Abstract

Actinomycetes  are   one  kind  of  the  microorganisms  that  very  important producer  of   secondary  metabolites  for  drugs.Active substances  of  microbial origin  have  been  searched   through  a  series  of  screening  methos  to   obtain novel  compounds  and  strains.  The  purpose  of  this  research  was  to  characterize the  antibacterial  coumpound   from  actinomycetes  associated  sponge  and identification  of  actinomycetes  base  on  morfology  and  fisiology  characteristic. Isolation of actinomycetes from sponge were done bypour and spreap plate. A total  of   actinomycetes  strain  were  isolated  from  sponges  collected   at  Barrang Lompo  island,  Makassar,  South  Sulawesi,   Indonesia. Abstract

Sauropus androgynus Merr. and uyup-uyup have been used as lactagogue traditional medicine since long time ago, but the pharmacological mechanism was unclear. This research has been done to determine prolactin induction of S. androgynus Merr and uyup-uyup as lactagogue agents. S. androgynus Merr leaf extract and uyup-uyup were given to the pigeon for 7 and 14 consecutive days, the epithelium tunica mucous of ingluvies glands induced by prolactin was then determine. The result showed that S. androgynus Merr. leaf extract and uyup-uyup have lactagogue effect as determined by prolactin induction. The induction of prolactin after S. androgynus Merr leaf extract administration (dose 54 mg/kg Body Weight (BW) for 14 days comparable to that of moloco B12 (dose 189 mg/kg BW) after 7 days administration (p<0.05).Key words : lactagogue, prolactin, tunica mucous

Abstract

Effects of drying methods in obtaining the extractive, phenolic content and antioxidant activity in Phyllanthus niruri Linn. herbs have been investigated. The drying  methods  tested  were  air-drying  at  ambient  temperature,  oven-drying  at 40  °C,   oven-drying  at  60  °C,  and  fresh  samples  as  control.  Results  revealed that  drying  of  the  fresh  plant  caused  the  decrease  of  extractive  obtainability, phenolic  content  and  antioxidant  activity.  There  were  significant  differences among drying  methods (p < 0.05). Abstract

Respiratory tract infections are the worldwide main cause of death in children aging less than 5 years. In developed countries; respiratory tract infections are the leading cause of morbidity, accounted for 20% of medical consultation, 30% of work absenteeism and 75 % of all antibiotic prescription. Upper respiratory tract infections in children, which often associated with viral infection, and in average about 40 – 60 % of children with upper respiratory tract infections were prescribed with antibiotics. The type of antibiotics also varies from hospital to another.The objectives of this study are to describe the pattern, outcome, and cost of antibiotic usage in pediatric respiratory tract infections. A prospective study of pediatrics admitted to Pediatric Ward, Penang Hospital for respiratory tract infections was carried out. Appropriate descriptive analysiswas used. The results showed that penicillin, erythromycin; amoxicillinclavulanate were the most commonly prescribed antibiotics for respiratory tract infections. The average duration of antibiotic therapy 3.0 ± 3.2 days, fever clearance time 2.2 ± 2.4 days, improvement of sign and symptoms 3.7 ± 2.5 days and mean length of stay in the ward was 4.9 ± 2.8 days. The average cost of antibiotics was RM 14.83 ± 83.13, and total cost of respiratory tract infections treatment was RM 98.88 ± 65.87.Key words: antibiotic, respiratory tract infection, pediatric, hospital

Abstract

Diabetes mellitus is a chronic disorder that has been recognized by Indonesian government as a major public health problem with far reaching consequences not just for its adverse impact on the health of Indonesian, but also for the economic burden it places on the health care systems. The objective of this study was to describe the therapeutic cost of outpatient diabetes mellitus type 2.A descriptive study was performed on patient who were admitted to the Department of Endocrinology of the Sardjito Hospital between July and August 2005. Patient were included if they were ambulatory during the time of the study. Exclusion criteria were patients with any other diseases. Only direct medical cost was considered (perspective from hospital). Direct costs to this analysis were cost of drugs, visit to the physician, laboratory examinations, and cost of diabetes-related complications and concomitant therapies.From the 100 examined patients, 44 percent were females and 56 percent were males. The average age of the patients was 60.75 ranging from 41 to 85 years of age. The average total costs per patient per month was Rp 208,500 with a maximum to be Rp 754,500. The largest cost was drug acquisition costs (59.5%), followed by cost of diabetes-related complications (31%). The control of blood glucose using combination therapy was more frequently attained in patients taking sulfonylurea and biguanid (44.62%). The combination of biguanid, α-glucosidase inhibitor, and insulin had the greatest expense, equal to Rp.571,000. Hypertension, neuropathy, and hyperlipidemia were the most frequently mentioned complications diseases. The cost for diabetes-related complication included hypertension and retinopathy had the greatest expense, equals to Rp 754,500.Key words: cost-identification, diabetes mellitus, Dr. Sardjito hospital

