Abstract

Contamination of Extended spectrum-betha lactamase (ESBL) producing Escherichia coli in milk can cause public health problems. The misuse of antibiotics in dairy has implications for the emergence of multi-resistant bacteria that can infect humans through food (foodborne diseases). The ESBL producing E. coli is resistant to ?-lactam antibiotics (including penicillins and 3rd and 4th derivatives of cephalosporins). This study aimed to analyze the phenotype of antibioticresistant ESBL-producing E. coli from livestock, farmer, dairy cows, milk, and pasteurized milk. The research was conducted on a dairy farm in Cijeruk District, Bogor. The method used was the ESBL EC tricycle for phenotype confirmation consisting of culture on tryptone bile x-glucuronide (TBX) agar and MacConkey agar (MCA) with the addition of the cefotaxime antibiotic 4 µg/mL for the presumption of ESBL-producing E. coli followed by confirmation of ESBL producing E. coli with indol biochemical tests with sulfite indole motility (SIM) and double disk diffusion test (DDST) on Mueller Hinton agar (MHA). The results from this study showed that ESBL producing E. coli was not found in water, effluent, milker hand swabs, teat swabs, and milk from the individual cows. ESBL producing E. coli was identified from fresh milk (2/10) and pasteurized milk (2/10) on the same farm. ESBL-producing E. coli were resistant to eight antibiotics (ampicillin, cefotaxime, ceftazidime, cefpodoxime, cephalothin, streptomycin, azithromicyn, enrofloxacin). Multidrugresistant ESBL-producing E. coli is one of the foodborne diseases that pose a health threat to the community.

Abstract

The use antibiotic growth promoters (AGP) can cause antibiotic residues and microorganism resistance, so it is prohibitied by the government. Jamu is an Indonesian herbal ingredient that can be used as an alternative in overcoming this problem. The aim of the study was to determine the effective concentration of a combination of ginger, curcuma, wild ginger, and honey herbs in improving the performance and profile of broiler chickens without changing the organoleptic properties of the meat. The study used 36 Cobb strain day old chicks (DOC) which were reared up to 32 days of age. The study used a completely randomized design with the addition of combination herbal medicine in drinking water as much as 0% (control), 1.25% (P1), 2.5% (P2), and 5% (P3). The variabels observed included performance (feed and drink consumption, body weight gain, final weight, feed conservation ratio (FCR), performance index (IP), mortality), organ profiles (weight of liver, hearts, lungs and spleen), and weight and length of the duodenum, jejeum, ileum of broiler chickens followed by organoleptic test on broiler chicken meat. The result showed that P1, P2, and P3 significanly (p<0.05) could increase the final weight and body weight gain. Carcass weight, abdominal fat weight, FCR, IP, mortality, and organ profile showed no significant difference (p>0.05) between treatment and control. Research shows that the combination of ginger, temulawak, lempuyang, and honey with concentration of 5% can improve performance without changing the organ profile of broiler chickens and organoleptic broiler chicken meat.

Abstract

The Kintamani dog was designated as the first Indonesian native dog by the Indonesian Kinology Association (Perhimpunan Kinologi Indonesia/Perkin) in 2006 and has been officially registered as an Indonesian native world dog by the Fédération Cynologique Internationale (FCI) on February 20 2019, under the breed name Anjing Kintamani – Bali. This study aims to evaluate the radiogram of the elbow joint in Kintamani dogs based on FCI assessment standards. The research samples used were radiograms of 34 Kintamani dogs aged 12 to 24 months, consisting of 19 male dogs and 15 female dogs. Forelimb radiographs were taken in two positions, mediolateral flexion 15° and craniocaudal pronation 15°. Radiograms were taken using computerized radiography (CR) and digital radiography (DR), then interpreted and analysed using Digimizer software. The interpretation of the elbow joint is done descriptively based on the presence of primary lesions and/ or osteoarthrosis in the elbow joint. A qualitative analysis was carried out regarding the normal anatomical structure of the elbow joint of the Kintamani dog according to the standards set by the FCI. Based on the FCI assessment score for the forelegs, 33 Kintamani dogs were assessed as normal (FCI grade 0). Only one Kintamani dog was assessed as having mild elbow dysplasia (FCI grade 1), as indicated by the presence of >2 mm osteophytes. Complete knowledge of the anatomy and radiography position is very necessary in determining the prognosis of elbow dysplasia in order to support efforts to breed the Kintamani dog as the first Indonesian native world dog.

Abstract

Bats are the natural reservoir and host for most human and animal viruses. There are more than 1,200 species of bats that have been identified, 20% or 239 species of which are spread across Indonesia. The high diversity in bats with the presence of various cell types and receptors has the potential to cause bats to become a source of transmission for various types of viruses, especially type-A influenza virus. Early detection of type-A influenza virus in bats is necessary considering the global distribution, species diversity and high population density of bats in Indonesia. and potential spread to other species. In this study, a total of 101 bat rectal swabs were collected from Cianjur Regency, West Java. Identification of bat species was carried out based on morphological and morphometric characteristics. From the identification results, four bat species were obtained, namely Rousettus amplexicaudatus Rhinolophus sp., Hipposideros sp. and Miniopterus sp. Influenza virus type-A was detected by reverse transcriptasepolymerase chain reaction (RT-PCR) based on the matrix gene. Of the 56 R. amplexicaudatus swabs rectal samples, 21 (37.5%) were positive for the matrix gene of influenza virus type-A. Of the 21 positive samples of type-A influenza, all of these samples were negative for the H5N1 influenza A virus subtyping test. Early detection of this virus is expected to minimize its destructive impact on global health by implementing control measures.

