Abstract

The accumulation of organic compound from waste feed and feces from intensive catfish culture can give negative impact to the water quality. On the other hand, the need of live feed for freshwater fish larvae are not sufficient. The aim of this research was to study the effectivity of intensively catfish culture wastewater as a media for Daphnia sp. culture. The experimental research design was completely randomized with five treatments, i.e: Using 5 g/l of chicken manure in the media culture of Daphnia as a control while treatments B, C, and D were using wastewater of intensively catfish culture. The concentration of wastewater of intensively catfish culture was 25%, 50%, 75%,and each treatment was replicated four times. Broodstock of Daphnia sp. used at least 1mm diameter with stocking density 20 ind / liter. Data on population growth of Daphnia sp. obtained were processed using analysis of variance with confidence interval of 95%, followed by least significant difference test (LSD). Furthermore, water quality parameters and density of plankton was examined as supporting data. The water quality data was observed every 4 days and plankton density observation was conducted at the beginning and the end of the study. The result of this research showed that the wastewater of an intensive catfish culture could be used as an alternative media for Daphnia sp. culture. Based on the analysis showed that the different media of Daphnia sp. culture significantly influenced to the population growth of Daphnia sp. The most effective media for Daphnia sp. culture in this research was treatment C and D i.e. 75 % and 100% of waste water of intensive catfish culture.

Abstract

Field dodder fCuscuta campestris Yunck.)is an obligate parasite plant (holoparasit) with twining stems that coil and fasten to host plants.The qualitative and quantitative studies were tested to 20 twining stems pieces, that infected on host plants (Durantha sp.).The results showed that the twining stems are very sensitive and responsive to the contact of host canopy, but it is not for grasses (Poaceae).A single piece has been found to grow at rate of more than 1 centimetres per day. The ability and speed of reaction of the twining stems by the contact stimulating to their host (haptotropism) were discussed.

Abstract

Microbial groups of entomopathogenic (fungi and bacteria) had been reported causing insect mortality. The aim of the study was to explore and characterized entomopathogenic from various host and locations. Fungal identification at genus and species level was caried out based on conidial morphology, hyphal growth, conidiophore and colony color; whilst for bacterial identification was based on standard Bergey’s manual for determinative bacteria. Sixteen entomopathogenic isolates that consisted of fungal and bacteria have been collected and preserved for further characterization. Of the 16 entomopathogen collected samples, five fungal genera was found i.e. Paecilomyces; Metarhizium, Beauveria, Hirsutella; and Cordyceps. Seven isolates belonging to six fungal isolates, and one bacterial isolate had been identified based upon ITS and 16S rDNA sequences, respectively. We confirmed that 6 fungal isolates belong to species of Paecilomyces reniformis, B. bassiana, M. anisopliae, M. anisopliae var acridum, Hirsutella thomsonii. One isolate of red pigmented bacteria Sm201102 have been identified was belonging to species Seratia marcescence. It was also obtained two fungal isolates from different host (spider and beetle) which confirmed by morphological character belong to Cordyceps sp.

Abstract

The problem in Lake Sentani area are degradation of watershead and land forest convertion, which cause decreasing water quality in Sentani Lake. For this reason, it is very important to analyze of water quality status in L. Sentani periodically in order to retain its sustainability. The analyzed result by using STORET method showed the quality statue of L.Sentani are ranged from moderate to heavy pollution in category since 2006.

Abstract

A study on seed germinations of Strombosia javanica BL. was conducted to observe morphological character and germination pattern of the seedling. As many as 134 seeds of the species, originally collected in Bogor Botanical Gardens, were germinated on medium contain soil, sands and compost in same quantity. Germination aspects such as germination patterns and developmental changes were monitored everyday until the first leaves appear. The seed germinations was typically hypogeal and totally spent 160-170 days, and consisted at least six stages: hypocotyl growth, radix primordial formatton, seedling root growth, seed ball lifting, seed ball removal and first leaves appearance. Rather intact seed ball remain exist at the end of hypocotyl vigorously mtil its removal, pmsumably one of special phenomena of the species germination associated to plant parasitism. Functionally, development of the cotyledons were abnormally suppressed by the existence of the seed ball. The cotyledon praphotosynthetic function seems to be replaced by greeny hypocotyl. First leaves development were never occurred before the seed ball removal. Seed coat elimination was able to increase germination initiation in a given time but facilitated damaging bacteria or fungi to contaminate the endosperm.

Abstract

Bacterial Leaves Blight (BLB) is one of the major diseases in rice. Among the disease resistance genes HDB, Xa7 gene still can compensate the diversity of Indonesian BLB pathogen in the field. One of the local rice germplasm, Parekaligolara (Indica, IRN: 1541) were selected based on allele mining research which contained a Xa7 alleles. The purpose of this study is to identify the alleles of the Xa7 gene contained in Indonesian local rice Parekaligolara germplasm in rice through the segregation testing based on phenotype and genotype performance of segregating population derived from the crossed with the susceptible control varieties, TN1. The segregation testing were conducted in F1 and F2 populations and analyzed on phenotypes of BLB resistance and genotypes using Xa7SNP markers. The analysis results showed that the Xa7 allele variation founded in the Parekaligolara germplasm are act as dominant alleles that contribute on the BLB resistance trait particularly to Race VIII of BLB pathogen. Xa7SNP8 is polymorphic markers for Xa7 alleles gene that are found in Parekaligolara. This marker could be used as a marker assisted for selection, if Parekaligolaras Xa7 alleles will use as a parent in the BLB disease resistant rice breeding program.

