Abstract

One of the traditional medicinal plants are believed to be as anti-ulcer effect is pegagan (Centella asiatica), pegagan extract thought can protect the gastric mucosa exposed to ethanol by increasing the integrity of the mucosal line of defense. This study aimed to determine the effect of Centella asiatica on rats gastric mucosal resistance (gastroprotective) which experience stress immobilizing. This research is experimental in vivo studies, conducted with 5 groups of male wistar rats treated with 6 replications. Group I is the normal diet control group; group II, normal diet control group mixed Centella asiatica 1 g / mm day; group III was a control group of mice that immobilization stress without giving prior pegagan, Group IV and V is the group that previously received pegagan diet for 3 days prior to immobilization stress with gotu kola concentration of 0.5 g / day and 1.0 g mm / dd day. After 2 days of treatment immobilization stress were measured both in the macroscopic parameters of extensive ulceration and bleeding and microscopic scores for parameters of edema, leukocyte infiltration, and necrosis of rat gastric tissue. The results of macroscopic observations showed that the area of ulceration and bleeding scoring groups I, II, III, there are differences in microscopic bemakna and proven for the parameters of edema, leukocyte infiltration and necrosis are also significant differences. As for group III, IV and V at all test parameters no significant difference.

Abstract

Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower (Hibiscus rosa-sinensis L.) with a concentration of 0.6% w/v has mucolityc activity which is equivalent to 0.1% acetylcysteine based on the capasity to decrease viscosity of the mucus. The use of hibiscus as a new cough medicine done traditionally. Syrup of ethylacetate insoluble fraction containing alkaloids was the appropiate liquid dosage form that has some advantages for examples easy to use and fast absorption of the active substance. The study aims to determine the optimum formulation syrup ethanolic fractions containing alkaloids from hibiscus flowers using the method of Simplex Lattice Design. Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower is obtained by using the Vacuum Liquid Chromatography (VLC). Simplex Lattice Design Methods used to optimize the formulation syrup ethylacetate insoluble fraction containing alkaloids from hibiscus flower with seven formulas with variations in the amount of glycerin, sorbitol solution 70%, and mucilago 0.5% CMC-Na. Physical stability was analized to find higher total response value (R total) as parameter of the optimum formula by Simplex Lattice Design Methods with Design Expert ® software version 8.0.2. Optimum formula syrup ethylacetate insoluble fraction containing alkaloids from the hibiscus flower obtained with the proportion of 25.376% glycerine, 51.985% of 70% sorbitol solution, and 22.639% of 0.5% CMC Na mucilago. Data obtained physical characteristics test compared with prediction value using Design Expert ® software version 8.0.2. Physical characteristics of optimum syrup ad the zero week compared with physical characteristics on the forth week using paired t-test. The result showed that physical properties of syrup formula optimum didn’t different significantly for easy for pouring and tasted response except for viscosity and acidity. Ethylacetate fraction syrup containing alkaloids from hibiscus flower optimization results are less stable during the four weeks of storage.

Abstract

Objectives To evaluate the effect of Curcuma xanthorrhyza Roxb extract, ginger, soybean and shrimp shell in combination compared to diclofenac sodium to liver and kidney function of patients with osteoarthritis. Methods twenty one subjects randomly allocated to different groups (Trial and control = diclofenac group). The trial group were treated by the combination of 50 mg temulawak extract, 100 mg ginger, 50 mg soybean and 100 mg shrimp shell three times daily while the control groupwere treated by 25 mg diclofenac sodium three times daily for a couple of week. Blood sample was obtained before and after two weeks therapy to determine the liver enzymes SGOT and SGPT and also serum ureum and creatinin in order to monitor the kidney function. The study was a prospective randomized open end blinded evaluation (PROBE). Result both trial and diclofenac group show significantly increase of SGOT levels (p<0.05) but the difference change among these groups are not significant (p=0.312). In the trial group there was decreasing of SGPT level (p=0.871) whereas there was an increase in level of SGPT in diclofenac group (p=1.000) although the difference between these two was not statistically significant (p=0.912). The change of serum ureum and creatinin level were not significantly different in trial group compared to diclofenac (p>0.05). In the trial group there was decreasing of serum ureum level although not significant (p=0.214). Both trial and diclofenac group showed no statistically significant increasing of creatinin serum levels (p>0.05). Conclusion there is no statistically difference effect between using the combination of temulawak extract, ginger, soybean and shrimp shell compared to diclofenac sodium toward liver and kidney function of osteoarthritis patients during this two weeks treatment.

Abstract

Sponges is one of the invertebrate Porifera phyla that produce active compounds with various structures and one of the biological activity as antimicrobial. The purpose of this research was to find out the sponge extracts that can inhibit microbial growth and potential of antimicrobial active compounds from sponge at Barrang Lompo Island. The microbes used were Escherichia coli, Salmonella typhii, Staphylococcus aureus, and Candida albicans. The sponge extracts were obtained by maceration 14 sponge samples with methanol, followed by partition using chloroform and methanol, then TLC-bioautography toward the active extract. The results showed that methanol extracts of sponge with code BRLP-009 and 010 have the most potential effect as antimicrobial agent.

