Abstract

Relapse in post orthodontic treatment is common in the community because resorption in the pressure side occurs excessively due to teeth being moved with an orthodontic device. Excessive resorption can be inhibited by using NaF which has been shown to inhibit osteoclast activity, thereby reducing osteoclast numbers. The purpose of this study was to analyze the effect of NaF administration on IL-1β and TNF-α expression of osteoclast in orthodontic teeth movement. The research used experimental-quantitative research type applying Randomized Posttest Only Control Group Design, by selecting the experimental group and the control group randomly. Animal model using 16 male Wistar rats given orthodontic force using NiTi closed coil spring adjusted to give 10 grams/cm2 of mechanical strength, were applied to the palatal in the maxillary incisors of the Wistar rat teeth. Rats were divided into two groups, the control group (C group) and treatment group (T group) given topical NaF application (11.75 ppm). Immunohistochemical examination was performed to calculate IL-1β and TNF-α expression on osteoclasts on the 7th day and 14th day. The administration of NaF reduces IL-1β and TNF-α osteoclast’s expression both on the 7th day (p <0.05) and 14th day (p <0.05). Data were tested by the Kolmogorov-Smirnov test, Levene test and the difference tested Annova followed by LSD (p <0.05). Topical administration of NaF application to the gingival sulcus of male Wistar rats in orthodontic tooth movement can reduce the number of osteoclasts that express IL-1β in the alveolar bone in the pressure area. Topical administration of NaF application to the gingival sulcus of male Wistar rats in orthodontic tooth movement can reduce the number of osteoclasts that express TNF-α in the alveolar bone in the pressure region. Decreased number of osteoclasts expressing IL-1β and TNF-α significantly occurred on the 7th day and 14th day. Keywords: IL-1β; TNF-α; NaF; osteoclast; orthodontic teeth movement

Abstract

Background: Stress has negative impact for oral health and can cause periodontal disease, because the secretion of cortisol increase when stress occur. The excessive secretion of cortisol assumed to decrease the number of lymphocytes and macrophages in gingiva which play a role on immunity of oral health. Objective: To determine the effect of electrical shock stressor on the number of lymphocytes and macrophages in gingival tissue of Sprague Dawley rats. Methods: Sprague Dawley rats were given electrical shock for 7 days (S1), 14 days (S2), and 28 days (S3). Lymphocytes and macrophages observed on histological preparation in interdental gingiva between first molar and second molar used binocular microscope, 1000x magnification. Result: The results showed that the number of lymphocytes in S1 group (3,67±0,816), S2 (3,83±0,752), and S3 (3,17±0,527), also macrophages in S1 group (0,83±0,752), S2 (1,00±0,894), and S3 (0,67±0,816) were lower than the number of lymphocytes (0,67±0,816) and macrophages (2,00±0,632) in control group. The number of lymphocytes (p = 0,002) and macrophages (p = 0,033 ) had significant difference between groups. Conclusion: The electrical shock stressor decreased the number of lymphocytes and macrophages in gingival tissue of Sprague Dawley rats that could cause the oral cavity risk to periodontal infection.   Keywords: electrical shock stressor, gingiva, lymphocytes, macrophages, stress

Abstract

Orthodontic treatment is one treatment that can improve the dental malocclusion. Bone remodeling involves the coordination of three cell types namely osteocytes, osteoclasts, and osteoblasts. Osteoblast cells is important in the process of bone remodeling, especially in the process of bone aposition in tension side. This study aimed to analyze the changes in number of osteoblasts cell in tension side of Sprague dawley alveolar bone which induced by orthodontic force during one weeks, two weeks, and three weeks. 36 rats were divided into 6 groups, the treatment group with apllied the orthodontic force for 1 weeks (P-1), 2 weeks (P-2), and 3 weeks (P-3) and thr control group without apllied the orthodontics force group for 1 weeks (K-1), 2 weeks (K-2), and 3 weeks (K-3). Wire ligature 0,20 mm was apllied on the right region of maxillary first Molar and Incicivus, then mesially activated with maximally tension gauge of Orthodontic appliance of Ni-Ti closed coil springs. Those samples were sacrified and sliced vertically on the regio right maxillary bone with mesiodistal position, then the preparation of histological section with Hematoxilyn Eosin staining (HE) was made. The number of osteoblast cells were observed in 3 areas of tension side using microscope with 400 magnification. The result showed that the osteoblasts number of 2- and 3-weeks treatment groups were greater significantly than control group. It concluded that the induction of orthodontic appliance mechanic increased the number of osteoblast on tension area. The increasing of osteoblast number is paralel with the duration of the orthodontic appliance mechanic induction.

