Abstract

Endophytic Fungi Isolation on Balinese Grape Plants (Vitis vinifera L. var.Alphonso Lavalle ) and its Effectivenese to Control Grey Mold Disease on Balinese Grape Gray mold rot is the disease caused by pathogen Botrytis cinerea. It is animportant disease in grapes (Vitis vinifera L.) since it can reduce the quantity andquality of grapes. Nowadays people use synthetic fungicides to control the pathogen,but the cost of synthetic fungicides is expensive and continuous usage will give anegative impact to the environment therefore endophytic fungus is an alternative waythat can be used to control the pathogen in grapes. The purpose of this study was toobtain endophytic fungi and obtain the best endophytic fungus to control gray moldrot pathogens. This study uses two methods which are in vitro endophytic fungalantagonist test and in vivo endophytic fungal antagonist test in the laboratory. Theresults showed that there were 10 endophytic fungi. The results of in vitro and in vivotests showed that the best endophytic fungus was Miselia sterilia A as confirmed bythe percentage of inhibition of Miselia sterilia A in the in vitro test of 82.14% and thesmallest percentage of pathogen attack in the in vivo test of 6%.

Abstract

Isolation and Selection of Antagonis Bacteria as an Alternative Control of Strawberry Wilt Diseases Strawberry (Fragaria sp.) is a plant that can produce fruit and a members of Rosaceae familia that entered Indonesia in the 1980s and began cultivativated in Candi Kuning Village, Bali at 1983. The farmer reported in 2017 at Candi Kuning Village, Bali, there are many strawberries plants are exposed to wilting disease caused by Fusarium oxysporum which can decreased strawberry production up to 80%. The purpose of this study is to find alternatives ways of chemical control that can cause environmental damage. Biological control, especially the use of antagonistic bacteria, has potential as a replacement because it is much more environmentally friendly and can create sustainable agriculture. The method used are isolation and selection, in vitro inhibitory test, and hypersensitive test on tobacco plants. Research activities included (1) sampling, (2) Fusarium oxysporum isolation from symptomatic strawberry plants, (3) isolation and selection of antagonistic bacterial candidates, (4) antagonistic candidate bacteria inhibitory test against Fusarium oxysporum by dual cultur method in vitro, and (5) hypersensitive test of antagonistic bacterial candidates on tobacco plant leaves. The result of this research is Fusarium oxysporum successfully isolated, obtained 24 candidates of antagonistic bacteria and only 4 candidates of antagonistic bacterial isolates capable of well inhibiting the development of Fusarium oxysporum with the average inhibitory percentage are more than 60 % which is the best inhibitory percentage of 90.17 % and in the hypersensitive test, the bacterial candidates tested were non-pathogenic to the plant.

Abstract

ABSTRACT The Diversity and Density of the Population Parasitoid who Associated with Plutella xylostella L. (Lepidoptera: Plutellidae) on Plantation Plants without and with Insecticide Application The research was conducted at the Laboratory of Integrated Pest and Disease  Management, Concentration of Plant Protection, Department of Agroecotechnology, Faculty of Agriculture Udayana University. The purpose of this research to find out diversity and density of parasitoids population associated with  Plutella xylostella on cabbage plantation without and with insecticide application. Sample was taken by using diagonal sampling. There are nine points sample on cabbage plantation without and with insecticide application and each point content 18 plants. The sample was taken every weeks, while application insecticide was done every 10 days. Data was analyzed by using T-test. The result of the research indicated that there is one parasitoid was found associated with P. xylostella on cabbage plantation without and with insecticide application. That parasitoid is Diadegma semiclausum. Population of P. xylostella and D. semiclausum not significant difference on cabbage plantation without and with insecticide application, and so it seems of parasitization rate of parasitoid.   Keywords: insecticide, Plutella xylostella, parasitoid, cabbage

