Abstract

Medicinal plants are one of the efforts used to control disease. The benefits of medicinal plants have been passed down from generation to generation by our ancestors. One plant that has medicinal properties is miana leaves (Plectranthus scutellaroides R.Br). In this research, the extraction method used was maceration, with 96% ethanol solvent. Miana leaf ethanol extract was screened for phytochemicals and toxicity tested using the Brine Shrimp Lethality Test (BSLT) method. Toxicity tests were carried out at various concentrations of 100 ppm, 250 ppm, 500 ppm, 750 ppm and 1000 ppm. The results of phytochemical screening on miana leaves show that miana leaves contain flavonoid compounds, tannins, saponins, steroids, but do not contain alkaloids and triterpenoids. The 96% ethanol extract of miana leaves (Plectranthus scutellaroides R.Br) has an LC50 value of 511.58 ppm, included in the toxic category with an LC50 value in the range of 30-1000 ppm.

Abstract

Miana (Coleus scutellarioides) contains flavonoids as its primary active components and is widely used in traditional medicine, particularly in Indonesia. In Bali, which is renowned for its traditional healing practices (Usada Bali), miana is commonly utilized as a herbal remedy. The leaves of miana possess various pharmacological activities, including anti-inflammatory, antiproliferative, antidiabetic, anthelmintic, and antipyretic effects. Geographical conditions, such as the altitude of the growing region, are among the factors that influence the flavonoid content in miana leaves. This study aimed to determine the total flavonoid content in miana leaves from three different altitudes in Bali: Bedugul (1273 masl), Klungkung (393 masl), and Sanur (11 masl). An experimental laboratory design was employed, using maceration with 96% ethanol as the extraction method. The total flavonoid content was quantified using UV-Vis spectrophotometry, with quercetin as the reference standard. The results showed that miana leaves from Klungkung exhibited the highest total flavonoid content (540.85 ± 7.97 mgQE/g), followed by Bedugul (488.68 ± 6.07 mgQE/g) and Sanur (368.53 ± 11.2 mgQE/g). Statistical analysis revealed that the total flavonoid content of miana leaves from Klungkung was significantly higher (P<0.05) than that from the other two regions. These findings indicate that the altitude of the growing region affects the total flavonoid content in miana leaves, with the highest levels observed at an intermediate altitude (393 masl).

Abstract

Abstract

Background: Peptic ulcer disease (PUD) was a persistent gastrointestinal disorder characterized by mucosal damage in the stomach or duodenum. Cyclea barbata leaves had been reported to contain bioactive compounds with gastroprotective potential. Objectives: This study investigated the protective effects of 70% ethanolic extract of C. barbata leaves on ethanol-induced gastric ulcers in rats. Methods: Thirty male rats were randomly allocated into six groups. The groups consisted of a normal control, a negative control (Na CMC 0.5%), a positive control (ranitidine 13.5 mg/kg BW), and three treatment groups that received C. barbata ethanolic extract at doses of 100, 200, and 400 mg/kg BW. Treatments were administered orally for 14 consecutive days, and the animals were sacrificed on day 15. Ulcer count, ulcer diameter, ulcer index, and gastric pH were assessed. Results: The negative control group exhibited an average of 6.4 ulcers. Administration of ranitidine and all tested concentrations of C. barbata extract significantly reduced the number of ulcers compared to the negative control. The most pronounced effect was observed at 400 mg/kg BW, yielding an average of 1 ulcer and a significantly lower ulcer index (p < 0.05). In contrast, gastric pH values showed no significant differences among the treatment and negative control groups (p > 0.05). Conclusions: The findings indicated that the ethanolic extract of C. barbata leaves exerted gastroprotective activity, with the 400 mg/kg BW dose demonstrating the strongest ulcer-preventive effect.

