Abstract

The Indonesian archipelago is an ideal setting for the study of speciation and biogeography. This archipelago is divided into three island groups based on zoogeography: Sundaland, Wallaceaand the Australian region. In this paper we used frogs in genus Fejervarya (Bolkay) to study biogeography and examine patterns of gene flow across proposed zoogeographic boundaries. Severalmolecular studies on Fejervarya species from Indonesia have been carried out, but comparative studies among members of the genus Fejervarya have yet to be performed. In order to elucidate genetic divergence and geographic distribution of these frogs, we conducted a molecular analysis of the mitochondrial 16S rRNA gene using 179 frogs from five Fejervarya species. In total we collected from 32 localities in Sumatra, Kalimantan (Indonesian part of Borneo), Java, Bali, Sulawesi and Lesser Sunda Islands in Indonesia. Molecular phylogenetic analysis recovered 35 haplotypes and showed that frogs in the genus Fejervarya were divided into two well-supported clades. The first group were of three species, F. limnocharis, F. iskandari and F. cf. verruculosa and the other group clade consisted of Fejervarya cancrivora and Fejervarya sp. (Sulawesi-type). The average sequence divergence among these four species ranged from 1.09 to 16.03% (mean = 11.29±2.83%). The present results clearly show that there are five Fejervarya species in the Indonesian archipelago. Fejervarya limnocharis and F. cancrivora are widely distributed and sympatric in Sumatra, Borneo and Java. Fejervarya iskandari is not endemic to Java and also occurs in the Lesser Sundas. Fejervarya cf. verruculosa and Fejervarya sp. (Sulawesi-type) are endemic to Lesser Sunda and Sulawesi Island, respectively. Abstract

Research on the Snakes Morphological Characteristics on Campus of Andalas University Limau Manih had been done. The research was conducted using survey method and Dissemination of Information to Public accompanied by morphometric measurements and descriptions. The results of the research that had been done caught 2 species that consist of one family namely Elapidae. This family is refer to snakes which has neurotoxin venom glands, no loreal scalation, and mostly teresterial. Maticora bivirgata flaviceps (Cantor, 1839) which has no loreal scale, red coloration on tail, and head; Naja sumatrana M?ller, 1887 which has no loreal scale, black coloration with yellow coloration on it?s hood, and yelowish head.

Abstract

Penelitian tentang variasi morfologi katak pohon Polypedates leucomystax Gravenhorst, 1829 (Anura; Rhacophoridae) di Sumatera Barat telah dilakukan dari bulan Maret sampai September 2014. Pengambilan sampel dilakukan di Dharmasraya, Padang Panjang, Pasaman Timur, Solok, Padang, Pesisir Selatan dan Kayu Aro dengan metode survey dan koleksi langsung di lapangan, kemudian dilanjutkan di Laboratorium Taksonomi Hewan Jurusan Biologi Fakultas MIPA Universitas Andalas untuk pengukuran karakter morfometrik dan meristik. Hasil penelitian menunjukkan bahwa P. leucomystax di Sumatera Barat memiliki variasi morfometrik yang tinggi, dan perbedaan antara populasi yang berada di dataran tinggi dan dataran rendah. Populasi dari Kayu Aro memiliki ukuran yang lebih panjang pada jarak moncong sampai mata (JMoM), jarak moncong sampai tympanum (JMoT), jarak hidung sampai tympanum (JHT) dan jarak hidung sampai mata (JHM) dibandingkan keenam populasi lainnya. Populasi P. leucomystax yang berada di dataran tinggi cenderung memiliki ukuran karakter kepala yang lebih panjang dibandingkan populasi dari dataran rendah, baik pada populasi betina maupun jantan.

Abstract

Bufonidae is widely distributed amphibian family around the world, including Indonesia (Sumatra) and Asia. Sumatra is an island which has separated because of sea level increase and create gene flows barrier for amphibian in Sumatra and Asia. The purpose of this research is to analyse Bufonidae phylogenetic which exist in several locations in Sumatra and Asia based on gene cytochrome b analysis. Samples were collected from six localities in West Sumatra. DNA extraction and amplification has been done in Genetics and Cell Biology Laboratory, Andalas University, whereas DNA sequencing were analyzed at MacroGen USA DNA Sequencing Laboratory, South Korea. About 14 tissue samples of Bufonidae from West Sumatra has been analyzed. The relationship of Bufonidae phylogenetics in Sumatra and Asia were divided into three main clusters. Duttaphrynus melanostictus in Sumatra is closer to Duttaphrynus melanostictus from India with 0,3-0,5% sequence divergence and it is a group of paraphyletic with Duttaphrynus melanostictus from Vietnam, Taiwan, and China.

