids are synthesized from a reaction between oxaloacetate and acetyl CoA, catalyzed by citrate synthase (CS) in the tricarboxylic acid cycle (TCA). Rhizosphere as microbial sources was obtained from Kruing (Dipterocarpus sp.), which was collected from ‘Carita’ Research Forest, Pandeglang, Banten, West Java. Citrate synthase gene-specific primers were designed based on citrate synthase gene sequences as depicted in Genbank. The isolation and amplification showed that citrate synthase can be detected and purified from Pseudomonas aeruginosa target and it consists of 1600 bp and encodes 509 amino acids. Based on BLAST (Basic Local Alignment Search Tool) analysis, CS genes that were successfully isolated had 92 % similarity with Pseudomonas aeruginosa type II citrate synthase. This CS gene is expected to be expressed in microalgae metabolism to divert the metabolism of carbohydrate formation into fatty acids.

Abstract

Tanaman transgenik adalah tanaman hasil rekayasa gen yang dibuat dengan cara disisipi satu atau sejumlah gen dari organisme lain, dengan tujuan untuk diperoleh sifat baru yang unggul dan diinginkan, misalnya resisten terhadap cekaman kekeringan, resisten terhadap hama, resisten terhadap herbisida. Produk teknologi DNA dikenal dengan Produk Rekayasa genetika (PRG), sebagai contoh adalah kedelai transgenik Tujuan penelitian ini untuk menganalisis potensi budidaya tanaman transgenik di Indonesia. Metode yang digunakan dalam studi ini adalah tinjauan pustaka (review) dengan menggali data dan informasi yang dikumpulkan dari berbagai sumber meliputi: undang-undang, peraturan pemerintah, surat keputusan, artikel dalam jurnal, laporan hasil penelitian. Hasil penelitian ini adalah aplikasi tanaman transgenik dilapangan perlu manajemen yang dapat mengatur pola tanam tanaman transgenik dan non transgenik, penyediaan bibit tanaman transgenik dikendalikan dalam bentuk paket kemasan, dan perlunya kepastian hukum dan dukungan pemerintah terhadap produk transgenik.

Abstract

Biodeversitas dan klasifikasi jenis laba-laba dapat menunjang ekowisata di Kabupaten Pandeglang, Provinsi Banten. Tujuan dari penelitian adalah untuk mengetahui jumlah jenis laba-laba (Araneae) di kawasan desa wisata Banyubiru. Pengumpulan sampel laba-laba dilakukan dengan metode ekplorasi. Analisis data keanekaragaman diuraikan secara deskriptif. Hasl penelitian menunjukkan bahwa keanekaragaman laba-laba Arachnida (Araneae) pada Desa Wisata Banyubiru Kecamatan Labuan Kabupaten Pandeglang diperoleh sebanyak 70 individu yang termasuk dalam 5 famili dan 10 spesies yaitu: A. argentata (Araneidae), Gasteracantha sp (Araneidae), A. apensa (Araneidae), A. trifasciata (Araneidae), W. saltabundus (Theridiidae), N. inaurata (Nephilidae), N. maculata (Nephilidae), L. reclusa (Sicaridae), P. paykulli (Salticidae) dan Hasarius sp (Salticidae). Keanekaragaman laba-laba Arachnida (Araneae) dari tiga stasiun di Desa Wisata Banyubiru Kecamatan Labuan Kabupaten Pandeglang menunjukkan indeks keragaman (H) berkategori sedang, indeks kemerataan tinggi dan indeks dominansi yang kecil. Ekosistem desa wisata masih lestari

Abstract

Mikroalgae dapat berperan sebagai bioindikator pencemaran air dan mempunyai banyak manfaat antara lain: sebagai komponen pangan, bahan obat, dan kosmetik. Tujuan penelitian ini untuk mengetahui kelimpahan spesies miroalgae di Situ Cibanten Ciomas Kabupaten Serang. Penelitian menggunakan metode purpossive sampling, sampel diambil pada pagi hari jam 08.00-12.00. Sampel disaring menggunakan plankton net dengan ukuran mesh 50 µm. Sampel kemudian ditambahkan dengan formalin 4 % dan diamati dengan menggunakan mikroskop di Laboratorium Terpadu Fakultas Sains, Farmasi, dan Kesehatan Universitas Mathla’ul Anwar Banten. Indeks diversitas tertinggi didapatkan di lokasi satasiun 1 dengan nilai 3.11. dan terendah pada stasiun 2 sebesar 2.360. Indek Kemerataan tertinggi pada stasiun 1 sebesar 0.978 dan terendah pada stasiun 2 sebesar 0.803. Indeks dominasi dari masing-masing spesies mendekati nilai 0, yang berarti nilai dominansinya rendah. Indeks kelimpahan relatif ditemukan pada spesies Spirogyrasp is of 17.7%, dan yang terendah adalah spesies Phacuslongicaida and Lemaneaannulata 0.4%. Situ Cibanten mengalami pencemaran sedang

