Abstract

Optimalization protoplast isolation of the leaves mesophyll orchid Paraphalaenopsis layockii. The obyectives of the research was to investigate the effect of combination treatment of sucrose concentrations and incubation time on protoplast isolation of the leaves mesophyll orchid of Paraphalaenopsis layockii. The factorial experiment consisted of two factors that was arranged by completely randomized design (CRD) with ten replications. The first factor was concentrations of sucrose which consists of three levels namely 0.4 M, 0.5 M and 0.6 M. The second factor was incubation time which consists of four levels namely 2 hours, 4 hours, 6 hours and 8 hours. Data were analyzed using Kruskal-Wallis test followed by Mann-Whitney test. The results showed that there was effect of treatment for all variables observed. The highest protoplast density and number of viable protoplasts achieved in the combined treatment of 0.6 M sucrose and 4 hours of incubation time.

Abstract

Hybrid Rice Planting Assessment in Purwakarta Hybrid rice planting assessment was conducted at lowland area in Wanasari village, Wanayasa subdistrict, Purwakarta district during second dry season 2008. Seven varieties of hybrid rice namely Mapan 02, Pioner PP II, SL 8 SHS, Pioner PP I, HIPA 3, HIPA 4 and HIPA 6 were evaluated to find out hybrid rice varieties that are suitable to the agroecosystem. The observations were done for number of tillers, number of panicle per hill, number of productive panicle per hill, panicle length, number of filled-grain per panicle, number of unfilled-grain per panicle, plant height, and yield. Descriptive statistics analysis showed that the most number of panicle was 17.67 on SL 8 SHS and the least was 8.67 on HIPA 3, while the highest number of productive panicle was 15.67 on Pioner PP I and the least was 6.3 on HIPA 3. The longest of panicle was 6.90 cm on Pioner PP I and the shortest was 6.36 cm on HIPA 3. The highest number of well-filled grain was 75.18 on Pioner PP I, but it has 195.80 unfilledgrain. The lowest number of well-filled grain was 52.88 on HIPA 3 and the lowest unfilled-grain was 108.40 on Mapan 02. The highest plant was HIPA 6 with the height of plant was 93 cm, and the lowest was Mapan 02 with the height of plant was 78.06 cm. While the most number of tillers was 21.8 on HIPA 6, and the lowest was 15.88 on Pioner PP II. The highest yield was 5.84 t ha-1 on Pioner PP I and the lowest was 3.76 t ha-1 on SL 8 SHS. According to agronomic characters and the yield, the most appropriate rice hybrid to plant at Wanasari village, Wanayasa sub-district, Purwakarta district during second dry season was Pioner PP I.

Abstract

Optimization of Rice Production in Tidal Swampland.The study aimed to obtain the optimumtechnology of tidal swamplands for rice farming sustainability, acquired some adaptive cultivars in tidalswamplands, assessed the effect of land salinity to support rice Planting Index. The experiment wasconducted inTelangrejo tidal land, South Sumatra from February to November 2011. The study wasconducted with a split plot design with three replications. As Main plot were Amelioration and fertilization( M ) ; M1. Cultivation techniques used by farmers, M2. No-tillage ( TOT ) with inputs (lime wash + 3t / ha + fertilizer ), M3. Minimum tillage with input ( leaching + lime 3 t / ha + husk 3 t / ha + ZnSO4 +fertilizer ). Sub Plot were rice varieties (V); V1. Sintanur, V2. Inpara 1, V3. Inpara 2, V4. Inpara 4,V5. Inpara 5. Observations were made: 1 ) Plants growing, 2 ) grain yield and yield components.Multiple regression analysis showed that there were highly significant component effect on grain yield;the amount of total grain , the grain emptiness, and the weight of filled grains of 1000, where these threeoutcome components were also influenced by the treatment given of Amelioration. Some of varietieswere tested have different yields. The highest grain yield was Inpara 4 of 3.42 t / ha, followed by Inpara2 of 2.89 t/ha, while Inpari 1 showed a higher response for tidal land improvements, there weredifferences in responses between varieties. Although that was not statistically significant, the Ameliorationwas affect to the growth of rice, but the provision of husk and the addition of ZnSO4 were not affect tothe tidal rice in Tegalrejo.

Abstract

This research aims to find the best condition for the cultivation of fragment of Caulerpa lentilifera, to determine the factors that influence its growth in-vitro, and to identify the types of microbes associated. The study was conducted at the Marine Tech Laboratory and Marine Biology Laboratory of Udayana University. The study begins with the in-vitro culture of C. lentilifera seaweed at the incubator and the tank. The observations of the morphology and the seaweed growth to determine the suitable conditions, the influens of growth factors and microscopic observation to determine the types of microbes were associated. The best results in in-vitro culture in incubator was achieved by provision of 2 ml of PES in 500 ml of sea water, 5000 lux of light intensity, 12 hours irradiation and 100% medium replacement once a week. Best results in-vitro culture in the using net method was achieved by provision of 3500 lux light intensity, 12 hours irradiation and 70% culture replacement once a week. The microbes associated with the C. lentilifera came from the protozoa phylum, flagellate, and cilliata classes.

