Abstract

Paracetamol has analgesic properties comparable to NSAIDs, but paracetamol have minimal side effects. Paracetamol is metabolized via sulfation and glucuronidation conjugation which is then excreted in the urine. A small part of the paracetamol has been changed to NAPQI. NAPQI will be detoxified by gluthathione. In high doses, there in an increase in NAPQI and a decrease in glutathione levels that results in oxidative stress and liver cell necrosis. Curcumin is often used as a traditional medicine to treat liver disease where it contains phenolic groups capable to scavenge free radicals. Curcumin extract can improve cellular responses to oxidative stress such as increasing the expression of Nrf2, SOD, and gluthathione. The purpose of this research was to know the effect of curcumin on the improvement of liver function in white rats (Rattus novergicus) induced by high dose  paracetamol. The design of this research was a descriptive research using literature studies from at least 15 international journals indexed by Scimago or national journals indexed by Sinta published in 2015-2020. Based on the journals used in this research, giving curcumin at a dose of 200 mg/kg BW/day for 2 weeks was effective in significantly increasing gluthathione levels in rats receiving high dose paracetamol. Giving curcumin at a dose of 100 mg/kg BW/day for 7 days can reduce AST and ALT activity in rats receiving high dose paracetamol, but the dose of curcumin that was more effective in reducing AST and ALT activity was 200 mg/kg BW/day for 2 weeks. This is because of curcumin which functions as a hepatoprotector that bind directly to the toxic metabolite of paracetamol, thereby reducing the use of glutathione and quench free radicals, so that oxidative stress in the liver decreased and gluthathione levels increased, AST and ALT activity decreased.

Abstract

Indomethacin is a nonsteroidal anti-inflammatory drugs (NSAIDs) that act to inhibit COX-1. The inhibition of COX-1 leads to inhibition of prostaglandin production. Prostaglandin is a regulator of gastric acid buffer secretion. Inhibition of prostaglandin decrease gastric and duodenum pH and damage the gastric and duodenum. Sea grapes (Caulerpa racemose var. cylindracea) are marcoalgae that contain flavonoids as antioxidants and anti-inflammatory. The aim of this study was to determine the effect of sea grapes on gastric and duodenum pH in rattus norvegicus induced by indomethacin. 32 male Rattus norvegicus were divided into 4 groups. Group K (-) without treatment. Group K (+) was induced by 30 mg/Kg BW indomethacin for 7 days. Group P1 was induced by 30mg/Kg BW indomethacin for 7 days followed by administration of 1g/100g BW sea grape extract for 14 days. Group P2 was induced by 30mg/Kg BW indomethacin for 7 days followed by administration of 2g/100g BW sea grape extract for 14 days. On the 29th day, rats were terminated, gastric and duodenal were isolated then the fluid pH was measured. One-way Anova test obtained p = 0,023. Post hoc test pH Gaster significantly different between group K (-) and K (+) (p= 0,005) and between group K (-) with P2 (p= 0,020). While in group K (+) with P1 and P2 there was no significant difference in pH.  Post hoc test of pH duodenum showed no differences in all group. The administration of sea grape extract did not show a differences of the gastric and duodenal pH significantly between rats induced by indomethacin.

Abstract

Hyperlipidemia is one of the main risk factors for heart disease caused by the formation of atherosclerosis. Eucheuma spinosum contains a natural antioxidant flavonoid, which has an antiatherogenic effect that can inhibit the formation of cholesterol in the liver so that it can prevent the development of atherosclerosis. This study aims to determine the effect of prophylactic administration of Eucheuma spinosum on the total cholesterol levels of Rattus novergicus induced by dexamethasone. This study was an experimental design with a post-test-only control group design. This study would use 24 white male Rattus norvegicus divided into three groups; a negative control group (K-) without treatment, a positive control group (K+) induced dexamethasone 8 mg/Kg BW on day 14 until day 20, treatment group (KP) given Eucheuma spinosum extract 200 mg/Kg BW on day 8 until day 20 as Prophylactic and on day 14 until day 20 induced dexamethasone 8 mg/Kg BW. After completing the treatment period, all groups were terminated and taken total calculation of cholesterol levels through the aorta of rats. Data was analyzed statistically computerized. The results of the study of total cholesterol levels that used Kruskal-Wallis test showed differences between groups (p<0.001). There was a decrease in total cholesterol levels in the treatment group (p<0.024) compared with the positive control group. The administration of Eucheuma spinosum extract as a prophylactic dose of 200 mg/Kg BW for 14 days could reduce the total cholesterol level of Rattus norvegicus induced by dexamethasone compared to those not given Eucheuma spinosum extract.
