Abstract

This study uses the sequencing method to determine the genetic diversity of the prolactin gene (PRL) in the exon four region in Bayang ducks. This study used 118 blood samples of Bayang ducks. Blood samples were isolated and then sequenced using a pair of primers L: 5'- GCA CAG TTG TTC TTA CTA GTT CG -3' and R: 5'- TCT GAG AAC TTT GCA GCT ATC T -3', which produced a 586 bp fragment in the Prolactin (PRL) gene exon 4. The amplification product was sequenced using First Base Singapore's services. The results showed that there were 16 polymorphisms in the exon four region and parts of introns 3 and 4 in the Bayang duck sample, namely two deletions at position -223C>del and -218A>del and 14 mutations at position -136G>A, 157G>A, +9T>G, +11G>A, +16T>C, +20A>C, +25T>C, +29A>C, +35T>A, +38A>G, +43C>A, +71G>A, +84G>A, +91T>A. Based on the study results, it can be concluded that the diversity of the prolactin gene (PRL) of Bayang ducks in the exon four region is polymorphic and can be used as a candidate for marker-assisted selection in Bayang ducks.

Abstract

Male Kamang ducks are one of the local ducks in West Sumatran germplasm originating from Tilatang Kamang and Kamang Magek Districts, Agam Regency, West Sumatera. The balance of metabolizable energy and crude protein (EM/P) in preparing the ration for Kamang ducks must be considered because it is closely related to the efficiency and growth of the ducks. Preparing the feed formulation for Kamang ducks with the right energy and protein balance will support their productivity. The research used male Kamang ducks in the grower phase, which is 3 to 7 weeks of age. Feed Treatment was metabolizable energy of 2800 kcal/kg with 17%, 18%, and 19% crude protein levels. The design used was completely randomized, with 3 treatments and 6 replications. The research showed no significant effect of treatments on feed consumption, body weight gain, or conversion ratio of male Kamang ducks in the grower phase; however, from the treatments, the highest income based on the IOFC value was ration B. In male Kamang ducks, good performance in balance of ME/P are 147.37 to 164.71 with the highest Income Over Feed and Cost (IOFC) value and lower fat abdominal at ME/P 155.55.

Abstract

This study aims to determine the diversity and correlation of egg production performance in Sikumbang Jonti ducks as a basis for making selection decisions. The materials used were 47 female Sikumbang Jonti ducks. In the production phase, Sikumbang Jonti ducks were intensively reared in individual battery cages for 24 weeks. The observed variables were body weight (g), age at first laying (days), egg production (eggs), and egg weight (g). Data on body weight, age at first laying, total egg production, and egg weight were analyzed descriptively by calculating the mean value, standard deviation, coefficient of variation, minimum value, and maximum value. Furthermore, data on body weight, age at first laying, egg production, and egg weight were also analyzed for correlation to see the direction and relationship between variables. The results of this study were that Sikumbang Jonti ducks have an average body weight of 1299g, age at first egg laying of 170.19 days, average total egg production of 36.62 eggs, and average egg weight of 59.06g. The diversity of body weight, age at first egg laying, and egg weight of Sikumbang Jonti ducks are moderate, while the diversity of egg production is high. The diversity of body weight, age at first laying, and egg weight of Sikumbang Jonti ducks are included in moderate diversity. In contrast, the diversity of egg production is included in high diversity. Based on the correlation of body weight, age at first laying, egg production, and egg weight of Sikumbang Jonti ducks, age at first laying is the selection variable that can be used as a good predictor of egg production performance. In conclusion, the study results show that a longer age of first laying can be used as a selection variable to increase the production and egg weight of Sikumbang Jonti ducks.

Abstract

Abstrak Itik Pitalah merupakan salah satu sumber daya genetik yang telah disahkan sebagai rumpun itik lokal asli Sumatera Barat. Keunggulan yang dimiliki oleh itik Pitalah adalah daya adaptasi yang baik. Follicle Stimulating Hormone (FSH) berperan dalam proses perkembangan folikel dan sekresi hormon steroid yang berfungsi dalam proses pembentukan kuning telur. Penelitian ini bertujuan untuk menganalisis keragaman gen Follicle Stimulating Hormone (FSH) pada itik Pitalah Sumatera Barat dengan menggunakan sampel darah dari 45 ekor itik Pitalah betina. DNA isolasi diamplifikasi dengan menggunakan sepasang primer forward  dan reverse yang didesain berdasarkan GeneBank DQ232890.1 dengan target fragmen 318 bp. DNA hasil amplifikasi disekuensing menggunakan jasa 1st Base Singapore untuk mengetahui keragaman gen Follicle Stimulating Hormone (FSH). Hasil penelitian menunjukkan bahwa terdapat 11 titik keragaman pada daerah target gen Follicle Stimulating Hormone (FSH). Keragaman gen Follicle Stimulating Hormone (FSH) ini bersifat polimorfik dan populasi itik Pitalah tidak berada dalam keseimbangan Hardy-Weinberg.Kata kunci: FSH; Itik Pitalah; Keragaman; Mutasi; Sekuensing

Abstract

AbstrakProlaktin merupakan hormon polipeptida yang disekresikan oleh kelenjar pituitary anterior dan secara spesifik kandidat gen yang mengontrol variasi jumlah produksi telur dengan reduksi biosintesis telur dalam beberapa periode mengeram. Tujuan penelitian ini  adalah untuk menganalisis keragaman gen Prolaktin pada itik Pitalah. Ekstraksi yang dilakukan pada sampel darah 45 ekor itik Pitalah dengan menggunakan protocol G-spinTM Total DNA Extraction Mini Kit dari Intron Biotechnology. Selanjutnya diamplifikasi dengan menggunakan sepasang primer forward 5’- CTG CAT CTG TGG ACA TTG CT-3’ dan reverse 5’GAA GCA GGT TTG GGA GTA CG-3’ dengan target fragmen 496 bp dan disekuensing menggunakan jasa 1sr Base Singapore. Data hasil sekuensing dianalisis menggunakan Seqmen DNASTAR dan MEGA 11 dan menunjukkan 14 keragaman pada daerah ekson 4 dan parsial intron 3 dan 4 pada titik mutasi INDEL 3703A, INDEL3939A, C3974A, A4030G, A4038C, T4056C, G4061A, T4091G, T4094A, T4098C, G4102A, C4106A, A4107C dan C4154G. Keragaman gen Prolaktin bersifat polimorfik dengan frekuensi genotype tidak dalam keseimbangan Hardy-Weinberg. Kata Kunci: Prolaktin, Itik Pitalah, Keragaman, Mutasi, Mutasi, Sekuensing

Abstract

Honey end egg yolk are one of the food supplements that contain antioxidants very high.This study aims to determine the quality of sentul male spermatozoa fed honey and egg yolk before artificial insemination on the quality of spermatozoa fertility hacchability of hacching eggs.This research was conducted by an experimental method with 4 levels 0 ml honey (control),3 ml honey,5 ml honey and 7 ml honey, Factor B giving egg yolk to sentul roosters with 4 levels 0 ml egg yolk (control),3 ml egg yolk,5 ml egg yolk, end 7 ml egg yolk.The results of the study showed that based on statistical analysis it was a very significant interaction between the combination of treatment levels of honey and egg yolk administraton on semen volume(0,85%),spermatozoa motility concentration(3,92x109),motility Spermatozoa(90,00%) spermatozoa abnormalities(5,00%) ,spermatozoa fertility eggs(91,67%),embryo mortality(7%),and egg hatchability(93%).From the results of the study it can be conluded that the administration of 5 ml honey and 5 ml egg yolk significantly gives the best results.