Abstract

Citrus spp conservation. in Bali Botanical Gardens and Potential UtilizationCitrus is one of the genus on Rutaceae. Plants who derived from East Asia and Southeast Asia has many benefits because in addition to having fruits rich with vitamin C, also produces essential oils in some parts of the body. Essential oil is widely used in the field of food flavoring industry, comestics and perfumery industries. Essential oil is widely used for treatment as aromatherapy. In plant breeding efforts the existence of various wild species is important to ensure the availability of genes, so that efforts to improve the quality of the plant can still be done. Several factors such as pests and diseases can cause the presence of Citrus spp. threatened sustainability. Therefore, it is necessary to do a conservation effort to protect and preserve it. Bali Botanical Gardens as one of the ex-situ conservation agencies, is in charge of preserving various types of plants from nature. Until the Year 2017 Bali Botanical Garden has collected 12 types of Citrus among others; C. aurantifolia, C. aurantium, C. amblycarpa, C. hystrix, C. grandis, C. maxima, C. medica, C. natsudaidai, C nobilis, C nobilis var. microcarpa C. papaya and Citrus sp. which originated from several regions in Indonesia. Judging from its content, essential oil from Citrus spp. potentially exploited as anti-insect and anti-cancer.

Abstract

Remote Sensing and Geographic Information System Application forAgroecosystem Based Agricultural Land Planning in Denpasar City This study aims to determine the level of potential agroecosystem land suitability, compile databases, build information systems and create maps of land useplanning for food crops and horticulture based on remote sensing and GeographicInformation System (GIS). This research took place from September 2018 - December2018 using an exploratory method which included analysis of satellite imagery, fieldsurveys, analysis of field data and laboratory data, land suitability classification,mapping of land suitability classes, compiling data bases, building informationsystems, commodity zoning, planning land use, and mapping of land use planning.Potential land suitability classes for irrigated rice fields are generally very suitable inall of Denpasar City (41 subak) but additional fertilizer is needed, especially N.Potential land suitability classes for horticultural (vegetable and fruit) commodities areclassified as appropriate (S2) through drainage improvements and fertilization.Climate factors are generally very suitable (S1) for food crops, and suitable (S2) forhorticulture plants. The train factor is generally classified as very suitable (S1) in foodcrops and horticulture unless the drainage factor is relatively slow and nutrientavailability is classified as moderate.

Abstract

ABSTRACT Landscape Planning of Republic of Indonesia Independence War Monument in Taman Mumbul Nusa Dua, Bali. Monument to Heroes of the War of Independence of the Republic of Indonesia in 1945, Taman Mumbul, Nusa Dua, Bali was built to commemorate and appreciate local Balinese who fought for freedom from colonialism in the past. This study aims to plan memorial landscape in Taman Mumbul Nusa Dua, Bali in harmony with the meaning of 1945 independence war monument of Republic of Indonesia and also has function as an urban park. The concept of the planning is to reinforces the meaning of the monument which is represented the struggle of the heroes to achieve independence in a green open space and to develope the site as an urban park for local people. This landscape planning harmonized the monument with the `heroism` philosophy and combined with the theme of `tapak tilas` on the main zone. The vegetation uses as an elements to appreciate the struggle of heroes in a green open space. The planning is also to create a place for recreation and socialize for local people or monument visitors. Keywords: landscape planning, historical monument, heroism, urban park.

Abstract

Identification of landslide potential in Baturiti District Tabanan Bali Districts Baturiti has most areas with a slope above 15%, land use as seasonal crop farming and rainfall is high enough to potentially the occurrence of landslide hazard. Based on the above, it is necessary to conduct research on potential landslide with the aim to know the level of landslide potential and map the potential spread of landslides in Districts Baturiti. This research was conducted from February to August 2017 with survey and scoring method. The study area has 12 units of land covering the low landslide potential class of 61764.2 hectares (77.1%). is present in the area represented by the land units 1,2,5,6 and 9. The medium landslide potential class of 9415,8 ha (11,7%) is in area which is represented by unit of land 3,7,10. The high landslide potential class of 8959.4 ha (11.2%) is in the area represented by the land unit 4,8,11,12.

