Abstract

Currently, infectious disease is increase in world wide. The African leaf (Vernonia amygdalina) – VA is used to antimicrobial treatment. It may protect the host against microbial attack in several ways. This plant has attracted the interest of researchers in recent decades because of the constituents have important roles in modulating immune system in bacteria infection. The aim of study is to analyze the prophylactic activity of VA's ethanol extract in modulating the levels of IL-6 and IL-10 as well as the number of bacteria in male Wistar rats that were (Staphylococcus aureus) – SA – infected. There were as many as 30 rats were divided into 5 treatment groups: negative control (NC) was treated by CMC Na 2% (w/v); positive control (PC) was treated by 9mg/200g body weight (BW) of cephadroxil; T1; T2; and T3 were respectively treated with ethanol extract of VA of doses 20mg/200g BW; 40mg/200g BW and 80mg/200g BW. After the oral treatment was administered, all the rats were infected with 0.25mL (3x108cfu) SA via intra peritoneal route. Their blood was drawn in order to identify the IL-6 and IL-10 levels by ELISA. Furthermore, their peritoneal fluid was also taken to count the number of survived bacteria by pour plate method. The results are showed median of IL-6 and IL-10 levels as well as bacterial number respectively in NC 370.530pg/mL; 67.044pg/mL; 7.4x103cfu/mL; in PC 234.556pg/mL; 42.839pg/mL; 6.8x103cfu/mL,; in  T1 164.019pg/mL; 17.240pg/mL; 1.1x104cfu/mL; in T2 49.291pg/mL; 2.961 pg/mL; 6.3x103cfu/mL and in T3 43.342pg/mL; 13.235pg/mL; 7.1x103cfu/mL. These results are implied that VA's ethanol extract is effective as a prophylactic agent to suppress the bacterial invasion at dose of 40mg/200g BW in Wistar rat particularly shown by the decrease level of IL-6 and the number of bacteria.

Abstract

Susahnya menumbuhkan jiwa wirausaha pada masyarakat Indonesia pada zaman sekarang termasuk yang dialami oleh peserta pengabdian kami yaitu petugas kebersihan di bawah PT. ISS Indonesia yang bekerja kampus Universitas Katolik Widya Mandala Surabaya. Hal itu disebabkan karena sebagian besar petugas kebersihan tersebut masih belum mendapatkan pembekalan dalam hal berwirausaha secara mandiri. Oleh sebab itu, pada kegiatan pengabdian ini, petugas kebersihan akan dibekali dengan ilmu berwirausaha dalam bentuk kegiatan pelatihan pembuatan produk minuman jamu yang dikemas secara modern dalam bentuk granul instan wedang pokak yang siap minum dan mudah diterapkan serta diimplementasikan secara sederhana. Keberhasilan kegiatan pembekalan menggunakan indikator perolehan nilai pre-test dan post-test. Hasil dari pembekalan yaitu para petugas kebersihan mampu meningkatkan pengetahuan dan wawasan tentang komponen bahan alam dalam formula jamu wedang pokak. Berdasarkan hasil evaluasi yang telah dilakukan, dapat disimpulkan bahwa kegiatan pengabdian yang dilakukan telah berhasil diterima dengan baik oleh para peserta yang dibuktikan melalui peningkatan pengetahuan berdasarkan nilai pre-test dan post-test yaitu sebesar 41%. Selain itu, kegiatan pengabdian ini telah berhasil dalam meningkatkan wawasan dan menambahkan ide bisnis bagi para peserta dalam berwirausaha dengan cara membuat sediaan jamu dalam bentuk granul instan wedang pokak secara mandiri yang memiliki nilai jual.

Abstract

The fox tail (Acalypha hispida) is known for its many benefits and has been used in traditional medicine. This aim of this research was determining the profile of specific and non specific standardization of fox tail leaves from three different areas (Batu, Bogor and West Sumba). The specific parameters consist of identity, organoleptic, microscopic, water soluble extract, ethanol soluble extract, phytochemical screening, chromatogram profile determination by thin layer chromatography (TLC), determination of infrared spectrum profile (IR), determination of UV spectrum profile and determination of secondary metabolite content (phenol and flavonoid). Non spesific parameters include shrinkage drying, ash content, acid insoluble ash, water soluble ash, percent foreign material and pH measurement. The result of standardization of dried powder fox tail leaves, have value of soluble ethanol extract ˃ 14%, water soluble extract ˃ 23%, contain flavonoids compounds, polyphenols, tannins, quinones and triterpenoids, total flavonoid levels > 0.2% and phenol > 0.7%, pectrophotometry IR result showed the presence of O-H, C-H, C = C and C-C groups, total ash content < 15.5%, acid insoluble ash < 2.5%, water soluble ash < 5.5%, drying shrinkage level < 10% and pH 4.5-5.

Abstract

Bakteri yang berperan dalam tumbuhnya jerawat diantaranya adalah Propionibacterium acnes dan Staphylococcus epedermidis. Pengobatan jerawat menggunakan antibiotik dalam jangka panjang dapat menimbulkan resistensi terhadap antibiotik. Kulit buah naga merah (Hylocereus polyrhizus) dapat digunakan sebagai alternatif antibakteri. Penelitian ini bertujuan untuk mengetahui golongan senyawa yang terkandung pada hasil fermentasi kulit buah naga merah dan aktivitas antibakteri terhadap Propionibacterium acnes. Pada penelitian ini kulit buah naga merah difermentasi selama 12 hari pada suhu kamar, hasil fermentasi diuji golongan senyawa dengan cara kromatografi lapis tipis dan uji aktivitas antibakteri dengan metode difusi cakram dengan parameter daerah hambat pertumbuhan (DHP). Hasil yang diperoleh uji aktivitas antibakteri hasil fermentasi kulit buah naga merah dengan konsentrasi 10%, 20%, 40%, 60%, 80% dan 100% tidak menunjukkan adanya hambat pertumbuhan terhadap bakteri Propionibacterium acnes. Hasil skrining golongan senyawa hasil fermentasi kulit buah naga merah memiliki kandungan flavonoid.
