Abstract

Exploration, Inventorize, and Characterization of Durian Kunyit (Durio zibethinus Murr) Germplasm in Sub-District of Pupuan, Tabanan District This research aimed to explore, inventorize, and characterize durian kunyit (Durio zibethinus Murr) plants in the sub-district of Pupuan. The research coveredthe exploration of durian kunyit in each village, the inventorize of the dispersal of durian kunyit, and the characterization of durian kunyit. The variables observed were the agronomic characters, morphology characters, and molecular character of durian kunyit. The results showed the discovery of 8 durian kunyit mother trees scattered at an altitude of 370 – 661 meters above sea level. The agronomic character of durian kunyit fruit are small–medium sized, fruit weight 1,1 kg, fruit length 17,3 cm, fruit diameter 15,0 cm, number of seeds 17 per fruit, aryl texture quite soft, watery condition, taste sweet little bitter, the flesh is quite thick, aril is yellow, the aroma is quite strong, not sticky in the hand and the production reaches 200 pieces per tree at the harvest. The morphology characters of durian kunyit were plants aged 10 – 80 years, plant height 16 m, tree circumference 165 cm, petiole length 17,8 mm, petiolewidth 2,9 mm, length leaf strands 14,2 cm, leaf blade width 4,9 cm. The molecular character of durian kunyit forms 8 specific banding patterns at 350 – 1600 basepair using the PKBT ISSR Primer 2.

Abstract

The Effect of Harvesting Age on Seeds Production and Seeds Quality of Long Bean Seeds (Vigna sinensis L.) This research was conducted in January 2018 until May 2018 in Subak Sedang, Angantaka Village, Abiansemal District, Badung, Bali. This research aimedto determine the effects of time of harvesting after flowering on Long Bean seed physiological maturity and quality. This research in a one factor research with 3 differences of age of harvesting. The three age of harvesting : P1 (23 days after flowering), P2 (26 days after flowering), and P3 (29 days after flowering). Theresearch used a randomized block design (RBD) with 9 replications. Observations variables were made on number of pods, number of pithy seeds, weight of seeds per hectare, weight of 1000 seeds, viability and vigor. The results showed that the treatments of age of harvesting P2 (26 DAF) had an effect on the seed production and quality.

Abstract

Effect of Hyacinth Organic Fertilizer and Biofertilizer on Biological Properties of Soil, Growth and Yield of Sawi Plants (Brassica juncea L.) This study aims to determine the effect of interactions between water hyacinth organic fertilizer and biofertilizer on the biological properties of soil, growth and yield of mustard greens. This research was conducted in April 2018 until June 2018 at Banjar Taman Tanda, Baturiti Village, Baturiti District, Tabanan Regency. The experiment used a randomized block design (RBD) with 2 factor, namely Eceng Gondok organic fertilizer 3 levels (P0, P1, P2), and biological fertilizer 3 levels (M0, M1, M2). 9 combination treatments and 3 replications so that there were 27 trial tests and data analysis using the Costat program and continued with the BNT test at 5% if the treatment was significantly. Parameters observed were pH, total N, organic C, microorganism population, plant height, fresh weight and dry weight of plants. the best treatment of water hyacinth organic fertilizer and biofertilizer on the yield of mustard plants was shown by a combination of 80 tons ha-1 of water hyacinth organic fertilizer and 60 ml of biological fertilizer (1% concentration solution) of 6,940 tons ha-1 and increased 24 % of control. This treatment provides the best response to green mustard plants.

Abstract

Grafting Method of Tomato using rootstock of eggplant to minimize problems caused by flooding and soil-borne diseases in Faculty of Agriculture Research Field Udayana University The study was conducted to determine the effectiveness of the Grafting Method of Tomato using rootstock of eggplant to minimize problems caused by flooding and soilborne diseases in Faculty of Agriculture Research Field Udayana University. The experimental design was used a Randomize bloc design (RBD), with 3 treatments including control and 9 replications. The treatments are, grafting, non-grafting and control usually use by the farmers. The first step of the research is grafting or tomato as a scion with eggplant as a rootstock. The grafting method was used is base one the AVRDC-method. The result showed the grafted tomato was free from wilting disease on the other hand the non-grafted tomato showed the symptom of wild diseases, the data indicated the grafted tomato was resistant by the soil-borne diseases including bacteria wilt as well as Fusarium wilt disease. For flooding we cannot compare the significantly result between grafted tomato and non-grafted tomato because the season is not wet season. The suggestion of this research is recommended to doing the research in wet season for more clear understand the effect of grafted tomato to the flooding.

