Abstract

D.I. HARTOTO. 1986. Local and spatial distribution of Puntius binotatus and Rasbora lateristriata in Ci Taman Jaya and Ci Binua, Ujung Kulon. Berita Biologi 3(6): 261 - 267. A study of local and spatial distribution of the "beunteur" (Puntius binotatus) and the "paray" (Rasbora lateristriata) on two streams at the vicinity of Ujung Kulon National Park was conducted during dry (August 1983) and rainy season (January 1984).This study as a part of a sets of autecological studies, was aimed to reveal the differences of local and spatial distribution pattern of the two species. The results indicated that the pattern of distribution of paray were not significantly differed from beunteur, however, it was clearly shown that there are certain centers of local distribution for each species. It was also shown that the upstream segments were dominated by the beunteur and the downstream/segments by the paray. Observation on the spatial .distribution yield that the beunteur dominated the calm water habitat resources and paray occupied the rapidly moving water habitat resources.The possibilities of control mechanism of each pattern of distribution from time. to time;including inter and intraspecific competition, substrate preferences and others; were briefly discussed.

Abstract

Pruatjan (Pimpinellapruatjan Molk.) is an Indonesian endangered medicinal plant that included in Appendix I based on CITES. Therefore it is a highly protected species. To avoid extinction of this plant, it is very important to conserve the plant. In vitro conservation is more suitable since this plant is difficult to be cultivated outside of its habitat. Cryopreservation technique may conserve this material for a long-term period. The objectives of this research were to find optimized treatments for pre culture, loading, and dehydration on cryopreservation of pruatjan. The research was conducted at Tissue Culture Laboratory in Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development, started from May to November 2007. Pre culture was conducted using DKW basal media that added by sucrose at the level of 0.3,0.4, and 0.5M for one and three days incubation. Loading was conducted in DKW basal media containing 2M glycerol and 0.4M sucrose for 15,30, and 45 minutes duration time. Dehydration was conducted in several cryoprotectants, namely PVS1 (22% glycerol + 13% propy lene glycol + 13% etylene glycol + 6% DMSO + 3% sucrose), PVS2 (30% glycerol + 15% etylene glycol + 15% DMSO + 0,4M sucrose), PVS3 (50% glycerol + 50% sucrose), and PVS4 (35% glycerol + 20% etylene glycol + sucrose 0.6M). Result showed that pruatjan could be preserved through cryopreservation by vitrification method. The best pre culture was using 0.3 M sucrose for one day, the best loading was 30 minutes, while the best cryoprotectant was PVS2 with 90% success before freezing and 40% after freezing. The success may be improved by applying pre growth treatment, optimizing temperature of thawing, modification of recovery media and incubation condition.

Abstract

Leaves anatomy of Averrhoa spp. in Indonesia have been studied for supporting morphological character which used to recognize four species: A. carambola, A. bilimbi, A. leucopetala and A. dolichocarpa. The letter two species have leave crowded terminally or near the top of stem similar to those leave of A. bilimbi, whereas the fruit shape similar to A. carambola.The result of this study indicated that, these four species showed differently in their thickness of lamina and epidermis cell as well.

Abstract

Nitrogen fixing dan phosphate solubilizing bacteria are important PGPR bacterial in addition of plant nutrients by increasing N and P, especially in saline soils. Experiments were conducted to observe the effects of  soil salinity and bacterial inoculant on growth eggplant (Solanum melongena) at green house of  Microbiology division, Research Center for Biology, The Indonesian Institute of Sciences,  Cibinong. Experiment laid out as factorial based randomized complete design  with three replications. Five levels of watering consisted of (1). Fresh water, (2). NaCl 0.1%, (3). NaCl 1%, (4). Sea water 75 %+25% fresh water, (5). Sea water 100%  as first factors. Five levels of inoculations/biofertilizers consisted of (1).control without fertilizer (K), (2). NPK (P), (3). Nitrogen Fixing bacteria (BPN1), all treatments was repeated 3 times  BPN2, mixed inoculation (BPN1 +  BPN2 + Phosphate Solubilizing Bacteria/BPF) as second factors.  Results showed that biofertilizer application increased growth and yield of eggplant in various levels of salinity at about 21.56%-53.35%  compared to uninoculated plants

Abstract

Penelitian mengenai aspek-aspek biologi remis marga Corbicula yang ada di Indonesia dapat dikatakan masih amat sedikit.Beberapa aspek yang telah dikerjakan antara lain mengenai sistimatiknya (Benthem Jutting 1953 dan Djajasasmita 1977) dan bebeiapa aspek ekologinya oleh Budiman & Djajasasmita (1979). Tulisan ini menguraikan hasil penelitian lanjutan Budiman & Djajasasmita (1979) dengan tekanan pada aspek-aspek sifat kelamin, pemijahan dan pertumbuhan larva remis Corbicula javanica dari perairan sekitar Bogor.

