Abstract

The sandalwood seedling has been planted in the nursery of Balitbang Kehutanan NTT to experience the destruction is about 20%, seedling is attacked by  damping-off. Biological control has been developed as alternate method against soil born diseases to eliminate  damage  on the environment. One of  the biological  agents  having high  antagonistic potential against soil born pathogen is Trichoderma spp. The direction of experiment  to search the cause of lodoh on the sandalwood  seedling and the application influences T. reesei on the control lodoh in the sandalwood seedling. Methods  of  the experiment was (1) isolation the sandalwood seedling of  painful and soil example  from NTT, (2) Postulat Koch test on the sandalwood seedling, (3) antagonistic test of T.  reesei  in vitro and (4) effectiveness  test of T. reesei against developing patogen in vivo. The result indicated that lodoh on the sandalwood seedling caused by Fusarium sp. T. reesei in vitro inhibited Fusarium sp. at average of 100% accordingly. The application of T. reesei as biological control agent against Fusarium sp. showed high  effectivity. The attact percentage of lodoh on sandalwood seedlings showed that the lowest one was growth on compost formulated with T. reesei (5%), whereas seedlings growth on compost without T. reesei,  unsteril  sands and soil samples  from NTT were 30%, 35% and 65%.

Abstract

Rhinoceros beetle (Oryctes rhinoceros) is one of the important pests of coconut tree. One of eco-friendly control applied for this pest is by using entomopathogenic fungiMetarhizium anisopliae. There is not much information about the variability and virulence of M. anisopliae toward O. rhinoceros. M. anisopliae isolates obtained from Biological Control Laboratory, Faculty of Agriculture, Universitas Gadjah Mada were cultured on PDA medium.M. anisopliae isolates was isolated from O. rhinoceros larvae (MaOr), Lepidiota stigma larvae (MaLs), Brontispa longissima beetle (MaBl).O. rhinoceros beetles were obtained from Kulon Progo, DIY. This study used molecular test, and virulence test toward 3rd stadium of O. rhinoceros larvae by using dipping method. Molecular test by sequence and phylogenetic analysis, showed that MaOr was located at different group (out group) with MaLs and MaBr. On the density 107 conidium/ml MaOr and MaLs were more virulent than MaBl towards 3rd stadium of O. rhinoceros larvae.INTISARIKumbang badak (Oryctes rhinoceros) merupakan salah satu hama penting pada tanaman kelapa. Salah satu upaya pengendalian yang ramah lingkungan adalah dengan menggunakan jamur entomopatogen, yakni Metarhizium anisopliae. Belum banyak diketahui mengenai keragaman dan juga virulensi dari M. anisopliae terhadap O. rhinoceros. Tujuan dari penelitian ini adalah untuk mengetahui keragaman genetik M. anisopliae dan virulensinya pada larva kumbang badak. Isolat yang digunakan berasal dari Laboratorium Pengendalian Hayati, Fakultas Pertanian, Universitas Gadjah Mada dalam bentuk kultur murni pada medium PDA. Isolat yang gunakan diisolasi dari larvaOryctes rhinoceros (MaOr), larva Lepidiota stigma (MaLs), dan kumbang Brontispa longissima (MaBl). Serangga yang diuji berasal dari daerah Kulon Progo, DIY. Pengujian secara molekuler dengan analisis sekuensing dan filogenetik, menunjukkan bahwa isolat MaOr terletak pada grup yang berbeda dengan MaLs dan MaBl berdasarkan pada urutan basa DNA. Pada kerapatan 107 konidium/ml isolat MaOr dan MaLs lebih virulen terhadap larva O. rhinoceros instar 3 dibandingkan dengan MaBl.

Abstract

Monitoring of the red flour beetle (Tribolium castaneum Herbst) population by using semiochemical. based bait is important for supporting integrated pest management program. The baits were formulated succesfully using wheat bran as carrier and vegetable oils such as corn oil, soybean oil and palm oil as kairomone, and 4,8 dimethyl decanal. (DMD) as aggregation pheromone. The beetles responded to all baits both in laboratory and warehouse. The level of attractancy on bait loaded DMD is higher than others. Therefore loading of DMD is obligatory.