Abstract

Identification and Characterization of Genetic Resources of Local Fruits inTabanan. The fruit is a source of vitamins and minerals that ensure the metabolic processesin the human body, some of which can also be used as medicines, cosmetics, SPA (massage)social and culture as a means to use for the material of offerings. Tabanan regency, as well asother areas in Bali has a variety of fruit but the resources still underutilized. This researchaims to identify the type of fruit genetic resources in Tabanan, profile the genetic resourcesregarding the morphology of fruit and agronomic characters, as well as to compile thegeographical spread map and diversity of fruits. Identification and characterization as well asthe distribution map is very important for the conservation, development and utilization ofgenetic resources in Tabanan Regency. The results of the research conducted in Tabanan,found 31 type and 52 subtype local fruit, with 6 different types of eminent fruit, 15 kinds ofrare fruits, 12 kinds of fruit that usually use for upakara and 4 types of fruits that are use asmedicine. Based on the results of the survey conducted in tabanan then it can be possible tocomposed 34 geographical local fruit spread map.

Abstract

Efforts to Increase Production and Quality of “Kristal” Guava Variety (Psidium guajava L. cv. Kristal) by Fertilization. Nowadays “Kristal” guava variety (Psidium Guajava L. cv. Kristal) is very popular for consumers because it has several advantages such as flesh fruit is crunchy, sweet-sour taste with slightly seeds or seedless, and shelf life of fruit relatively long that can be up to 1 month if stored at temperature of 10-15 oC. The challenge faced by farmers are the production and fruit quality still low, due to inadequate maintenance done. This research was conducted to study the effect of fertilizer to increase the production and quality of “Kristal” guava variety. Research was carried out in farmers garden at Pelaga Village, District of Petang, Badung Regency, Bali, from April-November 2016. The study was a randomized block design (RBD) with fertilizer treatment consisted of 4 levels and each repeated 6 times. The results showed that fertilization increase production and quality of “Kristal” guava fruit compared to the control. The highest weight of fruit per plant (18.28 kg / plant) were obtained on plants fertilized with compost 5kg/plant + urea 200 g/plant + KCl 250 g/plant + TSP 250 g/plant + gypsum 500 g/plant+ micro fertilizer CuSO4 and ZnSO4 1.5 g/plant, respectively, given once at the beginning of the study and significantly different compared to the lowest weight of fruit per plant which obtained on controls (8.99 kg/plant). The highest fruit diameter (8.72 cm) obtained on plants fertilized with compost 5kg/plant + urea 200 g/plant + KCl 250 g /plant + TSP 250 g / plant + gypsum 500 g/plant + micro fertilizer CuSO4 and ZnSO4 each 1.5 g/plant, respectively, given three times i.e at the beginning of the study, at the phase of anthesis, and at the maturity of fruit and significantly different compared to control (7,50 cm).

Abstract

The Effect of Organic and Inorganic Fertilizers on The Growth and Results of Green Mustard (Brassica rapa L.). Green mustard (Brassica rapa L.) is a vegetable that preferred by many consumers, this causes many farmers cultivate this vegetables. One of the important factor in the cultivation of green mustard is fertilizer type. Alternative to improve soil quality and fertility is by providing organic matter. A field research was conducted to evaluate the effect of organic and inorganic fertilizers on the growth and yield of green mustard, in Banjar Taman Tande, Baturiti Village, Bedugul from April to June 2018. This study was designed with a Randomized Block Design with nested patterns consisting of two factors. The first factor was the type of fertilizer, which was consisted of four types of organic fertilizer, namely: vermicompost, cow manure, chicken manure, goat manure and inorganic fertilizers (NPK Mutiara). The second factor was fertilizer dosage, which was consisted of three levels, namely: 0 tons/ha, 15 tons/ha, and 30 tons/ha, while for NPK Mutiara fertilizer dosages, namely: 0 kg/ha, 150 kg/ha, and 300 kg/ha.The results showed that the effect of organic and inorganic fertilizers were not significantly different on the growth and yield of green mustard plants and RAE values. The total N content of soil on vermicompost treatment was 0.23% equal to NPK Mutiara. The total population of soil microbes on vermicompost treatment under dosage of 15 tons/ha and 30 tons/ha were 1.4 x 106 cfu/g that was much higher compared to NPK Mutiara 150 kg/ha fertilizer (0.3 x 106 cfu/g).