Abstract

ABSTRACTPomegranate (Punica granatum L.) peel is a major agro-industrial byproduct rich in bioactive compounds, particularly polyphenols, which have attracted increasing interest for functional food and phytopharmaceutical applications. However, studies specifically analyzing the total polyphenol content of the local Balinese red pomegranate peel are still limited. This study aimed to determine the total polyphenol content and perform phytochemical screening of the red pomegranate peel collected from Bali, Indonesia. The research employed an experimental laboratory design. Red pomegranate peels were collected from Jadi Village, Tabanan Regency, Bali. The peels were dried, powdered, and extracted by maceration using 96% ethanol. Phytochemical screening was performed to detect major secondary metabolites. The total polyphenol content was determined using the Folin-Ciocalteu method with gallic acid as the standard, measured by UV-Vis spectrophotometry at 765 nm. All analyses were conducted in triplicate. The extraction yielded 25.78% of viscous ethanol extract. Phytochemical screening confirmed the presence of flavonoids, tannins, saponins, and steroids/triterpenoids. The standard gallic acid calibration curve showed good linearity with a correlation coefficient (R²) of 0.9916. The total polyphenol content was found to be 147.953 mg GAE/g at 1,000 ppm concentration and 34.136 mg GAE/g at 100 ppm. The findings demonstrate that the local Balinese red pomegranate peel contains significant polyphenolic compounds and secondary metabolites with antioxidant potential. This supports its potential for valorization as a source of natural antioxidants for functional food, nutraceutical, or sustainable packaging applications. Further studies are recommended to assess its antioxidant activity and to compare seasonal and varietal variations. Abstract

Drug distribution is an important stage in the pharmaceutical service system that plays a role in maintaining the quality, safety, and effectiveness of therapy to the hands of patients. Good Drug Distribution Practices (CDOB) are a reference in ensuring the quality of distribution, including in the preparation and implementation of Standard Operating Procedures (SOP) for delivery at Pharmaceutical Wholesalers (PBF). This study aims to evaluate the implementation of CDOB in the delivery SOP at PBF Enseval, with a focus on the role of the Responsible Pharmacist (RP). The study used a descriptive qualitative method with an evaluative approach through regulatory analysis and interviews. The results showed that most aspects of CDOB have been implemented, including the completeness of SOPs, documentation, training, and internal audits. However, there are still limitations in the involvement of RP at the SOP preparation stage, the real-time temperature monitoring for cold chain products is not optimal, and the Corrective and Preventive Action (CAPA) documentation is not fully digital. It is necessary to strengthen the role of RP, integrate temperature monitoring technology, and digitize documentation as an effort to improve the quality of drug distribution.

Abstract

Chlorella can produce MUFA and PUFA, such as linolenic acid, with an ideal ω-3: ω-6 ratio content (1:1). This study investigates the effects of centrifugation-induced shear stress on lipid metabolism and fatty acid composition in Chlorella sorokiniana and Chlorella vulgaris cultivated in CO2-enriched TAP medium modified with NaOH as an inorganic carbon source. Shear stress was applied during the exponential growth phase to assess its influence on biomass productivity, biochemical composition, and lipid accumulation under nutrient-rich (P1) and nutrient-deficient (P2) conditions. Results revealed that C. sorokiniana showed superior adaptability to shear stress compared to C. vulgaris, showing a 64.7% increase in lipid content and maintaining higher unsaturated fatty acid (UFA) proportions, particularly polyunsaturated fatty acids (PUFAs). In contrast, nutrient deprivation in P2 promoted saturated fatty acid (SFA) accumulation as an energy storage adaptation. Biomass recovery in P1 indicated the need for nutrient availability in sustaining growth following mechanical treatments. The fatty acid profile of C. sorokiniana was dominated by UFA (67.2%), including ω-3 and ω-6 PUFAs, whereas C. vulgaris showed a higher SFA (42.2%). These findings suggest that moderate shear stress can stimulate lipid biosynthesis and improve the nutritional quality of microalgal lipids without compromising cell viability, provided sufficient nutrients are available. The combination of shear stress induction and optimized nutrient management offers a promising strategy to enhance microalgal lipid production for functional food and nutraceutical applications.

Abstract

Polyherbal formulations have attracted increasing interest because combining plant extracts may improve phytochemical composition and antioxidant performance. This study aimed to optimize the proportions of ethanolic extracts of Cinnamomum burmannii bark, Gyrinops versteegii leaves, and Caesalpinia sappan wood using a simplex lattice design. Ten formulations were generated and evaluated for total phenolic content (TPC), total flavonoid content (TFC), total tannin content (TTC), and DPPH radical-scavenging activity (IC₅₀). The optimized formulation contained 10.00% C. burmannii, 21.51% G. versteegii, and 68.49% C. sappan, with experimental values of 79.27 mg GAE/g TPC, 197.64 mg QE/g TFC, 102.08 mg TAE/g TTC, and an IC₅₀ of 24.78 µg/mL. Experimental verification closely agreed with model predictions, with an average relative deviation of 2.95%. These findings demonstrate that simplex lattice design is an effective approach for optimizing polyherbal antioxidant formulations based on integrated phytochemical characteristics and antioxidant activity.