Abstract

Malocclusion is an occlusion that deviates from the normal state, so it needs to be treated using an orthodontic device. The formation of new alveolar bone after obtaining an orthodontic force will be surrounded by osteoid bones are not stable enough (relapse). Robusta coffee (Coffea robusta) has flavonoids which can increase lymphocyte proliferation. The purpose of this study was to analyze the effect of robusta coffee on the number of lymphocyte cells in the tension area of marmot’s (Cavia cobaya) periodontal ligament which induced by mechanical orthodontic forces. This type of research is laboratory experimental with the Post Test Only Control Group Design. The average number of lymphocyte cells in group A = 0.75, B = 1.45, C = 1.26, D = 1.54. One-way Anova test results showed that the number of lymphocyte cells in all groups showed a significant difference (p <0.05). The results of the post hoc LSD test showed that group A had significant differences with group B and group D (p <0.05). While group C did not have a significant difference with group A (p> 0.05). The conclusion obtained is that the provision of robusta coffee brew for 2 weeks can increase the number of lymphocyte cells in the tension area of marmot’s (Cavia cobaya) periodontal ligament which induced by orthodontic mechanical forces compared to without robusta coffee brew.

Abstract

Background.Orthodontic treatment using a bracket attached to the teeth dentofasial aiming to get a good look aesthetically. At the beginning of orthodontic bracket bonding systems to enamel surface through several stages of work so that they can spend time, includes teeth cleaning, basting phosphoric acid etching, washing with water, drying, basting material bonding / primer, and application of composite resin as a bracket adhesive material called total-etch technique. But as the times have found self-etch adhesive that further simplify the time and phases of work, the incorporation of phosphoric acid etching and bonding materials / primer in one package. Tensile strength is the strong adhesive tensile or the ability of an object to hold while receiving an attractive force and the force coming from the direction perpendicular to the surface of the object, for example when there is an attractive force toward the labial or buccal tooth driven. Purpose. This study aimed to determine differences in tensile strength and know the best adhesive that can be used between total-etch adhesive with a self-etch adhesive on orthodontic bracket bonding. Methods.This study used 16 pieces mandibular firstpremolars were divided into two groups, each consisting of eight teeth. The first group used a total-etch adhesive, while the latter uses self-etch adhesive. Bracket adhesive tensile strength was measured using Torsee's Digital System Universal Testing Machine. Results.Independent t-test showed a significant difference between groups in total-etch adhesive with a self-etch adhesive (p <0.05). Conclusions and suggestions.There is a significant difference in tensile strength measurements of total-etch adhesive with adhesive and self-etch adhesive total-etch proved better than a self-etch adhesive on orthodontic bracket bonding. Further research needs to be done with respect to the composite resin material used, the stages of research, and tools used tensiletest. Abstract

Habit is a repetitive act that is done spontaneously and commonly occurs in childhood. A habit in the oral cavity that can cause malocclusion is called a bad habit. Bad habits can affect the dentofacial functions such as the process of chewing, talking, tooth occlusion, periodontal tissue, and esthetic. If bad habits continue after six years old, they can cause malocclusion. The etiology of malocclusions can be classified into general and local factors. Bad habit is one of the general factors that play a role in the occurrence of malocclusion. Kinds of bad habits are sucking the thumb/finger, pushing the tongue, biting the lips and nails, wrong swallowing habits, breathing through the mouth, and bruxism. The study investigate the prevalence of bad habit and bad habit that mostly happened as etiology of Angle’s Class I malocclusion in patient of Orthodontic Clinic RSGM Jember University. This descriptional study with total sampling technique was conducted by identifying dental records and study models of Angle’s Class I malocclusion that caused by bad habit. Obtained data were tabulated and analysed so that conclusion could be made. Prevalence of Angle’s Class I malocclusion caused by bad habits was 9.4% with the largest distribution in thumb/finger sucking (38,7%), in the age group 9-10 years (50%), and in the male gender (53,6%). Abstract

Background: Orthodontic tooth movement depends on bone remodeling. VEGF plays an important role in bone remodeling in both pressure area and tension area. Robusta coffee contains caffeine, chlorogenic acid and caffeic acid. Caffeine may increase osteoclastogenesis, and caffeic acid has antioxidant effects that may reduce oxidative stress in osteoblasts Objective: To analysis of VEGF of orthodontic tooth movement post robusta coffeesteeping administration. Material and methods: The experimental laboratories research used 16rats  were divided into 2 groups. Group A: the rats were applied with  orthodontic mechanical force (OMF) and group B: OMF + coffee steeping of 20mg /100 g of BW. OMF was conducted by applying  ligature wire with a diameter of 0.20 mm on the molar-1 (M-1) and both incivus of right maxilla. Subsequently, M-1 of right maxilla was moved to mesial  with Niti closed coil spring. Observations were made on days 15 and 22 by taking the gingival crevicular fluid by putting paper point on the gingival sulcus of mesio-and disto-palatal area of M-1 of right maxilla to determine the levels of VEGF using ELISA method. Results: the administration of Robusta coffee steeping increased the levels of VEGF in both compression area and tension area (p <0.05).The levels of VEGF in tension area larger than the compression area(p >0.05).Conclusion: the Robusta coffee steeping administration increased the levels of VEGF oforthodontic tooth movement ,therefore it may improve alveolar bone remodeling process and it may be an alternative to accelerate orthodontic treatment.