Abstract

Red palm mite, Raoiella indica Hirst (Acari: Tenuipalpidae), has emerged as a new threat to coconut plantation in Indonesia. To control this mite, understanding on its biology is essential. The effects of three coconut varieties (Salak Dwarf, Local Sumenep Tall, and Hybrid coconuts) on the development and fecundity of R. indica were determined under laboratory conditions at 26−28oC, 80% RH, and photoperiod of 12:12 (L:D). The life cycle of R. indica consisted of the egg, larva, protonymph, deutonymph, and adult stages. Total development time of immature females was significantly influenced by coconut varieties. The shortest immature developmental time occurred at Salak Dwarf variety (26.07 days), whereas no significant difference of development time of immature female on Hybrid and Local Sumenep Tall varieties was found. Moreover, there was also no significant difference on the development time of immature male and on the fecundity of the mite among different coconut varieties. IntisariTungau merah kelapa, Raoiella indica Hirst (Acari: Tenuipalpidae), merupakan ancaman baru bagi perkebunan kelapa di Indonesia. Pemahaman mengenai biologi diperlukan untuk strategi pengendalian tungau tersebut di masa datang. Pengaruh tiga varietas kelapa yang berbeda (Genjah Salak, Lokal Sumenep, dan Hibrida) terhadap perkembangan dan keperidian R. indica diamati pada kondisi laboratorium dengan suhu 26−28oC, kelembapan nisbi 75−85%, serta pencahayaan 12 jam terang dan 12 jam gelap (L:D=12:12). Siklus hidup tungau terdiri dari telur, larva, protonimfa, deutonimfa, dan dewasa. Hasil pengujian menunjukkan bahwa waktu perkembangan betina pradewasa dipengaruhi secara nyata oleh varietas kelapa. Waktu perkembangan paling pendek terdapat pada kelapa Genjah Salak (26,07 hari), sedangkan waktu perkembangan betina pradewasa tidak berbeda nyata pada kelapa Hibrida dan Dalam Lokal Sumenep. Hasil lain pengujian menunjukkan tidak terdapat perbedaan nyata waktu perkembangan jantan pradewasa dan keperidian pada ketiga varietas.

Abstract

Geminivirus infection has caused severe losses on various economically important crops. The disease incidence on melon has been observed since 2004, and widespread at melon production centers in East Java and Special Province of Yogyakarta. In East Java and Special Province of Yogyakarta, the Disease Incidence of leaf curl on melon  reached 100% and 14.3 % respectively in 2008. Detection of the causal agent using primers of CPA5 and CPA2 resulted viral specific DNA fragment of 770 bp.

Abstract

One of the important disease on snakefruit (Salacca edulis) is fruit rot disease with white symptom. Control of snake fruit diseases is better when natural compounds rather than chemicals pesticide were used which might cause consumer hazard. Several of plant extract i.e. from Zingiberaceae which have antimicrobial activity had been studied. The purpose of this study were to identify the pathogen of white rot and to know the effect of torch ginger’s flower (Nicolaia speciosa) extract on disease development. The spores was identified under microscopic condition from diseased fruits and isolates. Torch ginger’s flowers was extracted in ethanol. After removal of the ethanol the crude extract was dissolved with distilled water. Food Poisoned Technique was done to evaluate its influence in vitro. Both detached and clustered fruits were sprayed with the extract then inoculated with the pathogen. The result showed that the pathogen was Chalaropsis sp. Detached snakefruit was more susceptible to Chalaropsis sp. infection than those in cluster. In vitro test showed 48–50 % of torch ginger’s flower extract in water inhibit 90% of colony growth. The extract inhibited the development of Chalaropsis sp. in detached snakefruit or in cluster ones. Concentrated extract protected snakefruit up to 100% from Chalaropsis sp. infection.

Abstract

Laboratory and field-cage experiments were conducted at the Universiti Pertanian Malaysia. Objective of the study was to determine the functional response of Menochilus sexmaculatus Fabricius (Coleoptera: Coccinellidae) predating on chilli aphid Aphis gossypii Glover (Homoptera: Aphididae). The larvae and adults of the predaceous coccinellid exhibited the Hulling's Type II functional response. A first instar of the coccinellid predating on single first instar of the aphid within 24.6 minutes in the laboratory and 16.8 minutes in the field. While a fourth instar and an adult of the coccinellid predating on single fourth instar of the aphid within 4.2 and 1.5 minutes respectively.

Abstract

The effectiveness of a synthetic-pheromone trap with active ingredient of ethyl-4-methyloctanoate in catching adults of Oryctes rhinoceros was believed to be influenced by the trap color. A RCBD trial with single factor and three block replicates was conducted during September to October 2008 at Oil-Palm-Trial Station at PTPN III, Labuhan Batu District, North-Sumatera Province. The single factor was trap color i.e. orange, green, white, blue, red, yellow, and pink. Results showed that total numbers and sex ratios of collected O. rhinoceros adults were not significantly influenced by the trap color. The total numbers of collected O. rhinoceros adult during 30 days on color of orange, green, white, blue, red, yellow, and pink were 136, 177, 126, 155, 129, 114, and 113 individuals with sex ratios ♂/♀ 0.47, 0.52, 0.62, 0.50, 0.60, 0.49, and 0.54, respectively. The synthetic pheromone significantly attracted more O. rhinoceros female (65%) rather than the male one (35%).

