Abstract

Penelitian dilakukan untuk mengetahui kualitas spermatozoa epididimis kerbau belang selama proses pembekuan (kriopreservasi). Bahan pengencer yang digunakan adalah AndroMed® sebagai kontrol (AM) serta kombinasi AndroMed® dengan 0,2% (S0,2) dan 0,4% sukrosa (S0,4). Hasil penelitian menunjukkan bahwa persentase motilitas spermatozoa epididimis kerbau belang setelah thawing dalam bahan pengencer AM (41%), nyata (P<0,05) lebih rendah dibandingkan dengan S0,2 (46%) dan S0,4 (46%). Demikian pula halnya dengan persentase hidup spermatozoa setelah thawing dalam bahan pengencer AM, S0,2, dan S0,4 secara berturut-turut adalah 52,2; 59,8 dan 60,8% (P<0,05). Sementara itu, tidak terdapat perbedaan nyata (P>0,05) pada persentase membran plasma utuh (MPU) setelah thawing dalam ketiga bahan pengencer. Persentase MPU setelah thawing perlakuan AM, S0,2, dan S0,4 adalah masing-masing 68; 68,8 dan 66,8%. Dapat disimpulkan bahwa penambahan sukrosa sebagai krioprotektan eksternal ke dalam bahan pengencer AndroMed® dapat meningkatkan kualitas spermatozoa epididimis kerbau belang setelah thawing.

Abstract

In this study, the concentration of prostaglandin F2 ? (PGF2?) in seminal vesicle fluid and seminal vesicle monolayer cell cultures of Bali cattle was determined. The seminal vesicle fluid was aspirated and the epithelial cells of the seminal vesicles were cultured in tissue culture medium (TCM) 199 growth medium containing 10% fetal calf serum (FCS) and 10% oestrus mares serum (EMS) with a density of 1.9 x 106 cells / ml medium. Following an incubation at 38.50 C in 5% CO2 atmosphere for 6 days and the level of PGF2 ? in the original seminal vesicle fluid and in the cell culture medium were determined by radioimmunoassay techniques (RIA). The results showed that the level of PGF2 ? in the non-extracted monolayer culture of seminal vesicle (1287,50 ± 3,39 pg/ml ) was significantly higher than that of detected in non-extracted seminal vesicle fluid (1,23 ± 0,79 pg/ml). In contrast, after extraction the level of PGF2 ? in seminal vesicle monolayer cell cultures (218,33 ± 2,87 pg/ml) significantly decreased as compared to seminal vesicle fluid (1750,83 ± 2,71 pg/ml). In conclusion the highest level of PGF2 ? was found in the extract of seminal vesicle fluid.

Abstract

A study was carried out to find out an alternative method of producing antitetanus antibody (IgY) in chicken and to evaluate its activity at different levels of pH, temperatures and proteolytic enzymes. Antitetanus IgY was produced by immunization of chickens with tetanus toxoid, three times weekly at gradual doses of 100, 200, and 300 Lf, respectively. Serum samples were collected 4 weeks following the last immunization. IgY was purified by ammonium sulfat precipitation and gel filtration chromatography (Sephadex G. 120).The purified IgY was then treated at different levels of temperatures and pH as well as proteolytic enzymes. Commercial antitetanus IgG was used as control. The activities of treated IgY and IgG were tested by enzyme linked immunosorbent assay (ELISA). IgY and IgG activities were significantly reduced at 80ºC and completely destroyed at 90ºC. Treatment with pepsin significantly reduced IgY and IgG whereas trypsin slightly reduced IgY activities and has no effect on IgG activities. IgY and IgG activities were reduced significantly at pH < 3 and and only sightly reduced at pH>10. It is evident that heating at >90oC, pH at <3 and treatment with pepsin significantly reduced IgY activities and it appears that IgG was more resistent to the efect of temperatures, pH and proteolytic enzymes

Abstract

A study was conducted to isolate and identify of lactic acid bacteria originated from gastric fluid of bali cattle, and to determine their potential as the candidates of biopreservative. Lactic acid bacteria were isolated by culturing the gastric fluid of bali cattle in de Mann, Rogosa, Sharpe (MRS) medium; screening the bacteria, and identification of bacteria species by Analytical Profile Index (API) 50 CHL Kit. The results showed that, the new species of lactic acid bacteria were isolated and identified as Lactococcus lactis spp lactis 1 (SR21 isolate) and Lactobacillus brevis 1 (SR54 isolate) that have broad spectrum antimicrobial activities. It is clear from this study that a potential lactic acid bacteria producing antimicrobial agent can be isolated from the gastric fluid of bali cattle.

Abstract

A study to detect Coxiella burnetii, an intracellular bacterium causing Q fever in human and livestock animals, was carried out in several ruminants in Bogor and Bali. The methods used for the detection was Nested-Polymerase Chain Reaction (Nested-PCR). Two pairs of primers, the first (OMP1 and OMP2) and the second (OMP3 and OMP4) were used to detect the genomic sequences and the conserved specific sequences of Coxiella burnetii, respectively. Organ samples such as liver and lung from 410 livestock ruminants, consisting of cattle (245 samples), sheep (105 samples) and goats (60 samples) were collected from several slaughter houses in Bogor and Bali. As many as 15 (6.12%) out of 245 cattle, 6 (5.71%) out of 105 sheep and none from goat were infected by Coxiella burnetii. Interestingly, 3 out of 15 infected cattle were Bali cattle. The results clearly indicate that Q fever is likely to be widespread among ruminant animals in Indonesia.

Abstract

A study to compare the physiological values (heart beat and respiration rate, body temperature, and electrocardiogram/ECG) between adult dugong (AD) and baby dugong (BD) was carried out. The dugong’s physiological values were examined by patient monitor. The results showed that heart beat rates of baby dugong (87.11+8.70 per minute) was higher than in adult dugong (45.73+4.10 per minute), but no significant difference was observed on the respiration rates between baby dugong (12.89+3.08 per minute) and adult dugong (10.11+3.44 per minute). The body temperature of baby dugong (29.26+0.77oC) was lower than that of adult dugong (31.42 + 0.41) oC. Regular heart beat rhythms was detected on Lead I, II, and III of ECG of both in baby and adult dugong. The ECG values of baby dugong was higher on voltages and shorter in duration (p<0.05) as compared than those of adult dugong. The axis values in adult dugong showed a position more on the right by (+ 90o)–(- 135o) with mean electricity axis (MEA) of +157,5o compared than axis value in baby dugong which appeared to be more on the left by (-32.7o)–(-76.7o) with mean MEA of -54.7o. The differences in cardiorespiration and body temperature values between adult dugong and baby dugong appeared to be due to differences in ages of the mammal, especially related with the differences on the metabolic rate and anatomical growth.

Abstract

A study was carried out to identify the blood groups of dogs and to determine their distribution among dog population as well as their possible relationship with hair color, sexes and location. Fifty dogs were used in this study and they were originated from Jakarta, Bekasi and Bogor, Indonesia. The blood groups of dogs were determined by monoclonal antibodies and a positive reaction was characterized by the presence of agglutination as a result of antigen-antibody reaction. Three blood groups, 1.1B (44.0%), 1.2B (42.0%) and 1(-)B (14.0%) were detected among the 50 dogs examined. No significant relationship between certain the blood groups and the hair color, sexes and their origin were detected in this study.
