Abstract

Propagation of mangosteen is challenging for many reasons, including limited seed set, slow rate of seedling growth, and difficulty with root formations. The objective of this research was to find the best combination of medium and plant growth regulator for in vitro growth and rooting of mangosteeen seed. Various types of explant (a whole seed; seed divided into 2, 3, and 4 cross sections; seed divided into 2, 3, and 4 longitudinal sections) were treated with five concentrations of benzyl amino purine (BAP; 0, 2.5, 5, 7.5, 10 mg/L) for shoot induction in ½ Nitrogen (N) Murashige and Skoog (MS) medium. The shoots were rooted on MS and woody plant medium (WPM) media with several combinations of indole butyric acid (IBA) and naphtalene acetic acid (NAA). Treatments for root induction were applied as follows: (i) low dose, given during induction of rooting, (ii) soaking the base of the shoots in medium treated with a high dose of auxin for 5 days, and then growing the shoots in MS ½ N with 1 mg/L NAA +  1 mg/L BAP medium. Our result show that BAP positively affected mangosteen bud growth. The best medium for mangosteen shoot regeneration was found to be  MS ½ N  + 5 mg/L BAP. This medium induced  the highest number of shoots from the seed explant cut into four cross sections. We found the best medium to induce in vitro rooting of mangosteen shoot was MS ½ N + 3 mg/L indole butiric acid (IBA) + 4 mg/L NAA medium. Some treatment negatively affected growth. Soaking the mangosteen shoot base in a medium with an overly high dose of auxin seemed to disrupt and inhibit growth of the mangosteen shoot.

Abstract

The objectives of this research were to composed organ from callus culture and to found the best concentration of plant growth regulator for organ growth from female flower explant of oil palm. This research has already done from June 2014 to May 2015 at Laboratory of Plant Physiology and Tissue Culture Department of Biology Faculty of Mathematics and Science University of North Sumatera. This research used Nonfactorial Completely Random Design. Explant was treated with five concentrations of 2,4-Dichlorophenoxy acetic acid (2,4-D; 99, 110, 120, 132, and 140 mg/L) for callus induction on Y3 medium (Eeuwens 1976). The result of this research showed that organ was formed from this treatment (basal segment of female flower explant) was root organ. 2,4-D plant growth regulator positively affected to growing of the root. The best result for time of callus induction, time of root growth, the highest percentage of explants that formed the root, fresh weight and dry weight of callus that has become the root generation was resulted from 99 mg/L 2,4-D.   Key words: Elaeis guineensis Jacq., female flower, plant growth regulator 2,4-D, organogenesis

Abstract

The objectives of this research are to analyze the students‟ misconceptions on topicphotosynthesis for each cognitive level and to know if the misconceptions remediation by usinginteractive multimedia and peer tutoringgive effect in decreasing students‟ misconceptionon topicphotosynthesis in SMA Swasta Nurul Iman Tanjung Morawa. Sample of this research are 56 studentsof science class XII. The types of this research are the descriptive research and pre-experimentalresearch with one group pretest-posttest design. The technique of data collecting was the two tiersmultiple choice closed reasoning diagnostic tests, consist of 45 questions which was used as pretestand posttest. The findingsshow that the students getting misconception on topic photosynthesis inhigh category of misconception by percentage 66%. The highest percentage of students‟misconception is at cognitive level 6, by percentage 76%. After treatment the results obtained thatmisconceptions remediation by using interactive multimedia and peer tutoring decrease students‟misconception as many as 47% which is from 66% to 19%. Hypothesis testing showed that tcount(3.969) > ttable (0.05)(55) (1.673), it means that H0 was rejected and Ha accepted. It can be concluded thatstudents‟ misconceptions remediation by using interactive multimedia and peer tutoring give effect indecreasing students‟ misconception on topic photosynthesis in science class XII SMA Swasta NurulIman Tanjung Morawa.Keywords: interactive multimedia, misconception, peer tutoring, photosynthesis