Abstract

Bacterial wilt disease caused by Ralstonia solanacearum destroys many crops of different plant families in South East Asia despite many researches about the disease, and the availability of developed control method in other parts of the world. There is no chemical available for the bacterial wilt pathogen and biological control is then chosen as an alternative to save the crops. Most of the biological control studies were based on antagonism between biological control agent and the pathogen. The biological control agents were intended to reduce the initial inoculum of the pathogen. The effort to minimize the initial inoculum of the pathogen by baiting with the use of hypersensitive host-plant was only reliable when conducted in the greenhouse experiments. Various microorganisms have been searched as possible biological control agents, for instance avirulent form of the pathogen, soil or rhizosphere bacteria (Bacillus spp. and fluorescent pseudomonads), actinomycetes (Streptomyces spp.), yeast (Pichia uillermondii, Candida ethanolica), and a consortium of microorganisms known as effective microorganisms (EM). None of these biological control agents has been used in field application and they need further investigation in order to effectively control bacterial wilt. Opportunities and challenges in developing biological control to combat bacterial wilt are discussed in the paper.Penyakit layu bakteri yang disebabkan oleh Ralstonia solanacearum menghancurkan banyak tanaman dalam famili yang berbeda di Asia Tenggara meskipun telah banyak penelitian tentang metode pengendaliannya. Penyakit ini sulit dikendalikan karena banyaknya variabilitas patogen dan belum tersedianya sumber ketahanan yang mapan. Di samping itu, sampai saat ini belum ada bahan kimia yang tersedia untuk patogen layu bakteri ini sehingga pengendalian biologi kemudian dipilih sebagai cara alternatif untuk menyelamatkan tanaman. Sebagian besar penelitian pengendalian biologi didasarkan pada antagonisme antara agen pengendalian biologi dan patogen. Agen pengendalian biologi tersebut dimaksudkan untuk mengurangi inokulum awal patogen. Upaya untuk meminimalkan inokulum awal patogen dengan umpan dengan menggunakan tanaman inang sangat rentan hanya dapat diandalkan ketika dilakukan dalam percobaan rumah kaca. Berbagai mikroorganisme telah diteliti kemungkinannya sebagai agensia pengendalian biologi seperti bentuk avirulen dari patogen, bakteri tanah atau bakteri rizosfer (Bacillus spp. dan pseudomonad fluorescen), actinomycetes (Streptomyces spp.), khamir (Pichia guillermondii, Candida ethanolica), dan konsorsium mikroorganisme yang dikenal sebagai EM (Effective Microorganisms). Meskipun demikian tidak satupun agensia pengendalian biologi ini sampai pada taraf aplikasi lapangan sehingga diperlukan penelitian lebih lanjut. Peluang dan tantangan dalam mengembangkan pengendalian biologi untuk memerangi penyakit layu bakteri dibahas pada tulisan ini.

Abstract

Plutella xylostella (L.) (Lepidoptera : Plutellidae) is key pest on cabbage. P. xylostella can be infected by Beauveria bassiana from other insects. The objective of this study was to determine the pathogenicity five isolates of B. bassiana on P. xylostella. Those isolates were BbUj1 (larvae of Lepidoptera), BbUj2 (rice brown plant hopper), BbUj3 (rice brown plant hopper), BbUj4 (rice seed bug), and BbUj5 (rice black bug). In the laboratory every isolate was inoculated 5 µL spore suspension at rate 107 spores/mL to third instar P. xylostella larva used micro liter syringe. Each treatment used 10 larvae and was replicated five times. The pathogenicity of B. bassiana was determined based on mortality and LT50 value. The potential isolate was tested to find LC50 subsequently tested on cabbage crop in the green house. The result of the study showed that BbUj1 from larvae of Lepidoptera was the potential isolate with LC50 of 9.49 x 10^5 spores/mL. LT50 was 2.67 days for the concentration 107 spores/mL. Green house application of B. bassiana at rate of 5 x 10^12 spores/ha caused 57 percent mortality of P. xylostella larva in seven days after treatment and LT50 was 5.67 days.