Abstract

African Swine Fever (ASF) is a disease in pigs of all ages caused by a virus from the Asfar virus family, and the virus is classified as an economically important pathogen group. This disease has caused the death of pigs in Nusa Tenggara Timur Province including Kupang Regency. The spread of ASF disease can occur when the animal health management is poor. Little was known about the health management of pigs in the district of Kupang. Therefore, this study was aimed to identify factors of health management which potentially affect the spread of ASF in the district of Kupang. This research was conducted in two selected sub-districts in Kupang Regency, namely East Kupang District and Amabi Oefeto District. The selection of these sub-districts was based on the results of discussions with the Head of the Livestock Service Office of Kupang Regency, where these two sub-districts became centers of pig production. The results showed that the health management of pigs in the selected sub-districts of Kupang Regency was still very poor among vehicle and people traffic control, disease control, livestock waste, sanitation and disinfection, pest control, pig traffic, pig marketing, health status and ASF, disposal system pig carcasses, and contact between pigs.

Abstract

Tyrosine kinase (TEK) is the subgroup of protein kinases class. Tyrosine kinase (TEK) is a protein in spermatozoa plasm that functions as a mediator during spermatozoa penetration to pellucide zone 3 (PZ3) of the ovum. The purpose of this study was to analyse the variation of receptor tyrosine kinase genes related to the fresh semen qualities of limousin bulls, using Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCR-RFLP) method with HindIII restriction enzyme in A/AGCTT restriction site. This study used 20 blood samples of limousin bull from BBIB Singosari Malang. DNA amplification carried out by PCR method using forward primer (TEK_F) 5'-TAGATTGTCGCTTGCCTGGG-3’ and reverse primer (TEK_R) 5'-CCTGTGCCGACAGGTTTACT-3’. The data analysis were obtained by counting the total of RFLP DNA fragments as the genetic variation. The correlation between the total of RFLP DNA fragments and the fresh semen qualities were analysed by Spearman correlation test with RStudio software. The polymorphic DNA fragments were on 136 bp, 88 bp, 72 bp, and 12 bp range. Monomorphic DNA fragment was on 39 bp range. Correlation coefficient between the RFLP DNA fragments total with semen pH was -0,158 (negative correlation) with the p-value 0,912; semen volume was -0.105 (negative correlation) with the p-value 0,659, sperm motility was -0,050 (positive correlation) with the p-value 0,831; and sperm concentration was 0,044 (positive correlation) with the p-value 0,852. The RFLP DNA fragment total with some fresh semen evaluation parameters had very low correlation with no significancy because of the p-value >0,05.

Abstract

Penelitian ini bertujuan untuk mengetahui kandungan kimia dan sedimentasi urin kucing lokal yang hidup bebas berkeliaran di sekitar Kota Denpasar. Sampel yang digunakan adalah urin kucing yang diambil secara acak dan digunakan sebagai latihan bedah cystocentesis dan kateterisasi. Sebanyak delapan ekor kucing jantan dan delapan ekor kucing betina dewasa sampel urinnya diambil dan dimasukkan ke dalam pot steril, lalu dilakukan pengamatan kimia urin dengan pemeriksaan menggunakan dipstick yang memiliki 10 paramater yaitu leukosit, nitrit, urobilinogen, protein, pH, darah, berat jenis (spesific grafity), keton, bilirubin, dan glukosa. Pemeriksaan dipstik dilakukan dengan cara mencelupkan stik ke dalam masing-masing sampel selama 0,5-1,0 menit hingga seluruh parameter terendam, kemudian diangkat dan ditaruh di media datar yang telah dilapisi tissue lalu ditunggu selama tiga menit hingga parameter berubah warna dan diulangi sebanyak tiga kali. Setelah dilakukan pemeriksaan kimia urin menggunakan dipstick, sampel urin kucing di dalam tabung reaksi diendapkan dengan menunggu dan mendiamkan sampel urin tersebut semalaman dalam tabung yang diposisikan secara tegak lurus. Setelah 20 jam diendapkan, kemudia endapan tersebut diambil dan diletakkan di atas objek glass lalu diperiksa menggunakan mikroskop cahaya dengan perbesaran 400 kali. Penelitian ini termasuk ke dalam penelitian observasional yang menekankan pada pengamatan kuantitas/ jumlah zat dan sedimentasi yang terdapat didalam sampel urin kucing. Dari 16 sampel yang diperiksa, semua sampel negatif terhadap pemeriksaan leukosit, nitri, urobilinogen, dan darah. Protein ditemukan pada 75% sampel kucing, Glukosa 1+ ditemukan pada 6,25%, dan masing-masing 100% positif pada pemeriksaan ketone dan bilirubin. Sedangkan rata-rata berat jenis urin kucing bernilai 1,022 dan pH urin bernilai 6,75. Selanjutnya dari hasil pemeriksaan sedimentasi urin, 11 sampel ditemukannya crystal dan casts urin yang terdiri dari Calcium oxalate monohydrate, triple phosphate, Magnesium Amonium Phosphate, Kumpulan Kristal Struvite, dan Lipid droplets. Dari pemeriksaan sampel tidak ada perbedaan signifikan antara kucing jantan dan betina.
