Abstract

In Indonesia, belimbing wuluh (Averrhoa bilimbi L.) has been used for traditional medicine, especially to heal stiff, mumps, rheumatism, ulcers, acne, tinea versicolor, high blood pressure, and dental pain and cough. This property is predicted comes from its secondary metabolites. Saponin is a compound group of secondary metabolites which is known to have antitussives and expectorants effect, antiinflammatory, antibacterial, and cytotoxic activity for cancer therapy. However, scientific information about the location for saponin accumulation in this plant is still limited. This information is needed for the use of saponin from A. bilimbi for herbal medicines development. The aim of this study was to know the content of saponin in fruits, leaves, and petioles of A. bilimbi. This work was initiated with sampling and preparation of samples, followed by the preliminary tests, including froth test and color test to show the saponin content qualitatively. Saponin was extracted and isolated by maseration using methanol solvent, and by thin layer chromatography (TLC) preparative, respectively. Saponin content was measured by spectrophotometer UV-Vis at 280 nm. Result shows that the highest saponin content, according to the absorbance value, is in the fruit organ. In A. bilimbi, saponin could be accumulated in the generative organ, although saponin synthesis is located in leaves. Based on the production of brown ring with Liebermann Burchard (LB) reagent test, it is suggested that triterpen saponin is the dominant saponin presents in A. bilimbi. Saponin content in A. bilimbi fruits is higher than saponin content in Musa paradisiaca var. sapientum L. stems. Therefore, triterpen saponin from A. bilimbi fruits is promising to be developed as commercial herbal medicines.

Abstract

Fertilizers used by farmers today is  artificial fertilizers (inorganic). The use of artificial fertilizer would cause the soil structure damaged due to artificial fertilizer will only provide nutrients to plants without regard to the soil fertility. Finally, food productivity will decrease and food security will be so weak. To overcome this, there needs to be a good fertilizer that meets nutrient for plants and soil. To overcome this, a good fertilizer that meets nutrient for plants and soil is very necessary. The use of biomass and farm waste to increase the value of agricultural wastes is so potential to be organic fertilizer. Composting organic materials can produce humic acid. Humic acid is able to absorb nutrients from the environment exploited by plant roots and transferred into the root cap. The purpose of study is to produce humic acid from agricultural and livestock waste. The method of this study is the extraction. Stages of this study are preparation, composting, humic acid isolation, and purification of humic acid. The results of this study showed the highest levels of humic acid is 48.08%.

Abstract

Toads (Phrynoidis aspera, Ingerophrynus biporcatus, Duttaphrynus melanostictus) and poisonous rock-frog (Odorrana hosii) are members of the class Amphibian from different Families. Previously, those three species toads were in the same genus called Bufo, but now they are clustered in a different genus based on the molecular systematic. Proteins secrete from the parotoid and skin glands are one of self-defense mechanisms against to predators. The aims of this study were to determine and compare the protein patterns in secretes from toad parotoid gland and skin gland of poisonous rock-frog. The specimens were obtained from sampling at several different locations according to their habitats. Secrete collections were done by stressing both physically and chemically. Secrete was extracted by the addition of acid and sonication. The secrete protein concentration was measured by the Bradford test, whereas protein patterns were analyzed by SDS-PAGE. The protein concentrations of secrete from toad’s parotoid gland are higher (Phrynoidis aspera 3.50 µg/ml; Duttaphrynus melanostictus 6.7 µg/ml Ingerophrynus biporcatus 1.32 µg/ml) than in skin gland secretion of poisonous rock- frog (1.07 µg/ml). There are 6 protein bands in parotoid gland secretion of Phrynoidis aspera, 5 protein bands in parotoid gland secretion Ingerophrynus biporcatus and 7 protein bands in secrete of Duttaphrynus melanostictus parotoid glands with molecular weight ranges 12.09-172.51 kDa. However, those proteins are different size of molecular weights for each species.  Furthermore, two protein bands (with different densities) are founded in skin gland secretion of poisonous rock-frog with molecular weight of 66.17 kDa and 78.73 kDa. The differences of genes, habitats and predators may influence the differences of protein patterns in secrete from toad’s paratoid gland and frog’s skin glands. Those protein patterns could be useful for further studies, especially on Bufo systematic, also for exploring the potential of protein secrete as antibacterial or antifungal.
