Abstract

A study on the community of butterfly fish (Chaetodontidae) has been conducted from April to June 2013 in two locations of coral reefs at Taman Nirwana coastal area, Padang, namely tourism zone and mangrove zone. This study aimed to determine composition and community structure of butterfly fish. This study used visual census method with limitation of 2.5 meters of view to the left and to the right along the line transects. The results found that nine species of butterfly fish which belong to two genera; Heniochus and Chaetodon. There were three species of Heniochus (i.e Heniochus pleurotaenia, H. varius, and H. Singularis) and six species of Chaetodon (i.e Chaetodon triangulum, C. collare, C. vagabundus, C. trifasciatus, C. rafflesii, and C. Kleinii). The abundance and species diversity of butterfly fish did not significantly differed between tourism and mangrove zones. In line with the two previous parameters, similarity indices were found very close between the tourism and mangrove zones. The low index values for all of the parameters may indicate a low quality of the coastal area community due to the human interferences.Key word : Chaetodontidae, composition, community structure, Nirwana Park beach-Padang

Abstract

An experiment on the reduction of stratification period with addition of GA3 hormone to strawberry (Fragaria x annanassa (Weston) Duchesne) seeds germination has been done from December 2013 to February 2014 in the Laboratory of Plant Physiology, Department of Biology, Faculty of Sciences, Andalas University, Padang. The experiment aimed to reduce the stratification period and determine the reliable concentration of GA3 on strawberries seeds germination. The experiment was conducted by using Split Plot Design which consisted of stratification levels (0 week, 1 week and 2 weeks) as main plots, while GA3 consentration (0 ppm, 25 ppm, 50 ppm and 75 ppm) as sub plots. The results from that 25 ppm GA3 combined to 0 week showed the best acceleration to  the emergence of sprouts, increased the number of seed germinations and sprouts length but did not significantly affected to the length of time required for the germination.Key word :GA3, germination, stratification, strawberry

Abstract

Research about partial optimization of thermophilic bacteria isolate M5-24 on protease production was conducted from May 2013 to December 2013 in Laboratory of Microbiology, Biology Department, Andalas University, Padang. Purpose of the research was to find out optimum condition of protease production by isolate M5-24. This study consisted of three steps. Step 1: Investigating the effect of temperature and pH used completely randomized design in factorial with two replications.The first factor was temperature (450C, 500C, 550C, 600C), the second factor was pH (7, 8, 9, 10). Step 2: Investigating the effect of inoculum doses and inducers used completely randomized design in factorial with three replications. The first factor was inoculum doses (1%, 5%, 10%) and the second factor was inducer (yeast extract 0,5%; pepton 0,1%;  kasein 1%; meat extract 0,5%). While Step 3: Investigating the effect of agitation (125 rpm, 150 rpm, 175 rpm, 200 rpm) used completely randomized design with six replications. The result showed that the best duration of growing was at 18th hours after incubation, the highest enzyme activity was at temperature 500 C;  medium’s pH at 8; the best of inoculum dose was at 5%; the best  inducer was casein 1% and optimum agitation was at 175 rpm. Extrinsic optimization can increase protease production of  thermophilic isolate M5-24 up to 1.7 times.Keywords : Optimization, Protease,  thermophilic, bacteria

Abstract

Dogs are the most popular pet animal although they could carry ectoparasites which can cause diseases in human. The purposes of this study were to identify ectoparasites and to determine their prevalence on pet dogs. This study was conducted from March to July 2013 using survey method and collecting ectoparasites directly from 15 pet dogs in Padang. Specimen processing and species identification were done at the Animal Taxonomy Laboratory, Biology Department, Andalas University. This study found 5 species of ectoparasites in pet dogs. There were Ctenocephalides canis Curtis, 1826, Dermacentor sp., Haemaphysalis sp., Rhipicephalus sanguineus Latreille, 1806 and one unidentified species sp. 1 from family Laelapidae. Rhipicephalus sanguineus has the highest prevalence (73,3 %) followed by Haemaphysalis sp. (46,7%), Dermacentor sp.(20%), Ctenocephalides canis (13,3%) and sp. 1 from Family Laelapidae (6,7%).Keywords : ectoparasite species, prevalence, pet dogs

Abstract

We experimentally studied the effect of organic mulches on weeds and yield of green beans (Phaseolus radiatus L.) have been conducted at green house and Laboratory of Plant Ecology, Departement of Biology, Faculty of Natural Sciences, Andalas University, Padang, from January to May 2014. The aims of this study were to determine the effect of different mulches (Mikania micrantha and Tithonia diversifolia) on weeds and yield of green beans. The experiment used nested design with eight treatments and three replications. The first factor (factor A) was two kinds of mulch (i.e Mikania micrantha and Tithonia diversifolia). While the second factor (factor B) was the given doses of mulch (i.e 0 g/polybag, 300 g/polybag, 600 g/polybag and 900 g/polybag). The results showed that Mikania micrantha mulch gave the stronger effect to reduce weed numbers. Mikania micrantha mulch on 900 g/polybag increased plant height and Mikania micrantha mulch on 300 g/polybag increased seed yield. Whereas Tithonia diversifolia mulch on 600 g/polybag increased plant height and the number of primary branches and Tithonia diversifolia mulch on 300 g/polybag increased seed yield.