Abstract

Sumatera merupakan salah satu pulau besar yang ada di Indonesia. Secara geografis pulau Sumatera terdiri dari pegunungan, lembah dan dataran rendah atau kawasan pantai, sehingga besar kemungkinan terjadi perbedaan pola adaptasi pada masing-masing daerah, kejadian ini akan menimbulkan tingginya variasi dan diferensiasi pada masing-masing populasi. Berdasarkan kondisi ini, dilakukan penelitian tentang diferensiasi morfometri Fejervarya limnocharis (Anura : Ranidae) Gravenhorst, 1829. Tujuan penelitian ini untuk mengetahui diferensiasi F. limnocharis (Anura : Ranidae) Gravenhorst, 1829 di Sumatera. Sampel dikoleksi pada beberapa daerah meliputi Langkat Sumatera Utara, Prapat Sumatera Utara, Padang Sidempuan Sumatera Utara, Padang Sumatera Barat, Alahan Panjang Sumatera Barat, Rokan Hilir Riau, Sungai Penuh Jambi, Muara Jambi, Semidang Bengkulu, Pagalaram Sumatera Selatan, Kota Bumi Bandar Bandar Lampung, kemudian dilanjutkan di Laboratorium Genetika dan Sitologi Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Andalas Padang. Hasil penelitian memperlihatkan bahwa terdapat perbedaan morfometri yang sangat signifikan antar populasi. Karakter morfometri yang memperlihatkan perbedaan adalah diameter tympanum, jarak moncong sampai tympanum, panjang kaki depan, panjang kaki belakang. Populasi F. limnocharis yang berasal dari Alahan Panjang memiliki kaki belakang lebih panjang dibandingkan populasi lain.Kata Kunci: Populasi, karakter, kaki belakang, habitat

Abstract

Biomaterial titanium mulai banyak digunakan sebagai bahan implan karena mempunyai kekuatan tinggi, lentur, tahan korosi dan biokompatibilitas yang baik. Namun demikian, titanium bersifat bioinert yang membuatnya tidak bisa berinteraksi dan menyatu dengan jaringan hidup. Untuk menutup kelemahan ini, titanium perlu dilapisi dengan bahan yang mempunyai bioaktivitas tinggi seperti biokeramik hidroksiapatit (HA). Pada studi ini, pelapisan HA telah dilakukan pada purwarupa implan berbentuk sekrup yang terbuat dari paduan titanium tipe β yang relatif baru dikembangkan, yakni Ti-29Nb-13Ta-4.6Zr (TNTZ). Proses pelapisan dilakukan dengan menggunakan metode Electrophoretic Deposition (EPD). Lapisan HA pada permukaan TNTZ meningkatkan bioaktivitas implan logam ini sehingga memicu proses penyatuan implan dengan jaringan hidup (osseointegration). Parameter yang digunakan untuk menentukan nilai osseointegrasi ini adalah besarnya gaya puntiran (torsi) yang dibutuhkan untuk melepaskan sekrup dari tulang dengan menggunakan alat removal torque tester (RTT). Untuk itu, sekrup TNTZ berukuran M3x0.5 yang tidak dilapisi HA (tanpa HA) dan yang sudah dilapisi HA (lapis HA) ditanamkan pada paha atas (tibia) hewan uji mencit Rattus norvegicus Wistar kemudian dipelihara selama 2 (dua) minggu. Setelah itu, hewan uji dimatikan, dan besaran torsi untuk melepaskan masing-masing sekrup dari tibia mencit diukur dengan alat RTT tersebut, dan dilanjutkan dengan analisis histopatologi pada jaringan bekas pemasangan implan. Hasil studi menunjukkan bahwa implan TNTZ dengan lapis HA memiliki nilai osseointegrasi yang jauh lebih tinggi (470%) dari implan tanpa HA. Analisis histopatologi menunjukkan bahwa proses pembentukan jaringan baru (osteogenesis) yang jauh lebih banyak pada jaringan tulang yang dipasangi implan TNTZ lapis HA dibandingkan dengan tanpa HA. Disamping itu, adanya lapisan  HA pada permukaan implan juga mampu mengurangi reaksi inflamasi yang berlebihan pada jaringan tulang hewan uji dalam waktu yang relatif singkat.Titanium biomaterials are starting to be widely used as implant materials because they have high strength, flexibility, corrosion resistance and good biocompatibility. However, titanium is bioinert which makes it unable to interact and blend with living tissue. To cover this weakness, titanium needs to be coated with a material that has high bioactivity such as hydroxyapatite (HA) bioceramic. In this study, HA coating was carried out on a screw-shaped implant prototype made of a relatively recently developed -type titanium alloy, namely Ti-29Nb-13Ta-4.6Zr (TNTZ). The coating process is carried out using the Electrophoretic Deposition (EPD) method. The HA layer on the TNTZ surface increases the bioactivity of these metallic implants thereby triggering the process of implant integration with living tissue (osseointegration). The parameter used to determine the osseointegration value is the amount of torsion required to remove the screw from the bone using a removal torque tester (RTT). For this reason, TNTZ screws measuring M3x0.5 which were not coated with HA (without HA) and which had been coated with HA (HA coated) were implanted in the upper thigh (tibia) of Rattus norvegicus Wistar mice and then reared for 2 (two) weeks. After that, the test animals were turned off, and the magnitude of the torque to remove each screw from the tibia of mice was measured with the RTT device, and continued with histopathological analysis of the implanted tissue. The results of the study showed that TNTZ implants with HA coating had a much higher osseointegration value (470%) than implants without HA. Histopathological analysis showed that the process of new tissue formation (osteogenesis) was much more abundant in bone tissue with HA-coated TNTZ implants compared to those without HA. In addition, the presence of an HA layer on the surface of the implant was also able to reduce the excessive inflammatory reaction in the bone tissue of the test animals in a relatively short time.