Abstract

Identification and Population Abundance of Leaf Miner (Liriomyza spp.) (Diptera: Agromyzidae) on Onion Plant (Alium ascalonicum L.) and WelshOnion (Allium fistolosum L.) This research was conducted in order to determine the species and abundance population of leaf miner which attacked the onion plant (Alium ascalonicum L.) andwelsh onion (Allium fistolosum L.). The sample in this research was taken by purposivemethod by taking 100 leaves per species of onion in Bangli and Tabanan Regencies.The leaf sample was reared in a bottle container and the emerged adult insects of theleaf miner were identified morphologically. The results showed the leaf miner speciesthat attacked onions plant and welsh onion were Liriomyza chinensis and Liriomyzahuidobrensis. The leaf miner species found on onion plant in Songan Village, BangliRegency was L. chinensis with an abundance of 100% (324 adult insects). Meanwhile,in Batusesa Village, Tabanan Regency, which is the center of welsh onion production,two species of leaf miner were found, namely L. chinensis and L. huidobrensis withan abundance of L. chinensis 97.0% (258 adult insects) and L. huidobrensis 3.0% (8adult insects). In generally, L. chinensis is a species of leaf miner which dominate theonion plantations.

Abstract

Initiation Of Embryogenic Callus Strawberries (Fragaria sp.) By Giving IBA (Indolebutyric Acid) and BAP (Benzylaminopurine)Micropropagation or multiplication method by vegetative that could produce seedling in the high number and in a short time, had the same character as its mains, with a progress that not depends on the weather.This research intend to find the effect of grow regulator IBA (Indolebutyric Acid) and BAP (Benzyl aminopurine) and to find the best combination on producing callus strawberry. This research done in Unit Pelaksanaan Teknis (UPT) kultur jaringan Program Study Agroekoteknologi, Faculty of Agriculture, Udayana University. The materials use by the base of strawberry leaf.Growth regulators IBA and BAP could grown a callus. Usage 0,5 ppm IBA + 0,5 ppm BAP is the most effective in deflexion. Relative fast in deflexion (2,6 days). Usage 1 ppm IBA + 1 ppm BAP is the most effective in expansion (6,3 days) but in 30 days mandating its not grown any callus.It require toned to study the sterilisasion method to minimize the contamination and browning in explants strawberry leaf and a tool that in use to do research more to get the right concentration with IBA and BAP combination to get the callus.

Abstract

ABSTRACT Detection of Cucumber Mosaic Virus(CMV ) and Chilli Veinal Mottle Virus (ChiVMV) on Weed Commelina spp. In Cropping Chilli Pepper(Capsicumspp.) Through Serology and Molecular Test This study aims to determine the type of weed Commelina spp. which became an alternative host Cucumber mosaic virus (CMV) andChili veinal mottle virus (ChiVMV) in pepper, as well as the detection of CMV and ChiVMV which infects the weed Commelina spp. use molecular methods. DAS-ELISA (Double Antibody Sandwich-Enzyme Linked Immunosorbent Assay) and molecular techniques through RT-PCR (Reverse Transcription-Polymerase Chain Reaction) are used. Research activities include (1) Survey mosaic symptoms and disease incidence in the field; (2) Identification the type of weed that grows on pepper plants area; (3) Preservation of samples Commelina spp. weed and chili which virussymptomatic; (4) Test through DAS-ELISA serology; (5) Molecular detection by RT-PCR. The research proves that the weed Commelina spp. can be alternate hosts ChiVMV and CMV, also CMV and ChiVMV can infect weedsCommelina spp in chilicropping. DAS-ELISA test results show samples of the weed Commelina spp. and chili infected with Cucumber mosaic virus(CMV) and Chili veinal mottle virus (ChiVMV). RT-PCR technique successfully applyingDNA target fragment size of 657 bp and 900 bp for CMV and ChiVMV accordance with the specific primers used.   Keyword :Chili pepper, CMV, ChiVMV, weeds commelina