Abstract

The study about effectivity of growth medium for multiplication Glomus spores isolated from the rhizosphere Pternandra echinata Jack. has been conducted from February to November 2013 in the greenhouse and Plant Physiology Research Laboratory Department of Biology, Faculty of Mathematic and Natural Science Andalas University. The aims of the study to determine the appropriate type of media for multiplication Glomus isolated from rhizosphere Ptenandra echinata. The study used a Completely Randomized Design with 3 treatments and 9 replications. The treatment consists of the type of media used are sand, zeolite, and zeolite sand. The results showed that statistically media used gives a significantly different effect on the increase in plant height and number of leaves. Meanwhile the spore density and dry weight of the spores will show results that are not significantly different. Percentage of degree infection maize in all growth medium showed high criteria. Generally, the growth medium sand, zeolite, zeolite sand effective for spore multiplication glomus isolated from Pternandra echinata Jack.Keywords : Effectivity, Glomus, Growth Medium

Abstract

The acute toxicity effects of leaf extract of dragon scales fern (Drymoglossum piloselloides (L.) Presl) on hematological values of male albino mice (Mus musculus L.) has been tested at the Laboratory Animal Physiology. The experiment tested extract dose from 50 to 200 mg/Kgbw on hematological values of male albino mice. The treatments  used a randomized block design method. The mice were injected by the leaf extract at several doses: i.e.  0 mg/Kgbw, 50 mg/Kgbw, 100 mg/Kgbw, 150 mg/Kgbw and 200 mg/ Kgbw. The blood were collected at 24 hours after treatment and hematological values have been examined including number of red and white blood cell, hematocrite, hemoglobin concentration (Hb), plasm protein concentration, absolute blood indices, diameter and morphology of eritrocyte. The results showed that the leaf extract of D. piloseloides gave no acute toxicity effect of hematological values.Keywords: Drymoglossum piloselloides, hematology, acute toxicity

Abstract

A study on composition and structure of phytoplankton community at Maninjau lake was conducted from April to July 2013. The planktons were sampled purposively at 11 stastions using vertically towed of plankton net. This study found a total 94 species of phytoplankton that mostly belong to Chlorophyceae. Density of phytoplankton varied significantly between sampling stations ranged from 1,083 to 5,119 ind/l. The highest density was found at Station where agriculture plant dominated the vegetations around the edge of the lake, while the lowest density found at station located near the centre of the lake. Those phytoplanktons with relative density higher than 5% were Synedra ulna, Cosmarium compressus, Cosmarium decoratum, Sturastrum playfairi, Chroococcus disperses, Chroococcus limneticus and Glenodinium quadridens. Diversity indiches of phytoplankton (H`) was ranged from 1,96-2,80 with highest diversity at Galapuang (near garbages disposal area and fish farms) and the lowest was at Pakan Rabaa (water inlet). Equitability indiches (E) ranged from 0.65 to 0.81. The Index of Similarity phytoplankton was range from 28.12-70.00%. The result of this study might suggest that composition of the phytoplankton in the lake was relatively high although the structure reflected some influencees of human activities.Key word :phytoplankton, community, diversity, and Maninjau Lake.

Abstract

An experimental study to figure out the effect of activated charcoal and coconut water to shoot encapsulation of Tetrastigma rafflesiae Miq. has been carried out from October 2013 to February 2014 at Plant Physiology Laboratory, Biology Department, Faculty of Sciences, Andalas University. The experiment used Completely Randomized Design with four treatments and six replications. Treatments consisted of 1g/L activated charcoal, 300 mL/L coconut water and combination between 1g/L activated charcoal and 300 mL/L coconut water. The result showed that addition of activated charcoal and coconut water decreased viability of T. rafflesiae encapsulation. The result indicated that the use of MS (Murashige Skoog) medium with 4% sodium alginate and 50 mM CaCl2.2H2O formed synthetic seeds with high germination percentage (83%).Keywords : Tetrastigma rafflesiae, encapsulation, activated charcoal, coconut water

Abstract

Histological analysis of liver of hard-lipped barb (Osteochilus hasseltii C.V.) was conducted from March to October 2013 at the Laboratory of Animals Structure and Development, Department of Biology, Faculty Sciences, Andalas University, Padang. Those fish samples were collected form Maninjau and Singkarak Lakes, West Sumatra. The liver tissue were isolated, fixated, dehydrated, planted on paraffin and colored by Haematoxilyn-eosin. Cell Structure and forms were described based on Microscopic tissue study. Results found that the histological damage of fish liver from Maninjau showed three storeys of Hypertrophy, lysis, and necrosis, while Singkarak showed two storeys of hypertrophy and lysis. Those results may indicate that hard-lipped barb from the two lakes had been conminated by some pollutants.Keywords: Osteochilus hasseltii, histopatology, liver, Singkarak Lake, Maninjau Lake.
