Abstract

A study on kidney histology of Silver Sharkminnow (Osteochilus hassletii) sampled from Maninjau and Singkarak Lakes, West Sumatra has been done from December 2012 to April 2013 at Animal Structure and Development Laboratory, Department of Biology, Andalas University, Padang. Direct observation and purposive sampling were conducted at four locations around Singkarak Lake (Sumani, Paninggahan, Sumpur, Ombilin) and one location in Maninjau Lake (Tanjung Sani). Histological samples of kidneys were isolated, fixated, dehydrated and embedded on paraffin and stained using Haematoxylin-Eosin. The results showed some histological alterations of kidney samples from all locations. Those alteration varied from cell hypertrophy, lysis, necrosis and emerging of scrape tissues. There were no significant differences on the degree of alteration among all populations. Abstract

A study on screening and characterization odf cellulolytic thermophylic bacteria from Sungai Medang hot spring was conducted on May to June 2012 using purposive sampling methods. This study aimed to find a cellulolytic thermophylic bacterium and to analyze its cellulolyties. Twenty eight cellulolytic thermophylic bacteria were isolated from Sungai Medang hot springs. The highest enzymatic activity (amylolytic and proteolytic activity) was shown by isolate MII2.1 from a location at which the temperature was 78oC. This isolate was characterized as an aerobic bacterium, gram negative and non motile. The form of colony was a circular margin, smooth shiny surface and elevation flat.Keywords: amylolytic, bacterium, cellulolytic, proteolytic, thermophylic

Abstract

A study on histological structure of gill and hemoglobin concentration of Silver Sharkminnow (Osteochilus hassletii C.V) in Singkarak and Maninjau Lakes, West Sumatera has been conducted from December 2012 to March 2013. This study aimed to determine the damage of histological structure of gill and haemoglobin concentration of O. hasseltii. We prepared slide preparation of gills to observe their damage structure, while hemoglobin concentration was counted/measured using Sahli method. The results showed various types of damage such as edema, loss of the epithelium on the secondary lamella, hyperplasia, fusion of secondary lamella, the loss of the secondary lamella structure, clubbing and thickening of elastic cartilage. Hemoglobin concentration of O. hasseltii ware 3.28% and 3.05 % for Singkarak and Maninjau respectively. There was no statistically significant difference between sampling sites for both of studied parameters.Keywords: Ostechilius hasseltii, gill histology, hemoglobin, Singkarak and Maninjau lakes

Abstract

The study of population density and distribution of Lizard (Mabuya multifasciata. Kuhl) have been carried from June to December 2012 at the Pasumpahan and Sirandah Islands. The survey method and purposive sampling were used in the research, and for direct collecting samples was used visual ecounter survey. Nine individual of M. multifasciata were observed in Sirandah Island with estimate density was 45 ind/ha. The lizard were dispersed through the island of sirandah but was absent in Pasumpahan Island. Abstract

An inventarisation of the Anuran species (Amphibia) at Harapan Rainforest, Jambi has been done from October 2011 to July 2012. We collected 127 samples from the field and made identification based on morphological characteristics. We identified 19 sepcies in which belong to five families i.e. Bufonidae (Phrynoidis asper Gravenhorst., Ingerophrynus parvus Boulenger., I. divergens Peters. and Pelophryne signata Boulenger.), Microhylidae (Kalophrynus pleurostigma Tschudi.), Dicroglossidae (Occidozyga sumatrana Peters., Fejervarya cancrivora Gravenhorst., F. limnocharis Boie., Limnonectes paramacrodon Boulenger., L. malesianus Kiew.), Ranidae (Hylarana erythraea Schlegel., H. parvaccola Peters., H. glandulosa Boulenger., H. nicobariensis Stolizka., H. baramica Boettger., H. siberu Mc.Cathy and Whitten., Odorana hosii Boulenger., Rana sp), Rhacoporidae (Polypedates leucomystac Boulenger.).Keywords: Amphibia, Anura, Identification key, Harapan Rainforest

Abstract

This study aimed to observe the ability of isolated bacteria from Sungai Medang hot spring in producing amylase and to determine the activity and characteristic of isolated amylolytic bacteria which gave the highest amylolytic index. The water samples were collectied by using purposive sampling based on different level of temperatures. This study has successfully collected 48 bacterial isolated from the hot spring. To find the highest amylolytic activity, we tested ten isolated bacteria which have high amylolytic indices. We found that MV2.7 isolated from 740C of hot spring gave the highest amylolytic activity. The isolated bacteria was positive proteolytic, cellulolytic and negative lipolytic. Abstract

The study about the effect of acidity adjusting of industrial waste of tea production on the mycelium growth of brown oyster mushroom (Pleurotus cystidiosus O.K. Miller) was conducted from January to April 2013 in the Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Sciences, Andalas University, Padang. The aim of this study is to determine the effect of setting the acidity of tea production industrial waste and the mycelium growth of brown oyster mushroom. This research was designed by completely randomized design (CRD) with five treatments, which were pH 4, pH 5, pH 6, pH 7 and pH 8 in 5 replication. The results showed that the treatment of pH 8 and pH 7 on industrial waste of tea before weathering was optimum for the growth of mycellium (18 – 19,2 days) . Abstract

An inventory of mammalian species at The Biological Education and Research Forest (HPPB) Andalas University was conducted from November 2011 to March 2012. We used six camera traps operated along forest paths where those predicted as regular tracks of the animals. We successfully identified ten species of mammals in which they belong to ten families and six orders. Those identified species were Hylomys suillus, Macaca nemestrina, Manis javanicus, Lariscus insignis, Hystrix brachyura, Ursus malayanus, Panthera tigris sumatrae, Sus scrofa, Tragulus javanicus and Muntiacus muntjak. The two species, Lariscus insignis and Hylomys suillus were new record from this area.Keywords: mammals, camera trap, Biological Education and Research Forest

Abstract

A study about species of plant consumed by Ungko (Hylobates agilis) at The Biological Education and Research Forest, Andalas University, has been done from July to November 2012. We used ad libitum observation to the gibbon’s feeding behavior and sampled those plants they ate. The samples were processed at Herbarium Andalas University (ANDA), Padang. Seventeen species of plants which belong to eleven families have been recognized as food resources of the gibbons. We described each species of the plants based on morphological characters primerily the part eaten by the gibbon. The results showed that most of the fruit eaten by ungko were fleshy, succulence, and easy to digest. The fruit types were bacca, legume, syconium, capsule and drupe. Abstract

Seven species of Zingiberaceae were investigated for their antimicrobial activities against Staphylococcus aureus, Escherichia coli and Candida albicans. Extract of grinded rhizome was centrifuged at 10.000 rpm for five minutes. This study used completely randommized design in nested pattern with three replications. The first factor was the tested microbe and the second was the fresh extract of the ginger rhizome. The result showed that fresh extract of red ginger (Zingiber officinale var. Rubrum) gave the highest inhibition to the growth of S. aureus (15.83 mm) and E. coli (15.33 mm) while elephant ginger (Z. officinale var. Roscoe) gave the highest inhibition growth of C. albicans (10.7 mm).Key words: antimicrobial, ginger, microbial pathogen
