Abstract

Abstract

Pedigree construction is one of the ways to seek the inheritance pattern of disease. Three generations pedigree has many benefit in genetic counseling session particularly in establishing a specific inheritance pattern such as autosomal recessive or dominant. This research aimed to analyze three generations pedigree in the family of ID and MCA patients in the Special school of Kendal. This is a descriptive observational study using the interview method in 33 student guardians or parents of ID and MCA students. Twenty-five of 33 respondents collects the complete data. This study showed that three generations of pedigree construction of 8 respondents could not conclude the inheritance pattern. The data showed that two pedigree are suspect of chromosomal abnormalities in the presence of family history with similar disorder, family history with ID and MCA, and family history with IUFD or stillbirth. Further investigation using cytogenetics, FISH, and microarray is needed to establish the diagnosis

Abstract

Total fish consumption during 2011-2015 showed an increase of 6.27%. The waste is estimated to be about 75% of the total weight of the fish. Thirty percent of the total 75% of fish processing waste is bone and skin. The collagen sources on the market is limited to cowhide and pig skin derived. Therefore, collagen from fish is expected to be an acceptable alternative source for all people because of its high availability and no risk of disease transmission. This study aimed to determine the difference of collagen content in manyung fish and snapper fish. Collagen extraction of manyung fish and snapper fish was done using alkaline and acidic solution. The soluble collagen concentration was measured by the standard Bradford solution using a UV-Vis spectrophotometer. The collagen concentration of manyung skin, snapper skin, manyung bone, snapper bone, and snapper scales obtained were 206.167; 202.833; 106.167; 77.833; and 24.50 ppm. The skin of manyung fish has the highest collagen content compared to other samples.

Abstract

Nile Tilapia (Oreochromis niloticus) is highly sought after by Indonesian consumers and is one of the leading commodities cultivated in freshwater. However, tilapia farming still frequently encounters challenges that lead to a decline in production, one of which is infection by pathogens such as Tilapia Lake Virus (TiLV). TiLV can infect fish in waters with temperatures above 25°C, with mass mortality peaking at 30°C, reaching a 50% death rate. Proper handling and control measures are essential to manage TiLV outbreaks effectively. TiLV detection in fish can be conducted using the Polymerase Chain Reaction (PCR) method. PCR-based virus detection is a diagnostic technique that identifies the presence of viral genetic material with high specificity and accuracy. This study aimed to detect TiLV infection in farmed Nile tilapia. The procedures included sample collection, necropsy, organ fixation, RNA extraction, PCR amplification, electrophoresis, and gel documentation analysis. Based on the PCR results, Nile tilapia samples tested positive for TiLV infection, as indicated by the presence of a 250 bp DNA band in the gel documentation system, aligning with the TiLV positive marker. Among the three fish showing suspected TiLV symptoms, two (66.6%) were confirmed infected.

Abstract

Abstract

Arabica coffee leaves (Coffea arabica L.) contain compounds known as secondary metabolites that have antibacterial properties, capable of suppressing the growth of Aeromonas hydrophila bacteria. The purpose of this study was to identify phytochemical compounds contained in Arabica coffee leaves (Coffea arabica L.), evaluate the antibacterial effectiveness of these leaves against Aeromonas hydrophila bacteria, and determine the concentration of Arabica coffee leaf extract that is most effective in inhibiting the growth of these bacteria. The method used in this research is the disc diffusion method. The concentrations of Arabica coffee leaf extract used were 25%, 35%, and 45%, and the data were analyzed using Kruskal Wallis test and Dunn's post hoc test. The results showed that the ethanol extract of Arabica coffee leaves had antibacterial activity. At a concentration of 45%, the extract showed an average inhibition zone of 15.95 mm against Aeromonas hydrophila, which is categorized as a very strong inhibition level. The Kruskal Wallis test yielded a significance level of 0.001 (p < 0.005), indicating a significant difference.