Abstract

A haploid breeding program was initiated to develop doubled haploid blast tolerance rice breeding via anther culture.Blast tolerant variety (Laka) was crossed with Kencana Bali (sensitive to blast).Anthers from F1 were cultured on two kind of media (combination of N6 macro salt and MS micro salt) containing 1 mg/l NAA + 0,1 mg/l kinetin called medium-1 and another one containing 1 mg/l 2,4-D + 0,1 mg/l kinetin called medium-2.Anthers were treated with cold shock (4 °C)for 5 and 10 days before cultured.Results of experiment showed that F1 plants derived anthers were able to form call Number ofplantlets were produced during anthers culture consisted of 482 green plants and 50 albinos. Most of green plants did not produced root.Root growth of plantlets were induced with 1.5-4.5 mg/l IBA in MS medium.

Abstract

This experiment aimed to find out the optimum density combinations of prawn (M. rossenbergii (de Man, 1879) and fish of kissing gouramy (Helostoma temminckii Cuvier, 1829) in the polyculture based aquaponic fish culture system. There are four treatments and three replications applied in the Completely Randomized Design experiment. The treatments were the combination of giant freshwater prawn (U) and kissing gouramy (T) densities for one square meter (m2), namely (A) 30 prawns and 100 gouramy (U30T100); (B) 20 prawns and 50 gouramy (U20T50); (C) 30 prawns and 50 gouramy (U30T50) and (D) 20 prawns and 100 gouramy (U20T100). An equal density of aquatic plant of water spinach Ipomoea reptans Poir was planted in each treatment units. The results from 60 days experiment showed that the highest growth of prawn were achieved in treatment C i.e 9,355 ± 1,096 g and survival rates of 75.02±0.67%. The highest growth of kissing gouramy were achieved at treatment D i.e 13,688 ± 1,382 g and survival rates of 86.66±0.83%. The water spinach was able to reduce ammonia, nitrite and nitrate. The lowest reduction was achieved at treatment A, i.e., amonia 6.54 %, nitrite 1,33% and nitrate 5,90%. While the highest reduction of ammonia is 11.72% found at treatment D, 3,52% for nitrite at treatment C and 11,72% for nitrate at treatment D. The highest reduction of Total N dan Total P by water spinach were found at the treatment C i.e. 32.54% and D i.e. 47.62%. According to the dominance and abundance index of plankton, Chlorophyceae and Cyanophyceae were recognized as the dominant class in all treatments.

Abstract

Tacca leontopetaloides (L.) O. Kuntze has the potential as food alternative source from its tubers and therefore selected for food diversification study.This research was conducted on Kangean island using open-ended interview methods,field observations,purposive sampling and chemical analysis.Tacca leontopetaloides still not much known and utilizated by the local people in East Java.The plant has different local names for each region, such as in Madura they known as "lorkong" and "oto'o", while in Kangean island,known as "to’toan". In Kangean island, local people utilized the tuber to make flour using simple process. The flour was used as a substitute for wheat flour mainly as an ingredient for porridge, cakes “serpot” and “eped-eped (omelette)”. The species was generally grow wild in teak garden, so they can be used as an alternative crop under the teak stands. Proximate analysis of Tacca leontopetaloides tubers was also carried out and showed that they contained: 2.67 – 2.71 % of ash; 6.73 - 7.84 % of protein; 0.43 – 1.90 % of fat; 0.41 - 0.60 % of crude fibre; 77.09 - 82.65 % of carbohydrate and available energy 352.36 - 365.83 Kcal/100g based on dry weight. Mineral content was also analysed and showed the following value: 173.50 - 173.67 mg/100g of magnesium; 4.00 - 8.69 mg/100g of iron; 69.89 - 87.72 mg/100g of calsium; 904.86 - 966.74 mg/100g of potassium; and 222.59 - 270.46 mg/100g of phosphor. Hence, Tacca leontopetaloides has sufficient nutrient contents from its carbohydrate and mineral.

Abstract

Degree of electrolyte leakage (EL) from leaf tissue after exposure to high temperature has been used as an indicator of heat tolerance. Inthe present study, EL was measured in an attempt to estimate heat tolerance of Viana umbellata (Thumb.) genotypes at three different growth phases. The degree of heat injury is significantly different among the three growth phases and among the genotypes as well as their interaction. However, the degree of heat injury tends to increase over the plant growth. The 18 genotypes of V. umbellata shows moderate genetic variability as shown by its value of genetic variance (46,25 ±16,35), phenotypic variance (85,37 ±27,60), and coefficient of genetic variation (26,36%).

Abstract

The trace element Selenium is toxic at high concentration.Most of organisms living in selenium rich environment are selenium resistant.One of the resistance mechanisms is methylation,in which selenium is methylated and transformed to non-toxic selenium compound.The methylation is catalyzed by seleno methyltransferase (SMT) coded by smt gene. The gene are expressed by selenium tolerant plants. However, there was no available report yet on such specific gene in the bacterial genome. This study was carried out to determine smt homologous gene on selected selenium accumulator bacteria, Geobacillus sp. 20k, The smt gene of was determined by amplifying target DNA and analyses its sequences through homology search (BLAST). The result showed that the DNA and its protein part of thermophilic enzyme involved selenium metabolisms.
