Abstract

Gene therapy involves the introduction of DNA or ribonucleic acid (RNA) into the target cell either to express or suppress the biosynthesis of proteins. The success of gene therapy is highly dependent on a suitable carrier system that can efficiently deliver genes in to specific desired cell with minimum cytotoxicity on target cells. Chitosan is interesting to be used as a gene carrier because it has a high positive charge and low toxicity to cells. Positive charge chitosan can form complexes with the plasmid. DNA complexes provide protection against enzymes degradation and promote internalization of plasmids that have been condensed. In this study the complex formation of chitosan-pEFneo-GFP and chitosan / TPP-pEFneo-GFP is by complex coaservation method. The results of complex analysis with a 0.8% agarose gel electrophoresis for 30 minutes 100Volt showed that the complex was stayed in the well and the plasmids did not migrate like plasmids which were dissolved in the water and buffer solvent. Stability evaluation in storage at room temperature for 14 days showed that the complex with chitosan concentration 0.02%-0.04% were the most stable, so that transfection analysis performed for the complex with chitosan concentration of 0.02%-0.04%. Transfection results showed that chitosan is able to protect plasmid and promote the internalization of plasmid into the nucleus and then expressed as a green luminescence. From these results chitosan potentially be developed as a carrier system in gene delivery.

Abstract

There has been dramatic progression in the adhesion of dental adhesives and resins to enamel and dentin in the 40 years since Buonocore1 introduced the technique of etching enamel with phosphoric acid to improve adhesion to enamel. The first dental adhesives bonded resins to enamel only, with little or no dentin adhesion. Subsequent generations have dramatically improved bond strength to dentin and the sealing of dentin margins while retaining a strong bond to enamel. The widespread demand for, and use of, dental adhesives has fuelled an intensive development of better and easier dental adhesives in rapid succession; dentists have literally been inundated with successive “generations” of adhesive materials. There are seven generatios in bonding agent and have a different characteristics. This article discusses the progression of dental adhesives up to the last generation.

Abstract

Aggressive Periodontitis is a form of periodontitis which is characterized with the presence of deep pocket, rapid loss of connective tissue attachment and alveolar bone. The rapid damage is caused by the dysfunction chemotaxis and phagocytosis from neutrophil.Calprotectin is a major cytosolic protein of neutrophils, monocytes and epithelial cells. The calprotectin level in gingival crevicular fluid is useful marker for evaluation the progression of inflammation in periodontitis disease. In this study, we investigated calprotectin level at different periodontal pocket depth from patient with aggressive periodontitis. This research involved twelve subjects who had been diagnosed as suffered from aggressive periodontitis. Their gingival crevicular fluid was taken from different periodontal pocket depths (slight ≤3mm, moderate 4-6mm, severe ≥ 7 mm), using sterile papperpoints. Level of calprotectin in gingival crevicular fluid were analysed by enzyme-linked immunosorbent assay (ELISA) procedures. The result showed that gingival crevicular fluids calprotectin level increased approximately in proportion to probing depth slight, moderate, and severe (p< 0,05), and showed a significant positive correlation with probing depth (R=0,776, p< 0,05). This research concluded that the calprotectin level in gingival crevicular fluid is positively related to clinical indicators (probing depth) and reflect the degree of gingival inflamation in patient with aggressive periodontitis

Abstract

Caffeine is a drug which is effective as an anti-cellulite. Caffeine works by slowing down the lipogenesis process and accelerating the lipolysis process. In this research, caffeine was formulated in Sodium CMC gel base and lactic acid (0, 2, 4, and 6%) as enhancers. The aims of these research was to know the effects of lactic acid as enhancer on pH, viscosity, spreadibility, flowability properties, and percutaneous penetration study of caffeine gel. The results obtained that all formulas have fullfilled of all requirements and it can be concluded that lactic acid lowered the pH, increased viscosity, and lowered spreadibility. The highest flux was obtained the caffeine gel on concentration of 6% lactic acid is equal to 5.43 g/cm2 menit.

Abstract

In dental care, tooth extraction without injection (topical anesthetic), extraction with injection (anesthetic injection), and drilling are condition triggering anxiety of patients. Difficulty in dental treatment for children can be overcome by understanding psychological aspect of children in a certain age. Some principles in handling children are good psychological approach, effective management technique of patient, empathy, and supporting circumstances. Thus, it is advisable to consider some techniques which are successfully-proved in psychology and may be applied in dental care such as attitude development, Tell-Show-Do, desensitization, reinforcement, modeling and sedation.

