Abstract

East Kalimantan is one of the regions that cultivates red dragon fruit (Hylocereus polyrhizus) in large quantities due to its promising health benefits. The peel of red dragon fruit has antioxidant activity comparable to its flesh, yet it is rarely utilized and often discarded as organic waste. This study aims to explore the potential of water and ethanol extracts from red dragon fruit peel as antioxidant sources that can serve as natural reducing and stabilizing agents in the biosynthesis of silver nanoparticles (AgNPs). Extraction using water and ethanol solvents was conducted to analyze the antioxidant compounds in the peel. The extract with the strongest antioxidant activity was used for AgNP biosynthesis, and the absorption peak was observed using UV-Vis spectrophotometry at a specific wavelength of 400–450 nm, which is characteristic of AgNP surface plasmon resonance. The results showed that the ethanol extract exhibited stronger antioxidant potential based on its lower IC₅₀ value and functioned effectively as a reducing and stabilizing agent at synthesis temperatures of 40–50°C, without degrading the extract's secondary metabolites. These findings support the utilization of red dragon fruit peel as a sustainable and eco-friendly raw material for the development of health-related products.

Abstract

Abstract

Biosynthesis of silver nanoparticles (AgNP) is a process of changing Ag+ ions to Ag0. The biosynthesis method in this research is green synthesis using natural ingredients to reduce silver, this synthesis is safer, environmentally friendly and economical. Based on this, this research aims to determine the optimal reaction conditions in the biosynthesis process using ethanol extract from cock leaves. The research method carried out pH optimization with variations of 8, 9, and 10 and then obtained the best pH, where the pH optimization will later be used in the AgNO3 optimization process with variations of 1mM, 2mM, and 4mM. After obtaining the best Ag optimization, the next stage was extract optimization using variations of 0.5%, 1% and 2%, the results of which will be seen based on the SPR peak of 400-500 nm which indicates the formation of AgNPs. The results of the research showed that AgNPs from cockang leaf ethanol extract were able to reduce at pH 8, AgNO3 2mM and 4mM, as well as 0.5% extract, and the contact time reaction showed a good formula for AgNP 1, AgNP2, AgNP3 which was indicated by the absorbance intensity at long waves that form AgNPs at the SPR peak of 400-500. Based on the results of SEM characterization, the morphology at a concentration of 2 mM has a surface that is not very smooth and the size is not very uniform, but at a concentration of 4 mM the surface is smooth and the particles are not uniform. With particle sizes in the range of 50-80 nm.

Abstract

ABSTRACTUtilization of Sweet Orange Peel Waste (Citrus sinensis) as a Base for Antibacterial Edible Film Formulation: Pectin Characterization Study and Extract Activity TestBackground: Sweet orange (Citrus sinensis) peel is an agro-industrial waste that has not been optimally utilized despite containing approximately 20–30% pectin (dry weight basis) and various bioactive compounds with potential applications as raw materials for biodegradable products. This study aimed to characterize pectin, evaluate the antibacterial activity of sweet orange peel extract, and establish a basic formulation of an antibacterial edible film as an environmentally friendly food packaging alternative.Method: This laboratory-based experimental study was conducted in 2024 and consisted of three stages: pectin extraction and characterization from sweet orange peel, extraction and antibacterial activity testing of sweet orange peel extract against Staphylococcus aureus and Escherichia coli, and development of a basic antibacterial edible film formulation. Pectin was characterized based on moisture content, ash content, equivalent weight, methoxyl content, degree of esterification, and solubility, while antibacterial activity was evaluated using the disc diffusion method. The characterization and antibacterial activity data were analyzed descriptively and compared with reference standards to determine the suitability of pectin and extract for edible film formulation.Result: The extracted pectin was classified as high-methoxyl pectin with good water solubility, high galacturonic acid content, and moisture content within the acceptable standard; however, it exhibited a low degree of esterification and ash content exceeding the recommended limit. The ethanolic extract of sweet orange peel demonstrated moderate to strong antibacterial activity against both tested bacteria. The optimum edible film formulation was obtained using 12% pectin and 40% ethanolic extract, producing the most favorable physical characteristics. Conclusion: Sweet orange peel has considerable potential as a source of pectin and natural antibacterial agents for the development of environmentally friendly edible films, thereby supporting organic waste valorization and the development of sustainable food packaging.
