Abstract

Diabetes mellitus (DM) is a metabolic disease with characteristics of chronic hyperglycemia and glucosuria. This occurs because of abnormalities of insulin secretion, insulin’s function or both. DM almost always followed by the increasing possibility of chronic complications. One complication encountered is hypercholesterolemia. Therefore we need a comprehensive DM therapy. Bitter melon fruit extracts (Momordica charantia lynn) shown to control the metabolism of glucose (hypoglycemic), This is because the extract contains pinsulin working a case of the hormone insulin. In addition there are several opinions and the results of studies suggest that the extract can lower blood cholesterol levels (hypolipidemic). This research carried out aiming to prove the presence or absence of decrease in blood cholesterol levels in diabetic male wistar rats (injected with alloxan) after given a bitter melon fruit extract. The samples in this study were divided into three groups: group I, group II and group III. All samples were injected alloxan to make the mice became diabetic. Blood cholesterol levels after administration of alloxan and three days later were measured and compared. On 3rd day until 6th day Group I was given 1% CMC Na as controlnegative, group II were given extracts of bitter melon fruit and the third group is given glibenclamide as a positive control. On ay-7 cholesterol levels were measured again. The results of data analysis showed that no significant differences in all outcome measures from all groups (Kruskall Wallis> 0.05). From this it was found that 62 hours after injection of alloxan did not have elevated levels of cholesterol and no significant decrease in cholesterol in male rats fed extracts of the fruit of bitter melon 700mg/200gr BB in 4 days.

Abstract

The leucocytes count and differential counting are routine blood test that have any significant role in diagnose and evaluation of disease or health. The aim of this study is to evaluated both in stress condition in animal model used Wistar male rat. Stress induced by electrical foot shock for 7 days. The result showed that leukocyte count in samples were higher than control (p<0.05). It was considered that stressor induce the adrenal cortex’s hormone secretion particularly glucocorticoid. This hormones caused movement of leukocyte in bone marrow storage to the vessel, leukocyte in marginating granulocyte pool moved to circulating granulocyte pool and and make the result of blood test is higher. This movement make the responds immunes to any injures worst and person with stress more susceptible to any infection.

Abstract

Relapse in post orthodontic treatment is common in the community because resorption in the pressure side occurs excessively due to teeth being moved with an orthodontic device. Excessive resorption can be inhibited by using NaF which has been shown to inhibit osteoclast activity, thereby reducing osteoclast numbers. The purpose of this study was to analyze the effect of NaF administration on IL-1β and TNF-α expression of osteoclast in orthodontic teeth movement. The research used experimental-quantitative research type applying Randomized Posttest Only Control Group Design, by selecting the experimental group and the control group randomly. Animal model using 16 male Wistar rats given orthodontic force using NiTi closed coil spring adjusted to give 10 grams/cm2 of mechanical strength, were applied to the palatal in the maxillary incisors of the Wistar rat teeth. Rats were divided into two groups, the control group (C group) and treatment group (T group) given topical NaF application (11.75 ppm). Immunohistochemical examination was performed to calculate IL-1β and TNF-α expression on osteoclasts on the 7th day and 14th day. The administration of NaF reduces IL-1β and TNF-α osteoclast’s expression both on the 7th day (p <0.05) and 14th day (p <0.05). Data were tested by the Kolmogorov-Smirnov test, Levene test and the difference tested Annova followed by LSD (p <0.05). Topical administration of NaF application to the gingival sulcus of male Wistar rats in orthodontic tooth movement can reduce the number of osteoclasts that express IL-1β in the alveolar bone in the pressure area. Topical administration of NaF application to the gingival sulcus of male Wistar rats in orthodontic tooth movement can reduce the number of osteoclasts that express TNF-α in the alveolar bone in the pressure region. Decreased number of osteoclasts expressing IL-1β and TNF-α significantly occurred on the 7th day and 14th day. Keywords: IL-1β; TNF-α; NaF; osteoclast; orthodontic teeth movement