Abstract

Preparation level prior to conduct dental filling results in smear layer. When smear layer is too thick, it will disturb the adhesion of restoration material to cavity wall, therefore it needs cleansing using dentin conditioner. One of cleansing materials for smear layer is acid with low concentration like citrate acid, hydrogen peroxide, and 10% of polyacrilate acid (Sutrisno, 2005). The use of natural materials in dental practice is an alternative and may minimize the side efect, one of them is belimbing wuluh (Averrhoa bilimbi L). This study is experimental laboratory aimed at analysing the potential of cleansing force of belimbing wuluh (Averrhoa bilimbi L) juice on the smear layer of cavity wall of tooth prepared. Ten samples of incisive teeth of cows were prepared with circle-shaped class 1 cavity design with 5 mm in diameter, 2,5 mm in depth in one third from the center of dental labial surface. Subsequently, 5 samples were polished with 10% of poliacrylic acid, the other 5 samples were polished 100% of belimbing wuluh juice, allowed for 10 seconds and irrigated with sterile aquades and dried. The samples were examined for their cleanliness from smear layer using SEM with 5000x magnifying. The result showed average value of cleanliness of the cavity bottom from smear applied with 10 % of polyacrilate acid was lower compared to the group with 100% of belimbing wuluh juice. Statistically, it showed a significant difference between the control group (10 % of polyacrilate acid) and treatment group (100% of belimbing wuluh juice). In conclusion, 100% of belimbing wuluh juicen has potential as dentin conditioner.

Abstract

This studi was aimed to isolate piperine from cabe jawa (Piper retrofractum Vahl) and to look at the effect of piperine and hexane-undissolved piperine free fraction on testosterone levels of male rats. The subject of this research were twenty five healthy male wistar rats., which every group consist of five rats. Every group was treated by orally administration with Andriol (Testosterone undecanoat) 0,72 mg , CMC-Na 1%, piperine 1,6 mg/kg bw, hexane-undissolved piperine free fraction 29,10 mg/kg bw and hexane-undissolved fraction 31,72 mg/kg bw twenty seven days period of time, once a day. At the end of treatment, testosterone concentration was measured using Enzime Linked-Immunosorbent Assay (ELISA) technique. Result were statistically evaluated by ANOVA followed by LSD test at the 0,05 significance level. The result of this studi showed that serum testosterone level was increased compared to the CMC-Na 1% group for hexane-undissolved piperine free fraction while piperine caused a decrease in the testosterone level. However, the increase and the decrease in values were not statistically significant (p>0,05).

Abstract

A simple, accurate, precise, and economic UV-spectrophotometric method was developed for the estimation of ofloxacin in phosphate buffer at pH 3,0; 7,4; and 12,0. Ofloxacin solubility is dependent on pH of the solvent due to its amphoteric properties. Ofloxacin has absorbance maxima at 295 nm in pH 3,0; 288 nm in pH 7,4; and 290 nm in pH 12,0. Linearity was obtained in the concentration range of 4-10 g/mL for each pH. Ofloxacin detection limit at pH 3,0 was 150 g/L, at pH 7,4 and pH 12,0 were 10 g/L, respectively. Recovery values at three pH conditions were in the range 89,93-104,25%. The results of analysis have been validated statistically and by recovery studies.

Abstract

Local anesthetics are believed to be the most frequently used drugs in clinical dentistry. Lidocaine and mepivacaine are commonly used in RSGM (Oral and Dental Hospital Jember University), although they are generally regarded as safe, some adverse reactions can be expected and do occur. This study was undertaken to know the effect of vital sign on mepivacaine in local anaesthetic on dental clinical use. Material and Methods: A total of 42 (14 males, and 28 females) healthy students of Faculty of Dentistry Jember University were included in this clinical trial. They were locally injected as block anaesthesia of inferior alveolaris nerve. Blood pressure, pulse, and respiration were examined pre and post injection. Result: Mean of respiration pre/post (18.7/1.8) p>0.05, pulse pre/post (79.8/79.9) p>0.05, and diastole pre/post (74.9/73.9) p>0.05, and systole pre/post (112.0/110.2) p>0.05. There were no clinical and statistical differences on vital sign between pre and post injection. Conclusions: Mepivacaine as local anaesthetic is clinically safe on vital sign.

Abstract

Level of oral hygiene have a close relationship with periodontal health status; where good oral hygiene will minimize the occurrence of periodontal disease. Aims of this study were to assess the oral hygiene status and periodontal health patients that come to Department of Periodontics of Jember University Dental Hospital at August 2009 until August 2010. This type of research is an observational study of clinical (cross sectional). A total of 155 sample (87 males and 66 females) were examined by using Oral Hygiene Index-Simplified (OHI-S) and Periodontal Index. Good oral hygiene was seen in 11% of total sample examined, 54,2% had fair oral hygiene and rest 34,8% showed poor oral hygiene. 0,6% of total sample examined had healthy periodontal, 18,1% had simple gingivitis, 41,3% had initial of periodontal disease destructive, 37,45 had periodontal disease destructive and 2,6% had terminal disease. Oral hygiene status and periodontal health examination revealed that majority of sample had fair oral hygiene and periodontal disease destructive.