Abstract

Fejervarya limnocharis are amphibians which generally can be found in agriculture area. The high intensity of pesticide application in agricultural area can lead to alteration of physiological aspects of organism inhabiting that area. Quantitative examination of hematology aspect can be conducted to evaluate physiological condition of species. This research was aimed to find out about hematology profile of F. limnocharis living in agriculture area using pesticide in West Sumatera. Thus, examinations of erythrocytes, leukocytes, hemoglobin concentrations, and hematocrit values collected from F. limnocharis living in three different areas have been conducted. The data gained have been compared with normal blood value of various amphibians species from previous research. The result acquired respectively for three locations show that total of erythrocytes were in normal range, which were 1,616 x 106/mm3; 1,539 x 106/mm3; and 1,712 x 106/mm3; hemoglobin concentration were lower than normal range, which constituted 4,06 g/dL; 4 g/dL; and 4,18 g/dL; hematocrit values have normal range, which made up 40,06 %; 40,14 %; and 45,7 %; total of leukocytes were higher than normal range, which were 27,450/mm3; 49,934/mm3; and 22,040/mm3. Usage of pesticide in agriculture area causes alteration of some hematological aspect.Keywords: Fejervarya limnocharis, hematology profile, pesticide, West Sumatera

Abstract

The true frog species, Odorrana hosii, found in West Sumatera Indonesia, has high morphological differentiation and also estimated has high genetic variation. A total of 35 O. hosii at a seven location have analyzed using DNA microsatellite markers. Genetic variation of five microsatellite loci provided the highest value of expected heterozygosity (He) for the population in Padang (He= 0.618), while the lowest was the population in Merapi (He= 0.427). There are genetic differences in moderate levels among populations of O. hosii in West Sumatra (FST= 0.108) with inbreeding intrapopulation value (FIS= -0.559), and high value of gene flow among the populations (Nm = 2.061). This study becomes the first molecular data for establishing effective population management conservation.

Abstract

Ploidization of Mystacoleucus padangensis Bleeker, 1852 (Pisces: Cyprinid) with heat shock induction has been carried out from January to September 2014 at Genetics and Cytology Laboratory, Biology Department of Faculty of Mathematics and Natural Sciences, Andalas University. This study aimed to find suitable temperature for ploidizatiom on M. padangensis. The study used Completely Randomized Design method with five treatments and five replications. The treatments were various water temperatures (i.e. 27oC as control, 36oC, 38oC, 40oC, and 42oC). Abstract

The research about potential of herpetofauna on traditional medicine in West Sumatera was conducted from March to August 2012. This research was conducted at several districts in West Sumatera such as Batusangkar, Padang, Payakumbuh, Pasaman, Painan and Sijunjung. The survey and quisioner method were used by interviewing informan about the species of herpetofauna which were used in traditional medicine. A total of 5 reptilian species were recognized as medicine such as Eutropis multifasciata (Bingkaruang), Naja sumatrana (Ula sendok), Phyton reticulatus (Piton), Chelonia mydas (Katuang), Dogania subplana (Labi-labi) and one species of amphibian i.e Hylarana erythrea (Koncek hijau) from Ranidae.Keywords: potential, herpetofauna, tradicional medicine, West Sumatera