Abstract

The success of orthodontic treatment can be achieved through tissue, cellular and molecular mechanisms due to mechanical stimuli formed by the pressure side and the tension side. Sodium Fluoride (NaF) is known to increase the bone formation process in the area of attraction in vitro and in vivo. The study aimed to determine sodium fluoride (NaF) against Caspase-3 by osteoblasts and IL-10 by osteoclasts in alveolar bone remodeling on orthodontic tooth movement. The research applied laboratory experiment using 16 male Wistar rats. Rats were divided into two groups; group C: given orthodontic strength without NaF; and group T: given orthodontic strength with NaF. Variable measurements were carried out after the 7th and 14th day. The data was statistically tested using One-Way Anova test analysis (p ≤0.05) to compare the mean number of expressions of both groups C and T. The number of osteoclasts expressing IL-10 was significantly higher in group T when compared to that of group C while the number of osteoblasts expressing Caspase-3 was smaller in group T when compared to that of group C. NaF can increase IL-10 expression in osteoclasts and decrease Caspase-3 expression by osteoblasts on orthodontic tooth movement. Keywords: Caspase-3; IL-10; sodium fluoride (NaF); immunohistichemistry

Abstract

Stress is a natural reaction that allows the human compelled to face the difficulties. Stimuli that causes stress can be physical and psychological. Physical stressors can occur continuously or sustained, which causes pathological processes in the body. This pathological process may be occur in liver and heart. One of parameter to assess characteristics of tissue can use Serum Glutamic Oxaloacetic transaminase (SGOT). The aim of this study was to prove differences level of SGOT in male Wistar rats (Rattus norvegicus) that exposed and control group were not exposed by pain stressor electrical foot shock for 28 days. Blood sampling was carried out on day 28 in intracardiac then tested the levels of SGOT. Statistical analysis showed that there was no significant change (p≤0,05) between control and treatment group. Liver damage from stressors may be not cause necrosis of liver cells. SGOT was not a single biomarker that was specific and sensitive for the diagnosis of liver disease because SGOT was also found in the heart.   Keywords: electrical foot shock, SGOT, stress

Abstract

Stress is the body response toward any kind of stressors that can lead to physiological changes in body systems. When it’s stresses, it will secretes stress hormones, such as cortisol, which can lead to elevate gluconeogenesis in liver. The cortisol blood levels increase much greater in rats exposed to 28 days stressors. It will force liver cells to perform continuous gluconeogenesis. These can cause damage to liver cells that can be characterized by increased levels of SGPT (Serum Glutamic Pyruvic transaminase). The aim of the study was to determine the differences in SGPT levels between male Wistar rats that were exposed by electrical foot shock stressors for 28 days and control rats. This was an experimental laboratory with post -test only with control group design, with sixteen rats were divided into two groups; control group and treatment group. The treatment group were exposed by electrical foot shock stressor for 28 days. On 28th day, serum samples were taken for examination of SGPT levels in both samples by kinetic methods . The results of T-test showed that there was no difference in SGPT levels between treatment and control groups. Keywords: electrical foot shock, serum glutamic pyruvic transaminase level.

Abstract

Background: A study with female animal models is important because the system immune of females is remarkably different from the male because of interaction between sex hormone and immune system. Interleukin-17 (IL-17) plays an important role in immune response toward Candida albicans (C. albicans) infection and Erythrocyte Sedimentation Rate (ERS) is an easy and sensitive test to assess the inflammation. Purpose: This study aimed to evaluate C. albicans infection, analyse the IL-17 levels and ESR in a female animal model of oral candidiasis. Methods: Female Wistar rats were used as oral candidiasis animal model. The rats divided into three groups (pre-treatment group (P0), 5th-day post-treatment group (P1) and 8th-day post- treatment group (P1). Each group consists of six rats. After the adaptation period, the P0 was sacrificed. The drinking water of the P1 and P2 was added tetracycline HCl 500mg /1L. On the day before and after inoculation C. albicans, Methylprednisolone was injected. Ten minutes before inoculation with 0.3 ml C. albicans 9.4 x 107/mL, rats were sedated by CPZ 0.7 mg IM. The rats in P1 group were sacrificed after five days and in P2 were sacrificed eight days after inoculation. The IL-17 plasma levels measured by enzyme-linked immunosorbent assay (ELISA), decreased on the 5th day but not on the 8th-day post-treatment. The obtained data were analyzed by parametric and non-parametric tests according to normality and homogeneity of the data with p<0.05. Results: The colony forming unit (CFU) of C. albicans collected over the mouth on increased almost 8-fold and on 8th-day post-treatment almost 3-fold compared with pre-treatment. The ESR increased on the 8th day but not on the 5th-day post-treatment. Conclusion: The IL-17 level was decreased on the 3rd day, ESR increased on eight days after inoculation of C. albicans in female rats’ model of oral candidiasis.
