Abstract

The purpose of this study was to evaluate Taenia saginata cystisercus antigen for the detection of bovine cysticercosis. Taenia saginata cysticercus antigen was derived from local isolates, obtained from the experimental infection of Taenia saginata tape worms from Bali. The research was done by ELISA (Enzyme Linked Immunosorbent Assay) optimized by determining the optimal concentration of antigen, the optimal dilutions of serum and the optimal dilutions of conjugate. The results showed that Taenia saginata cysticercus crude antigen (Bali isolate) are antigenic and can be used to detect cattle cysticercosis. Optimal concentration of antigen: 2 ug/ml, optimal dilutions of serum: 1:80 and optimal dilution of conjugate: 1:4000.

Abstract

The purpose of this study was to examine the effect of the extract of Moringa oleifera leave on Argulus sp infestation in comet fish, and determine an optimal concentration and time. Moringa oleifera leave was extracted using ethanol 96%. In total, 25 comet fishes (Carassius auratus auratus) were used in this research, in which infested by one to five individuals of Argulus sp per fish. This research used pretest and posttest design with four treatments of extract concentration of Moringa oleifera leave, namely, TI 25 mg/l, TII 37.5 mg/l, TIII 50 mg/l, TIV 62.5 mg/l, and T0 without extract as a control. Each treatment was imposed on five comet fishes. The parameter of this study was the amaount of Argulus sp released from the body of the fish. The results showed that the extract of Moringa oleifera leave had an effect on ectoparasite on comet fish. There was an indication that the more concentration of the extract of Moringa oleifera leave, the more Argulus sp released, although statistically, it was not significant (P>0.05). The optimal time of giving the extract of Moringa oleifera leave to Argulus sp infestation was 12-h.

Abstract

The purpose of this study was to evaluate the ELISA test using field serum and to determine the incidence rate of cysticercosis of T. saginata in Bali. Serum’s sample obtained from Bali cattle that slaughtered at the abattoir and Bali cattle owned by the farmers. The result of sera examination showed that 237 (87.7%) were positive of antibodies of cysticercus of T. saginata. Furthermore, as many as 90 (33.33%) of the 270 Bali cattles, their feces were also taken for examination of worm eggs. The results of stool examination showed that 80 (88.9%) were infected with trematodes and 14 (15.5%) were infected with a mixture of trematodes and nematodes. By comparing the results of ELISA and stool examination using the sensitivity and specificity approach or Table 2 x 2, the results showed that there was a cross-reaction between Cysticercus of T. saginata and trematode. Further effort to purify the antigen of cysticercus of T. saginata is still needed to improve its sensitivity and specificity.

Abstract

The observational study was directed to find out the number of populations, large of habitat, home range, and density of long tailed macaques (Macaca fascicularis) in Badung Peninsula. Population site was recorded using GPS program. Data were analyzed using ArcView 3.2 and GmapCatcher programs. There were four population sites of long tailed macaques, namely Pura Dalem Karang Boma, Pura Gunung Payung, Pura Batu Pageh, and Pura Luhur Uluwatu. The average of population density was 114 individuals per hectare and the average of home range was 852,5 hectare. The study indicates that the population densities of long tailed macaques in Badung Peninsula are very high and their home ranges are very large. It is suggested that the population growth of the long tailed macaque in Badung Peninsula must be controlled for decreasing the negative impact.

Abstract

Bali province has been declared as rabies endemic area. Bali government has already implemented several efforts to control rabies, but it does not have any significant impact to make Bali free from the disease. This condition is not caused by limited resources only, but also by factors of socio-demographic dog’s ownership. This observational study using cross sectional design has been done to determine the public's knowledge and dog’s ownership related factors in Karangasem Regency. Nine villages were selected based on the positive rabies cases, which was formerly confirmed by laboratory tests. The Villages were divided into three category based on the socio-economic and culture, namely urban, sub urban and rural area. Data were analyzed descriptively and by multivariate logistic regression analysis. The results showed that from the 408 households, 252 respondents (61.8%) reported having the dogs. The percentage of dog ownership in urban, sub urban and rural is 17.6%, 9.8% and 34.3% respectively. It can be suggested that the rural area should be a priority in controlling rabies in Karangasem Regency.

Abstract

Fluorescent Antibody Test (FAT) is a golden standard for rabies diagnosis, but it requires many sample preconditions before testing. Immunohistochemical technique (IHC) has the same principle as the FAT, however, it is more simple. This study was conducted to examine the sensitivity and specificity of IHC compare to FAT. The sensitivity and specificity were calculated using the formula of 2 x 2 contingency table and statistical analysis of the chi-square. Out of 40 samples that rabies positive on FAT, 27 samples (67.5%) showed rabies positive on IHC and 13 samples (32.5%) showed rabies negative on IHC. All samples that rabies negative on FAT were also rabies negative on IHC. The IHC has the sensitivity and specificity level 67.5% and 100% respectively. The IHC has a lower sensitivity than FAT, however, IHC has many advantages such as it can be performed on formalin fixed-samples. A further research is necessary to improve the sensitivity of IHC on detecting the rabies.