Abstract

The objective of this research was to examine the effect of sucrose in improving the quality of the plasma membrane intact and sperm motility of frozen semen of Garut ram. Semen was collected using artificial vagina weekly from six mature garut rams.   Immediately after initial evaluation, fresh semen was divided into four parts and diluted with Tr s extender without sucrose (T0), Tris extender + sucrose 0.2g/100 ml (T1), Tris extender + sucrose 0.4g/100 ml (T2) and Tris extender + sucrose 0.6g/100 ml (T3), respectively.  Results of this research showed that the percentage of sperm motility after thawing in T2 (49.00 ± 5.48%)  was significantly (P<0.05) higher than T0 (42.00 ± 2.74%) but was not significantly difference (P>0.05) than T1 (46.00 ± 4.18%)and T3 (48.00 ± 4.47%).   Evaluation of plasma membrane intact showed that T1 (62.33 ± 6.51%) was s gnificantly different (P <0.05) w h T0 (49.40 ± 2.19%) but was not significantly different (P> 0.05) than T2 (58.50 ± 4.97%) and T3 (56.40 ± 5.90%).  In conclusion, he addition of sucrose in semen extender  improved the quality of frozen semen of Garut ram.  Concentration of 0.2g / 100 ml is the op ma dose to improve the quality of the plasma membrane intact and motility of spermatozoa during the freezing process.

Abstract

 The effluent generated from aquaculture can be exploited through the application of aquaponics technology. The aim of this research was to evaluate the utilization of catfish culture waste by tilapia based on closed recirculation. Catfish and tilapia were used as fish test while water spinach was used as a biofiltration. Dissolved of nitrogen and phosphorus that obtained from catfish culture would be utilized by water spinach. Completedly Randomize Design with consisted of three treatments and three replicates were performed. The different stocked density on catfish culture were used as a treatment i.e. 500 individuals per pond (P1); 1000 individuals per pond (P2); and 1500 individuals per pond (P3). Different stocking density was also applied to tilapia culture as a treatment namely 50 individuals per m2 (Q1); 100 individuals per m2 (Q2); and 150 individuals per m2 (Q3). Result showed that catfish culture with stocking density of 1500 individuals per pond (P3) was better than (P1) and (P2) in terms of absolute weight growth, specific growth rate, and survival (P<0.05). Absolute weight growth, specific growth rate, and survival of tilapia cultured at Q2 (stocking density of 100 individuals per m2) and Q3 (stocking density of 150 per m2) were better than Q1 (stocking density of 50 individuals per m2) (P<0.05).

Abstract

A research to study the effect of intercropping on rehabilitated of physic nut (Jatropha curcas L.) was conducted in Muktiharjo Research Station, Pati, Central of Java, from January to December 2010. Plant materials were physic nut rehabilitated by grafting of main branch with IP-2A (1 year old) and intercrops (peanut, soybean, mungbean and sesame). The experiment arranged in randomized block design with 6 replications. The treatments were: 1) intercropping peanut + physic nut, 2) intercropping soybean + physic nut, 3) intercropping mungbean + physic nut, 4) intercropping sesame + physic nut, and 5) physic nut monoculture. Plot size was 8 m x 8 m, plant distance of physic nut 2 m x 2 m, and plant distance for peanut, soybean and mungbean were 25 cm x 25 cm and sesame was 50 cm x 25 cm. The result showed that jatropha seed yield on intercropping physic nut with soybean (980.05 kg/ha) and mungbean (974.22 kg/ha) were not significantly different from physic nut monoculture (1,094.84 kg/ha). Crop that appropriate for intercropped with rehabilitated physic nut (second year) were soybean and mungbean.

Abstract

Wild species of begonia existed in remnant forest of Cibodas Botanic Gardens, but their diversity, distribution and ecological preference were not much known. Data of condition and habitat characteristic of wild begonia are important in their cultivation. The aims of this study are to inventory the diversity of wild begonias and their distribution habitat preference in remnant forest of Cibodas Botanic Gardens. Sample were collected from four remnant forest using exploration method. Analysis of vegetation where wild Begonia existed was conducted using 1x1 m plot in around every individu of Begonia found. The principle component analysis (PCA) was carried out using R program version 2.11.0. The result showed that 50% of the mountain Begonia in West Java can be found in remnat forest area. The light intencity, soil humidity and air humidity were highly correlated to distribution of wild Begonia. The microclimate conditions in remnant forest area are appropriate with habitat of Begonia.