Abstract

Hewan ternak mamalia seperti: sapi, kambing, kerbau dan kelompok unggas seperti: ayam dan bebek memiliki peran penting, salah satunya untuk kebutuhan pangan. Tujuan  penelitian ini adalah untuk mengetahuidan mengidentifikasi telur cacing parasit pada feses hewan ternak Di Propinsi Banten. Metode yang digunakan dalam penelitian ini  adalah metode flotasi dan sedimentasi dengan menggunakan sampel feses hewan ternak (sapi, domba dan kerbau). Hasil penelitian menunjukan bahwa hewan ternak di Provinsi Banten terinfeksi Nematoda, Trematoda dan Cestoda pada fesesnya. Telur cacing parasit yang ditemukan sebanyak 9 jenis, yaitu: Haemochus, Trichostronylus, Toxocara, Cooperia,Trichiuris trichiura, Strongyloid sp.,Moniezia sp.,Fasciola sp.dan Paramphistomum sp. Prevalensi tertinggi  dan intensitas tertinggi dari jenis Haemochus. Infeksi pada ternak juga dapat terjadi secara tunggal atau campuran (terdiri atas dua atau lebih cacing parasit). Prevalensi tertinggi adalah infeksi tunggal Nematoda 56,25% dan infeksi campuran (kombinasi Trematoda dan Nematoda) sebanyak 28,75%. Tingkat prevalensi dan intensitas telur cacing parasit di hewan ternak di Provinsi Banten tergolong rendah.

Abstract

Studi tentang ekstraksi gelatin dari tulang ikan kakap merah dilakukan melalui perendamandalam asam klorida dengan konsentrasi 4, 5, dan 6% selama 48 jam. Tulang ikan kemudiandicuci dengan air mengalir sampai pH netral (6–7) dan diekstraksi dengan akuades pada suhu 80, 85, dan 90 oC selama 6 jam. Ekstrak gelatin yang dihasilkan disaring dengan kain saring, dan filtratnya dikeringkan dalam oven pada suhu 55 oC selama ± 48 jam sehingga diperoleh lembaran gelatin. Hasil ekstraksi gelatin ini kemudian diuji kadar rendemen, viskositas, pH, kekuatan gel dan karakternya secara organoleptik. Hasil analisis menunjukkan bahwa perendaman dalam larutan HCl 4% selama 48 jam dan ekstraksi pada suhu 80ºC selama 6 jam menghasilkan gelatin terbaik dengan rendemen 7,4%, viskositas 6,73 cPs, pH 5,05, kekuatan gel 226,8 g/bloom, dengan jumlah mikroba 2,5 x 10 3 koloni/g dan E. colinegatif. Hasil uji organoleptik terhadap warna gelatin yang dihasilkan tidak berbeda nyata dengan gelatin komersial, yaitu antara coklat kekuningan sampai kuning keputihan. Akan tetapi gelatin tulang ikan kakap merah sedikit berbau ikan sedangkan gelatin komersial tidak berbau (netral).

Abstract

Caries and dental plaque are oral and dental problems. Green spinach (Amaranthus hybridus L.) can be used as medicine and has antibacterial activity. It also benefits human health and is a nutrition source. The phytochemical results of the leaf extract showed that A. hybridus L. contains alkaloids, flavonoids, steroids, saponins, and tannins. This study aimed to determine the inhibitory action of A. hybridus L. leaf extract against the growth of Streptococcus mutans and to formulate A. hybridus L. leaf extract in a mouthwash. The disk diffusion method and brain heart infusion agar medium were used in this study, with five treatment groups: extract concentrations of 25%, 50%, and 100%, positive and negative control were studied. Physical tests, including organoleptic, pH, and viscosity tests, were performed. The pH of A. hybridus L. leaf extract in the mouthwash was 4.80, with a viscosity of 1.2456–1.378 cP. The inhibition zones of antibacterial activity against S. mutans in the extract concentrations of 25%, 50%, and 100% were 1.13, 1.73, and 2.90 mm, respectively. The ethyl acetate extract of A. hybridus L. leaves in mouthwash displayed antibacterial activity against S. mutans, with an average inhibition zone of 0.57 mm. The mouthwash formulation produced an inhibition zone with an average F1 of 0.57 mm and F2 of 1.67 mm.

Abstract

Sausage is a meat product processed that is popular food especially in Pandeglang, Banten Province. The importance of halal certificates or the existence of the MUI (Indonesian Ulama Council) halal logo for processed meat products makes Muslim people confident to consume them. The aim this research was to identify pig DNA contamination in sausage products in six districts in Pandeglang without the MUI halal labels using RT-PCR (Real Time-Polymerase Chain Reaction). RT PCR that can calculate to pig to fill these sample free from pig contamination. This research was divided into two stage, the first stage is extracted or carried out DNA and the second stage is RT PCR analysis. The results of the DNA purity test on sausage samples had DNA purity values ​​of 1.84-1.9 (A260 / A280) and resulted in sample concentrations ranging from 37.8 to 102.5 ng / µl. The only amplification on the FAM curve was in the positive control pig. the Cq value ranges from 30 - 31.29. The results of RT PCR on sausage samples in the district area in Pandeglang Banten did not detect the presence of pig DNA.