Abstract

There are two groups of antioxidant resources, these are synthetic and natural antioxidants. The most important natural antioxidant is phenolic compounds that are produced naturally from plants. Phenolic Antioxidants are potent antiradical, biologically active as they are able to donate hydrogens to free radicals and break the chain of lipid oxidation reactions in the early stages. This study is aimed to search natural antioxidant compounds from the leaves persimmon (Diospyros kaki L.F) that have been utilized traditionally to prevent from sickness. In order to detect the presence of antioxidant compounds in the leaves extract, a reagent of DPPH (2,2-diphenyl-1-pykrilhidrazine) in methanol was used. Initially, powder of persimmon leaves (115 g) is macerated 3 times with methanol, 24 hours each time at room temperature. The liquid extract obtained is evaporated by rotavapor till residue (methanol extract) (phase I) is obtained. The residue was then extracted with CHCl3 to give CHCl3 soluble (phase II) and insoluble fractions. The later is then extracted with water to give water soluble (phase III) and insoluble fractions. These fractions (phase I-III) are chromatophed by TLC and sprayed by DPPH. Phase II contains antiradical compounds as shown by yellow color on the violet background. By preparative TLC [SiO2, CHCl3-EtOAc (1-4 v/v)], antiradical substance is isolated and identified tentatively as substituted phenolic compounds. Antiradical potential is measured spectrophotometrically, the isolat shows IC50 at 107.7 μg/mL that is lower than that of vitamin C (3.04 μg/mL).

Abstract

The present study was to aimed to evaluated and compare in vitro antioxidant activities of 2 Indonesian herbal ingredients (A and B), determined total phenol content., cytotoxic and apoptosis induction activities on HL-60 cells. These data were providing some useful information for people healthy dietary and the new potential application of natural antioxidant containing food materials in functional foods and also as new cancer therapeutics promising candidates. The parameters were total antioxidant activity, amount of total cytotoxic effect on the growth of human promyelocytic leukemia cells (HL-60). Statistical comparison was perform with Student’s t-test at p<0.05. The correlation coefficient (r2) between the parameters tested was established by regression analysis. The scavenging effect of extracts herbal on DPPH radicals increased from 0.3-1.5 mg/ml, where is sample A 14.33% to 64.29% and sample B 9.09% to 57.53% was obtained. High content of total phenol compounds were in sample A (21.72 mg GAE/g), lower amounts were in sample B (17.53 mg GAE/g). Apoptosis of HL-60 cells from the morphological changes side (chromatin condensation). Chromatin condensation, a specific and distinct feature of apoptotic cells, was found in the majority of treated cells. The results indicated that the cell death receptor pathway was involved in the apoptosis induced by Indonesian herbal extracts.

Abstract

Peptic ulcer were been caused by an imbalance between the aggressive factors and a number of defense mechanisms. Pepino (Solanum muricatum Aiton) fruit predicted had antiulcer activity. This study is aimed to test the antiulcer activity of pepino fruit juice in Swiss-Webster mice model which induced by 50 µL concentrated acetic acid. The steps in this study consist of making of fruit juice, phytochemical screening, testing of antiulcer activity at 300, 500, dan 800 µL/20 g BW in doses, and data analyzed using ANOVA. The concentration of pepino fruit juice was 2.5 g/mL. Phytochemical screening showed that pepino fruit juice contain alkaloids, flavonoids, and tannins with protection ratio were 6.02, 23.15, and 63.29% at 300, 500, dan 800 µL/20 g BW, respectively.

Abstract

Inflammation is a protective action that role on defending mechanism to defeat agent causing cell injury. Glucoside of skunkvine (Paederia scandens) has anti-inflammation effect. The aim of this study was to know the anti anti-inflammatory effect of skunkvine (Paederia scandens) extract on artificial edema in Wistar rat. Thirty-five Wistar rats were divided into 7 groups, each group consisted 5 rats. In the control and treatment groups, aquadest (negative control), fenilbutazon (positive control), skunkvine extracts in dose 10mg/kgBW, 20mg/kgBW 30mg/kgBW, 40mg/kgBW, and 50mg/kgBW were administered per orally. In order to induce the edema, one hour after administration, in each group, right hind paw of the rats was injected subcutaneously with by 1% carrageenin. The volume measurement performed with one hour interval for 5 hours assessing with pletysmometer. The data were analyzed by ANOVA. The result of this study showed there was significant difference on group with skunkvine extract 20mg/kg BW to negative control and positive control. In conclusion, that the skunkvine extract 20 mg/kgBW has potency as anti-inflammation substance.