Abstract

Satyrium  nepalense  is  an  endangered  medicinal  herb  found at  the higher altitude of 2400-5000m. Local inhabitant used tubers of  Satyrium  nepalense  as  an  energetic  tonic  and  as  an  important medicine  to  cure  different  type  of  fever  in  traditional  health  care system  of  Uttarakhand  (INDIA).  Present  study  was  carried  out  to evaluate  the  phytochemical,  antibacterial  screening  against  four (Streptococcus  mutans,  Pseudomonas  aeruginosa,  Staphylococcus aureus  and  Klebsiella  pneumoniae)  microorganisms  in  methanolic extracts  and  habitat  studies  for  conservation  of  S.  nepalense. Tubers  of  Satyrium  nepalense  were  extracted  separately  with methanol  by  hot  extraction  process  using  soxhlet  apparatus.  The extracts  were  subjected  to  Lyophilization  to  get  dry  extract  and preserved  in  aseptic  condition.  The  different  group  reagents  used for  phytochemical  screening  as  dragendorffs’s  for  alkaloid, molisch’s  test  for  carbohydrates,  shinoda  test  for  flavonoids  etc. Antibacterial  study  was  carried  out  by  disc  diffusion  method.  The highest  zone  of  inhibition  was  recorded  as  15.0±0.00  mm  against Klebsiella  pneumonia  and  15.0±0.82  mm  against  Staphylococcus aureus.  Phytochemical  screening  shows  the  presence  of  alkaloids, carbohydrates/glycosides,  flavonoids  and  unsaturated  sterols/triterpenes  in  Satyrium  nepalense.  The  result  indicates  that methanolic  extract  of  Satyrium  nepalense  shows  potent antibacterial activity against all four bacterial strains.Key  words:  Uttarakhand,  Satyrium  nepalense,  medicinal  orchid,  antibacterial, phytochemicals

Abstract

Hydrotrope solutions provide safe and effective media for the microwave assisted extraction of andrographolide, the major bioactive chemical constituent of plant Andrographis paniculata. Microwave assisted extraction of androgpaholide was carried out by using hydrotropes, sodium benzoate and urea. The objective of this work were to determine the Minimum Hydrotrope Concentration (MHC) and to determine the effectiveness of each hydrotrope with respect to andrographolide at different system powers. The microwave assisted extractions of andrographolide were carried out at different concentration of hydrotropes (0.2-3M) and different system powers (39.9 and 119.7 Watt). Twenty grams of Andrographis paniculata dried powder were added to 200 ml of hydrotrope solutions and extracted in a microwave extractor for 15 minutes. The research result showed that the percentage of the microwave assisted extraction of andrographolide by using sodium benzoate and urea were up to 10.9% and 1.05% respectively. The Minimum Hydrotrope Concentration and the effectiveness of hydrotropes that was measured in term of Setschenow constant (Ks) were reported for two hydrotropes used in this study.

Abstract

Eurycomanone is a cytotoxic ingredient found in Eurycoma longifolia Jack. Previous studies have noted its activity against many epithelial cell lines. In this study, eurycomanone had obvious cytotoxic effect on HeLa cells by methylene blue staining assays. After HeLa cells were treated with eurycomanone, typical morphological changes, including cytoplasm shrinkage and decrease of cell volume were observed by light microscope. Chromatin condensation and nuclear fragmentation could be observed by fluorescence microscope after staining with Hoechst 33258 nuclear staining. DNA fragmentation and apoptotic body formation that is characteristic of apoptosis could be determined after treated cells were assayed with TUNEL (terminal deoxynucleotidyl transferase mediated d-UTP nick end labeling). The externalization of phosphatidyl serine (PS) could be determined by flow cytometry analysis using Annexin-V/PI double staining. The result suggested that eurycomanone exerted antiproliferative activity on HeLa cells by inducing apoptosis.Key words : eurycomanone, cytotoxic, apoptosis, HeLa

Abstract

Curcumin analogues 2,5-bis-(4’-hidroxy-3’-methoxy)-benzilidinecylopentanone (PGV-0) and 2,5-bis-(4’hidroxy-3’,5’-dimethyl)-benzilidinecylopentanone (PGV-1) have a potency to be developed as cytotoxic agent. The aims of this research are to elucidate the geometric isomer and to study the cytotoxic effect on T47D cells of both compounds. To establish the geometric isomer these compounds, they were elucidated by LC-MS, 1H-NMR, 13C-NMR, HMBC, HMQC, NOESY. Their cytotoxic effect were evaluated by MTT assay method on T47D cells. The results concluded that the geometric isomer of PGV-1 is zusammen-zusammen (Z-Z) and PGV-0 is entgegen-entgegen (EE). The IC50 of both compounds are 1.74 and 9.39 μM respectively.Key words: PGV-0, PGV-1, Cytotoxicity

Abstract

Initial characterization, the MIC, Fe uptake and detection of siderophores, of the resistance K.pneumoniae mutants against BRL41897A (KSL mutants) showed that there were differences amongs them. Therefore it was necessary to observe the OMP and IROMP proteins of the mutants especially which had different MIC’s to the WT galur (M10). Results from SDS-PAGE analysis showed that KSL19 produced 49kDa protein weakly and similar result was found in the 22 kDa protein production by KSL38, KSL52, KSL58 and KSL59. Using Fe-CAA media -media lack of Fe - showed that 3 mutants synthesized certain protein weaker than M10 galur, KSL19 in the production of the 88 kDa protein, KSL38 and KSL59 at the 80 kDa protein. We also observed that KSL19 synthesized new 88 kDa protein. This result showed that certain mutant which had decreased production of one protein could stimulate another weak protein.Key words: SDS-PAGE – IROMP – K.pneumoniae mutants – BRL 41897A.