Abstract

Histophatology Structure of Siam Citrus Petiole (Citrus nobilis L. var. microcarpa) Infected by Citrus Vein Phloem Degeneration (CVPD) at Mild and Severe Attack Rates The main cause of decreasing the production of Citrus nobilis L. var. microcarpa in Indonesia is the presence of CVPD disease (Citrus Vein PhloemDegeneration). Citrus nobilis L. var. microcarpa infected by CVPD showing anatomic and morphological changes. The main objective of this study was todetermine anatomic changes in petiole tissue of Citrus nobilis L. var. microcarpa infected by CVPD at the level of mild and severe attacks. The research wasconducted in Citrus field, Kintamani, Bangli-Bali and Biomolecular and Genetic Resources Laboratory, Faculty of Agriculture, Udayana University in August September 2018. The result for visual observation showed that the presentasion of CVPD symptoms in the field was 46% and attack intensity was 39.6%. Samples were identified using PCR metod with O11 and O12c markers. DNA visualization amplified on 1% agarose showed appearing of DNA band for samples with mild and severe attack rates. Samples gave positive reaction to Liberibacter asiaticus. It showed that the samples were positive infected by CVPD disease. Preparations of petioles were made to observe the histological structure microscopically. The result showed that there were changes on histological structure of petioles infected CVPD. Petioles infected CVPD at mild symptom and severe symptom showing cell wall thickening on epidermis, shape changes on cortex and there were an accumulation of starch, lysis and necrosis showing on floem with severe attack rate, and there were stretching between xylem cells.

Abstract

Application of Geograpic Information System and Remote Sensing for Mapping of Land Use and Suitability ini Batur Tengah Village Bangli This study aims to map land use and evaluate the suitability of land for horticultural, secondary and plantation crops. This research held from December2017 - March 2018 using a method consisting of, identification of satellite images (land use), field surveys, reinterpretation of land use maps, analysis of soil samples and land suitability classification. The results that there were eight types of land use in Batur Tengah Village with a total area of 474 ha. The actual class of land suitability of fruit horticulture plants is generally S3 (marginal suitability) with dry season limiting factor, soil texture, soil-CEC and N-total and slope, potential suitability class S3. The actual land suitability class of plantation (coffee) plants is generally S3 with a dry season limiting factor, soil texture, soil-CEC, N-total and slope. Potential land suitability class S3. The class of actual land suitability of vegetable horticulture plants is generally S3 (marginal suitability) with dry season limiting factors, soil texture, soil-CEC and N-total and slope, potential land suitability class S3. The actual land suitability class of palawija crops is generally N (not suitable) with dry season limiting factors, soil texture, soil-CEC and N-total and slope, potential land suitability class S3. The land suitability geographic information system is presented interactively which can display land suitability classes, agricultural commodities and land use types.

Abstract

The effectiveness analysis of neem plant (Azadirachta indica A. Juss.) to control leaf borer on plant Podocarpus neriifolius Plant maintenance and development cannot be avoided from pests and diseases. Losses due to pest or disease attacks encourage patients to control variousefforts. So far the pest and disease control efforts carried out are still based on synthetic pesticides. The use of various types of synthetic pesticides, on the other hand bring problems to the environment such as causing poisoning to humans, killing non-target organisms, secondary pest outbreaks, pest resistance, soil, water and air pollution. One alternative pest control that can be done is by organic pesticides. One of the plants reported to have the ability to control pests is the neem plant (Azadirachta indica). This study aims to determine the effectiveness of neem extract in controlling leaf borer caterpillar on Podocarpus neriifolius compared to vegetable pesticides Dane brand and synthetic pesticides. The results of the research conducted, it was found that the most effective dose of mimba extract was 80 g/liter. Both wet and dry extracts are equally effective in controlling pests. Control using dry leaf extract with dose of 80 g/liter is more effective than using the Dane brand pesticide in three times the dose.

Abstract

Histopatology Siamese Citrus Leaves (Citrus nobilis) Infected by Citrus Vein Phloem Degeneration (CVPD) at mild, moderate and severe attacks. Aims of this study is to identify the presence of the bacteria Liberobacter asiaticum, cause of CPVD (Citrus Vein Phloem Degeneration) disease and to knowthe differences in the histology of Siamese citrus plant tissue that is attacked by mild, moderate and severe attacks. This research was in the Laboratory of Plant Structure and Development of the Faculty of Biology, University of Gajah Mada and conducted at the UPT of the Genetic and Molecular Biology Laboratory of Udayana University. Field observations and sampling were carried out in the Wanasari Banjar, Kintamani Village, Kintamani Sub-District, Bangli District, Bali Province. There are several symptomatic citrus plants and attacked by CVPD with various levels of severity by visual appearance. On the observed land it were found that the attack percentage was 70% and the intensity of the attack was 46.4%. PCR (Poly polymeration chain reaction) identification method using OI1 and OI2 specific primers. The DNA band 1160 bp was found in mild, moderate, and severe chlorosis symptomatic samples. Leaves without symptoms do not change shape in the epidermis, cortex, xylem and phloem, while form changes occur in phloem, xylem and cortex in symptomatic citrus plants.