Abstract

This study aims to develop and determine the feasibility of a tissue culture textbook based on scientific literacy, conducted from November 2020 to May 2021. This study used the Dick & Carrey development model, modified to the formative evaluation stage. The instrument used is a questionnaire filled out by validators (material experts, learning design experts, layout design experts), lecturers of the Network Culture course, and students majoring in biology. The results showed that 77% of students had difficulty learning about somaclonal diversity and needed additional cultural application material. There were no scientific literacy-based tissue culture textbooks in the UNIMED library. Based on the survey, 76.9% of students said they liked somaclonal diversity material, 86.6% said it was challenging to study, 53.8% of the material described the interaction of science, environment, technology, and society, 30.8% did not explain the application of culture. As the results of textbook development, validator analysis shows 86% (very feasible) from material experts, 91% (very feasible) from learning design experts and 91% (very feasible) from layout design experts. The percentage of responses showed: 93% (very feasible) from lecturers, 86% (very feasible) from individuals, 90% (very feasible) from small groups and 89% (very feasible) from limited groups. Overall the textbook is very feasible to use.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi keberadaan miskonsepsi siswa terhadap materiGenetika di kelas XII IPA SMA Negeri 13 Medan. Metode penelitian ini merupakan penelitiandeskriptif. Populasi dalam penelitian ini adalah seluruh siswa kelas XII IPA SMA Negeri 13Medan yang berjumlah tujuh kelas. Teknik pengambilan sampel dilakukan dengan cara sampelkelompok (cluster sample) dan dipilih dua kelas, yaitu: kelas XII IPA 2 dan kelas XII IPA 7berdasarkan rekomendasi guru Biologi. Total sampel yang dipakai berjumlah 91 siswa. Teknikpengumpulan data dalam penelitian ini berupa instrumen tes diagnostik dua dimensi yang terdiri40 soal berbentuk pilihan ganda dengan tingkat keyakinan (CRI). Hasil penelitian menunjukkanterdapat miskonsepsi siswa terhadap materi Genetika sebesar 32,01%. Dari delapan indikatorpembelajaran terdapat tiga indikator dengan jumlah miskonsepsi yang tinggi, yaitu: indikator 8(mendeskripsikan proses sintesis protein, 38,46%), indikator 5 (mendeskripsikan struktur RNA,37,94%), dan indikator 2 (mendeskripsikan hubungan sel, kromosom, gen, dengan DNA,37,55%). Berdasarkan tingkat kognitif tes, siswa paling banyak mengalami miskonsepsi padaC5 (evaluasi, 41,94%).Katakunci: identifikasi miskonsepsi siswa, genetika

Abstract

This study was conducted in SMA Se-Langkat which aims to determine the level of difficulty biologyteacher (1) SMA As Langkat; (2) that have been certified (3) that has not been certified in high schoolBiotechnology As Langkat; (4) which is taught in public schools; and (5) are taught in private schools. Thesesamples included 40 teachers of biology of the population 40 teachers from 19 high school biology As Langkat.The research instrument is a non tests that documentation, questionnaire, interview and observation sheet.The results showed the difficulty level biology teachers: (1) SMA As Langkat classified as moderate (58.61%); (2)that have been certified very low (18.42%); (3) that has not been certified is low (41.68%); (4) who teach inpublic schools classified as very low (28.64%); (5) who teach in private schools classified as very low (29.54%).Results of this study mengiplikasi importance of addressing the difficulties faced by teachers withcommunication between teachers of biology utilizing discussion forum subject teachers (MGMP) and the needto facilitate schools with facilities and infrastructure that support the learning success.Keywords: Analysis of the Difficulties Teachers, Learning Tools, Biotechnology

Abstract

This study aims to detect the students difficulties in learning heredity topic and the factors thatinfluence on it. A descriptive research design was used in this study to describe the level of students difficultiesbased on cognitive and learning indicator aspect and also the causing factors. The population are taken fromwhole of XII Grade Students in SMA Swasta Harapan 1 Medan. There were 3 classes chosen as the sample byapplying cluster random sampling technique. The instruments used to obtain data were multiple choice test,essay test, concept map and questionnaire. The result of data analyzed showed : (1) On cognitive aspect, thedifficulties are categorized into very high difficulties from C1 until C6 level of cognitive which the highestpercentage is on C6 category as 80,21%, (2) On learning indicator aspect, the students have very highdifficulties in explaining the causing of pseudo deviation of Mendelian Law as 72,32%, (3) Society is the factorsthat categorized as influential in causing students learning difficulties which is 76,77%.Keywords : learning difficulties, cognitive aspect, learning indicator, heredity