Abstract

The objective of  research is  to determine the expression of osteocalcin on osteoblasts during orthodontic tooth movement after administration robusta coffee extract. Materials and Methods: 16 rats  were divided into 2 groups: group K: rats applied with orthodontic mechanical force (OMF) and  group P: OMF +  coffee robusta extract of 20mg /100 g of BW. OMF was conducted by applying  ligature wire on permanent maxillary right first molar and both permanent maxillary incisivus. Subsequently, the permanent maxillary right first molar  moved to mesial with Ni-Ti orthodontic closed coil spring. Observations were made on day 15 by immunohistochemical examination to determine the expression of osteocalcin. Results: Robusta coffee extract  improved the expression of osteocalcin in the compression and tension areas (p <0.05). Expression of osteocalcin in  tension area  larger than that in compression area (p <0.05). Conclusion: The administration of Robusta coffee extract  increases the expression of osteocalcin, which can increase alveolar bone remodeling.

Abstract

Background: Orthodontic tooth movement will be followed by periodontal ligament and alveolar bone remodeling. Orthodontic mechanical force (OMF) will be distributed through the teeth to periodontal ligament and alveolar bone and then will generate local pressure resulting in bone resorption and tension areas that will form new bone. Robusta coffee contains caffeine, chlorogenic acid and caffeic acid. Caffeine may increase osteoclastogenesis, and caffeic acid has antioxidant effects that may reduce oxidative stress in osteoblasts. Purpose: This study conducted to analyze the effect Robusta coffee steeping on levels of RANKL and TGF-β1 in orthodontic tooth movement. Method: 16 male rats were divided into 2 groups. Group C: rats given OMF, Group T: given OMF and coffee brew at 20 mg/ 100 g BW. OMF in rats was conducted by applying ligature wire on the molar-1 (M-1) and both incisivus of right maxilla. Subsequently, M-1 of right maxilla was moved to mesial with a Niti closed coil spring. Observations were made on days 15 and 22 by taking the GCF by putting paper point on the gingival sulcus of mesio- and disto-palatal areas of M-1 of right maxilla to determine the levels of RANKL and TGF-β1 using ELISA method. Result: The administration of coffee brew was effective to increase levels of RANKL and TGF-β1 in the compression and tension areas (p <0.05). RANKL levels in compression area were higher than in the tension area (p <0.05), while the levels of TGF-β1 in the tension area were higher than in the compression area (p <0.05). Conclusion: The administration of coffee brew was effective to increase the levels of RANKL and TGF-β, therefore it might improve alveolar bone remodeling process.

Abstract

Background: The application of orthodontic forces subjects blood capillaries to considerable pressure, resulting in hypoxia on the pressure side. Vascular endothelial growth factor (VEGF), expressed in osteoblasts represents an important mitogen that induces angiogenesis. Osteoblasts and blood capillaries play an important role in bone formation. Robusta coffee contains chlorogenic acid and caffeic acid both of which produce antioxidant effects capable of reducing oxidative stress in osteoblasts. Purpose: The aim of this study was to analyze the effects of Robusta coffee extract on the number of osteoblasts and blood capillaries in orthodontic tooth movement. Methods: This research constituted a laboratory-based experimental study involving the use of sixteen male rodents divided into two groups, namely; control group (C) consisting of eight mice given orthodontic mechanical force (OMF) and a treatment group (T) containing eight mice administered OMF and dried Robusta coffee extract at a dose of 20mg/ 100 g BW. The OMF was performed by installing a ligature wire on the maxillary right first molar and both maxillary incisors. In the following stage, the maxillary right first molar was moved to the mesial using Tension Gauze with a Nickel Titanium Orthodontic closed coil spring. Observation was subsequently undertaken on the 15th day by extracting the maxillary right first and second molar with their periodontal tissues. Thereafter, histological examination was performed using hematoxylin-eosin (HE) staining technique to measure the number of osteoblasts and blood capillaries on the mesial and distal periodontal ligaments of the maxillary right first molar. Results: The administration of Robusta coffee extract increases the number of blood capillaries and osteoblasts on both the pressure and tension sides were found to be significantly higher in the T group compared to the C group (p<0,05). Conclusion: Robusta coffee extract increase the number of osteoblasts and blood capillaries, thereby playing a role in improving the alveolar bone remodeling process in orthodontic tooth movement.
